Miniaturized Cultivation of Microbiota for Antimalarial Drug Discovery.

The ongoing search for effective antiplasmodial agents remains essential in the fight against malaria worldwide. Emerging parasitic drug resistance places an urgent need to explore chemotherapies with novel structures and mechanisms of action. Natural products have historically provided effective antimalarial drug scaffolds. In an effort to search nature's chemical potential for antiplasmodial agents, unconventionally sourced organisms coupled with innovative cultivation techniques were utilized. Approximately 60,000 niche microbes from various habitats (slow-growing terrestrial fungi, Antarct

Nasopharyngeal microbiota in healthy children and pneumonia patients.

Our study is the first to compare the nasopharyngeal microbiota of pediatric pneumonia patients and control children by 454 pyrosequencing. A distinct microbiota was associated with different pneumonia etiologies. Viral pneumonia was associated with a high abundance of the operational taxonomic unit (OTU) corresponding to Moraxella lacunata. Patients with nonviral pneumonia showed high abundances of OTUs of three typical bacterial pathogens, Streptococcus pneumoniae complex, Haemophilus influenzae complex, and Moraxella catarrhalis. Patients classified as having no definitive etiology harbored…

The contributory role of gut microbiota in cardiovascular disease.

Our group recently discovered that certain dietary nutrients possessing a trimethylamine (TMA) moiety, namely choline/phosphatidylcholine and L-carnitine, participate in the development of atherosclerotic heart disease. A meta-organismal pathway was elucidated involving gut microbiota-dependent formation of TMA and host hepatic flavin monooxygenase 3-dependent (FMO3-dependent) formation of TMA-N-oxide (TMAO), a metabolite shown to be both mechanistically linked to atherosclerosis and whose levels are strongly linked to cardiovascular disease (CVD) risks. Collectively, these studies reveal that…

Role of the enteric microbiota in intestinal homeostasis and inflammation.

The mammalian intestine encounters many more microorganisms than any other tissue in the body thus making it the largest and most complex component of the immune system. Indeed, there are greater than 100 trillion (10(14)) microbes within the healthy human intestine, and the total number of genes derived from this diverse microbiome exceeds that of the entire human genome by at least 100-fold. Our coexistence with the gut microbiota represents a dynamic and mutually beneficial relationship that is thought to be a major determinant of health and disease. Because of the potential for intestinal …

Bridging immunity and lipid metabolism by gut microbiota.

The human gut is a unique organ in which hundreds of different microbial species find their habitat and in which different host physiologic functions, such as digestion, nutrition, and immunity, coexist. Although all these players were studied separately for decades, recently, there has been an explosion of studies demonstrating the essential role for interactions between these components in gut function. Furthermore, new systems biology methods provide essential tools to study this complex system as a whole and to identify key elements that define the crosstalk between the gut microbiota, imm…

Human microbiota characterization in the course of renal transplantation.

Recent studies demonstrate that the human microbiota, the collection of microorganisms growing on and in individuals, have numerous bidirectional interactions with the host, influencing immunity, resistance to infection, inflammation and metabolism. Little has been done to study the potential associations between microbiota composition and transplant outcome. Here, we investigated the longitudinal changes in the blood, urinary, oral and rectal microbiota of renal allograft recipients before and at 1 and 6 months after transplantation. The results showed major changes in microbiota composition …

The genital econiche: focus on microbiota and bacterial vaginosis.

Ecological and evolutionary forces shaping the normal and abnormal microflora of the genital econiche are discussed, in particular those rel … ed a microbiological and immunological enigma. Immune tolerance to both normal and abnormal vaginal microbiota, mainly derived from gut microflora, as a result of coevolution with humans might explain the absence of inflammation, supported by short-chain fatty acids, known to modulate immune responses, that are produced in large quantities by anaerobes. Nod2 is required for the regulation of commensal microbiota in the intestine.

Mutations in the Nod2 gene are among the strongest genetic risk factors in the pathogenesis of ileal Crohn's disease, but the exact contributions of Nod2 to intestinal mucosal homeostasis are not understood. Here we show that Nod2 plays an essential role in controlling commensal bacterial flora in the intestine. Analysis of intestinal bacteria from the terminal ilea of Nod2-deficient mice showed that they harbor an increased load of commensal resident bacteria. Furthermore, Nod2-deficient mice had a diminished ability to prevent intestinal colonization of pathogenic bacteria. In vitro, intesti

Molecular analysis of the subgingival microbiota in health and disease.

This investigation provides molecular analyses of the periodontal microbiota in health and disease. Subgingival samples from 47 volunteers with healthy gingivae or clinically diagnosed chronic periodontitis were characterized by PCR-denaturing gradient gel electrophoresis (DGGE) with primers specific for the V2-V3 region of the eubacterial 16S rRNA gene. A hierarchical dendrogram was constructed from band patterns. All unique PCR amplicons (DGGE bands) were sequenced for identity. Samples were also analyzed for the presence of Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, and…

Spatial organisation of microbiota in quiescent adenoiditis and tonsillitis.

BACKGROUND: The reasons for recurrent adenotonsillitis are poorly understood. METHODS: The in situ composition of microbiota of nasal (5 children, 25 adults) and of hypertrophied adenoid and tonsillar tissue (50 children, 20 adults) was investigated using a broad range of fluorescent oligonucleotide probes targeted to bacterial rRNA. None of the patients had clinical signs of infection at the time of surgery. RESULTS: Multiple foci of ongoing purulent infections were found within hypertrophied adenoid and tonsillar tissue in 83% of patients, including islands and lawns of bacteria adherent t…

Microbiota of periapical lesions refractory to endodontic therapy.

The periapical microbiota of 36 teeth with refractory apical periodontitis was investigated. None of the teeth had responded to conventional endodontic or long-term (> 6 months), calcium-hydroxide treatment. Eight patients had received antibiotics systemically. After anaerobic culture, a total of 148 microbial strains were detected among 67 microbial species. One of the 36 lesions was culture-negative. Approximately half (51.0%) of the bacterial strains were anaerobic. Gram-positive species constituted 79.5% of the flora. Facultative organisms, such as Staphylococcus, Enterococcus, Enterobacte…

Predictive Functional Profiling of Allergy-Associated Microbiota with PICRUSt2.

Functional profiling of host-associated microbiota is essential for elucidating its role in immune dysregulation, such as in allergic diseases. Predictive bioinformatics tools like PICRUSt2 (Phylogenetic Investigation of Communities by Reconstruction of Unobserved States, version 2) enable the inference of microbial metabolic potential from 16 S rRNA gene amplicon data. This chapter describes the computational workflow for using PICRUSt2 to analyze allergy-associated microbiota. The protocol begins with a standardized amplicon sequence variant (ASV) table and its corresponding taxonomy and seq…

Gut microbiota-derived extracellular vesicles in Parkinson's disease.

The gut microbiota contributes to the pathophysiology of Parkinson's disease (PD), but the mechanism is not known. Extracellular vesicles (EVs), membrane-enclosed nanosized particles, contain various biomolecules, cross biological barriers, and reach sterile body compartments. We hypothesized that EVs released by the gut microbiota might influence PD pathophysiology. We isolated gut microbiota-derived EVs from the fecal samples of 20 patients with PD and 19 healthy controls. Patients with PD had higher concentrations of fecal EVs, and the mean particle size of EVs was larger. The 16S ribosomal…

Polyphenols, Gut Microbiota, and Exercise Triad Supports Performance.

Polyphenols are bioactive compounds commonly found in foods like fruits, vegetables, and cocoa. These compounds have the potential to impact the gut microbiota and metabolism to support exercise and recovery through both direct and indirect mechanisms. The purpose of this narrative review is to present evidence for a triad relationship between polyphenolic compounds, the gut microbiome, and exercise. This was achieved by reviewing and summarizing mechanistic and human intervention study outcomes to elucidate how polyphenols influence the triad relationship. Polyphenols exert a prebiotic effect…

Uric acid remodels gut microbiota to fuel obesity.

Obesity and hyperuricemia frequently coexist, yet researchers have long lacked a causal framework regarding the two disorders. In this issue of Cell Host & Microbe, Tian et al. address this gap by mapping a uric acid-driven liver-gut axis that remodels gut microbiota, thereby promoting dietary lipid absorption and further obesity.

Gastric microbiota-mediated immune remodelling in gastric cancer.

Increasing evidence indicates that the gastric microbiota plays crucial roles in regulating the tumour microenvironment (TME), influencing gastric tumourigenesis and progression. Several bacteria, including Streptococcus, Lactobacillus and Bacteroides, have shown robust immunomodulatory effects on TME. In this review, we summarise current understanding of the crosstalk between the gastric microbiota and TME in gastric cancer (GC). Functional alterations of the gastric microbiota from healthy mucosa to malignancy are delineated, with emphasis on the impacts of bacteria on different immune cell …

Gut microbiota-activated PLCγ2 monocytes drive atrial fibrillation.

Atrial fibrillation (AF) is the most common sustained arrhythmia, conferring risks of stroke and heart failure. Monocyte activation-associat … deletion of PLCG2 blocks atrial recruitment of these monocytes and reduces AF. Depletion of the gut microbiota with antibiotics reduces the number of phospho-PLCγ2+ monocytes in the atria, suppresses EndMT, and attenuates AF inducibility, and these effects are reversed by fecal microbiota transplantation (FMT). Mechanistically, gut microbiota-derived bacteri …
