Idiopathic cardiomyopathy and skeletal muscle abnormality.

The skeletal muscle of three cases presenting with idiopathic congestive cardiomyopathy has been studied, histologically, histochemically, ultramicroscopically, and electromyographically. In all three there is clinical evidence of skeletal muscle weakness, and in all three, pathologic changes were found in the muscle. These changes were different in each case and varied from mitochondrial myopathy to spinal atrophy to vacuolar myopathy. Other reported cases of cardiomyopathy demonstrating skeletal muscle pathology are discussed.

Carcinoid myopathy and treatment with cyproheptadine (Periactin).

A case of the carcinoid syndrome associated with a proximal myopathy is reported. Histology showed advanced atrophy of type II muscle fibres but no inflammation. Perinuclear acid phosphatase was increased. Electron microscopy revealed persistence of the Z-line until the muscle fibre had been severely disrupted. Similar lesions have been observed in the hereditary muscular dystrophy in mice, and also in these animals and in rats when injected with 5-hydroxytryptamine (5-HT). Treatment with cyproheptadine caused a documented response in the patient's debilitating diarrhoea and also produced symp

Structural and functional responses of myofibrillar proteins from yellow croaker to plasma-activated buffer: evidence from a simulated PAW system.

As a green non-thermal technology, plasma-activated water (PAW) generates reactive oxygen and nitrogen species (RONS) through plasma-liquid interactions and thereby affects the oxidation, conformation, and functionality of aquatic myofibrillar proteins (MPs). To investigate the regulatory effects of PAW on yellow croaker MPs, the original extraction buffer was replaced with plasma-activated buffer of different activation times, and the oxidation, conformation, and functional changes of MPs during chilled storage were evaluated. The results indicate that moderate PAW treatment induces mild oxid

Effects of pine needle ice structuring protein on the functional properties of myofibrillar proteins from pork patty subjected to freezing-thawing cycles.

This study investigated the effects of pine needle ice structuring protein (PNISP, 0-0.8%, w/w) on physicochemical and functional properties of Myofibrillar proteins (MP) from pork patties under repeated freeze-thaw (F-T) cycles. Repeated F-T cycles significantly impaired MP functionality, including reduced solubility and surface charge, increased aggregation, weakened viscoelasticity, and deteriorated gel and emulsion stability. PNISP supplementation effectively alleviated these impairments, with 0.6% PNISP showing the optimal effect. PNISP-treated samples exhibited higher solubility, lower t

Microstructural characterization of myocardial infarction with optical coherence tractography and two-photon microscopy.

Myocardial infarction leads to complex changes in the fiber architecture of the heart. Here, we present a novel optical approach to characterize these changes in intact hearts in three dimensions. Optical coherence tomography (OCT) was used to derive a depth-resolved field of orientation on which tractography was performed. Tractography of healthy myocardium revealed a smooth linear transition in fiber inclination or helix angle from the epicardium to endocardium. Conversely, in infarcted hearts, no coherent microstructure could be identified in the infarct with OCT Additional characterization

Effects of low-intensity swimming on radiation-induced leg contracture in mice.

OBJECTIVE: To evaluate the effects of low-intensity swimming on radiation-induced leg contracture. METHODS: Forty mice were randomly and equally divided into four groups: 1) irradiation; 2) swimming before irradiation; 3) swimming after irradiation; 4) swimming after contracture, and their left hind legs were exposed to gamma irradiation of 60 Gy. The mice were allowed to swim freely for 10 minutes, three times per day. For group 2, the mice were allowed to swim for only 1 week before irradiation. For group 3, the mice were allowed to swim immediately after irradiation until day 130, post-irr

Skeletal muscle repair in a mouse model of nemaline myopathy.

Nemaline myopathy (NM), the most common non-dystrophic congenital myopathy, is a variably severe neuromuscular disorder for which no effective treatment is available. Although a number of genes have been identified in which mutations can cause NM, the pathogenetic mechanisms leading to the phenotypes are poorly understood. To address this question, we examined gene expression patterns in an NM mouse model carrying the human Met9Arg mutation of alpha-tropomyosin slow (Tpm3). We assessed five different skeletal muscles from affected mice, which are representative of muscles with differing fiber-

The biomechanics and evolutionary significance of thermal acclimation in the common carp Cyprinus carpio.

The effects of thermal acclimation were investigated in the common carp Cyprinus carpio L. Acclimation and acute temperature effects were tested during ontogeny from larval [9.5 mm total length (L)] to juvenile (69.0 mm L) stages and between 8 and 21 degrees C. The myosin heavy chain (MHC) composition, myofibrillar Mg(2+)-Ca(2+)-ATPase activity, and muscle strains showed significant thermal acclimation effects. MHCs were only expressed in an acclimation temperature-dependent fashion in fish longer than 37 mm. During fast starts, the temperature had a significant effect on the white muscle stra

Serological markers and HLA-DQ2 haplotype among first-degree relatives of celiac patients. Catalonian Coeliac Disease Study Group.

Serologic markers, HLA-DQ2 haplotype, and clinical features suggestive of celiac disease were studied to assess their diagnostic value in a multicentric study to detect celiac disease in 675 first-degree relatives of 227 celiac probands. Serum IgA-class anti-endomysium and IgA-class anti-gliadin antibodies were positive in 5.8% and 1.9% of relatives, respectively. HLA-DQ2 haplotype was present in 64% of relatives, and the overall rate of celiac disease diagnosed by intestinal biopsy was 5.5%. The frequency of HLA-DQ2 in the celiac patients and controls was 93% and 18%, respectively. The most f

Neurogenic effects on the palatopharyngeal muscle in patients with obstructive sleep apnoea: a muscle biopsy study.

Muscle biopsies from the palatopharyngeal muscle of eight patients with obstructive sleep apnoea were performed during uvulopalatopharyngoplasty. Control biopsies were performed during tonsillectomy in seven control patients with no history of symptoms suggesting obstructive sleep apnoea. The diagnosis was based on the patient's history and a whole night recording of arterial oxygen saturation and respiration movements. The mean number of oxygen desaturations > or = 4% per sleeping hour was 39 (range 7-80) in patients with obstructive sleep apnoea. In the control patients the occurrence of mus

Left atrial ultrastructure in mitral valvular disease.

Light microscopic and ultrastructural observations were made on left atrial tissues obtained from 14 patients at the time of operation for correction of mitral valvular disease. Cardiac muscle cells varied in size but most frequently were hypertrophied. In fibrotic areas, present in all left atria, the muscle cells tended to be isolated from adjacent cells and exhibited degenerative changes of varying severity. These changes consisted or proliferation of Z-band material and cytoskeletal filaments, myofibrillar loss, proliferation of elements of free and extended junctional sarcoplasmic reticul

[Clinical, biochemical, morphological and electrophysiological studies of glycogenosis Type II in childhood with double deficiency of enzymes (author's transl)].

The clinical, biochemical, morphological and electrophysiological findings in a 13-month-old child, who died of glycogenosis type II, is presented. In addition to the deficiency of alpha-1,4-glucosidase, which is typical for the disease, a deficiency in hyaluronidase could be detected for the first time in the skeletal and heart muscles and in the liver. On the other hand, the beta-glucoronidase and beta-acetylglucosaminidase activity was highly increased. Deposits of a substance, most probably an acid mucopolysaccharide, which could be differentiated from glycogen by chromography and electron

Electrophysiological studies of the antrum muscle fibers of the guinea pig stomach.

The membrane potentials of single smooth muscle fibers of various regions of the stomach were measured, and do not differ from those measured in intestinal muscle. Spontaneous slow waves with superimposed spikes could be recorded from the longitudinal and circular muscle of the antrum. The development of tension was preceded by spikes but often tension appeared only when the slow waves were generated. Contracture in high K solution developed at a critical membrane potential of -42 mv. MnCl(2) blocked the spike generation, then lowered the amplitude of the slow wave. On the other hand, withdraw
