Recovery of retching after lesions involving the nucleus of the solitary tract.

The nucleus of the solitary tract (NTS) in the caudal brainstem receives various inputs that trigger vomiting. Chemical (kainic acid) NTS lesions in decerebrate, paralyzed cats temporarily suppressed fictive vomiting induced by different emetics. Subsequently, retching but not expulsion, resumed in 2-3.5 h. Since the NTS does not appear essential for retching, antiemetic drugs targeted at the NTS may not completely suppress vomiting.

Practical management of hyperthyroidism.

There are several causes of hyperthyroidism, and correct diagnosis is essential for management. Graves' disease is most commonly managed with radioactive iodine therapy (131I), antithyroid drugs or surgery. Toxic adenomas (single or multiple) may be treated with 131I or surgery. Most types of thyroiditis are managed expectantly. Pregnant women, children and the elderly deserve special consideration. Follow-up is vital to identify the later development of hypothyroidism.

[Drugs and vitamins].

Since most of the drugs are taken orally via the gastrointestinal tract, interactions with food ingredients may lead to alterations in the intake of essential dietary factors. With regard to vitamins the following effects have been demonstrated: early decomposition resp. inactivation, decreased absorption or a qualitatively and quantitatively modified metabolism. Examples of such changes are given with regard to water- and lipid-soluble vitamins.

Secondary Amine-Based Chemical Isotope Labeling Coupled with Mass Spectrometry for Simultaneous Quantification of Carboxylic Compounds during Hepatocellular Carcinoma Progression.

Carboxylic metabolites and drugs play essential roles in biological regulation and disease progression. Their simultaneous quantification remains analytically challenging due to their low ionization efficiency and poor chromatographic retention. Here, we synthesized a pair of secondary amine-based chemical isotope labeling reagents, N-(piperidin-4-ylmethyl)benzamide (PMBA) and d5-N-(piperidin-4-ylmethyl)benzamide (d5-PMBA), for the simultaneous labeling of carboxylic metabolites and drugs. The secondary amine moiety on PMBA/d5-PMBA can efficiently react with carboxyl groups on carboxylic compo

Unveiling RNA ligands: Harnessing NMR for hit generation.

In recent decades, there has been extraordinary progress in the discovery of biomacromolecules essential for the occurrence and progression of diseases, providing new and more effective perspectives for drug discovery. The discovery and development of new drugs are vital to healthcare, as most existing drugs target protein-based biomolecules such as enzymes and receptors. Nevertheless, only a small part of the genome encodes for proteins, while a large portion is transcribed into RNA. Moving beyond proteins, RNA is emerging as a promising target, offering a vast, largely unexplored chemical sp

Recent advances in the role of polysaccharides in liver diseases: a review.

Liver diseases are a serious health problem worldwide, especially with a sustained increase in the burden of it every year. However, drugs commonly used in patients have limited efficacy and serious adverse reactions associated with long-term use. Therefore, it is urgent to find effective and safe alternatives. Polysaccharides are currently considered promising alternatives to traditional drugs because of their extensive activity and low toxicity. This review investigated the studies on hepatoprotective polysaccharides over the past 6 years, detailing their hepatoprotective effects, potential

Epigenetics-targeted drugs: current paradigms and future challenges.

Epigenetics governs a chromatin state regulatory system through five key mechanisms: DNA modification, histone modification, RNA modification, chromatin remodeling, and non-coding RNA regulation. These mechanisms and their associated enzymes convey genetic information independently of DNA base sequences, playing essential roles in organismal development and homeostasis. Conversely, disruptions in epigenetic landscapes critically influence the pathogenesis of various human diseases. This understanding has laid a robust theoretical groundwork for developing drugs that target epigenetics-modifyin

Sedation for adult ICU patients: A narrative review including a retrospective study of our own data.

The optimization of patients' treatment in the intensive care unit (ICU) needs a lot of information and literature analysis. Many changes have been made in the last years to help evaluate sedated patients by scores to help take care of them. Patients were completely sedated and had continuous intravenous analgesia and neuromuscular blockades. These three drug classes were the main drugs used for intubated patients in the ICU. During these last 20 years, ICU management went from fully sedated to awake, calm, and nonagitated patients, using less sedatives and choosing other drugs to decrease the

Pharmacology and Adverse Events of Emergency-Use Authorized Medication in Moderate to Severe COVID-19.

Some effective drugs have been approved or issued an Emergency Use Authorization for the treatment of COVID-19 in hospitalized patients, but post-market surveillance is warranted to monitor adverse events. We reviewed clinical trials and case reports in patients with moderate-to-severe COVID-19 infection who received remdesivir, baricitinib, tocilizumab, or sarilumab. The drug-specific pharmacokinetics, toxicity, and drug interactions are summarized in this study. Remdesivir and baricitinib are small-molecule drugs that are mainly metabolized by the kidneys, while tocilizumab and sarilumab are

Effects of Drugs and Excipients on Hydration Status.

Despite being the most essential nutrient, water is commonly forgotten in the fields of pharmacy and nutrition. Hydration status is determined by water balance (the difference between water input and output). Hypohydration or negative water balance is affected by numerous factors, either internal (i.e., a lack of thirst sensation) or external (e.g., polypharmacy or chronic consumption of certain drugs). However, to date, research on the interaction between hydration status and drugs/excipients has been scarce. Drugs may trigger the appearance of hypohydration by means of the increase of water

Anti-epileptic drugs inhibit viability of synoviocytes in vitro.

INTRODUCTION AND OBJECTIVE: The hyperplasia of synovial fibroblasts is considered to be essential for the evolution of joint destruction in rheumatoid arthritis (RA). Previously, we reported that anti-rheumatic drugs, both COX inhibitors and disease-modifying anti-rheumatic drugs inhibit proliferation of synoviocytes in vitro. The presented study investigates the effect of anti-epileptic drugs on the viability and proliferation of synovial fibroblasts in vitro. METHODS: Experiments were conducted on human synoviocytes derived from an RA patient and rabbit synoviocytes cell line HIG-82. Cell p

Comparative genomics study for identification of putative drug targets in Salmonella typhi Ty2.

Typhoid presents a major health concern in developing countries with an estimated annual infection rate of 21 million. The disease is caused by Salmonella typhi, a pathogenic bacterium acquiring multiple drug resistance. We aim to identify proteins that could prove to be putative drug targets in the genome of S. typhi str. Ty2. We employed comparative and subtractive genomics to identify targets that are absent in humans and are essential to S. typhi Ty2. We concluded that 46 proteins essential to pathogen are absent in the host genome. Filtration on the basis of drug target prioritization sin

Mast cell stabilizers.

The precise mode of action of the well-studied cromolyn sodium and the newer nedocromil sodium has not been completely elucidated. Because the drugs do not pass the cell membrane and enter the cell, they are virtually not metabolized, do not exert a systemic action, and therefore are associated with only minimal systemic toxicity. To be effective either drug must be applied topically and directly to the nasal mucosa. Proper contact with the nasal mucosa is essential for efficacy; in patients with nasal congestion and secretions, vasoconstrictors or saline lavages are indicated before cromolyn
