A previous study described the establishment of human embryonic stem cell (ESC) lines from different sources of embryonic material, including morula, whole blastocyst and isolated inner cell mass. Using these methods, a repository of ESC lines has been established with different genetic abnormalities, which provides an unlimited source of disease cells in culture for undertaking research on the primary disturbances of the cellular processes in the genetically abnormal cells. ESC lines with genetic disorders were derived from the mutant embryos detected and avoided from transfer in the ongoing

The discovery of genetic exchange in African trypanosomes belonging to the Trypanosoma brucei group is an important development in our understanding of these organisms. Genetic exchange is a feature of major importance in relation to population structure and speciation. Furthermore, a convenient laboratory-based mating system would be of considerable value as a tool in trypanosomiasis research. It is now known that although cyclical development of trypanosomes within the tsetse fly does not require mating to occur, genetic exchange may take place under Conditions in which genetically distinct

The findings presented here are from a qualitative study in which data were gathered from 20 women who had received inconclusive genetic testing results for inherited breast cancer susceptibility. Before describing the significance, for them, of their genetic test results, all of the participants related what it was like to live with a strong family history of breast cancer. The focus of this article is the women's experience of living with a personal and strong family history of breast cancer. For these women, having such a history had become a fact of life that could not be ignored.Three the

Liver xenotransplantation has emerged as a potential solution to the shortage of deceased human donor organs and is now becoming a reality due to recent developments in genetic engineering and immunosuppressive therapy. Early efforts using non-human primates and genetically modified pigs faced significant challenges such as thrombocytopenia and graft rejection. Understanding the mechanism behind those challenges and using novel genetically engineered pigs enabled researchers to overcome some of the hurdles, but more research is needed. However, new advances might allow pig liver xenotransplant

The mammalian gut is colonized by trillions of microorganisms collectively called the microbiome. It is increasingly clear that this microbiome has a critical role of in many aspects of health including metabolism and immunity. While environmental factors such as diet and medications have been shown to influence the microbiome composition, the role of host genetics has only recently emerged in human studies and animal models. In this review, we summarize the current state of microbiome research with an emphasis on the effect of host genetics on the gut microbiome composition. We focus particul

The vestibular disorders, which generally manifest themselves as nausea, vomiting, and dizziness or vertigo, have troubled many people. Increasing evidence suggests that certain vestibular disorders are hereditary, and gene variants contribute to the onset and susceptibility of these disorders. This review summarizes the latest research progress, especially the molecular and genetic aspects of several common vestibular disorders, including Meniere's disease, vestibular migraine, benign paroxysmal positional vertigo, and motion sickness, with the aim of encouraging more fundamental research and

Autoimmune hepatitis (AIH) is liver parenchymal inflammation mediated by the autoimmune response of hepatocytes. Its clinical features include elevation of aminotransferases in serum, presence of various autoantibodies in circulation, hypergammaglobulinemia, and interface hepatitis in liver tissue. Although the etiology and pathogenesis of AIH have not been fully elucidated, a consensus has been reached on the theories of "genetic susceptibility" and "molecular simulation" , and "immunoregulatory disorder" remains a hot research topic for many scholars. This article reviews the research advanc

Calcineurin inhibitors (CNIs) are the mainstay of immunosuppression in kidney transplantation. Interpatient variability in the disposition of calcineurin inhibitors is a well-researched phenomenon and has a well-established genetic contribution. There is great diversity in the makeup of African genomes, but very little is known about the pharmacogenetics of CNIs and transplant outcomes. This review focuses on genetic variants of calcineurin inhibitors' metabolizing enzymes (CYP3A4, CYP3A5), related molecules (POR, PPARA) and membrane transporters involved in the metabolism of calcineurin inhib

Serotonergic dysregulation is thought to underlie much of the pathology in bulimia nervosa (BN). The purpose of this review is to expand the serotonergic model by incorporating specific and nonspecific contributions of estrogens to the development and maintenance of bulimic pathology in order to guide research from molecular genetics to novel therapeutics for BN. Special emphasis is given to the organizing theory of general brain arousal which allows for integration of specific and nonspecific effects of these systems on behavioral endpoints such as binge eating or purging as well as arousal s

Human tooth agenesis involves the absence of one or more teeth and often refers to a genetic or a developmental disorder. Identifying whether certain patterns of tooth agenesis are more prevalent than others may prove valuable for treatment planning, as well as for genetic research into its etiology and development. In the present article a method is described where unique values are assigned to patterns of tooth agenesis. Using the idea of binary arithmetic, the absence or presence of teeth are represented by 1 and 0, and translated into corresponding unique values, the Tooth Agenesis Code (T…

INTRODUCTION: Pharmacogenetics evaluates how genetic variations influence drug responses. Nowadays, genetic tests have advanced, becoming more affordable, and its integration is supported by stronger clinical evidence. Guidelines such as those from CPIC (Clinical Pharmacogenetics Implementation Consortium) and resources like PharmGKB facilitate genotype-based prescribing; and organizations like the FDA promote genetic testing before initiating certain medications. Preventive pharmacogenetic panels seem promising, but further research on biomarkers and diverse populations is needed. The aim of

Inflammatory mediators are usually defined as extracellular signals, yet intracellular regulators determine which signals are converted into cell-specific, durable inflammatory programmes. Interferon regulatory factor 4 (IRF4) is a stringent test of this broader concept because its effects vary with abundance, partner choice, chromatin accessibility, metabolism, cell lineage, tissue, and disease phase. We conducted a systematised narrative review and applied a causal-discrimination framework that separates lineage determination and activation from downstream amplification, necessary mediation,

BACKGROUND: Germline pathogenic and likely pathogenic variants (GPV) in cancer susceptibility genes may guide medical care. However, inaccurate recall of test results by patients may impact the uptake of appropriate medical care, underscoring the importance of accurate recall of results.
METHODS: Among individuals with a confirmed cancer susceptibility GPV that alters cancer risk management, individuals were categorized based on their accuracy of recall. Multiple logistic regression was performed to determine demographic and psychosocial factors associated with recall of test results.

OBJECTIVES: Despite the importance of disclosing a pathogenic/likely pathogenic (P/LP) variant related to hereditary cancer, fewer than half of index cases share their genetic test results with at-risk, biological relatives. Given this missed opportunity for cancer prevention, this study explored the barriers to family communication in Korea, from the perspective of women with hereditary breast and ovarian cancer syndrome.
METHODS: In-depth interviews were conducted with 22 women (17 affected, 5 unaffected) carrying P/LP variants in one the BRCA genes in Korea. Individual face-to-face and sma

BACKGROUND: Current practice for patients with breast cancer referred for genetic counseling, includes face-to-face consultations with a genetic counselor prior to and following DNA-testing. This is based on guidelines regarding Huntington's disease in anticipation of high psychosocial impact of DNA-testing for mutations in BRCA1/2 genes. The initial consultation covers generic information regarding hereditary breast cancer and the (im)possibilities of DNA-testing, prior to such testing. Patients with breast cancer may see this information as irrelevant or unnecessary because individual geneti

Holoprosencephaly (HPE) is a complex brain malformation caused by impaired or incomplete midline division of the prosencephalon. It's characterized by cerebral and facial anomalies of different levels of severity. Both genetic and environmental factors are known to cause HPE, but they cover only few cases. Genetic causes are responsible for about 20% of cases: they are chromosomal abnormalities and gene mutations: up to date, nine genes (SHH, ZIC2, SIX3, TGIF, PATCHED1, TDGF1/CRIPTO, FAST1, GLI2 and DHCR) are definitely associated with HPE, but many others candidate gene are under investigatio

Sickle cell disease (SCD) is a genetic blood disorder caused by abnormal hemoglobin that damages and deforms red blood cells (RBCs). The abnormal red cells break down, causing anemia, and obstruct blood vessels, leading to recurrent episodes of severe pain and multiorgan ischemic damage. SCD affects millions of people throughout the world and is particularly common among people whose ancestors come from sub-Saharan Africa. Sickle cell trait (SCT) is an inherited condition in which both normal hemoglobin and sickle hemoglobin are produced in the RBCs. SCT is not a type of sickle cell disease. P

In genetic epidemiological studies informative families are often oversampled to increase the power of a study. For a proband-family design, where relatives of probands are sampled, we derive the score statistic to test for clustering of binary and quantitative traits within families due to genetic factors. The derived score statistic is robust to ascertainment scheme. We considered correlation due to unspecified genetic effects and/or due to sharing alleles identical by descent (IBD) at observed marker locations in a candidate region. A simulation study was carried out to study the distributi
