Qingrequzhuo capsule alleviated methionine and choline deficient diet-induced nonalcoholic steatohepatitis in mice through regulating gut microbiota, enhancing gut tight junction and inhibiting the activation of TLR4/NF-κB signaling pathway.

Qingrequzhuo capsule (QRQZ), composed of Morus alba L., Coptis chinensis Franch., Anemarrhena asphodeloides Bunge, Alisma plantago-aquatica … ere measured through 16S rRNA sequencing. Finally, the effects of QRQZ on gut mucosal permeability, endotoxemia, and liver TLR4/NF-kB signaling pathway levels were examined. Our results showed that QRQZ significantly reduced the lipid accumulation in liver and the liver injury in NASH mice. In addition, QRQZ treatment decreased the levels of inflammatory cyto …

Angptl4 integrates dietary and microbial signals to disrupt gut barrier function in MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major contributor to liver morbidity, yet mechanisms linking gut barri … s with dysbiosis and metabolic dysfunction, capturing a gut barrier-related dimension distinct from endotoxemia or acute injury. Thus, intestinal Angptl4 emerges as a mechanistic hub linking diet, microbiota, and gut-liver dysfunction, supporting precision barrier-targeted strategies in MASLD.

From gut microbiota metabolism to microvascular injury: Exploring the role and mechanisms of gut microbiota in obesity-induced coronary microcirculation dysfunction.

Obesity, a global epidemic, drives coronary microvascular dysfunction (CMD), a precursor to cardiovascular disease, via gut microbiota dysbiosis. Clinical evidence shows a 47.7% reduction in coronary flow reserve among obese patients, highlighting obesity's pivotal role in CMD pathogenesis. This review elucidates molecular mechanisms linking obesity-induced dysbiosis to CMD, focusing on microbial metabolites: TMAO promotes lipid deposition and inflammation; reduced SCFAs impair endothelial function; bile acids modulate vascular tone via FXR/TGR5; LPS induces metabolic endotoxemia; BCAAs trigge…

Soluble, Insoluble, and Mixed Rice Bran Dietary Fibers Generate Distinct Host-Microbiota-Metabolome Responses in DSS-Induced Colitis.

Rice bran dietary fiber comprises soluble and insoluble fractions, but their differential host, microbial, and metabolic effects remain unclear. We compared RB-SDF, RB-IDF, and their mixture (RB-MIX) at two doses in DSS-induced colitis. All formulations alleviated symptoms, histological injury, barrier dysfunction, endotoxemia, and inflammation, but their relative advantages varied by end point and dose. RB-SDF showed histological protection and the greatest high-dose SCFA output, whereas RB-IDF produced distinct benefits for barrier-related and metabolic end points. RB-MIX remained effective …

Carotid body sensing of systemic inflammation contributes to recruitment of the splanchnic anti-inflammatory pathway.

Systemic inflammation engages autonomic reflexes that modulate immune responses, yet the peripheral sensors and central circuits responsible for recruiting sympathetic anti-inflammatory pathways remain incompletely defined. Here, we identify the carotid bodies as critical afferent sensors required for full engagement of a splanchnic sympathetic reflex during endotoxemia. Using in vivo splanchnic sympathetic nerve activity recordings, cytokine measurements, and brainstem neuronal mapping, we show that intraperitoneal injection of lipopolysaccharide (LPS) activates splanchnic sympathetic outflow…

Aptamer targeting HMGB1 attenuates inflammatory disease via domain-specific antagonism.

Damage-associated molecular patterns (DAMPs) are key mediators of inflammatory disease, among which HMGB1 is a prototypical extracellular al … hages. In vivo, ZH-1a reduced late-phase systemic inflammation and multiorgan injury in LPS-induced endotoxemia, improved survival in polymicrobial sepsis, and attenuated inflammatory responses and organ damage in an HMGB1-challenge model. In addition, ZH-1a alleviated joint inflammation and structural damage in collagen-induced arthritis, and further enhance …

Borrelia burgdorferi disrupts gut barrier functions and microbiota-antibody interactions.

Lyme disease, an emerging tickborne disease caused by Borrelia burgdorferi (Bb), can result in myriad symptoms of unknown causes in humans. Here, we show that Bb rapidly disrupts the gastrointestinal immune barrier in infected mice, inducing a "leaky gut" syndrome, characterized by increased gut permeability, systemic endotoxemia, elevated LPS binding protein and intestinal fatty acid binding protein, and altered blood leukocyte profiles. Patients with acute Lyme disease showed similar blood changes that correlated with disease symptoms and liver function measurements and largely resolved afte…

The endocannabinoidome-microbiota axis in obesity: a potential link to colorectal cancer development.

Colorectal cancer (CRC) is a multifactorial disease strongly influenced by genetic and environmental factors, as well as by immune and metab … esity, may influence colorectal tumorigenesis by impairing intestinal barrier integrity, increasing endotoxemia and immune dysregulation, altering microbial metabolite production, and activating NF-κB-mediated pro-tumorigenic signaling pathway. This is the first review to comprehensively integrate current evidence linking obesity-associated eCBome-microbiota …

TIGRIS and EUPHRATES eventually join and provide new evidence: a narrative review of the polymyxin B hemoperfusion.

Septic shock driven by endotoxemia is associated with high mortality despite advances in supportive care. Polymyxin B hemoperfusion (PMX-HP) selectively removes circulating endotoxin and has shown variable efficacy in randomized trials. While earlier studies, such as EUPHAS, suggested benefit, subsequent trials, including ABDO-MIX and EUPHRATES, yielded neutral results, partly due to heterogeneous patient selection. The recently completed TIGRIS trial addressed these limitations by enrolling septic shock patients with intermediate endotoxin activity (EAA: 0.60-0.89) and high multiple organ dys…

Inhibition of acute lung inflammation by a neuroimmune circuit induced by vagal nerve stimulation.

Vagus nerve stimulation (VNS) has been shown to limit immune cell activity across several pathologies ranging from sepsis to auto-immune diseases. While stimulation of vagal efferent neurons is known to reduce maladaptive host responses during endotoxemia, only selective vagal afferent neuron stimulation inhibited TLR7-induced macrophage activation and neutrophil recruitment to the lung. These anti-inflammatory actions are dependent on adrenal gland-derived epinephrine, as adrenalectomy or inhibition of epinephrine production eliminated the protection afforded by VNS. Selective afferent VNS in…

Succinate exacerbates mastitis in mice via extracellular vesicles derived from the gut microbiota: a potential new mechanism for mastitis.

BACKGROUND: A high grain diet causes an ecological imbalance in the gut microbiota and serves as an important endogenous trigger of mastitis in dairy cows, but the underlying mechanisms are unclear. Our previous study revealed that subacute rumen acidosis (SARA)-associated mastitis has distinct metabolic profiles in the rumen, especially a significant increase in succinate, but the role of succinate in the pathogenesis of mastitis remains unclear. RESULTS: Succinate treatment exacerbates low-grade endotoxemia-induced mastitis in mice. Specifically, succinate increased the production of gut mi…

Gut microbiota is involved in male reproductive function: a review.

Globally, ~8%-12% of couples confront infertility issues, male-related issues being accountable for 50%. This review focuses on the influence of gut microbiota and their metabolites on the male reproductive system from five perspectives: sperm quality, testicular structure, sex hormones, sexual behavior, and probiotic supplementation. To improve sperm quality, gut microbiota can secrete metabolites by themselves or regulate host metabolites. Endotoxemia is a key factor in testicular structure damage that causes orchitis and disrupts the blood-testis barrier (BTB). In addition, the gut microbio…

Westernized countries are becoming increasingly dependent on SFA-enriched diets, and a deeper understanding of SFA regulation of innate immune memory is imperative in this pandemic era.

Hyponatremia in a Patient With Vasodilatory Shock Due to Overdose of Antihypertensive Medications: A Case Report.

Vasodilatory shock can be caused by septic shock, neurogenic shock, anaphylaxis, drugs, and toxins. Vasopressin is commonly used for the res … n of vasopressors. She developed hyponatremia after initiation of vasopressin due to the absence of endotoxemia, and her serum sodium normalized once vasopressin was discontinued. We recommend monitoring for hyponatremia as a complication of vasopressin, especially in patients without sepsis.

Dietary Ursolic Acid Prevents Alcohol-Induced Liver Injury via Gut-Liver Axis Homeostasis Modulation: The Key Role of Microbiome Manipulation.

Ursolic acid (UA), a natural triterpenoid widely distributed within fruits and edible plants, has been proven to relieve alcoholic liver disease (ALD). However, the mechanisms involved largely remain unclear. This study investigated whether the beneficial effects of UA on ALD could be related to gut-liver axis (GLA) modulation. Special attention was paid to the contribution of gut microbiome manipulation. UA ameliorated intestinal oxidative stress and barrier dysfunction induced by alcohol. As a consequence of gut leakiness amelioration, the related endotoxemia-mediated liver toll-like recepto…

In vivo, acetate protects mice from NLRP3 inflammasome-dependent peritonitis and LPS-induced endotoxemia. Collectively, our research demonstrates that acetate regulates the NLRP3 inflammasome via GPR43 and Ca2+-dependent mechanisms, which reveals the mechanism of metabolite-mediated NLRP3 inflammasome attenuation and highlights acetate as a possible th …
