Stress and the menstrual cycle: relevance of cycle quality in the short- and long-term response to a 5-day endotoxin challenge during the follicular phase in the rhesus monkey.

The notion that stress activates central and peripheral pathways to inhibit the menstrual cycle is well accepted, but the initial processes through which this occurs have not been investigated. This study uses a relevant nonhuman primate model to document the cyclic endocrine effects imposed by a moderate short-term stress episode in the follicular phase. The stress paradigm is a 5-day inflammatory/immune-like challenge produced by the administration of bacterial endotoxin [lipopolysaccharide (LPS)], which, through the release of endogenous cytokines and other mediators, induces a physiopathol

Computational modeling of fear and stress responses: validation using consolidated fear and stress protocols.

Dysfunction in fear and stress responses is intrinsically linked to various neurological diseases, including anxiety disorders, depression, and Post-Traumatic Stress Disorder. Previous studies using in vivo models with Immediate-Extinction Deficit (IED) and Stress Enhanced Fear Learning (SEFL) protocols have provided valuable insights into these mechanisms and aided the development of new therapeutic approaches. However, assessing these dysfunctions in animal subjects using IED and SEFL protocols can cause significant pain and suffering. To advance the understanding of fear and stress, this st

Deficiency in the cell-adhesion molecule dscaml1 impairs hypothalamic CRH neuron development and perturbs normal neuroendocrine stress axis function.

The corticotropin-releasing hormone (CRH)-expressing neurons in the hypothalamus are critical regulators of the neuroendocrine stress response pathway, known as the hypothalamic-pituitary-adrenal (HPA) axis. As developmental vulnerabilities of CRH neurons contribute to stress-associated neurological and behavioral dysfunctions, it is critical to identify the mechanisms underlying normal and abnormal CRH neuron development. Using zebrafish, we identified Down syndrome cell adhesion molecule like-1 (dscaml1) as an integral mediator of CRH neuron development and necessary for establishing normal

Integrated biomarker profiling reveals synergistic toxicity of pesticide-nanoparticle mixtures in zebrafish.

Aquatic ecosystems are increasingly exposed to contaminant mixtures rather than single compounds, creating complex interactions that remain poorly understood in ecotoxicology. Agricultural pesticides and engineered nanoparticles are among the most pervasive stressors, often detected together in surface waters due to intensive farming practices and widespread industrial use. Spirotetramat (a systemic tetramic acid insecticide), methoxyfenozide (an ecdysone agonist), and zinc oxide nanoparticles (ZnO-NPs) were focused on as representative co-occurring contaminants with potential joint effects on

The price of protection: an integrated toxicological and modelling assessment of flubendiamide exposure in Anabas testudineus.

The widespread use of pesticides such as flubendiamide raises increasing concerns regarding their ecological impact on non-target aquatic organisms. This study evaluated the acute and chronic toxicity of flubendiamide in the freshwater fish Anabas testudineus by integrating hematological, biochemical, oxidative stress, and genotoxic biomarkers with advanced modelling approaches. Fish were exposed to sublethal concentrations (2.66 and 5.32 mg/L) for 45 days, and responses were assessed at multiple time points. Acute toxicity assays revealed a time-dependent decrease in LC50 values, confirming c

Volatile blends from MeSA-treated soybean plants modulate drought responses in neighboring receivers: evidence for volatile-mediated infopriming.

Drought stress triggers complex biochemical and physiological adjustments in plants, including the release of volatile organic compounds (VOCs) that can influence neighboring individuals. Here we show that such volatiles, particularly those induced by methyl salicylate (MeSA), can convey chemical information that modulates the drought response of nearby soybean plants. Using an emitter-receiver chamber system, we exposed receiver plants under water deficit to volatiles from emitters subjected to either drought, MeSA application, or both. Drought reduced total VOC profile emission of emitters p

Reframing Pathogenesis-Related proteins: from plant immune drivers to stress resilience integration nodes.

Plants are constantly exposed to biotic and abiotic stresses, but the molecular mechanisms integrating these signals remain underexplored. Traditionally viewed as defense proteins against pathogen infection, pathogenesis-related (PR) proteins comprise 19 diverse families encompassing defense-related enzymes (glucanases, chitinases, nucleases, proteases), antimicrobial peptides, enzyme inhibitors and reactive oxygen species (ROS)-generating proteins. While their significance during biotic stresses is well-documented, research has started to emerge on their dynamic regulation and functional role

Probiotics as a Treatment of Chronic Stress Associated Abnormalities.

Chronic stress is a widespread problem that significantly affects both physical and mental health, leading to numerous complications such as mood disorders, cognitive impairments, gastrointestinal issues, and chronic diseases. The dysregulation of the hypothalamic pituitary adrenal (HPA) axis and the gut-brain axis underlies several stress related disorders, leading to systemic inflammation, neuroinflammation, dysbiosis, and impaired gut barrier integrity. This review emphasizes the growing significance of probiotics as a potential treatment strategy for addressing chronic stress. Probiotics a

Molecular Characterization and RNAi-Based Functional Validation of Ache in the Neonicotinoid Stress Response of the Melon Fly, Zeugodacus cucurbitae.

The melon fly, Zeugodacus cucurbitae (Coquillett), is a destructive tephritid pest of cucurbit crops, and neonicotinoids remain important tools for its management. Although neonicotinoids primarily target nicotinic acetylcholine receptors (nAChRs), their possible effects on acetylcholinesterase (AChE)-mediated cholinergic homeostasis in Z. cucurbitae remain poorly understood. In this study, we molecularly characterized the Z. cucurbitae Ache gene and evaluated its expression and functional contribution under dinotefuran, acetamiprid, and thiamethoxam stress. Bioinformatics analyses confirmed c

Comparative identification of abiotic stress-responsive differentially expressed genes in chickpea using unsupervised machine learning, and traditional meta-analysis.

BACKGROUND: Climate change poses a growing threat to chickpea production through abiotic stresses such as drought, heat, and salinity. Understanding the molecular stress response of chickpea is critical for improving resilience and minimizing yield loss. OBJECTIVE: To identify robust, stress-responsive differentially expressed genes (DEGs) in chickpea under abiotic stress (drought, salt, and salinity) by comparing three complementary large-scale RNA-seq data analytical approaches. METHODS: This study employed three complementary approaches, traditional meta-analysis, conventional statistical

Genetic and biochemical approaches used for identification and mechanistic characterization of nitric oxide-responsive plant genes.

Nitric oxide (NO) is a multiregulatory signal molecule that integrates development and stress responses. To elucidate the molecular mechanisms of NO phytoeffects and to identify NO-associated genes, both genetic screens and genome-wide transcriptome analysis have been employed in numerous studies. Forward genetic screens have linked NO signalling to key biological processes, such as photosynthesis, cytokinin metabolism, stress adaptation, and cell cycle regulation. Reverse genetics has further characterized the role of NO-related genes involved in NO biosynthesis (e.g., NIA1/NIA2, NOA1), signa

Amygdala-specific changes in Cacna1c, Nfat5, and Bdnf expression are associated with stress responsivity in mice: A possible mechanism for psychiatric disorders.

The CACNA1C gene encodes the alpha-1c subunit of the Cav1.2 calcium channel, a regulator of neuronal calcium influx involved in neurotransmitter release and synaptic plasticity. Genetic data show a role for CACNA1C in depressive symptoms underlying different psychiatric diagnoses. However, the mechanisms involved still require further exploration. This study aimed to investigate sex and region-specific changes in the Cacna1c gene and behavioral outcomes in mice exposed to chronic stress. Moreover, we evaluated the Nuclear factor of activated T-cells 5 (Nfat5) and the Brain-derived neurotrophic

[The pathophysiology and diagnosis of anxiety disorder].

In addition to genetic factors, the role of epigenetic and other environmental factors in the promotion of anxiety disorder has attracted much attention in psychiatric research. When stress is encountered in the environment, the hypothalamus-pituitary adrenal system (HPA system) is activated and cortisol is secreted. CRHR gene function is closely related to this response. As a result of haplotype analysis of CRHR genes in depression and panic disorder patients, it was found that genetic polymorphism of CRHR1 and CRHR2 was related to both disorders. It is reported that abused children are more

Photoreceptors as central hubs of developmental plasticity and environmental adaptation.

Plant photoreceptors are well known for their roles in light-dependent developmental processes such as photomorphogenesis, shade avoidance, and circadian regulation. These canonical functions involve light perception through specific receptors, including phytochromes and cryptochromes, and downstream transcriptional reprogramming mainly in the context of early seedling growth. Beyond their classical roles, recent studies have revealed that photoreceptors also act as integrators of diverse environmental signals, contributing to abiotic stress adaptation, root development, and vascular different

AIBP Protects Müller Glial Cells Against Oxidative Stress-Induced Mitochondrial Dysfunction and Reduces Retinal Neuroinflammation.

Glaucoma, an optic neuropathy with the loss of retinal ganglion cells (RGCs), is a leading cause of irreversible vision loss. Oxidative stress and mitochondrial dysfunction have a significant role in triggering glia-driven neuroinflammation and subsequent glaucomatous RGC degeneration in the context of glaucoma. It has previously been shown that apolipoprotein A-I binding protein (APOA1BP or AIBP) has an anti-inflammatory function. Moreover, Apoa1bp-/- mice are characterized by retinal neuroinflammation and RGC loss. In this study, we found that AIBP deficiency exacerbated the oxidative stress

CRHR1 mediates the transcriptional expression of pituitary hormones and their receptors under hypoxia.

Hypothalamus-pituitary-adrenal (HPA) axis plays critical roles in stress responses under challenging conditions such as hypoxia, via regulating gene expression and integrating activities of hypothalamus-pituitary-targets cells. However, the transcriptional regulatory mechanisms and signaling pathways of hypoxic stress in the pituitary remain to be defined. Here, we report that hypoxia induced dynamic changes in the transcription factors, hormones, and their receptors in the adult rat pituitary. Hypoxia-inducible factors (HIFs), oxidative phosphorylation, and cAMP signaling pathways were all di

Comparative analysis of meningeal transcriptomes in birds: Potential pathways of resilience to repeated impacts.

The meninges and associated vasculature (MAV) play a crucial role in maintaining cerebral integrity and homeostasis. Recent advances in transcriptomic analysis have illuminated the significance of the MAV in understanding the complex physiological interactions at the interface between the skull and the brain after exposure to mechanical stress. To investigate how physiological responses may confer resilience against repetitive mechanical stress, we performed the first transcriptomic analysis of avian MAV tissues using the Downy Woodpecker (Dryobates pubescens) and Tufted Titmouse (Baeolophus b
