Changes in myofibrillar structure and function produced by N-terminal deletion of the regulatory light chain in Drosophila.

The similarity of amino acid sequence and motifs of the N-terminal extensions of certain class II myosin light chains, found throughout the animal kingdom, suggest a common functional role. One possible role of the N-terminal extension is to enhance oscillatory work and power production in striated muscles that normally operate in an oscillatory mode. We conducted small-angle X-ray diffraction experiments and small-length-perturbation analysis to examine the structural and functional consequences of deleting the N-terminal extension of the myosin regulatory light chain (RLC) in Drosophila flig

A knockout screen of genes expressed specifically in Ae. aegypti pupae reveals a critical role for stretchin in mosquito flight.

Aedes aegypti is a critical vector for transmitting Zika, dengue, chikungunya, and yellow fever viruses to humans. Genetic strategies to limit mosquito survival based upon sex distortion or disruption of development may be valuable new tools to control Ae. aegypti populations. We identified six genes with expression limited to pupal development; osi8 and osi11 (Osiris protein family), CPRs and CPF (cuticle protein family), and stretchin (a muscle protein). Heritable CRISPR/Cas9-mediated gene knockout of these genes did not reveal any defects in pupal development. However, stretchin-null mutati

An ensemble dose-response model for monoenergetic ions and doses relevant for space radiation carcinogenesis.

For future missions to the moon or Mars, exposure to space radiation is a significant hazard requiring accurate risk assessment tools to guide and evaluate mitigation strategies. NASA has developed a cancer risk assessment model to perform such projections, but uncertainties associated with radiation quality and dose-rate effects are substantial. Quality factors (QFs) are used to describe the enhanced effectiveness of radiation types found in space for inducing a health effect (carcinogenesis, in this case) compared to terrestrial radiation. Space radiation QFs are developed on the basis of do

Multiscale Modeling and Systems Biology in Microgravity Investigations.

Spaceflight imposes a unique combination of environmental stressors including ionizing radiation, microgravity, and prolonged isolation that disrupt biological systems from the genome to the proteome. These stressors contribute to a range of health effects such as musculoskeletal degradation, neuroimmune dysfunction, and psychological challenges. Advances in countermeasure development, including biomedical devices and therapeutics, have already yielded terrestrial benefits for conditions like osteoporosis and cardiovascular disease. To systematically characterize spaceflight-induced biological

Low-dose simulated galactic cosmic radiation exposure after hindlimb unloading has long-term impact on skeletal muscle metabolic proteins in female mice.

Spaceflight stressors, including microgravity-induced unloading and galactic cosmic radiation (GCR), acutely disrupt mitochondrial function and contribute to skeletal muscle atrophy. The long-term remodeling of skeletal muscle following combined unloading and radiation exposure remains poorly understood. We investigated protein abundance changes 9-mo postexposure to combined unloading and radiation exposure. Female, 6-mo-old, C57Bl/6J mice underwent 5 days of hindlimb unloading (HU) or weight-bearing (WB) conditions, followed by 0, 0.5, or 1.5 Gy of simulated GCR exposure using the simplified

Leveraging bioinformatics approaches for drug repositioning in space radiation protection.

The health effects of space radiation, primarily Galactic Cosmic Rays (GCRs), on humans remain largely unknown, with potential cardiovascular consequences posing a significant threat to astronauts on long-duration spaceflight missions. Currently, there are no established pharmacological countermeasures for GCR exposure. Drug repositioning offers a promising strategy to accelerate pharmaceutical research in space medicine. This study leverages existing bioinformatics techniques to identify and prioritize potential drug candidates associated with proteomic perturbations following simulated GCR e

Reduced Mechanical Tactile Stimulation Under Space Microgravity Affects Synaptic Signaling and Contributes to Neuromuscular Aging in Caenorhabditis elegans.

Although space travel is becoming more accessible, our understanding of how the space environment and microgravity (μG) affect biology, physiology, and human health remains incomplete. This study examined the effects of μG on synaptic signaling and neuromuscular aging in Caenorhabditis elegans. The D01 cohort, consisting of L4 larvae to young adults raised in μG, exhibited a downregulation of genes linked to synaptic signaling, dopamine response, locomotion, cuticle development, and mitochondrial metabolism. This was accompanied by altered synapse dynamics, reduced motility, and shorter body l

Impact of spaceflight on the murine thymus and mitigation by exposure to artificial gravity during spaceflight.

The environment experienced during spaceflight may impact the immune system and the thymus appears to undergo atrophy during spaceflight. However, molecular aspects of this thymic atrophy remain to be elucidated. In this study, we analysed the thymi of mice on board the international space station (ISS) for approximately 1 month. Thymic size was significantly reduced after spaceflight. Notably, exposure of mice to 1 × g using centrifugation cages in the ISS significantly mitigated the reduction in thymic size. Although spaceflight caused thymic atrophy, the global thymic structure was not larg

Insulin secretion and sensitivity in space flight: diabetogenic effects.

Nearly three decades of space flight research have suggested that there are subclinical diabetogenic changes that occur in microgravity. Alterations in insulin secretion, insulin sensitivity, glucose tolerance, and metabolism of protein and amino acids support the hypothesis that insulin plays an essential role in the maintenance of muscle mass in extended-duration space flight. Experiments in flight and after flight and ground-based bedrest studies have associated microgravity and its experimental paradigms with manifestations similar to those of diabetes, physical inactivity, and aging. We p

In vitro cultured cells as probes for space radiation effects on biological systems.

Near future scenarios of long-term and far-reaching manned space missions, require more extensive knowledge of all possible biological consequences of space radiation, particularly in humans, on both a long-term and a short-term basis. In vitro cultured cells have significantly contributed to the tremendous advancement of biomedical research. It is therefore to be expected that simple biological systems such as cultured cells, will contribute to space biomedical sciences. Space represents a novel environment, to which life has not been previously exposed. Both microgravity and space radiation

Microbiological Quality of Maize Silage in Relation to Agricultural Practices: A Four-Year Study.

Silage is a fundamental component of cattle feed, and its microbiological quality is critical for animal health and human safety. Improper ensiling conditions, such as oxygen exposure or inadequate acidification, can promote the growth of pathogens like Listeria monocytogenes, Clostridium botulinum, and Bacillus cereus. This study aimed to evaluate the microbial status of maize silages and identify pre-ensiling factors influencing its hygienic safety. Over a four-year period, 406 silage samples were collected from cattle farms across Poland. The research evaluated general hygiene indicators an

A new era of dermatophytes: emerging species, new risk factors, and taxonomic change.

SUMMARYThe traditional landscape of dermatophyte infections has changed over the past decade. The emergence and global expansion of Trichophyton indotineae have highlighted the impact of irrational and uncontrolled antifungal and topical corticosteroid use, presented treatment challenges, and highlighted the deficiencies of currently available diagnostic methods. Trichophyton mentagrophytes genotype VII, which has until recently been associated with animal reservoirs, is now associated with human-to-human transmission and is frequently related to sexual transmission. Infections associated with

Synthesis of a dehydrodieugenol B derivative as a lead compound for visceral leishmaniasis-mechanism of action and in vivo pharmacokinetic studies.

Leishmaniasis is a parasitic neglected tropical disease, affecting 12 million people. Available treatments present several limitations, with an increasing number of resistance cases. In the search for new chemotherapies, the natural product dehydrodieugenol B was used as a scaffold for the synthesis of a series of derivatives, resulting in the discovery of the promising analog [4-(4-(5-allyl-3-methoxy-2-((4-methoxybenzyl)oxy)phenoxy)-3-methoxybenzyl)morpholine, 1]. In this work, we investigated the effect of compound 1 on cell signaling in Leishmania (L.) infantum, culminating in cell death, a

Comparison of manual and automated methods in the hematological evaluation of a frugivorous bat captured in urban green areas.

Increasing and unplanned urbanization can intensify wildlife contact with humans and domestic animals, raising concerns about the spillover of zoonotic pathogens. Bats play a relevant ecological role due to their great niche diversity and wide distribution, and may act as reservoirs for various pathogens. Despite their importance, hematological values for non-hematophagous bats remain limited, hindering the assessment of their health and physiological responses. This study characterized hematological parameters and blood cell morphology in Artibeus planirostris captured in urban green areas of
