Cholesterol 25-hydroxylase inhibits Newcastle disease virus replication by enzyme activity-dependent and direct interaction with nucleocapsid protein.

Newcastle disease virus (NDV) is a significant enveloped virus within the Paramyxoviridae family, posing a major threat to the global poultry industry. Increasing evidence suggests that cholesterol-25-hydroxylase (CH25H) and its enzymatic product, 25-hydroxycholesterol (25HC), exhibit broad-spectrum antiviral activity properties by modulating lipid metabolism and various signaling pathways. However, the specific role of CH25H in regulating NDV infection and replication remains unclear. In this study, we demonstrate that CH25H significantly inhibits NDV replication by blocking viral entry throu

Cooperative dynamics of PARP-1 zinc-finger domains in the detection of DNA single-strand breaks.

The DNA single-strand break (SSB) repair pathway is initiated by the multifunctional enzyme PARP-1, which recognizes the broken DNA ends by its two zinc-finger domains, Zn1 and Zn2. Despite a number of experiments performed with different DNA configurations and reduced fragments of PARP-1, many details of this interaction that is crucial to the correct initiation of the repair chain are still unclear. We performed Molecular Dynamics (MD) computer simulations of the interaction between the Zn1/Zn2 domains of PARP-1 and a DNA hairpin including a missing nucleotide to simulate the presence of an

Pigeon pea penta- and hexapeptides with antioxidant properties also inhibit renin and angiotensin-I-converting enzyme activities.

Pigeon pea protein was sequentially digested with pepsin followed by pancreatin and the hydrolysate separated into 18 fractions using reversed-phase high-performance liquid chromatography. Fractions were analyzed for in vitro antioxidant properties (radical scavenging, metal chelation, and ferric iron reducing ability) in addition to inhibition of renin and angiotensin-converting enzyme (ACE). The most active fractions were analyzed by mass spectroscopy followed by identification of 10 peptide sequences (7 pentapeptides and 3 hexapeptides). All the peptides showed a wide range of multifunction

Comparative characterization of candidal cell surface-associated phosphoglycerate mutase (Gpm1) - fungal cell wall adsorption and host protein binding across pathogenic Candida species.

Some intracellular enzymes are repeatedly detected at the cell surface of fungal pathogens and, due to this noncanonical cellular localization and additional functions played thereon, are classified as multifunctional "moonlighting" proteins. Yet, the extent to which such behaviour is conserved across clinically relevant yeasts and how these proteins interact with the host defence systems remain incompletely recognized. In this work, we investigated the fungal surface accessibility, cell wall adsorption and the interaction with selected host proteins of a glycolytic enzyme - phosphoglycerate m

Deficiency of ECHS1 causes mitochondrial encephalopathy with cardiac involvement.

OBJECTIVE: Short-chain enoyl-CoA hydratase (ECHS1) is a multifunctional mitochondrial matrix enzyme that is involved in the oxidation of fatty acids and essential amino acids such as valine. Here, we describe the broad phenotypic spectrum and pathobiochemistry of individuals with autosomal-recessive ECHS1 deficiency. METHODS: Using exome sequencing, we identified ten unrelated individuals carrying compound heterozygous or homozygous mutations in ECHS1. Functional investigations in patient-derived fibroblast cell lines included immunoblotting, enzyme activity measurement, and a palmitate loadi

Double-stranded RNA-dependent RNase activity associated with human immunodeficiency virus type 1 reverse transcriptase.

Early events in the retroviral replication cycle include the conversion of viral genomic RNA into linear double-stranded DNA. This process is mediated by the reverse transcriptase (RT), a multifunctional enzyme that possesses RNA-dependent DNA polymerase, DNA-dependent DNA polymerase, and RNase H activities. In the course of studies of a recombinant RT of human immunodeficiency virus type 1 (HIV-1), we observed an additional, unexpected activity of the enzyme. The purified RT catalyzes a specific cleavage in HIV-1 RNA hybridized to tRNALys, the primer for HIV-1 reverse transcription. The cleav

New insights on familial colorectal cancer type X syndrome.

Familial colorectal cancer type X (FCCTX) is a heterogeneous colorectal cancer predisposition syndrome that, although displays a cancer pattern similar to Lynch syndrome, is mismatch repair proficient and does not exhibit microsatellite instability. Besides, its genetic etiology remains to be elucidated. In this study we performed germline exome sequencing of 39 cancer-affected patients from 34 families at risk for FCCTX. Variant classification followed the American College of Medical Genetics and Genomics (ACMG) guidelines. Pathogenic/likely pathogenic variants were identified in 17.65% of th

Extending the limits of quantitative proteome profiling with data-independent acquisition and application to acetaminophen-treated three-dimensional liver microtissues.

The data-independent acquisition (DIA) approach has recently been introduced as a novel mass spectrometric method that promises to combine the high content aspect of shotgun proteomics with the reproducibility and precision of selected reaction monitoring. Here, we evaluate, whether SWATH-MS type DIA effectively translates into a better protein profiling as compared with the established shotgun proteomics. We implemented a novel DIA method on the widely used Orbitrap platform and used retention-time-normalized (iRT) spectral libraries for targeted data extraction using Spectronaut. We call thi

Investigation of recessive ataxia loci in patients with young age of onset.

Autosomal recessive cerebellar ataxias are a phenotypically and genetically heterogeneous group of diseases. Major forms can be distinguished on the basis of clinical signs, age of onset, biochemical parameters or genotypes. To develop rational diagnostic strategies, phenotypic information, e.g., age of onset combined with population-specific disease frequencies could be highly favourable. We tested this hypothesis for single candidate loci and mutations in North European ataxia patients with juvenile and early adult onset. While we could prove that Friedreich ataxia (FRDA) is frequent in Germ

Eugenol-derived carbamates as sub-nanomolar dual cholinesterase inhibitors with neuroprotective activity against Alzheimer's disease: Structure-activity relationships and molecular dynamics insights.

Carbamate-based cholinesterase inhibitors represent a clinically validated therapeutic strategy for Alzheimer's disease (AD); however, sub-nanomolar AChE inhibitory activity within natural phenol-derived scaffolds has not been previously reported. To address this gap, a series of eleven eugenol-based carbamate derivatives (E1-E11) was rationally designed, synthesized, and evaluated as multifunctional anti-AD agents. Systematic variation of the carbamate N-substituent enabled fine modulation of inhibitory potency and isoform selectivity, revealing well-defined structure-activity relationships.

Synergistic bioremediation of metolachlor-contaminated soil by Penicillium oxalicum MetF1 and earthworms: Enhanced degradation, microbial community restructuring, and ring-opening metabolic activation.

The chloroacetamide herbicide metolachlor (MET) is a widespread environmental contaminant posing risks to aquatic and soil ecosystems. Although the fungal strain Penicillium oxalicum MetF1 efficiently degrades MET in liquid culture, its performance in complex soil environments remains unexplored. Recognizing the multifunctional roles of earthworms as soil ecosystem engineers-including their capacity to modulate microbial activity and pollutant bioavailability-we incorporated Eisenia fetida into a synergistic bioremediation system with P. oxalicum MetF1 in MET-contaminated soil. The synergistic

Drug Delivery Systems for Hepatocellular Carcinoma: Bridging Pathophysiological Barriers and Advanced Nanomedicine Strategies.

Hepatocellular carcinoma (HCC) is characterized by high heterogeneity, a complex tumor microenvironment, and multiple physiological barriers that limit conventional therapies and cause significant toxicity. Drug delivery systems (DDS), particularly nanoscale delivery systems, offer promising opportunities to overcome these obstacles through passive targeting, active targeting, and stimuli-responsive drug release. Nevertheless, despite substantial progress in preclinical research, the clinical translation of most DDS remains challenging. This review systematically outlines the pathophysiologica

Synthesis, characterization, in vitro and in silico studies of novel lophine clubbed acylthioureas.

In the present study, a chemical library of novel lophine clubbed acylthioureas (8a-l) was designed and synthesized through a multistep sequence. 2-(4-Nitrophenyl)-4,5-diphenyl-1H-imidazole (3) was synthesized from benzil, 4-nitrobenzaldehyde, and ammonium acetate via multicomponent reaction. Then the nitro group of the synthesized compound was reduced using zinc and hydrochloric acid to furnish 4-(4,5-diphenyl-1H-imidazol-2-yl) aniline (4). On the other hand, the acid chloride (6a-l) of substituted benzoic acids was prepared by reacting acids (5a-l) with thionyl chloride, followed by their re

Metal-Substituted Hydroxyapatite Nanoparticles as Antimicrobial and Osteogenic Biomaterials for Hard-Tissue Applications.

Bacterial colonization and biofilm formation on orthopedic and dental implants remain major clinical complications, while conventional systemic antibiotics are often limited by poor penetration into biofilms and infected bone. These limitations have motivated the development of biomaterials with intrinsic antibacterial activity. Hydroxyapatite (HA), a major inorganic component of bone and teeth, possesses excellent biocompatibility, osteoconductivity, and bone-bonding ability but exhibits limited inherent antibacterial activity. Incorporation of therapeutic metal ions, including Ag+, Cu2+, Zn2

Hydrothermal Synthesis of Light-Responsive Adhesive Protein Polymeric Dot Nanogel for Wound Healing.

Most bioadhesive gels are polymeric formulations synthesized through multistep procedures and critically lack intrinsic therapeutic properties, relying on preloaded drugs or antibiotics to promote wound healing. Here, we present a novel strategy for developing next-generation smart bioadhesive gels that overcome these limitations by combining simple synthesis, built-in therapeutic functionality, and superior wound-healing performance. Our approach employs a one-step, controllable hydrothermal carbonization process, eliminating the need for photoinitiator and complex multistep methods, enhancin
