[Sympatric Speciation of the Plague Microbe Yersinia pestis: Monohostal Specialization in the Host-Parasite Marmot-Flea (Marmota sibirica-Oropsylla silantiewi) System].

An ecological scenario of the origin of the plague microbe that is interpreted in the light of modern Darwinism (synthetic theory of evolution) is presented. It is shown that the plague microbe emerged from a clone of the psychrophilic saprozoonotic pseudotuberculosis microbe Yersinia pseudotuberculosis O:1b in the mountain steppe landscapes of Central Asia in the Sartan time, 22000-15000 years ago, in the monohostal Mongolian marmot (Marmota sibirica)-flea (Oropsylla silantiewi) host-parasite system. It was noted that the evolutionary process described corresponds to the sympatric form of spe

Detection of Wolbachia in the tick Ixodes ricinus is due to the presence of the hymenoptera endoparasitoid Ixodiphagus hookeri.

The identification of micro-organisms carried by ticks is an important issue for human and animal health. In addition to their role as pathogen vectors, ticks are also the hosts for symbiotic bacteria whose impact on tick biology is poorly known. Among these, the bacterium Wolbachia pipientis has already been reported associated with Ixodes ricinus and other tick species. However, the origins of Wolbachia in ticks and their consequences on tick biology (known to be very diverse in invertebrates, ranging from nutritional symbionts in nematodes to reproductive manipulators in insects) are unknow

Glutathione Peroxidase from Talaromyces marneffei Interacts with Host Cytoskeletal Proteins: Insights from Yeast Two-Hybrid and Molecular Dynamics Simulations.

Talaromyces marneffei is a dimorphic fungal pathogen that can subvert host immune defenses; however, the molecular mechanisms underlying its interactions with host cells remain incompletely understood. Glutathione peroxidase from T. marneffei (TmGpx1) has previously been identified as an antigenic protein that elicits antibody responses in patients with talaromycosis. To elucidate the contribution of TmGpx1 during human-fungal pathogen interaction, the yeast two-hybrid system was performed using TmGpx1 as bait to screen a cDNA library derived from non-induced human macrophage cells. Sixteen ca

Immunological fitness of echinocandin-resistant Nakaseomyces glabratus (former Candida glabrata): The impact on the management of candidemia.

Nakaseomyces glabratus (formerly Candida glabrata) (N. glabratus) is a commensal yeast that is increasingly associated with invasive candidi … gates in vitro. These results suggest that resistance-associated cell wall remodeling can influence host-pathogen interactions. These findings suggest that altered immune interactions and delayed clearance may confer additional fitness advantages to echinocandin-resistant N. glabratus, potentially impacting the pathophysiological interactions between host and pathogen.

Host-Brucella interactions and the Brucella genome as tools for subunit antigen discovery and immunization against brucellosis.

Vaccination is the most important approach to counteract infectious diseases. Thus, the development of new and improved vaccines for existing, emerging, and re-emerging diseases is an area of great interest to the scientific community and general public. Traditional approaches to subunit antigen discovery and vaccine development lack consideration for the critical aspects of public safety and activation of relevant protective host immunity. The availability of genomic sequences for pathogenic Brucella spp. and their hosts have led to development of systems-wide analytical tools that have provi

The Interface of Vibrio cholerae and the Gut Microbiome.

The bacterium Vibrio cholerae is the etiologic agent of the severe human diarrheal disease cholera. The gut microbiome, or the native community of microorganisms found in the human gastrointestinal tract, is increasingly being recognized as a factor in driving susceptibility to infection, in vivo fitness, and host interactions of this pathogen. Here, we review a subset of the emerging studies in how gut microbiome structure and microbial function are able to drive V. cholerae virulence gene regulation, metabolism, and modulate host immune responses to cholera infection and vaccination. Improve

Pathogen-epithelium interactions and inflammatory responses in Salmonella Dublin infections using ileal monolayer models derived from adult bovine organoids.

Salmonella enterica serovar Dublin (S. Dublin) is an important enteric pathogen affecting cattle and poses increasing public health risks. Understanding the pathophysiology and host-pathogen interactions of S. Dublin infection are critical for developing effective control strategies, yet studies are hindered by the lack of physiologically relevant in vitro models. This study aimed to generate a robust ileal monolayer derived from adult bovine organoids, validate its feasibility as an in vitro infection model with S. Dublin, and evaluate the epithelial response to infection. A stable, confluent…

Viral evolution during primary infection in immunocompromised hosts.

The immune response to viral infection is a delicate balance. By perturbing this balance, immunodeficiencies are expected to influence within-host viral evolution. Indeed, the presence of immunocompromised hosts has been argued to be a source of novel viral variants in some infectious diseases, including SARS-CoV-2. However, these arguments rest upon between-host models and so the role of immunodeficiencies on within-host evolution in primary infections is poorly understood. Using a mechanistic immunological model, here we consider how different immunodeficiencies shape the orchestration of th

Deciphering defensive mechanisms in Riemerella anatipestifer infection: pathogen adaptation, host immune response and emerging strategies.

Riemerella anatipestifer (RA) is an economically important avian pathogen that has historically caused significant disease in waterfowls but is increasingly recognized in various poultry production systems and wild bird species, highlighting its expanding host range and adaptive potential. Despite the availability of vaccines and antimicrobial interventions, RA remains a persistent challenge due to serotype diversity, limited cross-protective immunity, evolving antimicrobial resistance patterns, and an incomplete understanding of the host-pathogen interactions that govern disease outcome. Digital twin of Mycobacterium tuberculosis infection: Integrating immune dynamics and pathogen adaptation for precision therapy.

Mycobacterium tuberculosis remains one of the world's most persistent pathogens, responsible for over a million deaths each year despite effective drugs and decades of research. Disease progression varies widely among individuals, reflecting interactions between pathogen, physiology, host immunity, and pharmacological response. We propose a digital twin framework for tuberculosis that integrates clinical, immunological, and pharmacokinetic data into a continuously adaptive computational model. The twin would simulate host-pathogen dynamics from granuloma formation to systemic immune regulation

MAPK signaling pathways in host immune regulation during Mycobacterium tuberculosis infection.

Tuberculosis (TB) is one of the world's leading causes of death from a single infectious agent, with Mycobacterium tuberculosis (Mtb) primarily infecting the lungs via the respiratory tract. Following infection, immune cells such as macrophages and neutrophils phagocytose Mtb and initiate complex inflammatory and immune responses, driving the formation of granulomas and cavities within the lungs, ultimately leading to structural damage. In this intricate cascade, the MAPK signaling emerges as a critical regulator, orchestrating various cellular processes including inflammatory signaling, autop

Simulated Microgravity Reprograms Host Cell Architecture and Mitochondrial Function, Altering Toxoplasma gondii Growth.

Simulated microgravity (SMG or μG) influences Toxoplasma gondii growth and modifies the cellular structure and function of the host cells. As an obligate intracellular parasite, Toxoplasma gondii depends on the metabolic activity and mitochondrial function of its host. However, the impact of SMG on host-pathogen interactions remains unclear. We investigated the effect of SMG on host cell structure and mitochondrial functions and its impact on Toxoplasma gondii infection. Cultured human foreskin fibroblasts (HFF) and Vero cells under SMG formed stratified cell layers and exhibited morphological…

Artificial Intelligence Applications in Bacteriophage Genomics and Host Interaction.

Antibiotic-resistant bacterial infections remain a major public health challenge, contributing to an estimated 1.27 million deaths in 2019. Pathogens, such as Escherichia coli, Klebsiella pneumoniae, and Acinetobacter baumannii, cause a range of infections in clinical and food environments. As these bacteria rapidly develop resistance to multiple antibiotics, there is a growing need for alternative treatments. One promising approach is bacteriophage (phage) therapy. Phages are bacterial viruses that recognize, infect, and kill their hosts to propagate. Their relatively low isolation and produc

Autophagy and Mycobacterium Tuberculosis: the role of autophagy in antimicrobial immunity and therapy.

Tuberculosis (TB) remains one of the most severe infectious diseases worldwide, posing a persistent and increasingly serious threat to global public health. Cellular autophagy, a highly conserved innate immune mechanism, plays a crucial role in the elimination of intracellular pathogens, regulation of immune responses, and maintenance of cellular homeostasis, making it a key focus in TB research. This review systematically summarizes the types and regulatory mechanisms of autophagy, as well as its interactions with Mycobacterium tuberculosis (M. tb), and explores the potential applications of

Autophagy in Mycobacterium tuberculosis defense: unraveling complexities and reconciling contradictions.

Tuberculosis (TB), caused by Mycobacterium tuberculosis, remains a major global health challenge, exacerbated by prolonged treatment regimens, variable vaccine efficacy, and the emergence of drug-resistant strains. Beyond conventional pathogen-targeted antibiotics, increasing attention has focused on host-directed therapies (HDTs) that aim to enhance intrinsic immune mechanisms to improve disease control and treatment outcomes. Among these, autophagy, a conserved cellular process responsible for the degradation and recycling of damaged organelles, proteins, and intracellular pathogens, has eme

Understanding orthopoxvirus host range and evolution: from the enigmatic to the usual suspects.

In general, orthopoxviruses can be considered as falling into one of three host-utilization categories: highly specialized, single-host; broad host range; or 'cryptic', the last encompassing those viruses about which very little is known. Single-host viruses tend to exploit abundant hosts that have consistent patterns of interaction. For these viruses, observed genome reduction and loss of presumptive host-range genes is thought to be a consequence of relaxed selection. In contrast, the large genome size retained among broad host range orthopoxviruses suggests these viruses may depend on multi

TurboID-Based Proximity Labeling for Exploiting Effector Interacting Proteins in the Fungal Maize Pathogen Ustilago maydis.

TurboID-based proximity labeling (PL) is an advanced method for identifying proteins in the immediate molecular environment of a protein of interest under physiological conditions. TurboID enables the rapid and covalent biotinylation of proteins in proximity to the bait proteins in living cells, capturing both stable and transient interactions with high temporal resolution. Most plant pathogens secrete effectors that modulate host immunity and facilitate infection. The effector-targeted host proteins are commonly involved in plant resistance or susceptibility. Elucidating these proteins is mea
