Transcarboxylase 5S structures: assembly and catalytic mechanism of a multienzyme complex subunit.

Transcarboxylase is a 1.2 million Dalton (Da) multienzyme complex from Propionibacterium shermanii that couples two carboxylation reactions, transferring CO(2)(-) from methylmalonyl-CoA to pyruvate to yield propionyl-CoA and oxaloacetate. Crystal structures of the 5S metalloenzyme subunit, which catalyzes the second carboxylation reaction, have been solved in free form and bound to its substrate pyruvate, product oxaloacetate, or inhibitor 2-ketobutyrate. The structure reveals a dimer of beta(8)alpha(8) barrels with an active site cobalt ion coordinated by a carbamylated lysine, except in the …

Human cervicofacial actinomycoses: microbiological data for 1997 cases.

Actinomycoses are sporadically occurring endogenous polymicrobial inflammatory processes, in which fermentative actinomycetes of the genera Actinomyces, Propionibacterium, or Bifidobacterium act as the principal pathogens. Difficulties in diagnosing the disease in a timely and reliable fashion have led clinicians and microbiologists to grossly underestimate its medical importance. Therefore, we evaluated microbiological and selected clinical data derived from 1997 culture-positive cases of human cervicofacial actinomycoses examined in our laboratories during 1972-1999. The causative actinomyce…

The potential role of tranexamic acid and botulinum toxin in treating patients with acne vulgaris: a split face comparative study.

BACKGROUND: Many causes can lead to acne, such as increased sebum production, altered sebum lipid quality from increased androgen activity, abnormal keratinization of the infundibular epithelium, and Propionibacterium acnes (P. acnes) colonizing the follicles. AIM: The present study aimed to evaluate and compare the clinical efficacy of botulinum toxin-A vs. tranexamic acid intradermal injection in the treatment of acne vulgaris. METHODS: This split face study includes forty patients with varying grades of acne vulgaris. They were injected intradermally with tranexamic acid (20 mg/ml) on the…

Perturbations in the skin microbiome of infantile and adult seborrheic dermatitis and new treatment options based on restoring a healthy skin microbiome.

Seborrheic dermatitis (SD) is a common, multifaceted skin condition, but its undefined etiology hampers the development of effective therapeutic strategies. In this review, we describe the intricate relationship between the skin microbiome and the pathogenesis of SD, focusing on the complex interplay between three major groups of organisms that can either induce inflammation (Malassezia spp., Staphylococcus aureus) or else promote healthy skin (Propionibacterium spp.). We describe how the disequilibrium of these microorganisms in the skin microbiome can develop skin inflammation in SD patients…

Beneficial or deleterious effects of a preexisting hypersensitivity to bacterial components on the course and outcome of infection.

Priming with heat-killed Propionibacterium acnes enhances the sensitivity of mice to lipopolysaccharide (LPS) and other biologically active bacterial components. We show that P. acnes priming has protective and deleterious effects on a subsequent serovar Typhimurium infection. It may result in a complete protection or prolonged survival, or it may accelerate mortality of the infected mice, depending on the number of serovar Typhimurium bacteria administered and on the degree of LPS hypersensitivity at the time of infection. Both effects of P. acnes-induced hypersensitivity are mediated by gamm…

Susceptibility of anaerobic bacteria to antimicrobial agents.

The antimicrobial susceptibility of 1,117 clinical isolates of anaerobic bacteria was determined by the agar dilution technique. Metronidazole was the most active agent; only Propionibacterium acnes and Actinomyces sp. isolates were resistant. Clindamycin and chloramphenical were the next most effective agents. Beta-lactam antibiotics, with the exception of penicillin, were active against most anaerobes other than the Bacteroides fragilis group. At a breakpoint of 8 mg/l, 25% of Fusobacterium spp. and 30% of the non-fragilis Bacteroides spp. were resistant to penicillin. The highest resistance…

Bacterial aspects of chronic blepharitis.

Patients with all forms of chronic blepharitis were thoroughly evaluated. These patients were found to have evidence for a primary bacterial component in the disease process only in the clinical staphylococcal and mixed seborrheic/staphylococcal forms of chronic blepharitis. Evidence was found for a shared common pathway for Staphylococcus aureus, coagulase negative staphylococci, and Propionibacterium acnes to contribute to the disease process. These organisms were found to produce lypolytic exoenzymes including fatty wax esterase, cholesteryl esterase, and triglyceride lipase. Statistically …

Advances in understanding Cutibacterium acnes hemolysis: virulence factor or genomic trait?

BACKGROUND: Cutibacterium acnes is now recognized a major opportunistic pathogen causing a wide range of infections. Henceforth, this bacterium is largely involved in device-related infections especially prosthetic joint infections, spine infections but also cardiovascular infections, neurosurgical implant infections and breast infections. It has also been associated with acne and probably sarcoidosis and prostate tumors. We conducted a narrative review to better understand the phylotypes involved in these clinical entities and if hemolysis can be a diagnosis maker of pathogenicity. METHODS:

Effects of nutritive and non-nutritive sweeteners on acne vulgaris: A systematic review.

Acne vulgaris is a prevalent inflammatory skin disorder influenced by sebaceous gland activity, hormonal regulation, and microbial proliferation. Dietary factors, including sweetening agents, may modulate acne pathophysiology, affecting sebum production, inflammatory responses, and the skin microbiome. This systematic review summarizes the effects of nutritive and non-nutritive sweeteners on acne-related markers, with a focus on Cutibacterium acnes activity and associated inflammatory pathways. Non-nutritive sweeteners such as mannitol and xylitol show potential in regulating acne pathogenesis

Cutibacterium acnes: the Urgent Need To Identify Diagnosis Markers.

Cutibacterium acnes role is well described during acne but remains a mystery regarding its implication in bone and prosthesis or cerebrospinal fluid shunt infections. The main issue is that these low-grade symptom infections are difficult to diagnose and lead to irreversible and grave sequelae for patients. Consequently, there is an urgent need to find new biomarkers to accelerate the diagnosis of disease, an issue addressed by Beaver et al. thanks to a promising proteomic approach.

Antimicrobial and Antioxidative Activity of Newly Synthesized Peptides Absorbed into Bacterial Cellulose Carrier against Acne vulgaris.

The ongoing search for effective treatment of Acne vulgaris is concentrated, i.a., on natural peptides with antimicrobial properties. The aim of this work was the development of new amino acid derivatives with potential activity on dermal infections against selected microorganisms, including the facultative anaerobe C. acne. The peptides P1-P6 were synthesized via Fmoc solid phase peptide synthesis using Rink amide AM resin, analyzed by RP-HPLC-MS, FTIR, DPPH radical scavenging activity, and evaluated against C. acne and S. aureus, both deposited and non-deposited in BC. Peptides P1-P6 present

Humeral periosteal spur sign with osteolysis as an early radiographic indicator of low-grade Cutibacterium acnes prosthetic shoulder infection.

The early diagnosis of low-grade Cutibacterium acnes prosthetic shoulder joint infection is challenging due to the lack of clinical and laboratory signs. Patients present with atypical symptoms such as stiffness or failure to improve shoulder function. The diagnosis is often delayed with impact on long-term outcomes. We present the case of an 82-year-old man with a surgical site erythema occurring 7 weeks after reverse shoulder arthroplasty associated with a light raise of C reactive protein (20 mg/L). At 9 weeks, radiographs revealed a 'periosteal spur sign' (humeral calcar) and localised adj

Host-microbiome interactions and recent progress into understanding the biology of acne vulgaris.

Acne is one of the most common skin diseases worldwide and results in major health care costs and significant morbidity to severely affected individuals. However, the pathophysiology of this disorder is not well understood. Host-microbiome interactions that affect both innate and adaptive immune homeostasis appear to be a central factor in this disease, with recent observations suggesting that the composition and activities of the microbiota in acne is perturbed. Staphylococcus epidermidis and Cutibacterium acnes (C. acnes; formerly Propionibacterium acnes) are two major inhabitants of the ski…

Solar UV radiation and microbial life in the atmosphere.

Many microorganisms are alive while suspended in the atmosphere, and some seem to be metabolically active during their time there. One of the most important factors threatening their life and activity is solar ultraviolet (UV) radiation. Quantitative understanding of the spatial and temporal survival patterns in the atmosphere, and of the ultimate deposition of microbes to the surface, is limited by a number factors some of which are discussed here. These include consideration of appropriate spectral sensitivity functions for biological damage (e.g. inactivation), and the estimation of UV radi

The development of antimicrobial resistance due to the antibiotic treatment of acne vulgaris: a review.

OBJECTIVE: To review recent studies on the use of antibiotics in acne vulgaris which provide insight into the development of antimicrobial r … vulgaris can lead to antimicrobial resistance with serious and intractable problems not limited to Propionibacterium acnes (P. acnes), the skin and acne vulgaris themselves, but also to other bacterial species, with systemic consequences. These findings suggest that antibiotics should be prescribed in combination with benzoyl peroxide and/or topical retinoids and …
