Strigolactones regulate shoot elongation by mediating gibberellin metabolism and signaling in rice (Oryza sativa L.).

Strigolactones (SLs) are plant hormones that regulate diverse physiological processes including shoot elongation. However, little is known about the regulatory mechanism of SLs in rice shoot elongation. Our results demonstrate that defects in SL biosynthesis or signaling led to dwarfism, and the dwarf statures of SL-deficient mutant (d17) and SL-insensitive mutant (d14) were restored to wild-type (WT) by gibberellin (GA) treatment, indicating that their dwarfism was associated with decreased GA content or weakened GA sensitivity. Our results indicate that the bioactive GA1 contents in d17 and …

Diffuse large B-cell lymphoma chemotherapy reveals a combined immunodeficiency syndrome in cartilage hair hypoplasia.

Cartilage hair hypoplasia (CHH) is a rare autosomal recessive ribosomopathy characterised by skeletal and integumentary system manifestations. It may also present with varied forms and intensities of haematopoietic and/or immune disorders. We report a 27-year-old female who presented a picture of combined immunodeficiency after receiving an adriamycin-based chemotherapy regimen followed by autologous stem cell transplantation. Her medical history indicated neonatal dwarfism, recurrent ear, nose and throat and respiratory infections, and hypogammaglobulinaemia, which were suggestive of a primar…

The dwarf phenotype in GH240B mice, haploinsufficient for the autism candidate gene Neurobeachin, is caused by ectopic expression of recombinant human growth hormone.

Two knockout mouse models for the autism candidate gene Neurobeachin (Nbea) have been generated independently. Although both models have similar phenotypes, one striking difference is the dwarf phenotype observed in the heterozygous configuration of the GH240B model that is generated by the serendipitous insertion of a promoterless human growth hormone (hGH) genomic fragment in the Nbea gene. In order to elucidate this discrepancy, the dwarfism present in this Nbea mouse model was investigated in detail. The growth deficiency in Nbea+/- mice coincided with an increased percentage of fat mass a…

A hormonally controlled serum factor stimulating the thymidine uptake into lectin-activated lymphocytes.

The in vitro stimulation of [3H]thymidine uptake into lectin-activated lymphocytes in the presence of various sera was studied. The mean precision of the assay is 5%, and the study of the confidence intervals shows variations from 4% to 12%. Compared to a normal reference serum (fixed as 1 U/ml), the serum thymidine uptake stimulating activity (mean +/- SEM) was 1.04 +/- 0.07 U/ml in normal adult males, 2.63 +/- 0.48 U/ml in acromegalic patients, 1.51 +/- 0.13 U/ml in constitutional dwarfism and 0.37 +/- 0.04 U/ml in untreated hypopituitary dwarfism with a significant difference between the gr…

Disabilities and Handicaps of Patients with Laron Syndrome.

Background: Laron Syndrome (LS) is a rare hereditary form of dwarfism occurring, with few exceptions, in Jewish, Muslim, and Asian populations or their descendants spread over all continents. It is caused by deletions or mutations in the GH-Receptor gene, resulting in high serum levels of a structurally and biologically normal, but inactive GH and low-to-undetectable IGF-I. Aim: To summarize the disabilities and handicaps observed in patients with LS, from infancy through adult age. Results: Diagnosing, treating and following a cohort of 76 patients with LS (in many cases from infancy into adu…

Pseudoachondroplasia associated with os odontoideum and retro-odontoid mass: case-based update.

PURPOSE: Pseudoachondroplasia is a rare skeletal dysplasia caused by a mutation in the COMP gene. Infants with pseudoachondroplasia present with rhizomelic dwarfism. Pseudoachondroplasia can resemble achondroplasia, which also presents with a phenotype of rhizomelic dwarfism. The differentiation between these two conditions is important because while both can present with cervicomedullary compromise, the underlying pathophysiology and its management is entirely different. RESULTS: We present a case of a 13-year-old female patient with pseudoachondroplasia who presented with cervical myelopath…

Intermittent PTH (1-34) injection rescues the retarded skeletal development and postnatal lethality of mice mimicking human achondroplasia and thanatophoric dysplasia.

Achondroplasia (ACH) and thanatophoric dysplasia (TD) are caused by gain-of-function mutations of fibroblast growth factor receptor 3 (FGFR3) and they are the most common forms of dwarfism and lethal dwarfism, respectively. Currently, there are few effective treatments for ACH. For the neonatal lethality of TD patients, no practical effective therapies are available. We here showed that systemic intermittent PTH (1-34) injection can rescue the lethal phenotype of TD type II (TDII) mice and significantly alleviate the retarded skeleton development of ACH mice. PTH-treated ACH mice had longer na…

[Echographic screening: small, yes... but why?].

This study concerns a case of micromelus dwarfism discovered after 30 weeks of amenorrhea, during a second systematic sonography performed in a second pregnancy without pathological history and a normal first pregnancy. This dwarfism is complete, without any associated malformations, but on the contrary a moderate hypotrophy. In addition, the morphology of the long bones is normal, without any evidence of fracture; hand and feet are in normal position, with five digits. Radiography of the uterine contents confirms these findings and shows a normal bony transparency. The karyotype is female, wi…

Lethal short-limbed chondrodysplasia in early infancy.

Nineteen cases of chondrodysplastic short-limbed dwarfism in early infancy were studied in the context of current clinical classification based on established radiographic criteria. The histopathologic findings were sufficiently distinctive in most categories to provide additional diagnostic criteria and to contribute to the understanding and delineation of these disorders. 1. Homozygous achondroplasia is distinguished by markedly disturbed endochondral ossification that differs from the pattern seen in typical heterozygous achondroplasia. The physeal chondrocytes contain abundant granules of …

A Ser(365)-->Cys mutation of fibroblast growth factor receptor 3 in mouse downregulates Ihh/PTHrP signals and causes severe achondroplasia.

Missense mutations in fibroblast growth factor receptor 3 (FGFR3) result in several types of human skeletal dysplasia, including the neonatally lethal dwarfism known as thanatophoric dysplasia. An engineered Ser(365)-->Cys substitution in mouse FGFR3, which is equivalent to a mutation associated with thanatophoric dysplasia-I in humans, has now been shown to cause severe dwarfism but not neonatal death. The mutant mice exhibit shortened limbs as a result of markedly reduced proliferation and impaired differentiation of growth plate chondrocytes. The receptor-activating mutation also resulted i…

The bHLH Transcription Factor DkLHW Is a Positive Regulator of Plant Height in Persimmon.

Dwarfism is an important agronomic trait in perennial fruit trees, although its molecular regulatory mechanism in persimmon remains poorly understood. In this study, we identified a bHLH transcription factor, DkLHW, which functions as a positive regulator of plant height in persimmon. Yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), and pull-down assays demonstrated that DkLHW interacts with DkMYB117. Electrophoretic mobility shift (EMSA) assays showed that DkLHW enhances the DNA-binding ability of DkMYB117 to the DkGA2ox2 promoter. Dual-luciferase reporter assays furth…

Phytoplasma Effector SJP8 Suppresses Host Immunity by Promoting the Degradation of ZjMYB15 and ZjMYB86-like to Perturb Jasmonic Acid and Hydrogen Peroxide Homeostasis in Jujube.

'Candidatus Phytoplasma ziziphi' infection causes severe damage to jujube (Ziziphus jujuba), inducing symptoms such as branch clustering, dwarfism, chlorosis and phyllody, leading to significant economic losses. However, the pathogenic mechanisms underlying jujube witches'-broom (JWB) disease remain largely unknown. In this study, we identified SJP8 as a key secreted effector protein of 'Ca. P. ziziphi'. Overexpression of SJP8 in jujube recapitulated the characteristic JWB-like symptoms, including dwarfism and leaf chlorosis. Transcriptome analysis revealed that SJP8 suppressed host immunity b…

A New Function of MbIAA19 Identified to Modulate Malus Plants Dwarfing Growth.

The primary determinants of apple (Malus) tree architecture include plant height and internode length, which are the significant criteria for evaluating apple dwarf rootstocks. Plant height and internode length are predominantly governed by phytohormones. In this study, we aimed to assess the mechanisms underlying dwarfism in a mutant of Malus baccata. M. baccata dwarf mutant (Dwf) was previously obtained through natural mutation. It has considerably reduced plant height and internode length. A comparative transcriptome analysis of wild-type (WT) and Dwf mutant was performed to identify and an…

A mutation in the promoter of the arabinogalactan protein 7-like gene PcAGP7-1 affects cell morphogenesis and brassinolide content in pear (Pyrus communis L.) stems.

Dwarfing rootstocks and dwarf cultivars are urgently needed for modern pear cultivation. However, germplasm resources for dwarfing pear are limited, and the underlying mechanisms remain unclear. We previously showed that dwarfism in pear is controlled by the single dominant gene PcDw (Dwarf). We report here that the expression of PcAGP7-1 (ARABINOGALACTAN PROTEIN 7-1), a key candidate gene for PcDw, is significantly higher in dwarf-type pear plants because of a mutation in an E-box in the promoter. Electrophoretic mobility shift assays and transient infiltration showed that the transcription f…

Evaluation of the therapeutic potential of a CNP analog in a Fgfr3 mouse model recapitulating achondroplasia.

Achondroplasia (ACH), the most common form of dwarfism, is an inherited autosomal-dominant chondrodysplasia caused by a gain-of-function mutation in fibroblast-growth-factor-receptor 3 (FGFR3). C-type natriuretic peptide (CNP) antagonizes FGFR3 downstream signaling by inhibiting the pathway of mitogen-activated protein kinase (MAPK). Here, we report the pharmacological activity of a 39 amino acid CNP analog (BMN 111) with an extended plasma half-life due to its resistance to neutral-endopeptidase (NEP) digestion. In ACH human growth-plate chondrocytes, we demonstrated a decrease in the phospho…

Grebe syndrome with bilateral fibular hemimelia and thumb duplication.

Grebe syndrome is a rare recessively inherited form of short-limbed dwarfism. Among the skeletal anomalies reported in the past, complete fibular hemimelia and thumb duplication have not been documented. We report a case of Grebe syndrome with these associated anomalies and review the various skeletal anomalies reported in the literature related to this syndrome. Awareness of the skeletal anomalies that can occur in this syndrome should enable an accurate diagnosis.

Pseudoachondroplasia in a child with prolapse of the mitral valve.

Pseudoachondroplasia is an autosomal dominant variant of osteochondroplasia that results in mild to severe short-limb dwarfism and early-onset of osteoarthrosis. It has been linked to results from mutations in the gene for cartilage oligomeric matrix protein. We describe a 4-year-old boy with pseudoachondroplasia who also had prolapse of the mitral valve. To the best of our knowledge, this association has not previously been reported.
