Clonal plants enhance their fitness in heterogeneous environments through clonal integration, which enables resource sharing among interconnected ramets. However, how clonal integration shapes rhizosphere microbiome assembly and function remains poorly understood. In this study, we constructed patchy heterogeneous soils with cadmium-polluted and unpolluted substrates to simulate fragmented polluted habitats. Apical ramets were grown in cadmium-polluted soil and basal positions were cultivated in unpolluted soil, with stolons left either intact or severed. Rhizosphere and root-zone soils were s

BACKGROUND: Little is known about intestinal fungi in IBS patients whose gut bacteria have been investigated a lot. In order to explore causal relationship between IBS and gut mycobiome, and use gut fungi to diagnose or even treat IBS, further characterization of it in IBS is required.
METHODS: Fifty-five diarrhea-predominant IBS (D-IBS) patients fulfilling Rome III criteria, and 16 healthy controls (HC) were recruited. Fresh fecal samples were collected and used for 16s rRNA and ITS2 high-throughput sequencing. Diversity and composition of gut bacteria and fungi, as well as bacterial-fungal

A pot experiment was carried out to simulate soil contaminated by sulfamethoxazole at different concentrations. The community structure of soil microorganisms was investigated using Illumina high-throughput sequencing, and 64 subtypes of antibiotic resistance genes (ARGs) resistant to six classes of antibiotic were also analyzed by PCR and droplet digital PCR. The results showed that soil contamination with sulfamethoxazole had no significant effect on fungal diversity after 120 days (P>0.05) whereas bacterial diversity was significantly reduced (P<0.05). The microbial community structure of t

Inducible bronchus-associated lymphoid tissue (iBALT) is an ectopic lymphoid structure composed of highly organized T cell and B cell zones that forms in the lung in response to infectious or inflammatory stimuli. Here, we develop a model for fungal-mediated iBALT formation, using infection with Pneumocystis that induces development of pulmonary lymphoid follicles. Pneumocystis-dependent iBALT structure formation and organization required CXCL13 signaling. Cxcl13 expression was regulated by interleukin (IL)-17 family members, as Il17ra-/-, Il17rb-/-, and Il17rc-/- mice failed to develop iBALT.

Tropical montane forests are undergoing rapid climate change, with declining cloudiness and warming in higher elevations. While impacts on vegetation are increasingly documented, effects on belowground microbial communities and ecosystem processes remain poorly studied. We investigated how experimental drought and warming influence soil microbial communities and decomposition across a 900-m elevation gradient in the tropical montane forests of the Dominican Republic. Precipitation exclusion shelters simulated drought at high (2250 m a.s.l.) and low elevation (1350 m a.s.l.) and high elevation

Carbonic anhydrases (CAs) are a class of zinc-containing metalloenzymes widely present in the biological world, catalyzing the reversible hydration of CO2 to form HCO3- and H+. These enzymes play essential roles in pH homeostasis, gas exchange, metabolic regulation, and virulence expression in pathogens. In fungi, CAs mainly belong to the α- and β-classes and have undergone extensive diversification during evolution. In plant pathogenic fungi, the functions of CAs have extended beyond traditional metabolic roles, evolving into key "environmental adaptation and virulence regulatory factors." Th

Fungal biofilms represent a major therapeutic hurdle due to their drug resistance and immune evasion. Here, we report a cocrystal-inspired nano-assembly strategy that co-assembles glycyrrhizic acid (GA), a natural triterpenoid saponin, with azole antifungal agents into multifunctional nanoparticles. The GA-azole co-assemblies, stabilized via hydrogen bonding and hydrophobic interactions, exhibit cocrystal-like properties while preserving the bioactivities of both components. This hybrid nanoplatform improves azole solubility and permeability and leverages GA's intrinsic membrane-disrupting, RO

Storage temperature critically governs physicochemical deterioration and fungal community succession in paddy rice. This study examined quality changes and fungal dynamics in three Yongyou-series hybrid rice varieties stored for 240 days at 15 °C and 25 °C. Fatty acid value (FAV), malondialdehyde (MDA), and electrical conductivity increased progressively, while polyphenol oxidase activity and soluble albumin and globulin contents declined; all changes were more pronounced at 25 °C, indicating accelerated lipid oxidation, membrane damage, and protein structural alteration. Human mycoses cover a diverse field of fungal diseases from skin disorders to systemic invasive infections and pose an increasing global health problem based on ineffective treatment options, the hampered development of new efficient drugs, and the emergence of resistant fungal strains. Niclosamide is currently applied for the treatment of worm infections. Its mechanisms of action, which include the suppression of mitochondrial oxidative phosphorylation (also known as mitochondrial uncoupling), among others, has led to a repurposing of this promising anthelmintic drug for the therapy of furthe

Biofilm is a complex structure consisting of microorganisms such as bacteria, fungi and an extracellular matrix (ECM). Biofilms are involved in most microbial infections and show persistent resistance to antibiotic treatment and immune response. Both Aspergillus fumigatus and Streptococcus pneumoniae are colonizers that can form biofilms in the respiratory tract. These pathogens have been simultaneously isolated from the same patient, but their interaction is poorly understood. We observed morphological changes in single- and mixed-species biofilms prepared for confocal laser scanning microsco

The human respiratory tract is exposed daily to airborne fungi, fungal enzymes, and secondary metabolites. The endemic fungi Histoplasma capsulatum, Coccidioides spp, Blastomyces dermatitidis, and Paracoccidioides brasiliensis, and occasionally Aspergillus fumigatus, are primary pulmonary pathogens of otherwise healthy people. Such infections resolve in most people, and only a few infections lead to disease. However, many fungi are directly allergenic by colonising the respiratory tract or indirectly through contact with cell wall constituents and proteases, causing or exacerbating allergic di

Finger millet is an ancient African-Indian crop that is resistant to many pathogens including the fungus, Fusarium graminearum. We previously reported the first isolation of putative fungal endophytes from finger millet and showed that the crude extracts of four strains had anti-Fusarium activity. However, active compounds were isolated from only one strain. The objectives of this study were to confirm the endophytic lifestyle of the three remaining anti-Fusarium isolates, to identify the major underlying antifungal compounds, and to initially characterize the mode(s) of action of each compoun

We report the discovery of a novel dual inhibitor targeting fungal sterol 14α-demethylase (CYP51 or Erg11) and human 5-lipoxygenase (5-LOX) with improved potency against 5-LOX due to its reduction of the iron center by its phenylenediamine core. A series of potent 5-LOX inhibitors containing a phenylenediamine core, were synthesized that exhibit nanomolar potency and >30-fold selectivity against the LOX paralogs, platelet-type 12-human lipoxygenase, reticulocyte 15-human lipoxygenase type-1, and epithelial 15-human lipoxygenase type-2, and >100-fold selectivity against ovine cyclooxygenase-1 a

The pathomechanism of chronic rhinosinusitis with nasal polyposis (CRS/NP) seems to be unclear. Bacterial-, fungal- and combined biofilms might play a potential role in the pathogenesis of various inflammatory diseases and recently in CRS/NP. A prospective, blinded observational study was performed to confirm that the combination of conventional hematoxylin-eosin (HE) and Gram staining protocols could be used to detect bacterial and fungal biofilms in patients with CRS/NP. A total of 50 patients with CRS/NP undergoing endoscopic sinus surgery (ESS) were analyzed. The negative control group con

The echinocandin (candin) class of antifungal drugs inhibit beta-1,3-glucan synthase and block synthesis of beta-1,3-glucan , an important polysaccharide in fungal cell walls. Candins are used widely for treatment of systemic infections caused by Candida and Aspergillus because of their high potency and low toxicity to humans. The incidence of candin resistance has been rare compared to that of azole resistance, although candin-resistant clinical isolates of C. albicans, C. glabrata, C. krusei and C. tropicalis have been reported in the USA and Europe in recent years. These isolates possess hu

Fonsecaea pedrosoi is a fungal pathogen that produces melanin. The functions of melanin and its possible influence in the protective immunological response during infection by F. pedrosoi are not known. In this work, treatment of F. pedrosoi mycelia with proteases and glycosidases followed by a denaturing agent and hot concentrated acid left a black residue. Scanning electron microscopy demonstrated that this processed melanized residue resembled very closely the intact mycelium in shape and size. Melanin particles were also isolated from culture fluids of conidia or sclerotic forms of F. pedr

Heavy metal (HM) contamination arising from rapid industrialization poses critical threats to global ecosystem integrity and public health. Conventional physicochemical approaches are limited by high costs, incomplete removal, and toxic waste generation, necessitating sustainable alternatives. Mycoremediation, which harnesses the remarkable, diverse capacities of fungi to tolerate and mitigate HM stress through sophisticated biological mechanisms, has emerged as a promising and sustainable approach to address HM pollution. This review examines the sources and ecotoxicological impacts of HM pol

In Candida albicans, potassium (K+) channels fine-tune ionic balance under stress, contributing to host colonization. Fungal two-pore domain, outwardly rectifying potassium (TOK) channels remain insufficiently characterized despite evidence implicating them in growth and viability. Here, we describe the atomic-resolution structure of a fungal potassium channel, TOK1 from C. albicans (CaTOK), revealing an architecture defined by eight transmembrane helices and a membrane topology distinct from previously characterized K⁺ channel classes. The first four helices form a tetraspanin-like bundle res

Adenylyl cyclase (AC) is the central regulator of cAMP signalling in fungal pathogens, yet fungal ACs uniquely contain a protein phosphatase 2C (PP2C) domain whose function remains unclear. Here, we characterise the PP2C domain of the AC Cac1 in the human pathogen Cryptococcus neoformans. Structural and biochemical analyses show that Cac1-PP2C adopts a non-canonical scaffold but functions as a Ser/Thr-specific metalloenzyme. Deletion of the PP2C or AC catalytic (ACC) domain demonstrated that PP2C is required for full Cac1 activity, influencing melanin and capsule synthesis, sexual differentiat

Fungal pathogens have emerged as one of the most significant threats to global public health. Invasive fungal infections, characterized by high morbidity and mortality rates, have become one of the most severe diseases, posing a substantial threat to human health. In this study, a rational drug design strategy was employed, targeting lanosterol 14α-demethylase (CYP51). Using SCZ-14, a CYP51 inhibitor with moderate antifungal activity, as the lead compound, 27 novel 2-phenylthiazole derivatives were designed and synthesized through two rounds of structural optimization. Among these compounds, c
