Inhibitory mechanisms of antibody production by nitrogen oxides released from activated macrophages during the immune response: relationship to energy consumption.

We investigated the relationship between the sensitivity of mouse splenocytes in immune response to nitrogen oxides and energy consumption rate of the cells. Macrophage-like cells (Mm1) pretreated with IL-6 served as the source of the nitrogen oxides. The antibody production of both 2,4,6-trinitrophenyl-keyhole limpet haemocyanin-primed splenocytes and B cell hybridomas was markedly reduced; about 20-40% of splenocytes and B cell hybridomas were killed by co-culture with IL-6-treated Mm1. Cell viability and antibody production were completely restored by the addition of NG-monomethyl L-arginin…

Oxidized PDI promotes thrombus formation in oxidative stress.

Protein disulfide isomerase (PDI) functions in thrombus formation in vivo and represents a viable target for antithrombotic therapy. PDI is a redox sensor that can either reduce or oxidize substrates depending on the redox environment. However, whether PDI functions primarily as a reductase or an oxidase in the context of thrombus formation remains unknown. Here, we used pharmacological and genetic approaches along with PDI mutants to determine how the PDI redox state affects thrombus formation. LOC14, which inhibits PDI reductase activity and induces PDI oxidation, promoted thrombus formation

Platelets and Cardioprotection: The Role of Nitric Oxide and Carbon Oxide.

Nitric oxide (NO) and carbon monoxide (CO) represent a pair of biologically active gases with an increasingly well-defined range of effects on circulating platelets. These gases interact with platelets and cells in the vessels and heart and exert fundamentally similar biological effects, albeit through different mechanisms and with some peculiarity. Within the cardiovascular system, for example, the gases are predominantly vasodilators and exert antiaggregatory effects, and are protective against damage in myocardial ischemia-reperfusion injury. Indeed, NO is an important vasodilator acting on

Nitric oxide inhibits neonatal hepatocyte oxidative metabolism.

BACKGROUND/PURPOSE: Liver function is frequently impaired in neonates with sepsis. Nitric oxide (NO) is thought to be a mediator of organ dysfunction and liver oxidative metabolism during sepsis. The authors developed an in vitro model to investigate the effect of NO and the combined effect of NO plus H2O2 on neonatal hepatocyte oxidative metabolism. METHODS: Hepatocytes were isolated from neonatal rats. Oxygen consumption was measured polarographically. In Study A, cells were exposed to S-Nitroso-N-acetylpenicillamine (SNAP), an NO donor, at various concentrations. In study B, myxothiazol an

[Oxidative stress].

Increased oxidative stress has been paid attention to as an important causative factor for diabetic cardiovascular complications. Hyperglycemic state are supposed to induce the overproduction of reactive oxygen species (ROS) through various mechanisms including increased non-enzymatic glycation end products (AGEs) formation, increased polyol pathway flux, increased O2-production from mitochondrial and protein kinase C (PKC)-dependent NAD (P) H oxidase activation in cardiovasucular tissues. Oxidative modification of lipoprotein is critical for atheromatous lesion formation. O2- reacts with nitr

[Nitric oxide].

Nitric oxide was identified as the relaxing factor derived from the endothelium in 1987. Nitric oxide synthesis allows the vascular system to maintain a state of vasodilation, thereby regulating arterial pressure. Nitric oxide is also found in platelets, where it inhibits adhesion and aggregation; in the immune system, where it is responsible for the cytotoxic action of macrophages; and in the nervous system, where it acts as neurotransmitter. A deficit in endogenous synthesis of nitric oxide contributes to such conditions as essential arterial hypertension, pulmonary hypertension and heart di

Metal-Organic Framework Induced Prenucleation to Construct Solid Solution Semiconductor Metal Oxides With Synergistic Enhancement of Moisture Resistance and Gas Response.

Accurately resolving trace acetone against the highly humid background of exhaled breath remains a central obstacle to non-invasive diabetes diagnosis. Here we develop a metal-organic framework (MOF)-conversion-induced prenucleation strategy for the controlled construction of Fe2TiO5-δ (FTO) solid solutions. During the topological transformation of MIL-125(Ti), Ti-derived oxide nuclei form within the pores of a daughter Fe-based MOF, enabling the generation of FTO enriched with mixed-valence Fe3+/Fe2+ and Ti4+/Ti3+ centres and abundant oxygen vacancies. The optimized FTO-3.49 sensor selectivel

Spatial distribution of Mn oxides in paddy soil profiles regulates rice grain accumulation of Ni and Cr through adsorption and oxidation.

Paddy soils derived from basalt weathering contain high levels of Fe-Mn oxides, along with elevated nickel (Ni) and chromium (Cr), posing threats to rice safety. Unlike Fe oxides, Mn oxides exhibit both adsorption and oxidation capabilities, creating complex regulatory mechanisms for Ni and Cr. The environmental impacts of these oxides depend on their spatial distribution, though the mechanisms remain unclear. This study investigates the synergistic regulation of δ-MnO2 on the speciation transformation and bioavailability of Ni and Cr. Pot experiments were setup using δ-MnO2 distributed either…

Freeze-thaw cycles mediate the divergent impacts of iron-manganese oxides on the degradation of veterinary antibiotics in cold-region soils.

The persistence of veterinary antibiotics in soils, particularly in alpine regions, poses environmental risks. A critical knowledge gap exis … the OTC and CIP degradation rates by 66.7% and 43.0%, respectively. The influence of iron-manganese oxides varied depending on the antibiotic. They promoted OTC degradation, with maximum enhancement (70.8%) at 1.25% addition of the catalyst. For CIP, only 1.25% addition promoted degradation, whereas 2.5% and 5.0% additions suppressed the degradation rate …

Relationship between Mineralogically Complex Iron (Oxyhydr)oxides and Plutonium Sorption and Reduction: A High-Energy Resolution X-ray Absorption Spectroscopy Perspective.

To facilitate the continued use of commercial nuclear power and address environmental contamination, it is essential to understand the fate and transport of plutonium (Pu) in (sub)surface environments. Current geochemical models do not account for complexity in mineral assemblages, such as metal substitution or the role of nanoscale crystallite sizes. In this work, we studied mineralogically complex systems where Pu(V) was the sorbate and Al-substituted or nanoscale iron (oxyhydr)oxides were the sorbents. Using M4-edge and L3-edge high-energy resolution fluorescence detection X-ray absorption …

Characteristic and mechanism of biological nitrogen and phosphorus removal facilitated by biogenic manganese oxides (BioMnOx) at various concentrations of Mn(II).

Biogenic manganese oxides (BioMnOx) have attracted considerable attention as active oxidants, adsorbents, and catalysts. However, characteristics and mechanisms of nitrification-denitrification in biological redox reactions mediated by different concentrations of BioMnOx are still unclear. Fate of nutrients (e.g., NH4+-N, TP, NO3--N) and COD were investigated through different concentrations of BioMnOx produced by Mn(II) in the moving bed biofilm reactor (MBBR). 34% and 89.2%, 37.8% and 89.8%, 57.3% and 88.9%, and 62.1% and 90.4% of TN and COD by MBBR were synchronously removed in four phases,…

Generic phosphate affinity constants of the CD-MUSIC-eSGC model to predict phosphate adsorption and dominant speciation on iron (hydr)oxides.

Estimating the availability of phosphorus in soils and sediments is complicated by the diverse mineralogical properties of iron (hydr)oxides that control the environmental fate of phosphorus. Despite various surface complexation models have been developed, lack of generic phosphate affinity constants (logKPO4s) for iron (hydr)oxides hinders the prediction of phosphate adsorption to iron (hydr)oxides in nature. The aim of this work is to derive generic logKPO4s for the Charge Distribution-Multisite Complexation extended-Stern-Gouy-Chapman (CD-MUSIC-eSGC) model using a large phosphate adsorption…

Intermetallic Compound TiM (M=Co, Fe) with a Layered Structure Prepared by Deoxidizing Ilmenite-type Oxides in Molten LiCl-CaH2 Mixtures.

CsCl-type intermetallic compounds TiM (M=Co, Fe) were obtained by deoxidizing trigonal ilmenite-type MTiO3 with a reducing agent CaH2 in molten LiCl at 600 °C. X-ray diffraction, nitrogen adsorption, scanning electron microscopy, and transmission electron microscopy with energy-dispersive X-ray, and X-ray photoelectron spectroscopy analyses revealed the formation of nanoscale layered structures, which enhanced specific surface areas (approximately 20 m2 /g) in the intermetallic compounds. In the initial deoxidation stage, Li2 TiO3 -like compounds were observed as an intermediate, suggesting th

Rejuvenation of iron oxides enhances carbon sequestration by the 'iron gate' and 'enzyme latch' mechanisms in a rice-wheat cropping system.

The 'enzyme latch' theory believes that oxygen constraints on phenol oxidase can restrain the activity of hydrolytic enzymes responsible for … macroaggregates, microaggregates, and silt and clay particles. Variations in amorphous and free Fe oxides, Fe-bound organic carbon and phenol oxidase activity were examined. After nine years, compared with the initial soil, the activation degree of free Fe oxides increased by 1.3- to 1.6-fold and the topsoil SOC stock increased by 13-61% across all trea …

Effect of organic substrate and Fe oxides transformation on the mobility of arsenic by biotic reductive dissolution under repetitive redox conditions.

The mobility of arsenic (As) in soil is highly affected by the change in the form of iron oxides present in the soil, which has a strong correlation with the change in redox potential. In this study, the altered mobility of As under repetitive redox conditions and the effect of organic substrates (i.e., glucose) on such change during four anoxic-oxic cycles were studied. During the 1st anoxic period, 37.1% of soil As was released into the soil solution, but the As in the soil solution decreased to 25.2% after the 1st oxic period. Moreover, the As in the soil solution further decreased during t…
