Clinical characteristics and genetic variation in atopic dermatitis patients with and without allergic contact dermatitis.

In patients with atopic dermatitis (AD), the risk of contact sensitization may be higher as the disrupted skin barrier may increase the penetration of contact allergens. Therefore, it is necessary to screen for concurrent allergic contact dermatitis (ACD) in AD patients. To identify the clinical characteristics and genetic variation in AD patients with concurrent ACD. In total, 281 AD subjects who underwent patch testing were included. Subjects with a positive result were classified as "AD with ACD", while the others were classified as "AD only". Clinical characteristics and prevalence of gene

Phenotypical characteristics of the immune cells in allergic contact dermatitis, atopic dermatitis and pityriasis rosea.

Allergic contact dermatitis (ACD) is a cell-mediated, delayed type IV immunologic reaction. Atopic dermatitis (AD) is a chronic inflammatory skin disease that results from a complex interaction between immunologic, genetic, and environmental factors. Pityriasis rosea (PR) is a self-limited eruption of unknown etiology. Immune cell infiltrate is a constant feature in the inflammatory skin diseases. Here, we performed phenotypical characterization of the immune cells in ACD, AD and PR (ten cases each). We performed immunohistochemical stains for B cells (CD20), T cells (CD3), histiocytes (CD68)

Risk factors and temporal associations of progression of the atopic march in children with early-onset atopic dermatitis.

BACKGROUND: Risk factors and the temporal relationship between atopic dermatitis (AD) and atopic march remain understudied. OBJECTIVE: Determine risk factors for atopic march in early-onset AD patients and the temporality between AD and atopic march. METHODS: We used the MarketScan Research Database for our retrospective cohort analysis from 2010 to 2018, comparing infants diagnosed with AD before age 1 with controls without early-onset AD. Primary outcomes were hazard ratios (HRs) for the development of asthma, allergic rhinitis, and food allergy. RESULTS: Compared to 55,174 controls, high

Difference in the plasma level of miR-628-3p in atopic dermatitis patients with/without atopic keratoconjunctivitis.

INTRODUCTION: Some but not all patients with atopic dermatitis (AD) present with allergic conjunctival disease (ACD) including severe types such as atopic keratoconjunctivitis (AKC) with/without giant papillae. We hypothesized that different factors are involved in the severity of ACD and AD. Recently we reported that hsa-miR-628-3p could affect the balance of innate immunity by suppressing pathogen-associated molecular patterns such as toll-like receptor 3 (TLR3), RIG-I, and MDA-5. We also reported that TLR3 positively regulates ocular surface- and skin inflammation such as contact dermatitis

Exposure-response of ciclosporin and methotrexate in children and young people with severe atopic dermatitis: a secondary analysis of the TREatment of severe Atopic dermatitis Trial (TREAT).

This is a secondary analysis of a multicentre randomized controlled trial of ciclosporin and methotrexate in children and young people (CYP) with severe atopic dermatitis (AD). Longitudinal trough ciclosporin and erythrocyte methotrexate polyglutamate (MTX-PG) concentrations were measured to evaluate their associations with treatment response and adverse events. Both ciclosporin (4 mg kg-1 daily) and methotrexate (0.4 mg kg-1 weekly) led to a significant reduction in disease severity scores over the 36-week treatment period. Higher trough ciclosporin concentrations were associated with lower d

[Skin Microbiome and Dermatitis: Focusing on Two Distinct Skin Diseases, Atopic Dermatitis and Androgenetic Alopecia].

The skin of patients with atopic dermatitis (AD) is in a state of dysbiosis, having a microbiome of reduced diversity dominated by exacerbators such as Staphylococcus aureus and by fungal taxa such as Malassezia. As the symptoms of AD improve, microbial diversity increases and the level of colonization by exacerbators decreases. The level of skin colonization is correlated with scores on AD evaluation indices, thus the goal of AD treatment is to improve dysbiosis. Although Malassezia species exacerbate AD, they also secrete proteases that inhibit the formation of biofilm by Staphylococcus aure

Cumulative Disease Control with Upadacitinib in Moderate-to-Severe Atopic Dermatitis-A 52-Week Multicenter Real-World Study: IL-AD (Italian Landscape Atopic Dermatitis).

INTRODUCTION: Fixed-timepoint endpoints may not fully capture longitudinal treatment benefit in atopic dermatitis (AD). We evaluated the 52-week real-world effectiveness and safety of upadacitinib, focusing on cumulative disease control. METHODS: This multicenter retrospective analysis of prospectively collected LANDSCAPE registry data included patients with moderate-to-severe AD receiving stable upadacitinib 15 mg or 30 mg once daily. Patients who underwent dose modification from 15 to 30 mg or from 30 to 15 mg during the 52-week observation period were excluded from the present analysis to

Topical administration of testosterone ameliorates atopic dermatitis of KFRS4 rats.

Atopic dermatitis (AD) is a chronic inflammatory skin disorder influenced by genetic, environmental, and immunological factors, and its prevalence shows notable sex‑associated differences. KFRS4 rats spontaneously develop AD‑like dermatitis with higher severity in females, suggesting a role for sex hormones in disease modulation. This study investigated the contribution of gonadal hormones to dermatitis development in KFRS4 rats by examining the effects of gonadectomy and topical testosterone treatment. Ovariectomy did not alter dermatitis onset or severity in female rats, whereas orchiectomy

AI-Driven Digital Tools for Atopic Dermatitis: A Scoping Review.

BACKGROUND: Atopic dermatitis (AD) affects around 20% of children and up to 10% of adults. Its fluctuating course, severe pruritus, and impact on sleep, mental health, and daily functioning highlight the need for innovative approaches to diagnosis and disease management. Digital health technologies have rapidly expanded to fill this gap, yet their clinical readiness remains uncertain. OBJECTIVES: This scoping review aims to map AI-driven digital tools for AD, classify their functionalities, assess methodologies, and identify challenges and opportunities for real-world implementation. METHODS

A Subcellular Keap1-Nrf2 Disruption Strategy for Atopic Dermatitis Therapy.

Nuclear factor erythroid 2-related factor 2 (Nrf2) is a promising target for atopic dermatitis due to its role in skin barrier repair, yet effective modulators of Nrf2 are lacking. Here, we report an itaconate-iridium(III) complex conjugation strategy for the disruption of the Keap1-Nrf2 protein-protein interaction (PPI) for treating atopic dermatitis. The conjugate (1) specifically engages cytoplasmic Keap1 to restore Nrf2 activity, thus upregulating anti-inflammatory genes HO-1 and NQO1 while suppressing reactive oxygen species (ROS) in keratinocytes. It also reduces IL-13 production and boo

Comments on Dermatologists' TikTok Videos on Atopic Dermatitis: Content Analysis.

BACKGROUND: TikTok is one of the fastest-growing social media platforms in the world. It has become an important space for sharing information on a wide range of topics, including medical conditions such as atopic dermatitis (AD). Advice on skin conditions has become popular on TikTok, and most previous research in this area focuses on the credibility of the information being shared. However, little research has focused specifically on physician-created videos and their audience engagement and interaction. OBJECTIVE: Our study aimed to (1) characterize the audience's online response to board-

Salivary OMICS factors associated with atopic dermatitis in early infancy.

BACKGROUND: Atopic dermatitis (AD) often develops in early childhood and may persist into adulthood. Noninvasive measures to identify infants at risk for AD are limited. We investigated whether oral biomarkers and oral microbiota are associated with AD onset within the first 2 years of life. METHODS: In this prospective cohort study, we followed 119 infants residing in Upstate New York from birth to 24 months (2018-2023). AD diagnoses were obtained from electronic health records. Saliva samples were collected at seven time points (1, 2, 4, 6, 12, 18, and 24 months) and analyzed for 5 hormones

Cumulative Life Course Impairment in Atopic Dermatitis: A Comprehensive Review.

Background: Atopic dermatitis (AD) is a chronic inflammatory skin disease with a relapsing course and a substantial multidimensional burden. Conventional clinical and patient-reported outcome measures mainly provide cross-sectional evaluations and may not fully capture the long-term impact of the disease. The concept of cumulative life course impairment (CLCI) has been proposed to describe the progressive and lifelong consequences of chronic dermatologic conditions. Methods: This narrative review is based on a non-systematic literature search conducted in PubMed/MEDLINE and Embase up to July 2
