Lipoic (thioctic) acid increases brain energy availability and skeletal muscle performance as shown by in vivo 31P-MRS in a patient with mitochondrial cytopathy.

A woman affected by chronic progressive external ophthalmoplegia and muscle mitochondrial DNA deletion was studied by phosphorus magnetic resonance spectroscopy (31P-MRS) prior to and after 1 and 7 months of treatment with oral lipoic acid. Before treatment a decreased phosphocreatine (PCr) content was found in the occipital lobes, accompanied by normal inorganic phosphate (Pi) level and cytosolic pH. Based on these findings, we found a high cytosolic adenosine diphosphate concentration [ADP] and high relative rate of energy metabolism together with a low phosphorylation potential. Muscle MRS

Charge-Transfer Spectroscopy of Bisaxially Coordinated Iron(II) Phthalocyanines through the Prism of the Lever's EL Parameters Scale, MCD Spectroscopy, and TDDFT Calculations.

The position of the experimentally observed (in the UV-vis and magnetic circular dichroism (MCD) spectra) low-energy metal-to-ligand charge-transfer (MLCT) band in low-spin iron(II) phthalocyanine complexes of general formula PcFeL2, PcFeL'L″, and [PcFeX2]2- (L, L', or L″ are neutral and X- is an anionic axial ligand) was correlated with the Lever's electrochemical EL scale values for the axial ligands. The time-dependent density functional theory (TDDFT)-predicted UV-vis spectra are in very good agreement with the experimental data for all complexes. In the majority of compounds, TDDFT predic

Bacteriorhodopsin-based Langmuir-Schaefer films for solar energy capture.

The photovoltaic (PV) solar cell, converting incident solar radiation directly into electrical energy, today represents the most common power source for the earth-orbiting spacecraft, and the utilization of organic materials in this context is here explored in comparison with the present state of the art placing emphasis in organic nanotechnology. Poly[3-3'(vinylcarbazole)] (PVK) was synthesized by oxidative polymerization with ferric chloride of N-vinylcarbazole. The resulting polymer was then deposited on solid support by using the Langmuir-Schaefer (LS) technique. The pressure-area isotherm

Computational Insights into the Esterification of Palmitic Acid with Methanol in Water.

This work provides quantified explanations for the thermodynamic and kinetic characteristics of esterification in aqueous phase, and how phase transfer catalysts improve water phase esterification of fatty acids in a computational-experimental way. Self-catalyzed reaction mode with or without solvation effects, water participated reaction mode, and catalytic reaction mode (catalyzed by p-dodecyl benzene sulfonic acid, DBSA) are discussed. Our results show that the initial self-catalytic reaction mode undergoes the energy barrier of 100.1 kJ/mol, and rises to 148.9 kJ/mol when water molecule is

Alternative mitochondrial electron transfer for the treatment of neurodegenerative diseases and cancers: Methylene blue connects the dots.

Brain has exceptional high requirement for energy metabolism with glucose as the exclusive energy source. Decrease of brain energy metabolism and glucose uptake has been found in patients of Alzheimer's, Parkinson's and other neurodegenerative diseases, providing a clear link between neurodegenerative disorders and energy metabolism. On the other hand, cancers, including glioblastoma, have increased glucose uptake and rely on aerobic glycolysis for energy metabolism. The switch of high efficient oxidative phosphorylation to low efficient aerobic glycolysis pathway (Warburg effect) provides mac

Exploiting Metal-to-Metal Electron Transfer in a Ru(II) Polypyridine-Deferasirox Conjugate for Hypoxic Photodynamic Therapy.

The unique microenvironment of solid tumors, characterized by pathological hypoxia, remains a primary driver of treatment resistance and poor clinical outcomes in oncology. While photodynamic therapy has emerged as a promising treatment modality, its clinical efficacy is constrained by its dependence on molecular oxygen. Conventional photosensitizers typically operate by oxygen-dependent energy transfer, leading to a marked reduction or complete loss of therapeutic activity within the oxygen-depleted regions of solid tumors. Herein, we show that iron coordination can serve as a molecular switc…

A Dual Redox Mediator Concept to Catalyze Both Reaction Directions of the Two-Electron Transfer Path of MnO2 Cathodes for Aqueous Zn Batteries.

The dissolution/deposition energy storage path of MnO2 cathodes involves two-electron transfer and provides doubled theoretical capacity of the typical one-electron transfer reaction. However, its reaction activity and contribution in aqueous Zn batteries are seriously limited. Herein, we present a dual redox mediator (RM) concept to catalyze both of the reduction/oxidation reaction directions of this two-electron transfer path. Uniquely, the 2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) compound features two pairs of redox couples with potentials below and above that of the MnO2 cathode. TEMPO

From mitochondria to immune networks: new mesenchymal stem cell strategies to treat periodontitis.

Periodontitis is a chronic inflammatory disease that damages periodontal tissues and is mainly caused by immune dysfunction. Current treatments, such as mechanical debridement and adjunctive antimicrobial, work poorly; periodontal surgery brings pain and complications.Mesenchymal stem cells (MSCs) have a powerful ability to regulate immune responses and great potential for tissue regeneration, thus attracting extensive attention in the field of periodontal treatment. However, in the microenvironment of chronic periodontal inflammation, the therapeutic effect of MSCs is severely inhibited. Rece

Evidence for water ridges at oil-water interfaces: implications for ion transport.

Understanding ion transport across interfaces is of fundamental importance in many processes such as liquid-liquid extraction, phase transfer catalysis, enhanced oil recovery and emulsion stabilisation. However, the factors that control ion transport across interfaces are poorly known due to a lack of knowledge of structural changes at interfaces. We studied here the effects of ionic concentration and external force on the transport of ions across the decane-water interface using classical molecular dynamics simulations. The results show that the evolution of interfacial structures during ion

Cucurbit[8]Uril Achieved Single-Molecule Chiral Transfer to Supramolecular Circularly Polarized Luminescence.

Herein, cascaded supramolecular assembly was constructed from phenylalanine-modified biphenyl-pyridine derivatives (L/D-FPy), cucurbit[8]uril (CB[8]), and amphipathic sulfonatocalix[4]arene (SC4AD), which not only exhibits cascaded energy transfer and chiral transfer but also achieves amplified circularly polarized luminescence (CPL). The experimental results demonstrate that L/D-FPy and SC4AD directly assemble through electrostatic interactions to form particles, where the luminescence intensity and quantum yield (QY) were significantly enhanced by 140 and 8 times, respectively. The positivel…

Janus particle-type behavior of a model flavone derivative with distinct solvent-dependent competitive photoinduced processes.

In this article, Janus particle-type behavior, i.e. bi-faces character with respect to photoinduced charge transfer (CT) and proton transfer (PT) reactions, has been explored in the custom designed molecule 3-hydroxy-2-(4-(piperidin-1-yl)phenyl)-4H-chromen-4-one (HPPC), a molecule with both CT and PT sites. Solvent dependent distinct excited state CT and PT processes in HPPC have been explored using steady state and time resolved fluorescence spectroscopic measurements and quantum chemical calculations. The molecule HPPC shows only an excited state PT process in non-polar solvents like n-hepta

Monitoring collagen synthesis in fibroblasts using fluorescently labeled tRNA pairs.

There is a critical need for techniques that directly monitor protein synthesis within cells isolated from normal and diseased tissue. Fibrotic disease, for which there is no drug treatment, is characterized by the overexpression of collagens. Here, we use a bioinformatics approach to identify a pair of glycine and proline isoacceptor tRNAs as being specific for the decoding of collagen mRNAs, leading to development of a FRET-based approach, dicodon monitoring of protein synthesis (DiCoMPS), that directly monitors the synthesis of collagen. DiCoMPS aimed at detecting collagen synthesis will be

Spartin-mediated lipid transfer facilitates lipid droplet turnover.

Lipid droplets (LDs) are organelles critical for energy storage and membrane lipid homeostasis, whose number and size are carefully regulated in response to cellular conditions. The molecular mechanisms underlying lipid droplet biogenesis and degradation, however, are not well understood. The Troyer syndrome protein spartin (SPG20) supports LD delivery to autophagosomes for turnover via lipophagy. Here, we characterize spartin as a lipid transfer protein whose transfer ability is required for LD degradation. Spartin copurifies with phospholipids and neutral lipids from cells and transfers phos

Microsecond Barrier-Crossing Collapse in Cytochrome c Folding Revealed by Two-Dimensional Fluorescence Lifetime Correlation Spectroscopy.

Protein folding is initiated with a volume contraction called collapse. While the microsecond collapse of horse cyt c has been intensely studied, its molecular mechanism remains elusive. To characterize the structural change, we prepared three cyt c variants labeled with a fluorescent dye at distinct sites (D50, A83, and E104). We track their dynamics via FRET between the dye and the heme by two-dimensional fluorescence lifetime correlation spectroscopy (2D FLCS), which has single-molecule sensitivity and sub-microsecond time resolution. The results reveal that the collapse exhibits two-state

Intragenomic riboregulatory interaction modulates the conformation of the hepatitis C virus 3'X RNA.

The 3'X RNA of the hepatitis C virus (HCV) is a highly conserved noncoding element required for viral replication. This RNA is known to make intragenomic interactions with another highly conserved element, 5BSL3.2, located in the open reading frame of the HCV genome. This RNA-RNA interaction is part of a riboregulatory network that is hypothesized to control various aspects of viral replication. Here, we used single-molecule FRET to characterize the binding mechanism of this riboregulatory interaction. Binding affinities and equilibrium constants were determined under a wide range of experimen

A Standardized Prism-Based TIRF Platform for Quantitative Single-Molecule Fluorescence Studies of Biomolecular Dynamics.

Single-molecule Förster resonance energy transfer (smFRET) enables direct measurement of nanoscale conformational dynamics and heterogeneity in biomolecules, but quantitative interpretation of smFRET data critically depends on well-controlled excitation geometry, low background fluorescence, robust calibration, and reproducible data-analysis workflows. Prism-based total internal reflection fluorescence (pTIRF) microscopy provides important advantages for such measurements by physically separating excitation and emission paths and generating a highly confined evanescent field, yet practical gui…
