FpNuc1α regulates phosphate signaling and pathogenicity in Fusarium proliferatum infection of jujube fruit by targeting FpGit1 via a novel BSN motif.

Fusarium proliferatum is a postharvest pathogen responsible for severe rot in economically important fruits and vegetables, leading to food waste and safety concerns due to mycotoxin contamination. While the PHO signaling pathway is known to regulate phosphate homeostasis, its specific contribution to the virulence mechanisms of this foodborne pathogen remains unclear. In this study, we functionally characterized key components of the PHO pathway in F. proliferatum. Targeted deletion of PHO pathway genes (FpNuc1α, FpNuc1β, FpNuc2, FpPho80, and FpPho85) revealed their essential roles in vegetat…

Integrated Transcriptomic, Enzymatic, and Immunolocalization Analysis Reveals Pectin Remodeling-Mediated Defense Against Fusarium oxysporum f. sp. cubense Race 4 in Banana.

Fusariumoxysporum f. sp. cubense race 4 (Foc 4) causes Fusarium wilt by penetrating root cell walls, yet the molecular basis of cell wall-mediated resistance remains poorly understood. Here, we investigated the transcriptional, enzymatic, and cellular responses of the resistant banana cultivar Dongjiao No. 1 (DJ) and its susceptible mutant ke2 following Foc 4 infection. RNA sequencing revealed that DJ specifically upregulated a pectin degradation cassette comprising pectin methylesterase (PME3-Like), pectin acetylesterase (PAE1), and polygalacturonases (PG1, PG3, PG5, PG-Like-4) at the bud see…

Fusaspironols A-E: Steroidal spirostanols from the deep-sea-derived Fusarium inflexum ZEN8 with inhibitory activities against osteoclastogenesis and adipogenic differentiation.

Five previously undescribed polyhydroxylated spirostanols, fusaspironols A-E (1-5), along with eight known compounds (6-13), were isolated from the deep-sea-derived fungus Fusarium inflexum ZEN8. Their structures were elucidated using spectroscopic analysis, GIAO-based NMR calculations, and DP4+ probability analysis. Notably, this represents only the second report of spirostanols of fungal origin. A plausible biosynthetic pathway linking compounds 1-5 to the putative precursor cholesterol in this fungus was proposed. In biological assays, compound 8 significantly inhibited RANKL-induced osteoc…

Fusarium solani Skin and Soft Tissue Infection in a Patient With Chronic Sclerosing Skin Graft-vs-Host Disease: A Case Report.

BACKGROUND Fusariosis is an exceedingly rare infection most often attributable to the fungus Fusarium solani. It is a rare but known cause of invasive mold infections in immunocompromised patients post-hematopoietic cell transplant (post-HCT). Fusariosis can be extremely difficult to treat due to its tendency to disseminate in the context of underlying immunosuppression, and its high in vitro resistance to mainline antifungal agents. CASE REPORT Here we present a post-HCT patient undergoing immunosuppressive therapy for diagnosed chronic graft-versus-host disease (GVHD) of the skin who present…

Antioxidative Defence Mechanisms in Tomato Plants Induced by Biopriming with Trichoderma virens and Jasmonic Acid Against Fusarium Wilt and Damping-Off.

In plants, pathogenic infection causes oxidative damage to the host plants and eventually leads to the destruction of the plant's metabolism due to the oxidation of essential biomolecules. In this investigation, we monitored the effects of fusarium wilt and damping-off mediated generation of Reactive oxygen species (ROS) and their effective disposal by the antioxidative defence system in tomato plants. The main aim of the current research work was to assess the seed priming remedial effects of Trichoderma virens (Tv) (1 × 107 spores/mL) as a biocontrol agent and jasmonic acid (JA) (1 μM) as a …

Design, Synthesis, Structure-Activity Relationship, and Three-Dimensional Quantitative Structure-Activity Relationship of Fusarium Acid Derivatives and Analogues as Potential Fungicides.

In research related to fungicides, the development of compounds from natural products with high antifungal activity has attracted considerable attention. Fusaric acid (FA), an alkaloid isolated from the metabolites of Fusarium oxysporum, is an important precursor for developing pharmacologically active herbicides. In our previous work, we reported that FA has a wide range of inhibitory activities against 14 plant pathogenic fungi. In particular, it exhibited excellent antifugal effects on Colletotrichum higginsianum (EC50 = 31.7 μg/mL). Herein, to explore the practical application in the agric…

Ethylenediaminetetraacetic Acid Disodium Salt Acts as an Antifungal Candidate Molecule against Fusarium graminearum by Inhibiting DON Biosynthesis and Chitin Synthase Activity.

Fusarium fungi are the cause of an array of devastating diseases affecting yield losses and accumulating mycotoxins. Fungicides can be exploited against Fusarium and deoxynivalenol (DON) production. However, Fusarium resistance to common chemicals has become a therapeutic challenge worldwide, which indicates that new control agents carrying different mechanisms of action are desperately needed. Here, we found that a nonantibiotic drug, ethylenediaminetetraacetic acid disodium salt (EDTANa2), exhibited various antifungal activities against Fusarium species and DON biosynthesis. The infection of…
