Human embryonic stem cells and cardiac repair.

The muscle lost after a myocardial infarction is replaced with noncontractile scar tissue, often initiating heart failure. Whole-organ cardiac transplantation is the only currently available clinical means of replacing the lost muscle, but this option is limited by the inadequate supply of donor hearts. Thus, cell-based cardiac repair has attracted considerable interest as an alternative means of ameliorating cardiac injury. Because of their tremendous capacity for expansion and unquestioned cardiac potential, pluripotent human embryonic stem cells (hESCs) represent an attractive candidate cel

Haploinsufficiency of the mouse Forkhead Box f1 gene causes defects in gall bladder development.

The forkhead box f1 (Foxf1) transcription factor is expressed in the visceral (splanchnic) mesoderm, which is involved in mesenchymal-epithelial signaling required for development of organs derived from foregut endoderm such as lung, liver, gall bladder, and pancreas. Our previous studies demonstrated that haploinsufficiency of the Foxf1 gene caused pulmonary abnormalities with perinatal lethality from lung hemorrhage in a subset of Foxf1+/- newborn mice. During mouse embryonic development, the liver and biliary primordium emerges from the foregut endoderm, invades the septum transversum mesen

The mousetrap: what we can learn when the mouse model does not mimic the human disease.

In recent years, mouse models for human metabolic diseases have become commonplace because the information gained from in vivo study of biochemical pathways is invaluable, and many metabolic diseases are relatively easy to recreate in mice through gene knockout technology in embryonic stem cells. In certain cases, however, the knockout mice may reproduce only some of the human disease phenotype, may be more severely affected than human cases, or may have no clinical phenotype at all. Under these circumstances, the disease pathology can become more complex, causing the researcher to evaluate ba

Renal carcinogenesis, hepatic hemangiomatosis, and embryonic lethality caused by a germ-line Tsc2 mutation in mice.

Germ-line mutations of the human TSC2 tumor suppressor gene cause tuberous sclerosis (TSC), a disease characterized by the development of hamartomas in various organs. In the Eker rat, however, a germ-line Tsc2 mutation gives rise to renal cell carcinomas with a complete penetrance. The molecular mechanism for this phenotypic difference between man and rat is currently unknown, and the physiological function of the TSC2/Tsc2 product (tuberin) is not fully understood. To investigate these unsolved problems, we have generated a Tsc2 mutant mouse. Tsc2 heterozygous mutant (Tsc2+/-) mice developed

Melanin-containing hepatoblastoma with endocrine differentiation. An immunohistochemical and ultrastructural study.

BACKGROUND: The authors previously have reported that hepatoblastomas may exhibit endocrine differentiation. This report describes a hepatoblastoma in which a melanocytic component was present in addition to endocrine differentiation. METHODS: The tumor, which arose in a 15-month-old girl, was subjected to conventional histologic, histochemical, immunohistochemical, and electron microscopic investigation. RESULTS: The tumor had fetal and embryonal epithelial areas and osteoid. The presence of melanin could be suspected, even on the basis of gross examination. The melanin was found predominan

Quail fetal antigen (QFA): identification and comparison with chicken fetal antigen.

Quail fetal antigen (QFA), an analogous hematopoietic antigen to chicken fetal antigen (CFA), was identified and shown to be a developmental antigen on the blood cells of Japanese quail and other avian species. Rabbit antiserum against 14-day embryonic quail red blood cells (RBCs) was specifically adsorbed to achieve fetal specificity and to eliminate any cross reactivity with the CFA system. Complement-mediated microcytotoxicity and hemagglutination assays were used to detect the presence of QFA on hematopoietic cells. QFA was found on 100% of the peripheral RBCs from 10-day quail embryos but

Engineering the Developmental Continuum: From Embryo-Like Structures to Functional Tissue Interfaces.

Advanced in vitro developmental models, including embryo-like structures, organoids, and organ-on-chip platforms, have emerged as powerful systems for studying human development under controlled conditions. These models are enabled by advances in stem cell biology and bioengineering, allowing multicellular self-organization that recapitulates selected aspects of embryonic patterning and tissue formation. Key engineering parameters-such as stem cell source, signaling control, geometric confinement, matrix composition, and microfluidic regulation-define the boundary conditions that guide develop

Targeted cell interconversions reveal inner hair cell control of organ of Corti cytoarchitecture.

The mammalian organ of Corti is a precisely intercalated mosaic of two types of mechanosensory hair cells (HCs) and six types of supporting cells (SCs) arranged in 11 parallel rows. Differentially specialized SCs surround inner HCs (IHCs) and outer HCs (OHCs). To elucidate the developmental roles of IHCs and OHCs in the formation, differentiation, and assembly of the various SCs, we genetically switched HC identities at several developmental stages and also generated mice lacking IHCs. We find that IHCs promote or induce the (i) differentiation of one SC type (outer pillar) at the expense of a

Generating an organ-deficient animal model using a multi-targeted CRISPR-Cas9 system.

Gene-knockout animal models with organ-deficient phenotypes used for blastocyst complementation are generally not viable. Animals need to be maintained as heterozygous mutants, and homozygous mutant embryos yield only one-fourth of all embryos. In this study, we generated organ-deficient embryos using the CRISPR-Cas9-sgRNAms system that induces cell death with a single-guide RNA (sgRNAms) targeting multiple sites in the genome. The Cas9-sgRNAms system interrupted cell proliferation and induced cell ablation in vitro. The mouse model had Cas9 driven by the Foxn1 promoter with a ubiquitous expre

Cell-Mediated Branch Fusion in the Drosophila Trachea.

The Drosophila trachea is an interconnected network of epithelial tubes, which delivers gases throughout the entire organism. It is the premier model to study the development of tubular organs, such as the human lung, kidney, and blood vessels. The Drosophila embryonic trachea derives from a series of segmentally repeated clusters. The tracheal precursor cells in each cluster migrate out in a stereotyped pattern to form primary branches. Thereafter, the neighboring branches need to fuse to form an interconnected tubular network. The connection between neighboring branches is orchestrated by sp

To Stick or Not to Stick: Adhesions in Orofacial Clefts.

Morphogenesis requires a tight coordination between mechanical forces and biochemical signals to inform individual cellular behavior. For these developmental processes to happen correctly the organism requires precise spatial and temporal coordination of the adhesion, migration, growth, differentiation, and apoptosis of cells originating from the three key embryonic layers, namely the ectoderm, mesoderm, and endoderm. The cytoskeleton and its remodeling are essential to organize and amplify many of the signaling pathways required for proper morphogenesis. In particular, the interaction of the

A Case of Concurrent Disseminated Coccidioidomycosis and Embryonal Carcinoma When Lice and Fleas Coexist.

Coccidioidomycosis (CM) is a fungal infection endemic to the southwestern United States with a wide range of clinical presentations depending on the infected organ systems. Most infections are asymptomatic. Coccidioidomycosis causes a primary pulmonary infection and when symptoms occur, they most often resemble community-acquired pneumonia. One percent of cases disseminate, typically via hematogenous or lymphatic spread. It is in these cases that more severe symptoms may present and potentially overlap with those characteristics of other systemic illnesses. This is a case of CM disseminated to

The genetics and development of mandibles and hypopharyngeal sclerite and cornua in larvae of Drosophila gaucha.

The genetics and epigenetic processes associated with morphological organization are a principal aim of biology, ranging from cohesion between cells to shape and size of organisms. We investigate the post-embryonic development of Hypopharyngeal sclerite and cornua HPC and mandibles M of Drosophila gaucha larva. Integrated functioning of these Cephalopharyngeal skeleton parts of D. gaucha larva is essential for food acquisition, participating in locomotion and microhabitat selection. We examined two isolates by recording the growth of the HPC and M every 24 h for 8 days in parental, F1, F2 and

Congenital Heart Disease Genetics Uncovers Context-Dependent Organization and Function of Nucleoporins at Cilia.

Human genomics is identifying candidate genes for congenital heart disease (CHD), but discovering the underlying mechanisms remains challenging. In a patient with CHD and heterotaxy (Htx), a disorder of left-right patterning, we previously identified a duplication in Nup188. However, a mechanism to explain how a component of the nuclear pore complex (NPC) could cause Htx/CHD was undefined. Here, we show that knockdown of Nup188 or its binding partner Nup93 leads to a loss of cilia during embryonic development while leaving NPC function largely intact. Many data, including the localization of e

Teratogenic activity of HDAC inhibitors.

Modification of the terminal tails of histones is considered one of the documented mechanisms for epigenetic control of gene expression. Histone deacetylase inhibitors (HDACi) lead to a state of hyperacetylation of histone, a condition that can affect normal gene transcription. Furthermore, HDACi have many other protein targets involved in regulation of gene expression, cell proliferation and cell death. For these properties some HDACi are nowadays used as anticancer drugs with promising results. Several molecules with HDACi properties (valproic acid, trichostatin A, apicidin, MS-275, sodium b

[Biologic stress: possible role in the pathogenesis of depression].

This paper presents a review of biologic mechanisms of stress and analyzes their role in the pathogenesis of depressive disorder. Recent investigations point to the fact that apart from hereditary predisposition in the pathogenesis of depression different biologic mechanisms, included into the regulation of organism response to stress, have an important role. These findings are in harmony with the well known fact from the clinical practice, that exposure to stressor may lead to depression in persons who do not normally react depressively. The newest findings point out the significance of biolo

[Comparative analysis of chromosomal nucleolar organizing region activity in spontaneous and medical abortuses].

Comparative analysis of functional activity of silver stained nucleolar-organizing regions (NOR) activity was carried out in human fibroblasts of 70 spontaneous and 50 medical abortuses. The NOR activity was significantly higher in spontaneous abortuses compared with medical ones. This phenomenon which was observed in female, but not in male abortuses, did not depend upon the tissue origin of fibroblasts (embryonic or extraembryonic) and was due to increase of the NOR activity of the all acrocentric chromosomes to the level at which individual differences in the NOR activities among some acroc

Signaling pathways promoting epithelial mesenchymal transition in oral submucous fibrosis and oral squamous cell carcinoma.

Epithelial-mesenchymal transition (EMT) is a critical process that occurs during the embryonic development, wound healing, organ fibrosis and the onset of malignancy. Emerging evidence suggests that the EMT is involved in the invasion and metastasis of cancers. The inflammatory reaction antecedent to fibrosis in the onset of oral submucous fibrosis (OSF) and the role of EMT in its malignant transformation indicates a hitherto unexplored involvement of EMT. This review focuses on the role of EMT markers which are regulators of the EMT mediated complex network of molecular mechanisms involved in

The plastic pancreas.

Pancreas homeostasis is based on replication of differentiated cells in order to maintain proper organ size and function under changing physiological demand. Recent studies suggest that acinar cells, the most abundant cell type in the pancreas, are facultative progenitors capable of reverting to embryonic-like multipotent progenitor cells under injury conditions associated with inflammation. In parallel, it is becoming apparent that within the endocrine pancreas, hormone-producing cells can lose or switch their identity under metabolic stress or in response to single gene mutations. This new v

Beta-cell replacement for insulin-dependent diabetes mellitus.

Beta-cell replacement is considered the optimal treatment for type 1 diabetes, however, it is hindered by a shortage of human organ donors. Given the difficulty of expanding adult beta cells in vitro, stem/progenitor cells, which can be expanded in tissue culture and induced to differentiate into multiple cell types, represent an attractive source for generation of cells with beta-cell properties. In the absence of well-characterized human pancreas progenitor cells, investigators are exploring the use of embryonic stem cells and stem/progenitor cells from other tissues. Once abundant surrogate
