TLR-mediated B cell defects and IFN-α in common variable immunodeficiency.

Common variable immune deficiency (CVID) B cells have impaired responses to TLR7 and TLR9 agonists including poor cell proliferation, loss of cytokine production, and failure to produce IgG or IgA. We show that TLR7- or 9-activated B cells from CVID subjects with >0.5% peripheral isotype-switched CD27(+) B cells (group 2) have increased mature Cγ1 and Cγ2 heavy-chain mRNA transcripts compared to subjects who have <0.5% isotype-switched cells (group 1). While TLR-stimulated CVID plasmacytoid dendritic cells for all subjects had impaired IFN-α production, TLR7 or TLR9 stimulation in the presence

Anti-IgA antibodies in common variable immunodeficiency (CVID): diagnostic workup and therapeutic strategy.

Common Variable Immunodeficiency (CVID) patients who are seropositive for anti-IgA antibodies have a predisposition for anaphylactoid reactions to intravenous immunoglobulin replacement therapy (IVIG). Among 88 CVID patients, we identified eight with IgG anti-IgA antibodies (9%). All eight completely lacked IgA (<0.0009 g/l). Five of them had a history of anaphylactoid reactions to IVIG. However, four of these five patients tolerated subcutaneous immunoglobulin replacement therapy (SCIG). To identify predisposing factors for anti-IgA antibodies and related anaphylactoid reactions, we analyzed

[Cerebral toxoplasmosis in subjects with acquired immunodeficiency syndrome].

Cerebral toxoplasmosis is the most common cause of focal CNS disease complicating AIDS and may afflict 10% to 33% of such patients. We present the clinical, neuroradiological and immunological findings in 5 cases with cerebral toxoplasmosis complicating the acquired immune deficiency syndrome. All patients had focal neurological signs and symptoms; CT scan findings included single or multiple lesions with ring contrast enhancement, mass effect and oedema. CSF analysis showed increased protein levels, decreased glucose levels and pleocytosis; CSF IgG antitoxoplasma antibodies were positive in 4

[Immunological indices and the acid phosphatase activity of the blood serum in patients with suppurative and destructive complications of acute pneumonia during the treatment/process].

The content of T and B lymphocytes, IgA, IgM and IgG as well as blood serum acid phosphatase (AP) activity were examined in 55 patients with purulent destructive complications of acute pneumonia. The patients were distributed into 2 groups depending on the disease gravity. The patients with a grave disease showed the depression of T and B immunity systems, marked deficiency of serum IgA, IgM and IgG. The percentage and absolute number of O cells was increased according to the disease gravity. In second group patients with a less graver disease, the reduction in the analogous immunity indicator

Serum immunoglobulin elevations in the acquired immunodeficiency syndrome (AIDS): IgG, IgA, IgM, and IgD.

The exact nature of the immune defect in the acquired immune deficiency syndrome (AIDS) is not known. Most studies have focused on abnormal T-cell functions which occur in AIDS. Although polyclonal elevation of serum immunoglobulins is also consistently reported in AIDS, there have been no statistical studies measuring the isotypes (classes) comprising this hypergammaglobulinemia. Quantitative serum immunoglobulin levels of IgG, IgA, IgM, and IgD in patients with AIDS (n = 33) were compared to healthy homosexual (n = 71) and healthy heterosexual (n = 32) controls. Serum IgD levels are increase

Human faecal immunoglobulins in healthy infants and children, and in some with diseases affecting the intestinal tract or the immune system.

IgA, IgG and IgM is faeces were quantified by single radial immunodiffusion using extracts of freeze-dried faeces. IgA in small specimens of faeces seemed to mirror the total amound of IgA secreted into the gut at the time of sampling. Presumptive normal values of faecal IgA concentrations in infants and children were established. Agglutinins to rabbit erythrocytes served as markers for the antibody activity. Infants and children just recovered from enteritis of probably infectious origin had higher concentrations of both IgA and agglutinins in faeces. Faeces from three out of five patients wi

MARCH2/3 target FcγRI for K27-linked polyubiquitination and degradation to restrict the inflammatory response.

IgG Fc gamma receptor I (FcγRI) belongs to the immunoglobulin superfamily and plays a pivotal role in immune regulation. The post-translational regulation of FcγRI and its effects on immune regulation are unclear. In this study, we identified the membrane-associated RING-CH-type finger (MARCH) E3 ubiquitin ligases MARCH2 and MARCH3 as physiological regulators of FcγRI. MARCH2 and MARCH3 associate with FcγRI and mediate its K27-linked polyubiquitination at K336 and K368, respectively, leading to subsequent lysosomal degradation. While deficiency of either MARCH2 or MARCH3 modestly increases Fcγ

Type I IFN-dependent FcγRIV signaling in murine monocytes promotes lethal anaphylaxis during viral infections.

Anaphylaxis is a life-threatening hypersensitivity reaction. Clinical observations suggest heightened susceptibility during viral infections, yet the mechanisms remain poorly defined. Here, we show that both active and passive IgG-mediated anaphylaxis were exacerbated in the setting of acute viral infection. In mice, this enhancement was driven predominantly by FcγRIV, the homolog of human FcγRIIIa. FcγRIV crosslinking induced anaphylactic symptoms selectively in infected animals, with no effect in naive conditions. Among leukocytes, inflammatory monocytes emerged as the principal drivers of t

Short Vi-polysaccharide abrogates T-independent immune response and hyporesponsiveness elicited by long Vi-CRM197 conjugate vaccine.

Polysaccharide-protein conjugates have been developed to overcome the T-independent response, hyporesponsiveness to repeated vaccination, and poor immunogenicity in infants of polysaccharides. To address the impact of polysaccharide length, typhoid conjugates made with short- and long-chain fractions of Vi polysaccharide with average sizes of 9.5, 22.8, 42.7, 82.0, and 165 kDa were compared. Long-chain-conjugated Vi (165 kDa) induced a response in both wild-type and T cell-deficient mice, suggesting that it maintains a T-independent response. In marked contrast, short-chain Vi (9.5 to 42.7 kDa

Circadian clock cryptochrome proteins regulate autoimmunity.

The circadian system regulates numerous physiological processes including immune responses. Here, we show that mice deficient of the circadian clock genes Cry1 and Cry2 [Cry double knockout (DKO)] develop an autoimmune phenotype including high serum IgG concentrations, serum antinuclear antibodies, and precipitation of IgG, IgM, and complement 3 in glomeruli and massive infiltration of leukocytes into the lungs and kidneys. Flow cytometry of lymphoid organs revealed decreased pre-B cell numbers and a higher percentage of mature recirculating B cells in the bone marrow, as well as increased num

Some epidermolysis bullosa acquisita sera react with epitopes within the triple-helical collagenous domain as indicated by immunoelectron microscopy.

BACKGROUND: Epidermolysis bullosa acquisita (EBA) autoantibodies recognize epitopes predominantly within the N-terminal noncollagenous (NC)-1 domain of type VII collagen. Recently, some EBA cases with reactivity to other domains, i.e. the triple-helical (T-H) collagenous domain and the NC-2 domain, have been reported. OBJECTIVES: To investigate the ultrastructural localization of epitopes for sera from five patients with EBA that were unreactive by immunoblotting with the NC-1, NC-2 and collagenous domains of type VII collagen. METHODS: Immunogold postembedding indirect immunoelectron micros

Immunogold localization of SP-A in lungs of infants dying from respiratory distress syndrome.

Prematurely born infants can develop the neonatal respiratory distress syndrome (RDS) because of a deficiency of pulmonary surfactant. This lipoprotein complex synthesized by type II pneumocytes has different ultrastructural forms--intra- and extracellular lamellar bodies, which within the alveoli are transformed into tubular myelin, and this in turn gives rise to the surface monolayer, the functionally active form of surfactant. We have previously shown that at autopsy RDS lungs lack tubular myelin and have decreased immunoreactivity for antisera to surfactant protein A (SP-A), an important c

T- and B-cell functions in IgA-deficient patients.

In vitro lymphocyte function of 60 IgA-deficient patients (IgAdp) were studied. In the mitogen-induced lymphocyte activation test, peripheral blood mononuclear cells (PBMC) of IgAdp showed a weaker response to pokeweed mitogen (PWM) than those of controls (P less than 0.05), and the responses to phytohaemagglutinin (PHA) and concanavalin A (Con A) were normal. The amounts of IgA, IgG, and IgM secreted by PBMC from controls and IgAdp were measured in vitro by enzyme immunoassay. B cells were activated by PWM with or without hydrocortisone (HC) to inhibit HC-sensitive suppressor cells. Lymphocyt

Protective immunity in toxoplasmosis: correlation between antibody response, brain cyst formation, T-cell activation, and survival in normal and B-cell-deficient mice bearing the H-2k haplotype.

Correlations of Toxoplasma gondii-specific immunoglobulin M (IgM) and IgG production, antigen-specific T-cell activation, and the number of brain cysts were compared in immunocompetent CBA/J (H-2k), C3H/He (H-2k), and B-cell-deficient CBA/N (H-2k) mice. Almost all of the C3H/He mice (94%) survived in comparison to CBA/J (71%) and CBA/N (53%) mice following infection with 20 cysts of Me 49, an avirulent strain of T. gondii. The mortality in susceptible mice was reduced by treatment of the animals with sulfadiazine during the acute stage of infection. Decreased mortality in CBA/J and C3H/He mice

A 15-year-old boy with severe combined immunodeficiency, fungal infection, and weight gain.

Hematopoietic stem cell transplantation (HSCT) outcomes in X-linked severe combined immune deficiency are most effective when performed with patients <3 months of age and without coexisting morbidity, and with donor cells from a matched sibling. Even under such favorable circumstances, outcomes can be suboptimal, and full cellular engraftment may not be complete, which results in poor B or natural killer cell function. Protein losing enteropathies can accompany persistent immune deficiency disorders with resultant low serum globulins (immunoglobulin A [IgA], IgG, IgM) and lymphopenia. Patients

[Antibody deficiency without decrease in the level of serum immunoglobulins].

The paper reports the case of a 14 year-old boy, born at full term with intra-uterine growth retardation (1,640 g) who presented with short stature, generalized eczema and recurrent infections. During the first years of life, hypogammaglobulinemia with antibody deficiency occurred. The in vitro T lymphocyte function was normal. The infections have become less severe. The plasma levels of IgA, IgG, IgM and IgE are normal. There is hyperimmunoglobulinemia D. The deficiency of antibodies against most of the tested antigens persists. T and B lymphocyte counts are normal. The in vitro lymphocyte pr

Case Report: DOCK8 Deficiency Without Hyper-IgE in a Child With a Large Deletion.

Autosomal recessive (AR) DOCK8 deficiency is a well-known actinopathy, a combined primary immune deficiency with impaired actin polymerization that results in altered cell mobility and immune synapse. DOCK8-deficient patients present early in life with eczema, viral cutaneous infections, chronic mucocutaneous candidiasis, bacterial pneumonia, and abscesses, together with eosinophilia, thrombocytosis, lymphopenia, and variable dysgammaglobulinemia that usually includes Hyper-IgE. In fact, before its genetic etiology was known, patients were described as having a form of Hyper-IgE syndrome, a na

Clinical significance of anti-protein Z antibodies in patients with lupus anticoagulant.

INTRODUCTION: Protein Z serves as cofactor for the inactivation of factor Xa by the plasma protein Z-dependent protease inhibitor. Deficiency of protein Z was reported to exhibit a clinical manifestation like lupus anticoagulant characterised by thrombosis and fetal loss. As anti-protein Z antibodies may be associated with low protein Z levels, we hypothesised that anti-protein Z antibodies might play a role in lupus anticoagulant (LA). MATERIALS AND METHODS: Anti-protein Z antibodies were measured by commercially available ELISA in 102 LA-patients (69 with and 33 without thrombosis) and 33 h

CD36 is differentially expressed on B cell subsets during development and in responses to antigen.

Of a number of mAbs made by immunization with sort-purified marginal zone (MZ) B cells, one was shown to recognize the mouse scavenger receptor CD36. Although CD36 is expressed by most resting MZ B cells and not by follicular and B1 B cells, it is rapidly induced on follicular B cells in vitro following TLR and CD40 stimulation. In response to T-independent and T-dependent Ag challenge, we found that CD36 was expressed on IgM+ plasma cells, but down-regulated on isotype-switched plasma cells in vivo. Although development, localization, and phenotype of MZ B cells in CD36-/- mice appeared norma
