The Precuneus Region Drives Brain Network Changes in Tremor-Dominant Parkinson's Disease: Insights from a Morphological Causal Analysis.

The tremor-dominant (TD) subtype of Parkinson's disease (PD) is characterized by prominent tremor symptoms. However, the temporal and causal relationships between brain structural alterations in TD patients remain unexplored. A total of 61 TD patients and 61 matched healthy controls (HCs) were included in this study. The gray matter volume (GMV) of the bilateral precuneus (PCUN) was significantly reduced in TD patients. A structural covariance network analysis seeded with the left pallidum (PAL.L), which had the most significant differences, revealed a substantial reduction in covariance with

Predicting visual field global and local parameters from OCT measurements using explainable machine learning.

Glaucoma is characterised by progressive vision loss due to retinal ganglion cell deterioration, leading to gradual visual field (VF) impairment. The standard VF test may be impractical in some cases, where optical coherence tomography (OCT) can offer predictive insights into VF for multimodal diagnoses. However, predicting VF measures from OCT data remains challenging. To address this, five regression models were developed to predict VF measures from OCT, Shapley Additive exPlanations (SHAP) analysis was performed for interpretability, and a clinical software tool called OCT to VF Predictor w

Multi-shell connectome DWI-based graph theory measures for the prediction of temporal lobe epilepsy and cognition.

Temporal lobe epilepsy (TLE) is the most common epilepsy syndrome that empirically represents a network disorder, which makes graph theory (GT) a practical approach to understand it. Multi-shell diffusion-weighted imaging (DWI) was obtained from 89 TLE and 50 controls. GT measures extracted from harmonized DWI matrices were used as factors in a support vector machine (SVM) analysis to discriminate between groups, and in a k-means algorithm to find intrinsic structural phenotypes within TLE. SVM was able to predict group membership (mean accuracy = 0.70, area under the curve (AUC) = 0.747, Brie

Artificial intelligence in spatial transcriptomics: a review of methods, datasets, and directions.

Spatial transcriptomics (STs) measures gene expression while retaining the location of transcripts or cells within tissue sections. Unlike bulk RNA sequencing, which averages expression across cell populations, and single-cell RNA sequencing, which generally requires tissue dissociation, ST links molecular profiles to tissue architecture. This spatial information supports the analysis of tissue heterogeneity, cellular organization, and potential intercellular interactions. The spatially resolved nature of ST has unlocked diverse applications in fields such as cancer, neuroscience, reproductive

NSCLC EGFR Mutation Prediction via Random Forest Model: A Clinical-CT-Radiomics Integration Approach.

Non-small cell lung cancer (NSCLC) is the leading cause of cancer-related mortality worldwide. Accurate determination of epidermal growth factor receptor (EGFR) mutation status is essential for selecting patients eligible for tyrosine kinase inhibitors (TKIs). However, invasive genotyping is often limited by tissue accessibility and sample quality. This study presents a non-invasive machine learning model combining clinical data, CT morphological features, and radiomic descriptors to predict EGFR mutation status. A retrospective cohort of 138 patients with confirmed EGFR status and pre-treatme

Interpretable machine-learning prediction of PSEN1 missense variant pathogenicity based on multi-omics enrichment in six core Alzheimer's disease genes.

BACKGROUND AND OBJECTIVE: The clinical interpretation of Alzheimer's disease (AD) is frequently complicated by the prevalence of missense variants designated as being of uncertain significance within associated genes. Conventional computational prediction tools often overlook disease-specific pathophysiological contexts and lack pertinence and interpretability. Therefore, the present study aimed to develop a novel, interpretable framework for predicting the pathogenicity of AD missense variants by integrating transcriptomic and proteomic data enrichment patterns with machine learning methods.

Enhancing Anesthetic Depth Assessment via Unsupervised Machine Learning in Processed Electroencephalography Analysis: Novel Methodological Study.

BACKGROUND: General anesthesia comprises 3 essential components-hypnosis, analgesia, and immobility. Among these, maintaining an appropriate hypnotic state, or anesthetic depth, is crucial for patient safety. Excessively deep anesthesia may lead to hemodynamic instability and postoperative cognitive dysfunction, whereas inadequate anesthesia increases the risk of intraoperative awareness. Electroencephalography (EEG)-based monitoring has therefore become a cornerstone for evaluating anesthetic depth. However, processed electroencephalography (pEEG) indices remain vulnerable to various sources

Linking mixtures of air pollution exposures and preterm birth with a self-organizing map.

BACKGROUND: Exposure to nitrogen dioxide (NO2), ozone (O3), fine particulate matter (PM2.5), and heat has previously been associated with preterm birth. However, individuals are often exposed to a mixture of pollutants, which may exacerbate health effects. Unsupervised neural networks are highly suitable for characterizing high-dimensional exposures, but few studies of environmental mixtures have utilized these algorithms. OBJECTIVES: To implement a novel epidemiologic machine learning framework for linking high-dimensional mixture exposures and health outcomes, with an application in preterm

The presence, nature and network characteristics of behavioural phenotypes in temporal lobe epilepsy.

The relationship between temporal lobe epilepsy and psychopathology has had a long and contentious history with diverse views regarding the presence, nature and severity of emotional-behavioural problems in this patient population. To address these controversies, we take a new person-centred approach through the application of unsupervised machine learning techniques to identify underlying latent groups or behavioural phenotypes. Addressed are the distinct psychopathological profiles, their linked frequency, patterns and severity and the disruptions in morphological and network properties that

Associations between per- and polyfluoroalkyl substances and dental caries in the US adults.

Per- and polyfluoroalkyl substances (PFAS) are recognized environmental pollutants that have negative health effects. The association between PFAS and untreated root and coronal caries among adults is unknown. This study focused on the blood levels of various PFAS utilizing data from the National Health and Nutrition Examination Survey, which included a sample of 3340 individuals from the 2015 to 2020 cycles. The association between an individual's PFAS and their risk of untreated root and coronal caries was examined in this study using multivariable logistic regression, which adjusted for fac

Shared and distinct patterns of dynamic functional connectivity variability of thalamo-cortical circuit in bipolar depression and major depressive disorder.

Evidence has indicated abnormalities of thalamo-cortical functional connectivity (FC) in bipolar disorder during a depressive episode (BDD) and major depressive disorder (MDD). However, the dynamic FC (dFC) within this system is poorly understood. We explored the thalamo-cortical dFC pattern by dividing thalamus into 16 subregions and combining with a sliding-window approach. Correlation analysis was performed between altered dFC variability and clinical data. Classification analysis with a linear support vector machine model was conducted. Compared with healthy controls (HCs), both patients r

Electroconvulsive Therapy Induces Cortical Morphological Alterations in Major Depressive Disorder Revealed with Surface-Based Morphometry Analysis.

Although electroconvulsive therapy (ECT) is one of the most effective treatments for major depressive disorder (MDD), the mechanism underlying the therapeutic efficacy and side effects of ECT remains poorly understood. Here, we investigated alterations in the cortical morphological measurements including cortical thickness (CT), surface area (SA), and local gyrification index (LGI) in 23 MDD patients before and after ECT. Furthermore, multivariate pattern analysis using linear support vector machine (SVM) was applied to investigate whether the changed morphological measurements can be effectiv

Topical exposure to di(2-ethylhexyl) phthalate aggravates atopic dermatitis: an integrated network toxicology, bioinformatics, and experiment analysis.

Di(2-ethylhexyl) phthalate (DEHP) is one of the most widely used plasticizers; however, whether cutaneous DEHP exposure affects atopic dermatitis (AD) remains unclear. We combined network toxicology, transcriptomic analysis, machine learning, in vivo and in vitro experimental validation, and molecular docking to investigate whether cutaneous DEHP exposure exacerbates AD. ADMETlab 3.0 profiling of DEHP and its major metabolites predicted skin sensitization as one of the higher-probability toxicity endpoints among the evaluated computational outputs. We subsequently intersected DEHP-related targ

Predicting adverse cardiac events in sarcoidosis: deep learning from automated characterization of regional myocardial remodeling.

Recognizing early cardiac sarcoidosis (CS) imaging phenotypes can help identify opportunities for effective treatment before irreversible myocardial pathology occurs. We aimed to characterize regional CS myocardial remodeling features correlating with future adverse cardiac events by coupling automated image processing and data analysis on cardiac magnetic resonance (CMR) imaging datasets. A deep convolutional neural network (DCNN) was used to process a CMR database of a 10-year cohort of 117 consecutive biopsy-proven sarcoidosis patients. The maximum relevance - minimum redundancy method was

Altered task-modulated functional connectivity during emotional face processing in euthymic bipolar patients: A whole-brain psychophysiological interaction study.

BACKGROUND: Patients with bipolar disorder (BD) show deficits of facial emotion processing even in the euthymic phase. However, the large-scale functional brain network mechanism underlying the emotional deficit of BD remains unclear. Specifically, it is of importance to understand how the task-modulated functional connectivity (FC) was alternated over distributed brain networks in BD. METHODS: In this study, we analyzed functional MRI data of a face-matching task from 29 euthymic BD patients and 29 healthy controls (HC), and performed whole-brain psychophysiological interaction (PPI) analysi
