Scalable generation of T lymphocytes from human pluripotent stem cells in suspended organoids.

Human pluripotent stem cells (hPSCs) are a promising cell source for producing T cells for regenerative medicine and immunotherapy. However, it is challenging to develop a scalable suspension culture system for generating functional T lymphocytes from hPSCs. Here, we developed a Matrigel-based artificial thymic organoid (Gel-ATO) system by encapsulating hPSC-derived induced hematopoietic progenitor cells (iHPCs) with MS5-hDLL4 stromal cells. The resulting Gel-ATOs produced functional iT cells capable of antigen-specific cytotoxicity in vitro and tumor suppression in vivo. Building on this syst

Endothelialized callus organoids drive rapid regeneration of critical-size segmental long bone defects.

Large bone defects remain a major clinical challenge, as current treatments cannot effectively regenerate them and fail to restore functionality. To recapitulate the cellular complexity of the early fracture callus, we engineered human endothelialized callus organoids (hECOs) by co-culturing periosteum-derived skeletal progenitors with endothelial cells. This co-culture resulted in cellular self-assembly, leading to spatially organized, callus-like structures. Endothelial cells promoted skeletal progenitor cell expansion, extracellular matrix maturation, and progression toward hypertrophic car…

Colorectal neuroendocrine neoplasm organoids reveal the potential therapeutic value of targeting BRAF mutations.

BACKGROUND: Colorectal neuroendocrine neoplasms (NEN) are a rare entity lacking effective pharmacologic therapies. We aimed to address the paucity of preclinical models for colorectal NEN that facilitate the exploration of novel therapeutic strategies. METHODS: We used 3-dimensional culture techniques to establish colorectal NEN organoids. These organoids were subjected to pathological examination and sequence analyses to verify their neuroendocrine nature. Subsequently, the organoids were subcutaneously implanted into mice to create patient-derived organoid xenograft (PDOX) models. In vitro …

Multicellular rosette formation guides epithelial tissue assembly in pancreatic ductal adenocarcinoma cell organoids.

Mesenchymal-to-epithelial transitions are essential for epithelial tissue formation and thus the development of functional organs. Here, we demonstrate that the transition from a disordered mesenchymal state to a columnar epithelial structure in branched pancreatic ductal adenocarcinoma organoids is associated with rosette formation. We show that fluctuations in acto-myosin contractions on the emerging apical side of branches with high cell density create a tug-of-war mechanism, leading to regular spacing of rosettes. The distance between adjacent rosettes depends on the branch diameter, which…

Investigation of ABCA4 Missense Variants and Potential Small Molecule Rescue in Retinal Organoids.

PURPOSE: ABCA4-related retinopathy is the most common monogenic eye disorder in the world and is currently untreatable. Missense variants in … hy, into control induced pluripotent stem cells (iPSCs). The iPSCs were differentiated into retinal organoids, characterized and treated with small molecules. RESULTS: The expression level of ABCA4 missense proteins was reduced compared to WT ABCA4 suggesting the variants were degraded in a photoreceptor-like environment. The localization of the missense v …

Generation of vascularized retinal organoids containing microglia based on a PDMS microwell platform.

Retinal organoids (ROs) offer a biomimetic in vitro model for investigating human retinal development and disease. However, current ROs face several limitations, such as the absence of vascular networks and microglial cells (MGs). Here, we developed a vascularized retinal organoids (vROs) model by coculturing vascular organoids (VOs) with ROs in a V-bottom polydimethylsiloxane (PDMS) microwell platform. Through coculturing for 30 to 120 days, we observed the presence of tubular blood vessels at the center of vROs. Transcriptomic analysis revealed that the vascularization in ROs was associated …

Ceritinib inhibits growth and ACTH production of PitNETs: Insights from patient-derived organoids.

ACTH-secreting pituitary neuroendocrine tumors (PitNETs), also known as Cushing's disease (CD), are a rare condition lacking effective pharmacological treatments. Drug development for CD has been limited by the absence of suitable preclinical research models. Here, we established a patient-derived organoid (PDO) model and used it for screening tyrosine kinase inhibitors for CD. Ceritinib emerged as a promising therapeutic candidate due to its potent and consistent efficacy across PDOs from different patients in vitro, as well as in vivo in a xenograft mouse model. Multi-omics analyses and mole

Progress of iPSC-derived retinal organoids in the study of inherited retinal diseases.

Inherited retinal diseases (IRDs) constitute a complex and heterogeneous group of rare disorders characterized by significant genetic diversity. These conditions often lead to the degeneration of photoreceptor cells, resulting in severe visual impairment. A major challenge in the study and treatment of IRDs is the lack of appropriate preclinical models for investigating their pathogenesis and evaluating potential therapeutic interventions. In recent years, advances in retinal organoids (ROs) culture technology have provided promising new avenues for IRDs. This review systematically elaborates …

Intravital Multimodal Imaging of Human Cortical Organoids for Chronic Stroke Treatment in Mice.

Chronic stroke leads to enduring neurological deficits and remains a major clinical challenge. Human induced pluripotent stem cell (hiPSC)-derived cortical organoids (COs) offer promise for regenerative therapies, yet their application in chronic stroke remains understudied, in part due to limited tools for monitoring graft in vivo. Here, we present a Multimodal Imaging Platform for Organoid Tracking (MIPOT), which integrates MRI, bioluminescence imaging, light microscopy, and two-photon fluorescence microscopy to noninvasively track transplanted COs in the post-stroke brain over time. Using M…

Filtration-Based Rapid ECM-Hydrogel Elimination Enables Robust MALDI MSI of Intestinal Organoids.

The rapid development of matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) offers a powerful approach for the 2D metabolic profiling of intestinal organoids. However, its application remains constrained by challenges in sample preparation. Conventional extracellular matrix (ECM)-based hydrogel removal methods, such as repeated centrifugation or enzymatic digestion, are labor-intensive and time-consuming, often causing structural disruption of organoids and degradation of metabolites, thereby compromising the spectral quality and metabolite coverage. In this stud…

Small molecule treatment alleviates photoreceptor cilia defects in LCA5-deficient human retinal organoids.

Bialleleic pathogenic variants in LCA5 cause one of the most severe forms of Leber congenital amaurosis, an early-onset retinal disease that results in severe visual impairment. Here, we report the use of gene editing to generate isogenic LCA5 knock-out (LCA5 KO) induced pluripotent stem cells (iPSC) and their differentiation to retinal organoids. The molecular and cellular phenotype of the LCA5 KO retinal organoids was studied in detail and compared to isogenic controls as well as patient-derived retinal organoids. The absence of LCA5 was confirmed in retinal organoids by immunohistochemistry…

Robust generation of photoreceptor-dominant retinal organoids from porcine induced pluripotent stem cells.

Outer retinal degenerative diseases (RDDs) and injuries leading to photoreceptor (PR) loss are prevailing causes of blindness worldwide. While significant progress has been made in the manufacture of human pluripotent stem cell (hPSC)-derived PRs, robust production of pluripotent stem cell (PSC)-PRs from swine, a popular preclinical large animal model, would provide an avenue to collect conspecific functional and safety data to complement human xenograft studies. Toward this goal, we describe the highly efficient generation of PR-dominant porcine induced PSC (piPSC)-derived retinal organoids (…

Chronic low-dose cadmium exposure induces neurodevelopmental impairment in hESC-derived retinal organoids.

Cadmium, a heavy metal environmental toxin known for its neurotoxic effects on the central nervous system, is implicated in multiple eye diseases, including cataracts, glaucoma, and age-related macular degeneration. However, due to the lack of suitable experimental models, there is limited data on the impact of cadmium exposure on human retinal development. Human retinal organoids accurately mimic the 3D architecture of early fetal retinal tissue, making them suitable for investigating developmental retinal toxicity in the context of maternal cadmium exposure. This study found that cadmium exp…

Otic organoids: A model to study spiral ganglion neuron characteristics in Tmprss3-deficiency.

Organoids are valuable models to study human diseases. Cochlear implants (CIs) electrically stimulate spiral ganglion neurons (SGNs) to enable severely hearing-impaired people vocal communication. However, some studies found in patients with mutations in the TMPRSS3 gene that speech comprehension with CI was lower than for other etiologies. The reduced CI performance might be associated with reduced SGN excitability, the causes for which are largely unclear. We refined a protocol for generating SGN-like cells in otic organoids from human induced pluripotent stem cells (iPSC) and confirmed thei…

Advances in liver organoids: replicating hepatic complexity for toxicity assessment and disease modeling.

The lack of in vivo accurate human liver models hinders the investigation of liver-related diseases, injuries, and drug-related toxicity, posing challenges for both basic research and clinical applications. Traditional cellular and animal models, while widely used, have significant limitations in replicating the liver's complex responses to various stressors. Liver organoids derived from human pluripotent stem cells, adult stem cells primary cells, or tissues can mimic diverse liver cell types, major physiological functions, and architectural features. Recent advancements in the field have sho…

Human placental organoids as a model to probe early gestation maternal immune dynamics.

During pregnancy, the human placenta establishes tolerance toward fetal allogeneic tissue, where specialized trophoblast subtypes play a complex role in local and peripheral immunomodulation. However, due to inadequate models to study the early gestation of the human placenta, each trophoblast subtype's role in modulating the maternal immune response has remained elusive. Here, we derived human placental organoids from early gestation trophoblast stem cells to (1) identify patterns of immunomodulatory protein expression by trophoblast subtype and (2) evaluate the effects of the placental organ…

Hepatobiliary organoids differentiated from hiPSCs relieve cholestasis-induced liver fibrosis in nonhuman primates.

There is an urgent need for novel therapies to treat end-stage liver disease due to the shortage of available organs. Although cell transplantation holds considerable promise, its availability is limited due to the low engrafted cell mass and lack of unifying cell transplantation strategies. Here, we optimally established human induced pluripotent stem cell-derived functional hepatobiliary organoids (HBOs) based on our previous research and transplanted them into a monkey model via liver subcapsular and submesenteric transplantation routes to assess their potential clinical application.
