ABC transporters and beta-tubulin in macrocyclic lactone resistance: prospects for marker development.

Macrocyclic lactones (MLs) are highly lipophilic anthelmintics which are known to bind to and open ligand-gated ion channels. However, these anthelmintics, and particularly the avermectin members of the ML class of endectocides, are potent substrates for ABC transporters and these transporters may regulate drug concentration in both the host and the parasite. There is accumulating evidence that ivermectin (IVM), and to a lesser extent moxidectin (MOX), selects for certain alleles of P-glycoprotein and other ABC transporter genes, selects for constitutive overexpression of some of these gene pr

Spontaneous and gamma-aminobutyric acid (GABA)-activated GABA(A) receptor channels formed by epsilon subunit-containing isoforms.

A new gamma-aminobutyric acid (GABA)A receptor (GABAR) subunit class, epsilon, has recently been cloned and shown to form functional channels when coexpressed with both alpha and beta subunits. We report that the combination of alpha1beta3epsilon subunit subtypes expressed in L929 cells produced functional chloride ion channels that were both spontaneously active and gated by the application of extracellular GABA. When cells were voltage-clamped at -75 mV in the whole-cell configuration, holding currents of 50 to 300 pA associated with increased noise were consistently recorded. The applicatio

Aptamer-functionalized solution-gated graphene transistor for rapid detection of trace scandium.

The growing exploitation of rare earth resources has led to the migration and accumulation of Scandium (Sc) within food systems, giving rise to emerging food safety concerns. Conventional techniques for Scandium (III) ions (Sc3+) detection are limited by labor-intensive sample preparation, high instrumentation costs, and poor suitability for rapid on-site screening. Here, we report a highly sensitive and selective biosensor for Sc3+ that integrates a Sc3+-specific nucleic acid aptamer (Sc-1) with a solution-gated graphene transistor (SGGT). The sensing mechanism relies on aptamer conformationa

Non-canonical pore architecture underlies constitutive gating of human retinal TRPM1.

Transient receptor potential melastatin 1 (TRPM1), a Ca²⁺-permeable nonselective cation channel essential for retinal ON bipolar cell signaling and night vision, and implicated in congenital night blindness, has remained structurally and functionally poorly characterized. Here we report the cryo-electron microscopy structure of human TRPM1, revealing that although it assembles as a tetramer, it adopts an unexpected clockwise domain-swapped pore module with rotational geometry inverse to that observed in previously characterized 6-TM tetrameric channels. This inverted topology is accompanied by

AMP-activated protein kinase inhibits TREK channels.

AMP-activated protein kinase (AMPK) is a serine/threonine kinase activated by conditions that increase the AMP : ATP ratio. In carotid body glomus cells, AMPK is thought to link changes in arterial O(2) with activation of glomus cells by inhibition of unidentified background K(+) channels. Modulation by AMPK of individual background K(+) channels has not been described. Here, we characterize effects of activated AMPK on recombinant TASK-1, TASK-3, TREK-1 and TREK-2 background K(+) channels expressed in HEK293 cells. We found that TREK-1 and TREK-2 channels but not TASK-1 or TASK-3 channels are

Isolation of myocardial L-type calcium channel gating currents with the spider toxin omega-Aga-IIIA.

The peptide omega-agatoxin-IIIA (omega-Aga-IIIA) blocks ionic current through L-type Ca channels in guinea pig atrial cells without affecting the associated gating currents. omega-Aga-IIIA permits the study of L-type Ca channel ionic and gating currents under nearly identical ionic conditions. Under conditions that isolate L-type Ca channel currents, omega-Aga-IIIA blocks all ionic current during a test pulse and after repolarization. This block reveals intramembrane charge movements of equal magnitude and opposite sign at the beginning of the pulse (Q(on)) and after repolarization (Q(off)). Q

Functional stoichiometry of Shaker potassium channel inactivation.

Shaker potassium channels from Drosophila are composed of four identical subunits. The contribution of a single subunit to the inactivation gating transition was investigated. Channels carrying a specific mutation in a single subunit can be labeled in a heterogeneous population and studied quantitatively with scorpion toxin sensitivity as a selection tag. Linkage within a single subunit of a mutation that removes the inactivation gate to a second mutation that affects scorpion toxin sensitivity demonstrates that only a single gate is necessary to produce inactivation. The inactivation rate con

A patch-clamp study on the muscarine-sensitive potassium channel in bullfrog sympathetic ganglion cells.

1. A voltage-independent K+ channel was characterized and effects of muscarine were studied in cultured bullfrog sympathetic ganglion cells using the cell-attached patch-clamp configuration. 2. Three types of single-channel current were recorded from 2- to 10-day-old cultured cells in the presence of tetraethylammonium (2-20 mM), tetrodotoxin (1-2 microM), Cd2+ (0.1 mM) and apamin (20 nM). 3. The most frequently observed channel was a voltage-independent K+ channel which was open at the resting membrane potential and had a conductance of 52.6, 78.9 and 114.9 pS at a [K+]o of 2, 40 and 100 mM,

Potassium channel in rabbit corneal endothelium activated by external anions.

The apical membrane of the rabbit corneal endothelium contains a potassium-selective ionic channel. In patch-clamp recordings, the probability of finding the channel in the open state (Po) depends on the presence of either HCO3- or Cl- in the bathing medium. In a methane sulfonate-containing bath, Po is less than 0.05 at all physiologically relevant transmembrane voltages. With 0 mM [HCO3-]o at +60 mV, Po was 0.085 and increased to 0.40 when [HCO3-]o was 15 mM. With 4 mM [Cl-]o at +60 mV, Po was 0.083 and with 150 mM Cl-, Po increased to 0.36. Low Po's are also found when propionate, sulphate,

Voltage-gated potassium channels and the control of membrane potential in human platelets.

1. Human platelets were studied using a combination of patch-clamp and fluorescent indicators of membrane potential and [Ca2+]i. 2. Whole-cell and cell-attached patch recordings showed voltage-gated channels selective for K+ (IK(V]. These channels were activated by depolarization at a threshold close to the platelet resting potential and were blocked by the venom charybdotoxin (CTX; 10-20 nM). Several different conductance states were observed, ranging from 5 to 34 pS, with isotonic KCl in the patch pipette and bath. 3. Measurements with the potential-sensitive dye 3,3'-dipropylthia-dicarbocya

Mechanism of charybdotoxin block of a voltage-gated K+ channel.

Charybdotoxin block of a Shaker K+ channel was studied in Xenopus oocyte macropatches. Toxin on rate increases linearly with toxin concentration in an ionic strength-dependent fashion and is competitively diminished by tetraethylammonium. On rate is insensitive to transmembrane voltage and to K+ on the opposite side of the membrane. Conversely, toxin off rate is insensitive to toxin concentration, ionic strength, and added tetraethylammonium but is enhanced by membrane depolarization or K+ (or Na+) in the trans solution. Charge neutralization of charybdotoxin Lys27, however, renders off rate v

Cardiovascular toxicity associated with supplement use.

BACKGROUND: Supplement use is prevalent and appears to be increasing over time. In the United States, regulation by the Food and Drug Administration is limited largely to post-marketing surveillance, raising safety concerns. A variety of supplements have been associated with cardiovascular toxicity, which can occur via adulteration, substitution, or as a result of intrinsic toxicity of the supplement. Cardiovascular toxicity due to supplement use may arise via several different mechanisms, and has been reported with supplements that act as central nervous system stimulants, via poisoning of ca

Spontaneous inhibitory synaptic currents mediated by a G protein-coupled receptor.

G protein-coupled receptors (GPCRs) affect many physiological processes by modulating both intrinsic membrane conductances and synaptic transmission. This study describes spontaneous miniature inhibitory postsynaptic currents mediated by vesicular dopamine release acting locally on metabotropic D2 receptors leading to the activation of a G protein-coupled inwardly rectifying potassium conductance. Thus, individual exocytotic events result in spontaneous GPCR-mediated transmission, similar to synaptic activation of classical ligand-gated ion channels.

sigma-1 receptor modulation of acid-sensing ion channel a (ASIC1a) and ASIC1a-induced Ca2+ influx in rat cortical neurons.

Acid-sensing ion channels (ASICs) are proton-gated cation channels found in peripheral and central nervous system neurons. The ASIC1a subtype, which has high Ca2+ permeability, is activated by ischemia-induced acidosis and contributes to the neuronal loss that accompanies ischemic stroke. Our laboratory has shown that activation of sigma receptors depresses ion channel activity and [Ca2+](i) dysregulation during ischemia, which enhances neuronal survival. Whole-cell patch-clamp electrophysiology and fluorometric Ca2+ imaging were used to determine whether sigma receptors regulate the function

Engineering- nanochannels with pH-responsive gates for direct detection of glucose in human blood serum.

BACKGROUND: An abnormal blood glucose (Glu) level is a key signal of diabetes. The gluconic acid produced by Glu catalytic oxidation can cause changes in pH value. RESULTS: Inspired by nature, in which organisms use pH as a chemical gate to regulate ion transport through cell membranes, we report a pH-gated electrochemical luminescence (ECL) sensing system for Glu detection based on the relationship between Glu metabolism and pH. The pH gate was designed on a TiO2 nanochannel membrane (NM) by modifying the channel entrance with polystyrene-b-poly(4-vinylpyridine) (P4VP) chains that convert fr

Electrophysiological data showing the acute alcohol effect on neuronal excitability in the murine posterior parietal cortex.

It is well established that alcohol interacts with multiple molecular targets, including ligand- and voltage-gated ion channels and intracellular signal proteins, leading to molecular and cellular neuroadaptations in various brain regions, including the cerebral cortex, hippocampus, amygdala, and cerebellum [[1], [2], [3]]. Despite the extensive literature, little is known about the effects of alcohol in the posterior parietal cortex (PPC), a cortical region involved in multiple cognitive and executive functions [4,5]. Here, we report data showing the acute alcohol effects on neuronal excitabi

Cell size induced bias of current density in hypertrophic cardiomyocytes.

Alterations in ion channel expression and function known as "electrical remodeling" contribute to the development of hypertrophy and to the emergence of arrhythmias and sudden cardiac death. However, comparing current density values - an electrophysiological parameter commonly utilized to assess ion channel function - between normal and hypertrophied cells may be flawed when current amplitude does not scale with cell size. Even more, common routines to study equally sized cells or to discard measurements when large currents do not allow proper voltage-clamp control may introduce a selection bi

Field-Programmable Bimodal Switching in a Hybrid-Dual-Gated MoS2 Transistor.

The ultrahigh surface-to-volume ratio and expandable interlayer spacing of van der Waals solids allow their channel bodies to strongly and dynamically interact with foreign molecules. However, controlling such multiple molecular interactions within a single, integrated platform has remained a technical challenge. Herein, we introduce a so-called hybrid-dual-gated voltage-controlled bimodal switch demonstrated on a single MoS2 transistor by cointegrating high-k solid and ionic liquid electrolytes as dual-gate dielectrics. Upon applying the synchronized dual-gate voltages, it results in two dist
