Progress in Studies on Structural and Remedial Aspects of Newly Born Coronavirus, SARS-CoV-2.

The article highlights an up-to-date progress in studies on structural and the remedial aspects of novel coronavirus 2019-nCoV, renamed as SARS-CoV-2, leading to the disease COVID-19, a pandemic. In general, all CoVs including SARS-CoV-2 are spherical positive single-stranded RNA viruses containing spike (S) protein, envelope (E) protein, nucleocapsid (N) protein, and membrane (M) protein, where S protein has a Receptor-binding Domain (RBD) that mediates the binding to host cell receptor, Angiotensin Converting Enzyme 2 (ACE2). The article details the repurposing of some drugs to be tried for …

New Advances in Small Molecules Targeted at Viral Capsid-Genome Binding.

The capsid protein plays a crucial role in the viral life cycle. By interacting with the viral genome, it facilitates the assembly of the nucleocapsid, ultimately leading to the formation of the viral particle. Therefore, interfering with or disrupting the interaction between the capsid protein and viral genome can effectively inhibit viral replication and infection. This review focuses on elucidating the binding mechanisms between the capsid protein and the viral genome, as well as their potential applications as therapeutic targets. In particular, it summarizes the research progress on small…

Electrochemical Capillary Driven Immunoassay for Detection of SARS-CoV-2.

The COVID-19 pandemic focused attention on a pressing need for fast, accurate, and low-cost diagnostic tests. This work presents an electrochemical capillary driven immunoassay (eCaDI) developed to detect SARS-CoV-2 nucleocapsid (N) protein. The low-cost flow device is made of polyethylene terephthalate (PET) and adhesive films. Upon addition of a sample, reagents and washes are sequentially delivered to an integrated screen-printed carbon electrode for detection, thus automating a full sandwich immunoassay with a single end-user step. The modified electrodes are sensitive and selective for SA…

Intrinsic disorder in the core proteins of flaviviruses.

Hepatitis C virus and related viruses in the Flaviviridae family (such as dengue virus, yellow fever virus or West Nile virus) are amongst the most important human pathogens, causing substantial morbidity and mortality world-wide. The production of viral progeny in Flaviviridae is orchestrated by the small, multifunctional core protein, which coats and condenses the viral genomic RNA during Nucleocapsid formation. In addition to their structural role, mounting experimental evidence links core proteins to viral persistence and pathogenesis, by virtue of their promiscuous interactions with host …

Inhibition of the intracellular transport of influenza viral RNA by actinomycin D.

In primary chicken embryo cells infected with fowl plague virus addition of actinomycin D at defined times during the infection cycle has different consequences on viral replication. If actinomycin D is added immediately after infection with a concentration, which inhibits viral RNA synthesis only partially, it interferes with the nucleo-cytoplasmic transport of all viral RNA species (mRNA and vRNA) so far tested. If actinomycin D is present during infection (adsorption, penetration and uncoating) no viral RNA is synthesized, and the nucleocapsid of the infecting virus does not reach the nucle…

Rapidly progressive subacute sclerosing panencephalitis: an ultrastructural and immunoperoxidase study.

A case of subacute sclerosing panencephalitis (SSPE) in a 20-year-old male who died within 2 months, is described. Light microscopy revealed massive neuronal loss, reactive gliosis, perivascular cuffing and intranuclear (Cowdry type A) and intracytoplasmic inclusions. Immunocytochemical stain with the complement-fixing measles antibody was positive for intranuclear and intracytoplasmic inclusions. The reactive glia cells were positive for glial fibrillary acidic protein. Electron microscopy revealed a paramyxovirus-like structure of the nucleocapsids, occasionally showing a 'fuzzy' appearance …

Towards a viral etiology for Paget's disease of bone.

The etiology of Paget's disease of bone has long remained obscure. The recent discovery in electron microscopy of specific inclusions in the cytoplasm and nuclei of osteoclasts in tissue from patients with Paget's bone disease has been the starting point of investigations into a possible viral origin. The inclusions, made up of microcylinders, described by several authors as being found only in osteoclasts in Paget's bone disease, present a close morphological analogy with nucleocapsids of paramyxovirus of the measles group. Immunocytological methods have provided a complementary approach to t…

Rescue of a cytopathic paramyxovirus from peripheral blood leukocytes in subacute sclerosing panencephalitis.

An infectious, cytopathic paramyxovirus was rescued from the peripheral blood leukocytes of a patient with subacute sclerosing panencephalitis (SSPE) by cocultivation procedures. Cells infected with the virus (1) demonstrated polykaryocytosis, (2) contained cytoplasmic inclusion bodies of paramyxoviral nucleocapsids, (3) hemadsorbed monkey erythrocytes, and (4) reacted with fluorescein-conjugated antisera to measles virus. On the basis of these observations, the virus was initially believed to be measles-like virus similar to those that have been rescued from other patients with SSPE. However,…

Acute measles encephalitis in children with immunosuppression.

Four cases of encephalitis occurring in children treated for lymphatic malignancies by immunosuppressive drugs are reported. Measles virus was isolated from the brain in one case and identified immunologically in another. Nucleocapsids identical to those seen in subacute sclerosing panencephalitis were demonstrated in three cases. Severe immunosuppression was evidenced in two patients by failure of rosette formation and low phytohemagglutinin tests. Pathologically, the inflammatory reaction was absent in one brain and moderate in two. Clinically, epilepsia partialis continua was a prominent fe…

Systemic lupus erythematosus.

Recent investigations in systemic lupus erythematosus (SLE) have disclosed the presence of paramyxo-virus-like nucleocapsids in the cytoplasm of renal endothelial cells of patients with this disease. Elaborate techniques have failed to demonstrate conclusively that these are indeed viruses. Concomitantly, there is increasing evidence that SLE patients have defective cell-mediated-immunity. A 3-year-old child with SLE exhibited clinical evidence of defective cellular immunity. At postmortem, paramyxoviruslike tubular structures were demonstrated in her kidneys. Additional findings included the …

Genetic and Pathologic Characterization of a Novel Recombinant TC07-2-Type Avian Infectious Bronchitis Virus.

Avian infectious bronchitis viruses (IBVs) with the TC07-2 genotype have spread rapidly in East Asia since they were first reported in China in 2007. In 2015, an IBV with the TC07-2 genotype (designated KrD1515) was isolated from layer chickens with severe respiratory symptoms in Korea. In the present study, the full-length open reading frames of the spike (S) and nucleocapsid (N) genes of the virus were sequenced and analyzed. S1 gene phylogenetic analysis revealed that the KrD1515 virus clustered with viruses with the TC07-2 genotype, whereas N gene phylogenetic analysis revealed that the Kr…

Molecular epidemiology of rabies virus isolates from South Korea.

A molecular epidemiological study was performed on 13 Korean virus isolates, which were collected from wild and domestic animals diagnosed as rabid between 1998 and 2004. Seven samples were from domestic animals such as dogs and cattle infected by rabid raccoon dogs (Nyctereutes procyonoides koreensis), and the rest of the six samples were from raccoon dogs in the wild. The study was carried out based on the comparison of nucleotide and amino acid sequences of nucleoprotein (N) and glycoprotein (G) coding regions and nucleotide sequence of the G-L intergenic (Psi) non-coding region of the isol

Alphaherpesvirus-associated disease in greater bilbies (Macrotis lagotis).

CASE REPORT: A captive breeding colony of 9 greater bilbies (Macrotis lagotis) exhibited mild upper respiratory signs and sudden deaths with 100% mortality over a 2-week period. Histologically, acute necrotising and erosive epithelial lesions throughout the upper respiratory system and bronchi were associated with eosinophilic intranuclear inclusion bodies. Inclusions were also present in hepatocytes and adrenocortical cells, but were not always associated with necrosis. Transmission electron microscopy of lung sections revealed nucleocapsids forming arrays within some nuclei. A pan-herpesviru…

Cerebral endothelial cell infection by measles virus in subacute sclerosing panencephalitis: ultrastructural and in situ hybridization evidence.

Infection of vascular endothelium plays a central role in the pathogenesis of acute measles virus infection outside the central nervous system (CNS) but has not been described in the human CNS. An ultrastructural survey was made of blood vessels in five cases of subacute sclerosing panencephalitis (SSPE) to determine whether or not infection of cerebral vascular endothelium occurred in this persistent fatal CNS disease caused by measles virus. Morbillivirus nucleocapsids were found in a few endothelial cells in three necropsy cases but not in the limited tissue available from two biopsies. In …
