Microbial Ethylene Glycol Metabolism: Pathways, Metabolic Engineering, and PET Upcycling.

Ethylene glycol (EG) is an attractive non-sugar carbon source because it can be obtained from petrochemical routes, renewable production platforms, and the depolymerization of polyethylene terephthalate (PET), thereby linking plastic upcycling with sustainable biomanufacturing. In recent years, substantial progress has been made in elucidating EG catabolism, including the initial oxidation of EG, native glyoxylate assimilation pathways, synthetic route design, and chassis engineering. Nevertheless, efficient and robust EG bioconversion is still limited by unfavorable thermodynamics, low cataly…

Harnessing Machine Learning for Enzyme Enantioselectivity: Toward Intelligent Biocatalysis Engineering.

Enzyme enantioselectivity remains a central challenge in the biocatalytic synthesis of chiral pharmaceuticals and fine chemicals. Conventional evolution methods, constrained by prolonged experimental cycles and computationally intensive processes, struggle to comprehensively explore complex protein sequences. Although machine learning (ML) has been successfully applied to optimize enzymatic properties, such as catalytic efficiency and stability, its potential in enantioselectivity engineering remains underexploited. This review systematically evaluates how machine learning integrates multimoda…

Engineering a stable high-copy episomal plasmid in Yarrowia lipolytica.

Episomal plasmids are widely used in yeast metabolic engineering for their flexibility and high transformation efficiency. However, their application in Yarrowia lipolytica is often limited by low copy number and insufficient genetic stability. In this study, a robust episomal plasmid that balanced genetic stability and protein expression capacity was developed in Y. lipolytica. The autonomously replicating sequence (ARS) elements were selected, and ARS1 was identified as a favorable element that balanced genetic stability and gene expression capacity. To improve plasmid copy number, different…

Structure-Property Engineering in Ag23 Nanoclusters for Electrocatalytic Nitrate Reduction.

Electrocatalytic nitrate reduction reaction (NO3RR) provides a sustainable pathway for ambient ammonia synthesis; however, achieving concurrent high activity, high selectivity and appropriate stability of catalysts remains a substantial challenge. Here we report the synthesis and structure-property engineering of a class of atomically precise silver nanoclusters via geometric tuning and copper substitution. Compared with the parent nanocluster Ag23(PET)18(PPh3)8 (abbreviated as Ag23-1), a Cu-substituted Ag19Cu4(SCH2PhCl)18(PPh3)4 nanocluster (i.e., Ag19Cu4) modulates the electronic structure a…

Structured Pearcey-prime Gaussian beams and phase-controlled trajectory engineering.

We present a unified framework for generating structured Pearcey-prime Gaussian beams (PePGBs) with controllable spatial profiles and propagation dynamics. By introducing a generalized radial mapping function, diverse beam structures, including circular, elliptical, petal-shaped, oscillator-like, star-shaped, and lattice configurations, are constructed at the input plane through controlled redistribution of transverse spatial frequencies. Phase modulation is further employed to engineer the beam trajectory without altering its initial structure. The results show that the beams propagate along

Controlled Presodiation of Sodium-Ion Cathode via Electron Cloud Engineering.

Chemical presodiation offers a promising pathway to compensate sodium deficiency in cathode and anode materials, thereby enhancing the usable energy density of sodium-ion batteries. This approach is particularly critical for P2- and tunnel-type cathodes, such as Na0.44MnO2, where the as-synthesized materials typically exhibit a 50%-60% sodium deficiency. However, conventional presodiation reagents possess excessively high reductive potential, often leading to oversodiation, structural degradation, and compromised electrode reversibility. Here, we report a rational electron-cloud engineering st…

Oncolytic reovirus: tumor-microenvironment remodeling, engineering constraints, and translational strategies.

Oncolytic reovirus (ReoV) has emerged as a promising cancer immunotherapy platform because of its natural tumor tropism and immunostimulatory properties. Reovirus enters susceptible cells through receptors including junctional adhesion molecule-A and exploits oncogenic signaling and impaired antiviral responses to preferentially replicate in tumor cells. Viral replication induces apoptosis and other context-dependent forms of programmed cell death, while tumor lysis promotes antigen release and immune activation. However, reovirus monotherapy has shown limited clinical efficacy because of circ

Molecular Engineering of Hemicyanine Scaffold for Peroxynitrite-Activated Chemiluminescence Imaging.

Chemiluminescence eliminates interference from real-time excitation light, thereby maximizing the signal-to-noise ratio of imaging. However, the reported chemiluminescent scaffolds remain limited, and the development of novel and efficient chemiluminescent scaffolds is critical and urgent. Herein, we synthesized a series of peroxynitrite (ONOO-)-activated chemiluminescent probes based on a hemicyanine scaffold through molecular engineering, and systematically investigated optical properties as well as their response mechanisms. Upon reaction with ONOO-, the vinylene units in the hemicyanine sc…

Strategies for tailoring clusteroluminescence: from aggregation control to molecular engineering.

Clusteroluminescence (CL), light emission from non-conventional systems via through-space interaction (TSI), offers unique photophysical properties distinct from traditional luminophores. This review outlines dual-level strategies for tailoring CL: aggregation regulation and molecular engineering. Controlling aggregation via crystalline or amorphous packing, spatial confinement, and microenvironment design modulates cluster formation and TSI efficiency. Molecular approaches, such as heteroatom incorporation, substituent tuning, and conformational control, enable precise manipulation of electro…

Ferromagnetic Interface Engineering of Spin-Charge Conversion in RuO_{2}.

Spin-orbit torque efficiency is conventionally fixed by bulk materials. D-wave altermagnets introduce an additional nonrelativistic spin-charge conversion channel beyond the inverse spin-Hall effect. Using prototypical candidate RuO_{2} as an example, we show that the adjacent ferromagnet alone can dictate both the magnitude and sign of spin-charge conversion. Spin-pumping measurements on RuO_{2}/Y_{3}Fe_{5}O_{12} (YIG) and RuO_{2}/Ni_{80}Fe_{20} (Py) bilayers yield opposite effective spin-Hall angles that persist across crystalline and polycrystalline RuO_{2}.
