Effects of levodopa and dopamine of plasma glucose concentration in mice.

In nialamide-treated, fasted mice levodopa produced a dose-dependent hypoglycaemic response. The response was also seen in pargyline-treated mice but not in animals which were not treated with a monoamine oxidase inhibitor. Dopamine did not affect plasma glucose under these conditons. In doses which were ineffective when injected i.v., both levodopa and dopamine produced hypoglycemia when injected intracerebroventriculary (i.c.v.). The hypoglycaemic response to levodopa was prevented by the dopamine antagonists, haloperidol and pimozide. The possible involvement of 5HT in the hypoglycaemic res…

The amphetamine-induced inhibition of dopamine biosynthesis in rat striatum.

The dopamine biosynthetic machinery of intact synaptosomes of rat striatum showed a 5-fold increase in development from 3-day-old neonates to adults, and it was fully developed between 2-3 weeks after birth. Concurring with this development was the appearance 2 weeks after birth of a regulatory mechanism(s) through which amphetamine in vivo induced an inhibition of dopamine biosynthesis. The inhibition was not appreciably reversed when haloperidol, in addition to amphetamine, was administered.

Effect of drugs used in psychoses on cerebral dopamine metabolism.

1. Chlorpromazine 15 mg/kg, given daily to cats for 2 weeks, produced a rise in homovanillic acid (HVA) content of the caudate nucleus, whereas the same dose of thioridazine lacked this effect. Of these two drugs, only chlorpromazine causes a high incidence of drug-induced Parkinsonism in man.2. In the mouse, chlorpromazine, thioridazine and haloperidol increased striatal concentrations of HVA and accelerated the disappearance of dopamine (DA) after inhibition of catecholamine synthesis with alpha-methyltyrosine. Low doses of the three compounds increased, whereas high doses reduced, the conce…

The developmental dopamine trajectory hypothesis of ADHD across the lifespan.

Attention-deficit/hyperactivity disorder (ADHD) is increasingly recognized as a lifespan neurodevelopmental disorder, yet the mechanisms und … y can coexist with developmental changes in clinical expression. Here, we propose the Developmental Dopamine Trajectory Hypothesis, which posits that DAT related dopaminergic dysregulation may interact with the sequential maturation of frontostriatal, mesolimbic, and mesocortical circuits to contribute to developmental changes in ADHD symptom expression ac …

The heterogeneity of dopamine-mediated vasodilation in human intrarenal arteries.

INTRODUCTION: Adequate renal perfusion is critical for maintaining kidney function, and its impairment contributes significantly to acute kidney injury (AKI) and related cardiovascular complications. Dopamine (DA) is clinically employed to improve renal perfusion, but its efficacy remains controversial due to paradoxical vasoconstriction at higher doses. OBJECTIVES: This study systematically evaluates DA-induced vasomotor responses in human intrarenal arteries and elucidates the underlying molecular mechanisms. METHODS: Human intrarenal artery segments, including interlobar (IA, ∼1 mm), arcu…

Dopamine signatures of excessive and compulsive cocaine and fentanyl use.

Excessive and compulsive drug use despite adverse consequences is a hallmark of substance use disorder, yet individuals differ markedly in their vulnerability to develop these behaviors. Drugs of abuse are long known to alter endogenous dopamine (DA) signaling, but shared principles for how DA dynamics impact compulsive use among individuals and across drug classes are lacking. Here, we monitored DA release in the medial shell of nucleus accumbens (NAc) during cocaine and fentanyl self-administration, with or without coincident punishment, in large cohorts of mice. Contingent cocaine and fenta…

Shared mechanisms of dopamine and ATP transmission in the nucleus accumbens.

Dopamine (DA) neurons of the midbrain project throughout the striatum, including the nucleus accumbens core (NAc) and are thought to co-release ATP with DA from vesicles. The mechanisms of evoked NAc ATP release and clearance and t heir relationship to exocytotic DA transmission are largely unexplored and the focus of the present work. Using fast scan cyclic voltammetry (FSCV), we measured simultaneous ATP and DA transmission in response to pharmacological manipulations of release and reuptake cellular machinery. ATP transmission is tightly coupled to that of DA, though ATP release concentrati…

Neuroendocrine Effects of Hyperprolactinemia: Focus on Dopamine and Serotonin Systems.

Hyperprolactinemia is characterized by unusually high levels of prolactin, a hormone primarily known for regulating reproductive functions. However, studies have shown that prolactin has also a significant impact on various neurotransmitter systems in the brain. This review examined the intricate relationship between hyperprolactinemia and neurotransmission, with a particular emphasis on the dopaminergic and serotonergic systems. Prolactin affects these systems in ways that can notably influence mood, behavior, and cognitive functions. One significant consequence of elevated prolactin is the d…

An axonal brake on striatal dopamine output by cholinergic interneurons.

Depolarization of axons is necessary for somatic action potentials to trigger axonal neurotransmitter release. Here we show that striatal cholinergic interneurons (ChIs) and nicotinic receptors (nAChRs) on mouse dopamine axons interrupt this relationship. After nAChR-mediated depolarization, dopamine release by subsequent depolarization events was suppressed for ~100 ms. This suppression was not due to depletion of dopamine or acetylcholine, but to a limited reactivation of dopamine axons after nAChR-mediated depolarization, and is more prominent in dorsal than in ventral striatum. In vivo, nA…

Dopamine D2 receptors in the cerebellum: modulation of social behavior.

This review presents a systematic analysis of the cerebellar dopaminergic system and its role in regulating social behavior. It examines the mechanisms of dopamine synthesis and transmission, the main types and signaling pathways of dopamine receptors, and data on the localization and functional significance of these receptors in cerebellar cells, particularly D2 receptors. Special attention is given to experimental results demonstrating that modulation of D2 receptors in the cerebellum affects social preferences and memory in animals. The connections between the cerebellum and dopaminergic zo…

Quantitative chemoproteomics reveals dopamine's protective modification of Tau.

Dopamine (DA) is one of the most important neurotransmitters. Its oxidation leads to electrophilic quinone, which covalently modifies nucleophilic residues in proteins, resulting in 'dopamination'. Individual dopaminated proteins have been studied, most of which were functionally damaged by dopamination. Here, we developed a quantitative chemoproteomic strategy to site-specifically measure proteins' dopamination. More than 6,000 dopamination sites were quantified. Half-maximal inhibitory concentration values for 63 hypersensitive sites were measured. Among them, hypersensitive dopamination of …

[Art and creativity in Parkinson's disease: the mysterious effects of dopamine].

The article is devoted to the phenomenon of the emergence or enhancement of creative abilities during therapy in some patients with Parkinson's disease (PD). The article discusses the possible pathogenetic causes of this phenomenon, the connection with impulsive-compulsive disorders (ICD). All PD patients who are addicted to one type of creativity are characterized by increased motivation, forcing them to spend hours doing what they love, the time of which coincided with the start of dopaminergic therapy. There is a fine line between the emerging «passion» for art and CAVIAR, which is determin…

Are There Connections between the Neuroendocrine and Dopamine Systems in Bipolar Disorder?

BACKGROUND: Bipolar disorder (BPD) represents a frequent and disabling disease, characterized by the occurrence of extreme mood swings leading to episodes of depression or mania. Although dysfunctions in dopamine (DA) neurotransmission are increasingly recognized as key determinants of BPD, little attention has been given to the biological factors which may shape such a cyclicity of mania and depression. SUMMARY: We propose that BPD may result, at least in part, from skewed connections between the neuroendocrine system, the DA system, and the hypothalamic clock center. First, we provide a bri…

Magnesium-based micromotors for electrochemical detection of dopamine in blood.

In the fields of neuroscience and medicine, the accurate capture and quantitative analysis of dopamine (DA) levels in the blood are of great clinical significance for the early diagnosis of neurological dysfunctions and other pathological conditions. In this study, we developed a laccase (La) sensor based on micromotor technology to accurately detect DA content in blood. Firstly, the magnesium-based micromotor was used to endow the sensor with autonomous motion performance, which significantly increased the contact frequency between La and DA molecules in blood. Secondly, the micromotor-loaded…
