Mucolipidosis IV: ultrastructural diagnosis of a recently defined genetic disorder.

In nine cases of a new genetic disorder mucolipidosis IV clinical findings included early or congenital corneal cloudiness and mild to severe psychomotor retardation in all patients. The diagnosis can be established by electron microscopy of the conjunctiva, which along with several other tissues shows typical ultrastructural changes. All of the known patients described were descendants of parents of Jewish-Ashkenazi origin, and it seems that mucolipidosis IV is another "Jewish" genetic disease. Prenatal diagnosis can be made by finding the typical ultrastructural changes in cells of the amnio

Psychological studies in dystonia musculorum deformans.

Sixty-eight patients with DMD underwent psychometric assessment before or after thalamic surgery. Statistical comparisonss between categories of patients were undertaken as a function of religion, family history, and age at onset of symptoms as independent variables. It was found that Jewish patients, with negative family history, and age at onset of symptoms from 9 to 13 years, scored significantly higher on IQ tests than did all other groups. Other psychological test data revealed no specific personality patterns in the DMD patients, particularly no pattern of conversion hysteria or hysteria

Differences in SMN1 allele frequencies among ethnic groups within North America.

BACKGROUND: Spinal muscular atrophy (SMA) is the most common inherited lethal disease of children. Various genetic deletions involving the bi-allelic loss of SMN1 exon 7 are reported to account for 94% of affected individuals. Published literature places the carrier frequency for SMN1 mutations between 1 in 25 and 1 in 50 in the general population. Although SMA is considered to be a pan-ethnic disease, carrier frequencies for many ethnicities, including most ethnic groups in North America, are unknown. OBJECTIVES AND METHODS: To provide an accurate assessment of SMN1 mutation carrier frequenc

BRCA1 and BRCA2 germline mutation analysis among Indian women from south India: identification of four novel mutations and high-frequency occurrence of 185delAG mutation.

Mutations in the BRCA1 and BRCA2 genes profoundly increase the risk of developing breast and/or ovarian cancer among women. To explore the c … rlier. Interestingly, 185delAG, a BRCA1 mutation which occurs at a very high frequency in Ashkenazi Jews, was found at a frequency of 16.4% (10/61). There was one novel mutation (4866insT) and one reported mutation in BRCA2. Thus, our study emphasizes the importance of mutation screening in familial breast and/or ovarian cancers, and the potential impl …

Characterization of seven novel mutations causing factor XI deficiency.

BACKGROUND AND OBJECTIVES: Factor XI (FXI) deficiency is a rare autosomal recessive disorder, the main manifestation of which is injury-related bleeding. The disorder is rare in most populations, but common among Jews in whom two mutations, E117X and F283L, account for 98% of cases. Other mutations, C38R and C128X, are prevalent in French Basques and Britons, respectively. Additional sporadic mutations have been described in most parts of the world. The objective of this study was to identify the mutations in 15 unrelated FXI-deficient patients and characterize missense mutations by expression…

Characterization of BRCA1 and BRCA2 mutations in a large United States sample.

PURPOSE: An accurate evaluation of the penetrance of BRCA1 and BRCA2 mutations is essential to the identification and clinical management of families at high risk of breast and ovarian cancer. Existing studies have focused on Ashkenazi Jews (AJ) or on families from outside the United States. In this article, we consider the US population using the largest US-based cohort to date of both AJ and non-AJ families. METHODS: We collected 676 AJ families and 1,272 families of other ethnicities through the Cancer Genetics Network. Two hundred eighty-two AJ families were population based, whereas the …

Child injury in Israel: emergency room visits to a children's medical center.

The object of this study was to provide data for policy making and prevention program planning in Israel. The study examined all visits to the Department of Emergency Medicine at the Schneider Children's Medical Center in 1996 (41,279 visits in total). Approximately 22.6% of the emergency room patients were admitted following injury. Most (97%) were unintentional injury. Approximately 42% of the patients were less than 4 years old and about 20% were 2 years old. In all age groups, the rate of boys was double. Approximately 92% were Jews. Despite this low rate of non-Jewish patients, however, t…

Identification and functional analysis of two novel mutations in the multidrug resistance protein 2 gene in Israeli patients with Dubin-Johnson syndrome.

Dubin-Johnson syndrome (DJS) is an inherited disorder characterized by conjugated hyperbilirubinemia and is caused by a deficiency of the multidrug resistance protein 2 (MRP2) located in the apical membrane of hepatocytes. The aim of this study was to identify the mutations in two previously characterized clusters of patients with Dubin-Johnson syndrome among Iranian and Moroccan Jews and determine the consequence of the mutations on MRP2 expression and function by expression studies. All 32 exons and adjacent regions of the MRP2 gene were screened by polymerase chain reaction and DNA sequenci…

Euthanasia--the twenty-sixth specialty?

Twenty-five medical specialties currently offer board certification in the United States. The question is, should there be a 26th specialty- … By 1942, the Nazified physicians were ready to cure the nation by killing off "useless eaters," and Jews, the "cancer" of the Nordic race. If we are taught that history repeats itself, how can we condone the helping of one patient and assisting another with ending his life? Perhaps it is fitting that Dr. Kevorkian was a pathologist, not directly involv …

Fiberglass or silica exposure and increased nephritis or ESRD (end-stage renal disease).

The U.S. multiplant cohort mortality study of workers producing manufactured mineral fibers is finding increasing mortality from nephritis a … a. The Beduin, thought to be a population with maximal exposures, have higher rates of ESRD than do Jews in the age groups over 60 years. Although high blood concentrations of silica are found in persons with renal failure, the close association with elevated creatinine has been interpreted as evidence that the buildup of silica is due to renal failure …

Elevated risk of postpartum depressive symptoms among Arab women in Israel: The role of maternal role satisfaction and psychological flexibility.

OBJECTIVES: Postpartum depressive symptoms (PPDS) are more prevalent among ethnic minority women, yet the psychological processes contributing to this disparity, particularly potential protective factors, remain insufficiently understood. Guided by Lazarus and Folkman's (1984) stress and coping model, this study examined the contribution of maternal role satisfaction and psychological flexibility to PPDS among Jewish and Arab mothers in Israel. In addition, we tested whether maternal role satisfaction mediates, and psychological flexibility moderates, the association between ethnicity and PPDS
