Discovery of 7-alkyloxy- [1,2,4] triazolo[1,5-a] pyrimidine derivatives as selective positive modulators of GABAA1 and GABAA4 receptors with potent antiepileptic activity.

A series of 7-alkoxy - [1,2,4] triazolo [1, 5-a] pyrimidine derivatives were designed and synthesized. Maximal electroshock (MES) and pentylenetetrazole (PTZ) tests were utilized to access their anticonvulsant activity. Most of the series of compounds exhibited significant anti-seizure effects. Further studies demonstrated that the anticonvulsant activity of these compounds mainly depended on their allosteric potentiation of GABAA receptors. Among them, compound 10c was picked for the mechanism study due to its potent activity. The compound is more sensitive to subunit configurations of synapt…

Synthesis, anticonvulsant, and antinociceptive activity of new 3-(3-methyl-2,5-dioxo-3-phenylpyrrolidin-1-yl)propanamides and 3-phenyl-butanamides.

A focused library of new 3-(3-methyl-2,5-dioxo-3-phenylpyrrolidin-1-yl)propanamides and their nonimide analogs were synthesized and tested for anticonvulsant activity. These compounds were obtained through the coupling reaction of the starting carboxylic acids with appropriate amines. The initial anticonvulsant screening was performed in mice (intraperitoneal administration) using the maximal electroshock seizure (MES) and the subcutaneous pentylenetetrazole (scPTZ) seizure models. The most promising compound 6 showed more potent protection in the MES and scPTZ tests than valproic acid, which …

Synthesis and anticonvulsant activity of new N-Mannich bases derived from 3-(2-fluorophenyl)- and 3-(2-bromophenyl)-pyrrolidine-2,5-diones. Part II.

Synthesis and anticonvulsant activity of new N-Mannich bases of 3-(2-fluorophenyl)- and 3-(2-bromophenyl)-pyrrolidine-2,5-diones have been described. Initial anticonvulsant screening was performed in mice after intraperitoneal administration in the maximal electroshock seizure test (MES) and subcutaneous pentylenetetrazole seizures test (scPTZ). The neurotoxicity was determined applying the rotarod test. The in vivo results in mice showed that majority of compounds were effective in the MES test. Only seven molecules showed protection in the scPTZ test. The quantitative evaluation in the MES s…

Synthesis, physicochemical and anticonvulsant properties of new N-[(4-arylpiperazin-1-yl)alkyl]-3-phenyl- and 3-(3-methyl-phenyl)-pyrrolidine-2,5-diones.

The series of N-[(4-arylpiperazin-1-yl)-alkyl]-3-phenyl- and 3-(3-methylphenyl)-pyrrolidine-2,5-diones [VIII-XXV] were synthesized and evaluated for anticonvulsant and neurotoxic properties. Initial anticonvulsant screening was performed in mice, using intraperitoneal (ip) maximal electroshock-induced seizures (MES) and subcutaneous pentylenetetrazole-induced (scPTZ) seizure threshold tests. The neurotoxicity was determined applying the rotorod screen. Compounds VIII-XXV revealed protection only in the electrically induced seizures or were inactive. The most active were Mannich bases of 3-(3-m…

Carbonic anhydrase inhibitors. Inhibition of isoforms I, II, IV, VA, VII, IX, and XIV with sulfonamides incorporating fructopyranose-thioureido tails.

A series of aromatic/heterocyclic sulfonamides incorporating 2,3:4,5-bis-O-(isopropylidene)-beta-d-fructopyranosyl-thioureido moieties has b … nd hCA XIV (K(I)s of 21-3500nM). Two of the new compounds showed anticonvulsant action in a maximal electroshock seizure test in mice, with the fluorosulfanilamide derivative being a more efficient anticonvulsant than the antiepileptic drug topiramate.

3-Sulfamoylmethyl-1,2-benzisoxazole, a new type of anticonvulsant drug. Pharmacological profile.

The anticonvulsant and neurotoxic properties of 3-sulfamoylmethyl-1,2-benzisoxazole (AD-810) have been demonstrated. AD-810 suppressed electrically and chemically induced maximal seizures but did not prevent minimal seizures in experimental animals. In rats, rabbits and dogs, the anticonvulsant activity of AD-810 against maximal electroshock seizures was more potent than those of diphenylhydantoin and carbamazepine. In rats, AD-810 showed more rapid onset as well as longer duration of anticonvulsant activity than the above two drugs. The anticonvulsant effect of AD-810 was reduced but not abol…

GABA-ergic mechanisms in the anticonvulsive activity of newly synthesized barbiturates. II. Correlation of HB-7 with GABA-ergic agents.

The effect of the newly synthesized barbiturate HB-7 (2-hydroxylamino-5-ethyl-5-sec. pentylbarbituric acid) was investigated in experiments on mice in comparison with the effect of pentobarbital (PB), as well as their correlations with agents potentiating GABA-ergic transmission: GABA, aminooxyacetic acid, nipecotic acid, di-n-propylacetate (depakin, DPA), muscimol and baclofen. After the test applied (electroshock convulsions) 50 per cent of the animals in the control group had a tonic convulsion. HB-7 and all the other agents were administered in threshold doses. GABA-ergic agents were found…

Blood pressure responses to captive bolt and electrical stunning in sheep and cattle: A systematic review.

Cattle and sheep are often slaughtered after electrical stunning or captive bolt stunning utilizing pneumatic or cartridge-driven devices. While the procedure is effective, a percentage of animals regain consciousness after the initial shock. One factor that has not been considered to explain this recovery of consciousness is the effect on systemic arterial blood pressure (BP) - the major determinant of cerebral perfusion. A systematic literature search identified papers measuring BP after captive bolt or electrical stunning. BP increases were observed in all studies except a protocol which us

Electroconvulsive therapy is associated with increased immunoreactivity of neuroplasticity markers in the hippocampus of depressed patients.

Electroconvulsive therapy (ECT) is an effective therapy for depression, but its cellular effects on the human brain remain elusive. In rodents, electroconvulsive shocks increase proliferation and the expression of plasticity markers in the hippocampal dentate gyrus (DG), suggesting increased neurogenesis. Furthermore, MRI studies in depressed patients have demonstrated increases in DG volume after ECT, that were notably paralleled by a decrease in depressive mood scores. Whether ECT also triggers cellular plasticity, inflammation or possibly injury in the human hippocampus, was unknown. We her

The effects of chlordiazepoxide HCl administration upon punishment and conditioned suppression in the rat.

Rats were trained to lever press for sucrose on a random interval (RI) 64-s schedule. During a 55 min session there were four 3 min "intrusion periods" signalled by a flashing house-light. In experiment 1 there were two groups matched for baseline response rate. During the intrusion periods one group received response-independent footshock on an independent RI 64 schedule; the other group received response-contingent shock on this schedule. Shock intensity was varied for each rat to match degree of response suppression between the two groups. Chlordiazepoxide HCl (CDP) in doses 0.5-5 mg/kg all

Convenient synthesis of N-alkoxymethylbarbituric acids and N-alkoxymethylhydantoins.

A simple one-step method for the N-alkoxymethylation of barbituric acids and hydantoins is presented. The compounds are N-methoxymethylated or N-ethoxymethylated using phosphorus pentoxide and dimethoxymethane or diethoxymethane, respectively, in a chlorinated solvent. 1-Methoxymethyl-3-ethyl-5-phenylhydantoin showed significant anticonvulsant activity in the subcutaneous pentylenetetrazol test, while 1,3-bis(methoxymethyl)-5-ethyl-5-(p-tolyl) barbituric acid was inactive.
