[Ketamine-associated urological symptoms].

Ketamine is used as an anaesthetic and in third-line pain management. Furthermore, recreational use of the drug is becoming increasingly popular due to its dissociative and hallucinogenic effects. Ketamine can affect the urothelium, possibly with long-term damage to the bladder and kidneys. An otherwise healthy, 22-year-old female smoker was referred to our clinic. Shortly after starting recreational ketamine she experienced gross haematuria, urgency, frequency and dysuria. There had been no febrile episodes or flank pain. Her urine cultures were sterile. Physical examination, blood tests, uri

Interrater reliability of the Psychiatric Research Interview for Substance and Mental Disorders in an HIV-infected cohort: experience of the National NeuroAIDS Tissue Consortium.

The interrater reliability of the Psychiatric Research Interview for Substance and Mental Disorders (PRISM) was assessed in a multicentre study. Four sites of the National NeuroAIDS Tissue Consortium performed blinded reratings of audiotaped PRISM interviews of 63 HIV-infected patients. Diagnostic modules for substance-use disorders and major depression were evaluated. Seventy-six per cent of the patient sample displayed one or more substance-use disorder diagnoses and 54% had major depression. Kappa coefficients for lifetime histories of substance abuse or dependence (cocaine, opiates, alcoho

A review of chemical agents in the pharmacotherapy of addiction.

Chemical substance abuse has tormented mankind throughout history. A number of chemical approaches have been employed in an attempt to treat chemical addiction. Unfortunately, most of these have proven unsuccessful though several chemical entities have been shown to be moderately effective. The naturally occurring alkaloid ibogaine has been reported to interrupt the cravings for alcohol, cocaine and opiates. Other alkaloids from Tabernanthe iboga, such as ibogamine and tabernanthine, provide insight into the structure activity relationship at the different receptors believed to be involved in

Application of a radioimmunoassay screening test for detection and management of phencyclidine intoxication.

A radioimmunoassay procedure has been developed to monitor patients suspected of phencyclidine (PCP) intoxication. Symptoms in 11 patients suspected of phencyclidine intoxication included violent, aggressive behavior with delusions, hallucinations, agitation, and other signs of toxic psychosis. In five subjects serum concentrations of PCP ranged between 0.5 and 40 ng/ml. For laboratory confirmation, ascorbic acid should be administered to the patient after collecting the initial urine specimen. The initial urine and the first and second specimen after acidification should be collected and subm

Comparison of the visually evoked response in drug-free chronic schizophrenic patients and normal controls.

Thirteen cooperative male drug-free chronic schizophrenic patients, and 11 mentally normal male controls were studied. The VER was recorded from scalp leads O1, O2, Oz, C3 and C4 to combined ear reference (A1--A2). The stimulus was an unpatterned flash of single intensity. Compared to normal controls, there were no consistent differences in wave peak latencies or amplitudes for chronic schizophrenics in any brain area tested. When the chronic schizophrenic patients were separated on the basis of high and low tryptophan uptake, using the Frohman--Gottlieb criteria, the high uptake group exhibit

Chronic cerebral effects of alcohol and drug abuse.

A minority of alcohol abusers develop severe cerebral dysfunction in the form of Wernicke-Korsakoff syndrome. There is also evidence to suggest that cerebral dysfunction, particularly impaired abstracting ability, occurs in that larger population of heavy drinkers who do not go on to develop the Wernicke-Korsakoff syndrome. There is no consistent evidence that long-term marijuana, hallucinogen, or sedative use causes lasting neuropsychological disturbance. The deficits in abstract thinking reported by some LSD studies are similar to deficits others have reported among alcoholics. Since the LSD

Apparent hallucinations in monkeys during around-the-clock amphetamine for seven to fourteen days. Possible relevance to amphetamine psychosis.

Schizophrenia-like symptoms have been experimentally produced in humans by a single, large dose of amphetamine or by relatively low level, but continuous administration of the drug. In animal studies of the psychotomimetic properties of amphetamine, high doses and, in particular, repeated daily-injection drug schedules have often been used. However, amphetamine psychosis is not always a prominent effect of repeated intake drug schedules in humans and available clinical evidence suggests that psychosis develops more readily when the drug is taken in a continuous fashion over longer periods. The

Cardiovascular safety of psilocybin in psychiatric practice: a narrative review.

PURPOSE: Psilocybin-assisted interventions are moving from efficacy trials toward psychiatric implementation. Cardiovascular interpretation is central to this transition because trial participants are generally medically selected, receive standardized pharmaceutical-grade doses, and are monitored more intensively than many patients encountered in routine care. This review translates the cardiovascular evidence into a risk-stratified psychiatric decision framework. METHODS: We conducted a structured narrative review of clinical trials, systematic reviews, meta-analyses, cardiovascular pharmaco

Psychedelics for substance use disorders: are women being addressed? A scoping review.

INTRODUCTION: Substance use disorders (SUDs) are a prevalent health issue that affects individuals globally. Research is currently being conducted on the potential use of psychedelics as a new approach to treating SUDs. Women have been shown to respond differently than men to SUDs treatments and women-specific approaches have been recommended. This scoping review explores the ways in which studies examining the potential therapeutic effect of psychedelics on SUDs-related outcomes have taken into account women's specificities. METHODS: On February 19, 2024, we searched four databases-Embase, P

Effects of psychedelic microdosing on cognitive functions: A systematic review and meta-analysis.

Microdosing - the practice of consuming extremely low doses of classical psychedelic substances that do not elicit overt psychedelic effects - has gained significant attention as a potential method for enhancing cognitive performance. However, findings from controlled studies remain mixed and inconclusive. This preregistered meta-analysis examined the cognitive effects of classical psychedelic microdosing in 14 different studies (N = 1614), analyzing 59 effect sizes across multiple cognitive domains, spanning both acute (on-drug) and post-acute (off-drug) assessments. Results show a significan

The effects of psychedelic-assisted therapy on illness and death anxiety: A systematic review and meta-analysis.

Illness anxiety (IA) is characterized by worry or fear in response to living with a chronic illness or a preoccupation with having or acquiring a serious disease in conjunction with absent to mild somatic symptomatology. Death anxiety (DA) may underpin IA and is more common in individuals with life-threatening illness. Treatments for DA and IA, such as psychotherapy and antidepressant drugs, are limited. Serotonergic psychedelic assisted therapy (PT) is a promising treatment alternative as it acts more rapidly than antidepressants and has been shown to decrease anxiety associated with life-thr

Psychedelics, Spirituality, and Fundamentalism: A Brain Network Approach to Cognitive Flexibility and Rigidity.

This viewpoint reconceptualizes mysticism and fundamentalism as brain network disorders, with psychedelics like psilocybin, lysergic acid diethylamide, and N,N-dimethyltryptamine offering potential to modulate these states. By disrupting rigid neural patterns, psychedelics may foster cognitive flexibility, challenge inflexible belief systems, and offer therapeutic value for extremism and mental health disorders.

Are we hallucinating or can psychedelic drugs modulate the immune system to control inflammation?

Psychedelic drugs that activate 5-HT2A receptors have been long used for cultural, medicinal and recreational purposes. Interest in psychedelics for treating psychiatric disorders has resurged recently and is well documented; less well recognised are their anti-inflammatory properties. Growing evidence now demonstrates that psychedelics modulate immune responses, including inhibiting pro-inflammatory cytokine release. Furthermore, in vivo studies demonstrate that psychedelics, like (R)-DOI, reduce inflammation in animal models of acute and chronic inflammatory disease such as asthma. Likewise,

Psilocybin in the treatment of eating disorders: a systematic review of the literature and registered clinical trials.

BACKGROUND: Fluoxetine remains the only pharmacological treatment approved for Bulimia Nervosa, and no other drugs have been approved for eating disorders (EDs). The rationale for exploring psilocybin as a treatment for EDs is compelling, both from biological and psychological perspectives. Moreover, its safety profile in healthy individuals appears favorable. This systematic review aims to examine original research articles and registered clinical trials to assess the current psilocybin's therapeutic potential in EDs. METHODS: Systematic review following the indications of the Preferred Repo

Clinical pharmacology of ayahuasca: potential applications and future considerations.

INTRODUCTION: In the last few years, interest is growing around the topic of possible therapeutic properties of substances classified as psychedelic, especially for the treatment of mental disorders. Ayahuasca is a classic psychedelic with a complex composition, which has been used in traditional contexts for centuries. AREAS COVERED: In this review, we explore the current evidence and limitations around ayahuasca use for the treatment of depression, anxiety, posttraumatic stress and substance use disorders, with a focus on clinical and observation studies. EXPERT OPINION: Similar to what ha

Eligibility for psychedelic therapy: Clinical trial medical exclusion criteria and their implications for hospital practice.

Clinical research in psychedelic-assisted therapies is flourishing following the resumption of human trials exploring classic psychedelics and their therapeutic application to a range of mental health conditions. Translation from research to the clinic is underway in Australia and appears imminent in several other countries. Classic psychedelics function primarily as agonists of a subgroup of serotonin (5-HT) receptors but also act to varying degrees at other receptor and transporter systems. Some physiological responses to psychedelics may present risks to susceptible individuals that must be

Reduction of alcohol use and increase in psychological flexibility after a naturalistic psychedelic experience: a retrospective survey.

INTRODUCTION: Alcohol use can be significantly associated with negative social, professional, and health outcomes. Even more so, alcohol use disorder (AUD) is a critical public health issue and major avoidable risk factor. This study aimed to examine the effect of a naturalistic psychedelic experience on alcohol use and related measures. METHODS: A retrospective online survey was conducted on 160 individuals who reported a psychedelic experience and a concomitant drinking habit but did not necessarily have an AUD. Demographic data, characteristics of the psychedelic experience, and changes in
