Molecular Insights into the Action Mechanism, Resistance Development, and Ecological Risks of Cyantraniliprole.

Cyantraniliprole, a second-generation diamide insecticide, exhibits broad-spectrum efficacy against numerous insect pests due to its selective activation of insect ryanodine receptors (RyRs). This activation triggers uncontrolled calcium release from the sarcoplasmic reticulum, resulting in sustained muscle contraction, paralysis, and ultimately death. Its unique mode of action, which is different from that of organophosphates, carbamates, pyrethroids, and neonicotinoids, helps minimize cross-resistance, making it a valuable component of integrated pest management (IPM). However, continuous fi

Pirimicarb exposure disrupts gut microbiome composition in honey bees (Apis mellifera L.): Seasonal differences and impaired colony recovery.

Pesticide exposure poses a significant threat to the honey bee's health, yet the effects of chemicals like pirimicarb on honey bee gut microbiomes remain poorly understood. This study investigates the impact of pirimicarb on honey bee microbiota, with a particular focus on how host physiology (spring vs. winter bees) modulates microbial responses to pesticide exposure. Through a series of controlled experiments, we demonstrate that pirimicarb is associated with selective dysbiosis resulting in taxon-specific microbial shifts without altering total bacterial load. Key bacterial taxa such as Sno

Analysis of neonicotinoid pesticides and their metabolites by liquid chromatography-electrospray ionization-tandem mass spectrometry.

Neonicotinoid insecticides were first introduced in the 1990s as an alternative to older, more toxic types such as organophosphates, carbamates, and chlorinated hydrocarbons and have since become the most widely used class of insecticides globally. However, their use comes at a significant cost due to high toxicity to pollinators such as bees, beneficial predatory insects, and even insectivorous birds. To further the study of neonicotinoid effects, we developed a method for the LC-ESI-MS/MS (liquid chromatography-electrospray ionization-tandem mass spectrometry) analysis of imidacloprid, cloth

Successful Cryoprotectant-Free Vitrification of Honey Bee (Apis mellifera) Drone Sperm With Royal Jelly Supplemented Extender.

In trying to protect honey bee species and maintain genetic diversity, employing effective approaches for drone sperm conservation is crucial. Based on literature, drone sperm cryopreservation extenders and methods have not been fully optimized. Our research aim was to enhance drone bee sperm preservation by incorporating royal jelly (RJ) in the extender of the cryoprotectant-free vitrification method. Drone sperm was collected from adult drone bees (36 days old) using the manual inversion method. Different concentrations of RJ (0%, 0.5%, 1%, 2.5% and 5%) were added to the extender. Diluted sp

Friends without benefits: Extensive cytotype sympatry and polyploid persistence in an African geophyte.

PREMISE: Polyploidy is a major factor in plant adaptation and speciation. Multiple mechanisms contribute to autopolyploid frequency within populations, but uncertainties remain regarding mechanisms that facilitate polyploid establishment and persistence. Here we aimed to document and predict cytotype distributions of Oxalis obliquifolia Steud. ex A. Rich. across Gauteng, South Africa, and test for evidence of possible mechanisms, including morphological, phenological, and reproductive traits, that may potentially facilitate polyploid persistence. METHODS: Over 320 O. obliquifolia plants from

Transferrin-mediated iron sequestration suggests a novel therapeutic strategy for controlling Nosema disease in the honey bee, Apis mellifera.

Nosemosis C, a Nosema disease caused by microsporidia parasite Nosema ceranae, is a significant disease burden of the European honey bee Apis mellifera which is one of the most economically important insect pollinators. Nevertheless, there is no effective treatment currently available for Nosema disease and the disease mechanisms underlying the pathological effects of N. ceranae infection in honey bees are poorly understood. Iron is an essential nutrient for growth and survival of hosts and pathogens alike. The iron tug-of-war between host and pathogen is a central battlefield at the host-path

Molecular characterization of viruses found in honeybee (Apis mellifera) colonies infested with Varroa destructor and Nosema cerana in Egypt.

Honey bees (Apis mellifera) have a vital role as pollinators of various crops in the global food supply. Honeybee colonies in Egypt have recently experienced an unexplained rise in annual loss due to a phenomenon known as Colony Collapse Disorder (CCD). In the current study, honey bees were collected from 14 sites from eight governorates in Egypt. The genetic diversity among the collected honey bee populations was investigated using the mitochondrial DNA cytochrome oxidase subunit I (COI). The amplified COI regions were sequenced, analyzed and aligned with other GenBank entries. The nucleotide

An Opto-electronic Sensor-ring to Detect Arthropods of Significantly Different Body Sizes.

Arthropods, including pollinators and pests, have high positive and negative impacts on human well-being and the economy, and there is an increasing need to monitor their activity and population growth. The monitoring of arthropod species is a time-consuming and financially demanding process. Automatic detection can be a solution to this problem. Here, we describe the setup and operation mechanism of an infrared opto-electronic sensor-ring, which can be used for both small and large arthropods. The sensor-ring consists of 16 infrared (IR) photodiodes along a semicircle in front of an infrared

Anthrax toxin component, Protective Antigen, protects insects from bacterial infections.

Anthrax is a major zoonotic disease of wildlife, and in places like West Africa, it can be caused by Bacillus anthracis in arid nonsylvatic savannahs, and by B. cereus biovar anthracis (Bcbva) in sylvatic rainforests. Bcbva-caused anthrax has been implicated in as much as 38% of mortality in rainforest ecosystems, where insects can enhance the transmission of anthrax-causing bacteria. While anthrax is well-characterized in mammals, its transmission by insects points to an unidentified anthrax-resistance mechanism in its vectors. In mammals, a secreted anthrax toxin component, 83 kDa Protective

Importance of IgG4 determination in in vitro immunotherapy follow-up of inhalant allergens.

Choosing the right parameters to recommend immunotherapy in allergologic diagnosis is very important. Therefore, other parameters which are independent of the improvement of clinical manifestations and which indicate the evolution of asthma are very useful. Although a decrease in skin reactions was observed in 20% to 25% of patients in previous studies, since the in vitro techniques appeared, their evolution has been observed. Total and antigen-specific IgE evolution, as well as histamine release from basophils, in immunotherapy were followed as we had presented in previous studies. In this wo

Pollen Exposed to Aerial Pesticide Spray Is a Major Exposure Pathway for Stingless Bees.

Testing possible exposure routes of pesticides is essential for assessing the risks to pollinators. In Australia, the pesticide Bacillus thuringiensis var. israelensis (Bti) is widely used in mosquito control programs and applied aerially to mangrove and saltmarsh habitats. Bti formulations contain both bacterial spores and toxins, allowing detection using DNA-based methods, whereas bee foraging behaviour can be quantified by pollen DNA metabarcoding. We combined these approaches to investigate both the foraging behaviour of stingless bees and their potential exposure to Bti in coastal mosquit

Seasonal Susceptibility of Winter and Summer Honeybee Workers (Apis mellifera L.) to Oral Exposure of Selected Insecticides Under Laboratory Conditions.

To cope with severe seasonal changes in climate and food availability, colonies of Apis mellifera L. generate two distinct worker phenotypes during winter and summer. The aim of this research was to evaluate the effect of oral exposure to insecticides on the toxic susceptibility of winter and summer honeybee workers (Apis mellifera L.) under laboratory conditions. In this work, under all tested conditions and seasons, emamectin benzoate was the most toxic compound and used as the standard (Toxicity Index = 100) for comparison with other compounds. It had the lowest LC50 (Lethal Concentration r

Assessing the potential of bee-collected pollen sequence data to train machine learning models for geolocation of sample origin.

UNLABELLED: Pollen is a robust and widespread substance that captures a historical snapshot of a specific time and place, and it can be used to track movements through space by examining the pollen deposited on various objects. Palynology, the study of pollen, is used across fields such as conservation, natural history, and forensics, where it is particularly useful for tracing the origin and movement of objects. However, pollen has remained underutilized due to the difficulty of distinguishing many pollen taxa beyond the family level and limited pollen reference material to support location p

Host plant-specific volatiles of Beauveria bassiana-colonized plants initiate trophic plant-aphid-predator cascades.

BACKGROUND: Entomopathogenic ascomycetes (EAs) have efficacy in insect pest control through direct contact and indirectly as plant endophytes. As endophytes, they lead indirectly to pest mortality, enhance plant resilience to stresses, promote plant growth and alter plant volatile emissions. These changes can influence plant interactions with beneficial fauna, such as predators, parasitoids and pollinators. However, variation in endophytic colonization across plant species and EA strains raises questions about cultivar-specific responses within the same crop species. Here we evaluated the impa

Impact of drought stress memory and variations in type and degree on root characteristic, yield stability and drought tolerance in Orchardgrass.

BACKGROUND: As climate change intensifies, perennial plants face more frequent drought periods throughout their lifespan. Drought stress memory in certain plants significantly enhances their adaptability to challenging environmental circumstances. However, in open-pollinated crops, this process is influenced by population plasticity due to the type and degree of genetic diversity, and inbreeding depression. To understand how pre-exposure to moderate drought enhances the plants' response to subsequent severe stresses, four synthetic poly-crossed populations were constructed using parents with c

Plant Resource Use and Pattern of Usage by the Naturalized Orchid Bee (Euglossa dilemma: Hymenoptera: Apidae) in Florida.

The Neotropical orchid bee Euglossa dilemma was found to be naturalized in southern Florida in 2003, and, by 2022, it had colonized the southern half of Florida. Observations of the bee's collection of plant resources, primarily flowers, were made from 2003 through to 2022 to document its plant usage and understand the patterns of its plant usage. The bee utilized 259 plant taxa, 237 species, and 22 horticultural forms, in 156 genera and 56 families in 263 total uses. Of 247 taxa of flowers, 120 were visited primarily for nectar, 46 for both nectar and pollen, 60 for pollen, including 42 buzz-

Within- and between-population comparisons suggest independently acting selection maintaining parallel clines in Scots pine (Pinus sylvestris).

Parallel clines in traits related to adaptation in a species can be due to independent selection on a pair of traits, or due to selection in one trait resulting in a parallel cline in a correlated trait. To distinguish between the mechanisms giving rise to parallel adaptive population divergence of multiple traits along an environmental gradient we need to study variation, correlations, and selective forces within individual populations along the gradient. In many tree species, budset timing (BST) forms a latitudinal cline, and parallel clinal variation is also found in other seedling traits,
