PHILOPHTHALMUS HECHINGERI N. SP. (DIGENEA: PHILOPHTHALMIDAE), A HUMAN-INFECTING EYE FLUKE FROM THE ASIAN MUD SNAIL, BATILLARIA ATTRAMENTARIA.

Two cases of human philophthalmosis have been reported in Japan. Gravid flukes removed from the eyes of the patients were broken, but their morphological characteristics suggest that an unknown species of the genus Philophthalmus is involved as a pathogen for humans. The mitochondrial DNA barcode of the human eye fluke enabled us to discover its larval stage from the Japanese mud snail, Batillaria attramentaria. The discovered cercaria had previously been temporarily described as "Philophthalmid sp. I." In this study, we examined the infection status of B. attramentaria with Philophthalmid sp.

Preclinical Research in Glycogen Storage Diseases: A Comprehensive Review of Current Animal Models.

GSD are a group of disorders characterized by a defect in gene expression of specific enzymes involved in glycogen breakdown or synthesis, commonly resulting in the accumulation of glycogen in various tissues (primarily the liver and skeletal muscle). Several different GSD animal models have been found to naturally present spontaneous mutations and others have been developed and characterized in order to further understand the physiopathology of these diseases and as a useful tool to evaluate potential therapeutic strategies. In the present work we have reviewed a total of 42 different animal

Discovery of GnIH and Its Role in Hypothyroidism-Induced Delayed Puberty.

It is known that hypothyroidism delays puberty in mammals. Interaction between the hypothalamo-pituitary-thyroid (HPT) and hypothalamo-pituitary-gonadal (HPG) axes may be important processes in delayed puberty. Gonadotropin-inhibitory hormone (GnIH) is a newly discovered hypothalamic neuropeptide that inhibits gonadotropin synthesis and release in quail. It now appears that GnIH is conserved across various mammals and primates, including humans, and inhibits reproduction. We have further demonstrated that GnIH is involved in pubertal delay induced by thyroid dysfunction in female mice. Hypothy

Ypel1: a novel nuclear protein that induces an epithelial-like morphology in fibroblasts.

BACKGROUND: Embryonic development depends on the regulation of cell morphology and behaviour to carry out morphogenesis. One example of this is the development of the face, which is constructed from independent blocks of tissue that must grow in a coordinated way and then fuse to form a continuous tissue. RESULTS: We have isolated a novel gene of unknown function from mouse and quail embryos in a search for genes involved in craniofacial development. Sequence analysis of this gene, known as Ypel1, demonstrates a striking level of amino acid conservation between vertebrates and invertebrates b

The regulation of tyrosinase gene transcription.

Tyrosinase is one of the key enzymes essential for melanogenesis. The control of its activity rests in part at the level of transcriptional regulation. The 5' promoter regions of the human, mouse, chicken, quail, snapping turtle, and frog tyrosinase sequences have been isolated and the mechanisms regulating the activity of these sequences are beginning to be elucidated. This review provides an update on the following aspects of tyrosinase gene regulation: basal promoter elements that determine the site of transcription initiation for RNA polymerase II; the cis-acting elements and DNA-binding f

The neural crest: its relations with APUD and paraneuron concepts.

The neural crest cells give rise to a large variety of derivatives including neural, mesenchymal, APUD and/or paraneuron cell types. A better knowledge of these derivatives was partly achieved through studies using Le Douarin's quail/chick marker system. We review here evidences which were thus provided for a neural crest origin of calcitonin containing cells, carotid body, aortic paraganglia, adrenomedulla, and against a neurectodermal origin of enterogastric and respiratory tract endocrine cells. The role of neural crest cells in Pearse's APUD system is discussed. The results implicate that

A Spatio-Temporal-Dependent Requirement of Sonic Hedgehog in the Early Development of Sclerotome-Derived Vertebrae and Ribs.

Derived from axial structures, Sonic Hedgehog (Shh) is secreted into the paraxial mesoderm, where it plays crucial roles in sclerotome induction and myotome differentiation. Through conditional loss-of-function in quail embryos, we investigate the timing and impact of Shh activity during early formation of sclerotome-derived vertebrae and ribs, and of lateral mesoderm-derived sternum. To this end, Hedgehog interacting protein (Hhip) was electroporated at various times between days 2 and 5. While the vertebral body and rib primordium showed consistent size reduction, rib expansion into the soma

Genetic evidence for variant selection in the course of dilute passaging of mumps vaccine virus.

Mumps vaccine viruses, Leningrad-3 (L-3) strain, harvested at the 8th (8P) and 38th (38P) passage levels, were compared by nucleotide sequencing of the fusion (F) and the phosphoprotein (P) genes, and for replication efficiency in cell culture. Sequencing revealed only one clear base substitution throughout the entire F gene, and no substitutions in the variable 183-nucleotide-long region of the P gene. However, the 8P virus, unlike the 38P variant, contained multiple "ambiguous" nucleotide regions, i.e., additional bases positioned at the level of the principal ones. The 38P variant replicate

Morphological and immunohistochemical study of perivascular macrophages: Key findings for angiogenesis.

As a possible target for pro- or anti-angiogenic therapy, the process of angiogenesis has become more widely recognized in disease and is essential in normal growth and development. The current study examined the immunological activities of macrophages during angiogenesis in quail embryos. CD68 was used to identify perivascular macrophages, which also retained expression of some stem cell markers, including CD34 and CD117, and also CD21. The macrophages also expressed vascular endothelial growth factor (VEGF), a key promoter of angiogenesis, and matrix metalloproteinase-9 (MMP-9), which degrad

Anaphylaxis to duck and goose eggs with preserved tolerance to hen's eggs: A case report.

BACKGROUND: Selective allergy to eggs from specific avian species, while maintaining tolerance to hen's eggs, is rare and may present diagnostic challenges. CASE REPORT: We report the case of a 75-year-old woman with no history of atopic disease who experienced episodes of anaphylaxis after consuming duck and goose eggs, while tolerating hen's eggs. Skin prick tests with commercial hen's egg extracts were negative. Serum testing revealed very low total IgE levels (5 kU/L) and low specific IgE to ovomucoid (Gal d 1), with negative results for other egg proteins. Prick-to-prick testing showed p

Immunohistochemical Detection of Serum Amyloid A-Derived Amyloid Deposits in Poultry Using a Synthetic Peptide-Based Antibody.

Avian amyloidosis, mostly caused by the abnormal deposition of amyloid fibrils derived from serum amyloid A (SAA), is histologically diagnosed through the morphological identification of eosinophilic amyloid deposits in tissues. Congo red staining is a popular special staining method for detecting amyloid; however, immunohistochemical staining would allow precise identification and confirmation of SAA-derived amyloid by targeting its protein composition. The purpose of this study was to establish an immunohistochemical method for detecting amyloid deposits in poultry. A rabbit polyclonal antib

Acute toxicity of diphacinone in Northern bobwhite: effects on survival and blood clotting.

The anticoagulant rodenticide diphacinone was slightly toxic (acute oral LD50 2014 mg/kg) to Northern bobwhite (Colinus virginianus) in a 14-day acute toxicity trial. Precise and sensitive assays of blood clotting (prothrombin time, Russell's Viper venom time, and thrombin clotting time) were adapted for use in quail, and this combination of assays is recommended to measure the effects of anticoagulant rodenticides. A single oral sublethal dose of diphacinone (434 mg/kg body weight) prolonged clotting time at 48 h post-dose compared to controls. At 783 mg/kg (approximate LD02), clotting time w

Cranial nerve growth in birds is preceded by cholinesterase expression during neural crest cell migration and the formation of an HNK-1 scaffold.

The expression of the neural crest cell (NCC) markers acetylcholinesterase (AChE) and the HNK-1-epitope is compared from the emigration of cephalic NCC until the formation of the cranial nerves V-X in chicken and quail hindbrain. We show that NCC transiently express acetylcholinesterase (AChE) activity during their emigration; NCC migrate into butyrylcholinesterase (BChE)-positive areas of the cranial mesenchyme. Along these migratory tracks that foreshadow the course of later projecting cranial nerves, BChE increases strongly in cells that may represent immature Schwann cells. Both AChE and B

The neural crest is a powerful regulator of pre-otic brain development.

The role of the neural crest (NC) in the construction of the vertebrate head was demonstrated when cell tracing techniques became available to follow the cells exiting from the cephalic neural folds in embryos of various vertebrate species. Experiments carried out in the avian embryo, using the quail/chick chimera system, were critical in showing that the entire facial skeleton and most of the skull (except for he occipital region) were derived from the NC domain of the posterior diencephalon, mesencephalon and rhombomeres 1 and 2 (r1, r2). This region of the NC was designated FSNC (for Facial

Immunohistochemical localization of insulin-like growth factor I and II in the endocrine pancreas of birds, reptiles, and amphibia.

Immunoreactive insulin-like growth factors I and II (IGF-I, IGF-II) were sought in the endocrine pancreas of representative birds, reptiles, and amphibia using antisera specific for mammalian IGF-I and IGF-II and the classical islet hormones insulin (INS), glucagon (GLUC), somatostatin (SOM), and pancreatic polypeptide (PP) in double immunofluorescence. Both IGF-I and IGF-II immunoreactivities were present in the endocrine pancreas of all species. IGF-II immunoreactivity was exclusively found in INS-immunoreactive (-IR) cells, indicating evolutionary conservation of the islet IGF-II system. In

Development of high-affinity polyclonal antibodies against chicken CKAP4 and its application in infectious bursal disease.

Infectious bursal disease (IBD), a highly infectious and acute illness triggered by infectious bursal disease virus (IBDV), is notorious for causing immunosuppression in chickens, presenting a huge challenge for global poultry production. Cytoskeleton-associated protein 4 (CKAP4), an endoplasmic reticulum-resident tumorigenesis regulator, remains unexplored in IBDV pathogenesis and lacking anti-chCKAP4 antibodies hindered research. To address the critical lack of species-specific reagents, we generated anti-chCKAP4 polyclonal antibodies with a titer of 1:102400 via prokaryotic expression and a

Mixed infection with reovirus and Chlamydophila in a flock of budgerigars (Melopsittacus undulatus).

Eleven budgerigars (Melopsittacus undulatus) from a zoological collection presented at necropsy with emaciation and splenomegaly or hepatomegaly or both. Polymerase chain reaction assays performed on liver and spleen samples were positive for Chlamydophila psittaci in 2 of 3 birds tested, and histologic findings in 2 additional birds were compatible with chlamydiosis. The aviary was subsequently closed to the public, and a 45-day treatment regimen with doxycycline in the seeds was initiated. No further deaths of birds with hepatomegaly or splenomegaly were observed after the first day of treat

The role of retinoic acid in the morphogenesis of the neural tube.

We have examined the role of the signalling molecule, retinoic acid, in the process of neurulation and the subsequent growth and differentiation of the central nervous system using quail embryos that have developed in the absence of retinoic acid. Such retinoic acid-free embryos undergo abnormal neural tube formation in terms of its shape and structure, but the embryos do not display spina bifida or exencephaly. The neural tubes have a wider floor plate, a thicker roof plate and a different dorsoventral shape. Phalloidin staining and electron microscopy revealed alterations in the actin filame
