Follicular dendritic cell sarcoma of the tonsil: report of a rare case.

Follicular dendritic cell tumors are extremely rare. Only 17 cases have been previously described in the literature, and only three of them involved primary tumours of the oral cavity. We describe a new case of the latter, which occurred in a 51-year-old man who sought evaluation for a painless enlargement of his left palatine tonsil. The tonsil was excised, and histologic examination revealed that the tumor was a primary sarcoma that had arisen from the dendritic reticulum cells of the palatine tonsil. Postoperatively, the tumor site was treated with percutaneous irradiation (total dose: 70 G

Formation of filopodia-like bundles in vitro from a dendritic network.

We report the development and characterization of an in vitro system for the formation of filopodia-like bundles. Beads coated with actin-related protein 2/3 (Arp2/3)-activating proteins can induce two distinct types of actin organization in cytoplasmic extracts: (1) comet tails or clouds displaying a dendritic array of actin filaments and (2) stars with filament bundles radiating from the bead. Actin filaments in these bundles, like those in filopodia, are long, unbranched, aligned, uniformly polar, and grow at the barbed end. Like filopodia, star bundles are enriched in fascin and lack Arp2/

Dynamics of calcium and its roles in the dendrite of the cerebellar Purkinje cell.

The calcium ion (Ca2+) serves as an important cellular messenger with spatio-temporally highly dynamic patterns. Not only Ca2+ entering from the plasma membrane but also Ca2+ released from intracellular store sites play crucial roles in neurons as well as in other cell types. The cerebellar Purkinje cell shows a variety of spatio-temporal Ca2+ dynamics in its rich arborization, and the Ca2+ release mechanism mediated by IP3 receptors and ryanodine receptors has been shown to be crucial for the induction of long-term depression, a form of synaptic plasticity, in this neuron. Purkinje cells in a

CD83+ dendritic cells in inflammatory infiltrates of Churg-Strauss myocarditis.

Churg-Strauss syndrome (CSS) is characterized by a granulomatous vasculitis of multiple organs with the cardiovascular system being commonly affected. The initiation of the disease is not well understood, but immunologic phenomena are thought to contribute at least partially to the syndrome. We have studied endomyocardial biopsy specimens from a patient with CSS and eosinophilic myocarditis for characterization of infiltrating immune cells by immunofluorescence staining techniques and found a major population to be composed of CD83+CD14-CD19-CD56-HLA-DR+ dendritic cells (DCs). Further phenotyp

Mycobacteria target DC-SIGN to suppress dendritic cell function.

Mycobacterium tuberculosis represents a world-wide health risk and immunosuppression is a particular problem in M. tuberculosis infections. Although macrophages are primarily infected, dendritic cells (DCs) are important in inducing cellular immune responses against M. tuberculosis. We hypothesized that DCs represent a target for M. tuberculosis and that the observed immuno-suppression results from modulation of DC functions. We demonstrate that the DC-specific C-type lectin DC-SIGN is an important receptor on DCs that captures and internalizes intact Mycobacterium bovis bacillus Calmette-Guér

Enhancement of antitumor immunity by prolonging antigen presentation on dendritic cells.

Vaccination with dendritic cells (DCs) pulsed with antigenic peptides derived from various tumor antigens has great, but as yet significantly unrealized, potential in cancer treatment. Here, we describe a strategy for prolonged presentation of an MHC class I-restricted self-peptide on DCs through linkage of it to a cell penetrating peptide (CPP). DCs loaded with a peptide derived from tyrosinase-related protein 2 (TRP2) covalently linked to a CPP1 sequence retained full capacity to stimulate T cells for at least 24 h, completely protected immunized mice from subsequent tumor challenge, and sig

In situ localization of CD83-positive dendritic cells in psoriatic lesions.

BACKGROUND: Dendritic cells (DC) are considered to be the most potent antigen-presenting cells, and CD83 is expressed at a high level on immune-competent, activated and mature DC. Although changes in the number or localization of mature and activated CD83+ DC could be expected in psoriasis, there is little information on such changes. AIM: Morphological identification of CD83+ DC in psoriatic skin lesions. MATERIALS AND METHODS: Immunohistochemical staining was performed in 5 specimens of psoriasis vulgaris and 6 specimens of pustular psoriasis. Formalin-fixed, paraffin-embedded sections wer

Phenotypic characterization of five dendritic cell subsets in human tonsils.

Heterogeneous expression of several antigens on the three currently defined tonsil dendritic cell (DC) subsets encouraged us to re-examine tonsil DCs using a new method that minimized DC differentiation and activation during their preparation. Three-color flow cytometry and dual-color immunohistology was used in conjunction with an extensive panel of antibodies to relevant DC-related antigens to analyze lin(-) HLA-DR(+) tonsil DCs. Here we identify, quantify, and locate five tonsil DC subsets based on their relative expression of the HLA-DR, CD11c, CD13, and CD123 antigens. In situ localizatio

Two subsets of dendritic cells are present in human cerebrospinal fluid.

Little is known about the presence of dendritic cells in the human CNS. To investigate the occurrence of dendritic cells in the CSF, paired blood/CSF samples from patients with multiple sclerosis, acute optic neuritis, Lyme neuroborreliosis, other inflammatory neurological diseases and non-inflammatory neurological diseases were examined using flow cytometry. Almost all CSF samples contained myeloid (lin-CD11c+HLA-DR++CD123(dim)) and plasmacytoid (lin-CD11c-HLA-DR+CD123(high)) dendritic cells. In non-inflammatory neurological diseases, dendritic cells of either subset only constituted up to 1%

A common pathway for dendritic cell and early B cell development.

B cells and dendritic cells (DCs) each develop from poorly described progenitor cells in the bone marrow (BM). Although a subset of DCs has been proposed to arise from lymphoid progenitors, a common developmental pathway for B cells and BM-derived DCs has not been clearly identified. To address this possibility, we performed a comprehensive analysis of DC differentiative potential among lymphoid and B lymphoid progenitor populations in adult mouse BM. We found that both the common lymphoid progenitors (CLPs), shown here and elsewhere to give rise exclusively to lymphocytes, and a down-stream e

Dendritic cells and the control of immunity: enhancing the efficiency of antigen presentation.

BACKGROUND: Relevant antigens often are known for diseases that involve the immune system. Yet purified antigens by themselves do not control immunity, especially T-cell immunity. For example, many antigens have been defined for HIV-1 and melanoma, but good HIV-1 vaccines and melanoma immune therapies are lacking. Dendritic cells (DCs) are important intermediaries between antigens and better control of the immune system. METHODS: Some properties that allow DCs to control immunity are reviewed, followed by new studies using DCs as adjuvants in humans. An emerging area is then detailed, the spe

Infection of dendritic cells by murine cytomegalovirus induces functional paralysis.

Cytomegalovirus (CMV), measles and HIV are the main human pathogens known to induce immunosuppression. Unlike measles and HIV, and despite the availability of a well studied animal model, little is known about the mechanisms that control CMV-induced immunosuppression. We hypothesized that dendritic cells (DCs), which are crucial in generating and maintaining immune responses, represent a target for CMV and that the transient, but profound, immunosuppression that accompanies CMV infection results from viral interference with DC functions. Here we show that DCs were permissive to murine CMV infe

Dendritic cell migration controlled by alpha 1b-adrenergic receptors.

Dendritic cells (DC) bring Ags into lymphoid organs via lymphatic vessels. In this study, we investigated the possibility that the sympathetic neurotransmitter norepinephrine (NE) influences DC migration. Murine epidermal Langerhans cells mobilization is enhanced by systemic treatment with the alpha(2)-adrenergic antagonist yohimbine and inhibited by local treatment with the specific alpha(1)-adrenergic antagonist prazosin (PRA). Consistently, NE enhances spontaneous emigration of DC from ear skin explants, and PRA inhibits this effect. In addition, local treatment with PRA during sensitizatio

Signaling between the actin cytoskeleton and the postsynaptic density of dendritic spines.

The dendritic spine may be considered a fusion of a specialized actin-based structure akin to filopodia and lamellopodia, with an excitatory postsynaptic density containing glutamate receptors and signal-transducing machinery. This specialized neuronal microdomain is the site of the majority of excitatory synaptic contacts in the mammalian brain. Regulation of spine morphology, composition, and stability are likely to contribute to long-lasting changes in synaptic efficacy. Thus, understanding the function and regulation of dendritic spines is a fundamental problem ranging from molecular throu

Cells expressing dendritic cell markers are present in the rheumatoid nodule.

OBJECTIVE: To determine if dendritic antigen-presenting cells (DC) are present in rheumatoid nodules, as has been reported in the synovial lesions of rheumatoid arthritis. METHODS: Nodules (n = 14) were examined with monoclonal antibodies (Mab) recognizing the DC differentiation/activation markers CD83, CMRF44, and CMRF56 and an antibody recognizing the CD1a antigen present on epithelial tissue associated DC. Results. Cells expressing CMRF44 were common in rheumatoid nodules, comprising 22% of nucleated cells versus 13% in synovial membranes (n = 10). Cells positive for CD1a (5%) and CD83 (2%

Accumulation of macrophages with dendritic cell characteristics in the pulmonary response to Listeria.

Pulmonary immunity reflects a balance between proinflammatory and immunosuppressive factors in the lung. To determine the immune activities of exudate macrophages in the pulmonary immune response, Lewis rats were injected intratracheally with heat-killed Listeria (HKL), labeled ex vivo with the lipophilic dye PKH-26. At 24 h, macrophages from bronchoalveolar lavage fluid were purified on the basis of their surface membrane expression of RMA, a macrophage-specific antigen, which is brightly expressed by resident alveolar macrophages but dimly expressed by monocytes. Pulmonary macrophages were a

Dendritic cells and macrophages in the uveal tract of the normal mouse eye.

BACKGROUND/AIMS: Dendritic cells (DC) and macrophages are components of the immune cell populations in the uveal tract whose density, distribution, turnover, and function may play a role in the maintenance of immunological homeostasis in the eye. Little is known of these cells in the mouse eye despite this being the predominant experimental model in many studies of ocular immune responses and immunoinflammatory mediated eye diseases. The aim of the present study was to obtain further immunophenotypic data on resident tissue macrophages and DC populations in the mouse uveal tract. METHODS: Pie

Direct Ni2+ antigen formation on cultured human dendritic cells.

The possible direct antigen formation of Ni2+ on antigen-presenting cells (APCs) was studied with cultured human dendritic cells (DCs) obtained from 10 subjects contact allergic to Ni2+ and six non-allergic control individuals. All contact allergic subjects showed a significantly increased peripheral blood mononuclear cell (PBMC) response in vitro to Ni2+. DCs were expanded from the plastic-adherent cell fraction of PBMCs by culturing with granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4) for 7 days to obtain immature DCs, and with the addition of monocyte-cond
