Pleural mesothelioma (PM) is characterized by poor prognosis and limited therapeutic options. Y-box-binding protein 1 (YB-1) was shown to drive growth and migration of PM cells. Here, we evaluated the effect of genetic and pharmacological targeting of YB-1 on PM growth and response to cisplatin and radiation treatment. YB-1 knockdown via siRNA resulted in reduced PM cell growth, which significantly correlated with wt BAP1 and mutant NF2 and P53 status. Entinostat inhibited YB-1 deacetylation and its efficacy correlated with YB-1 knockdown-induced growth inhibition in 20 PM cell lines. Tumor gr

UVB radiation is the most mutagenic component of the UV spectrum that reaches the earth's surface and causes the development of DNA damage in the form of cyclobutane pyrimidine dimers and 6-4 photoproducts. UV radiation usually results in cellular death, but if left unchecked, it can affect DNA integrity, cell and tissue homeostasis and cause mutations in oncogenes and tumour-suppressor genes. These mutations, if unrepaired, can lead to abnormal cell growth, increasing the risk of cancer development. Epidemiological data strongly associates UV exposure as a major factor in melanoma development

The gusmps/gusmps mouse has no beta-glucuronidase activity and develops murine mucopolysaccharidosis type VII (MPS VII). The clinical and pathologic abnormalities are similar to those found in humans with severe MPS VII. Mutant mice are dysmorphic, dwarfed, and have a shortened life span. Pathologic findings include widespread lysosomal storage. To determine whether bone marrow transplantation (BMT) corrects these abnormalities, genetically identical mutant animals were given syngeneic bone marrow transplants using cells from +/+ mice. Initial experiments showed that levels of beta-glucuronida

A genetic analysis of an array of mitomycin-induced rearrangements in immature Drosophila oocytes is reported. Induced aberrations were recovered representing detachments of the compound-X chromosome, Y chromosome fragments, X chromosome loss and mosaicism. The spectrum of rearrangements induced by mitomycin C was very similar to that induced by X-ray treatment of immature oocytes. This work suggests that mitomycin C has two models of action. The drug is radiomimetic for it induces the types of aberrations recovered after X-irradiation. Mitomycin C also seems to have a delayed effect which is

Genetic consequences of radioactive fall-outs of the Chelyabinsk plant producing plutonium (1949-1952) and the Chernobyl accident have been analysed. Three powerful radioactive fall-outs caused a population genetic dose of 682,801 cSv per 217,750 persons (the average dose was 2.25 cSv). Individual variations were from 1-2 mSv to 1.2 Sv or more. The population genetic dose from the Chernobyl accident was higher (32 x 10(6) man/cSv), but the individual dose was lower (mainly no more than 1 cSv). Progenic analyses of residents of radiation contaminated areas showed no increase in the incidence of

Socioeconomic and demographic factors, lifestyle and cultural characteristics may play an important role in the development of genetic damage. This damage represents a potential health risk to an individual and increases the risk of developing negative outcomes. The aim of this study was to investigate the association of a set of factors and genetic damage by gathering data from previously studied populations in southern Brazil. This study analyzed data related to genetic damage and socioeconomic, demographic, and lifestyle variables of 514 individuals aged 18 to 64 years, residing in 8 cities

UNLABELLED: The risk of breast cancer increases decades after ionizing radiation exposure, thereby linking aging intrinsically to the evolution of cancer. We hypothesized that radiation accelerates aging and carcinogenesis through similar pathways, specifically low-grade systemic inflammation. In this study, we used the radiation-genetic mammary chimera model to examine the differential expression of 532 plasma proteins in BALB/c female mice between radiation exposure and experiment termination at 18 months. Mice were sham irradiated or irradiated with 50 cGy prior to being orthotopically tran

BACKGROUND: Radiation has been discussed as a potential causative factor for Multiple Sclerosis (MS). However, it is unresolved whether radiation increases the aggressiveness of the immune system or whether it alters the nervous tissue to become vulnerable to a pre-existing autoimmune attackdisposition. We report a patient with an MS-like disease confined to the irradiated part of his central nervous system (CNS).
CASE REPORT: within the course of a year, a 29 year-old man developed three relapses that were distinguishable regarding their neurological signs and symptoms. Clinically and on MRI

Colorectal cancer (CRC) arises from genetic, environmental, and lifestyle factors. Recognizing these risks helps clinicians identify high-risk patients and guide prevention. Risk factors are grouped as nonmodifiable (age, family history, race/ethnicity, prior abdominal/pelvic radiation, or inflammatory bowel disease) and modifiable (obesity, diet, inactivity, alcohol, or tobacco). The growing incidence of early-onset CRC (diagnosed before 50) is a particular concern and highlighted throughout this section.

In the Western population, 1 out of every 50 individuals will develop melanoma. The incidence of melanoma is increasing faster than any other malignancy. The development of melanoma is multifactorial arising from an interaction between genetic susceptibility and environmental exposures. Sixty to seventy percent of melanomas are thought to be caused by ultraviolet radiation. Most cutaneous melanomas are of increased risk. Prevention strategies involve mitigating the environmental risk factors and identifying individuals with phenotypic risk factors for increased surveillance.

The author reports on the molecular biological site of action for cancerogenic substances. He shows how UV-radiation may lead to malignomas of the skin in the hereditary disease xeroderma pigmentosum and how ionising rays may cause changes of the DNA, the place of the genetic information. Since the virus genesis of malignant diseases has become more important lately, the author tries to point out the molecular biological connections of an eventual involvement of virus in the development of malignant diseases.

Fibrodysplasia ossificans progressiva (FOP) is one of the genetic and developmental forms of heterotopic ossification. We report a case of FOP on the volar surface of the distal radius, located close to the median nerve and radial artery with neurologic symptoms secondary to median nerve entrapment. The patient underwent surgical excision of the heterotopic lesion followed by radiation therapy. He had no signs of recurrence with more than 1 year of follow-up. Careful microsurgical dissection of the heterotopic mass must be performed to prevent the formation of new painful lesions and iatrogeni

Lymphoedema is the build-up of lymphatic fluid leading to swelling in the tissues. Most commonly it affects the peripheries. Diagnosis is based on clinical assessment and imaging with lymphoscintigraphy. Treatment is supportive with compression garments, massage, good skin hygiene and prompt use of antibiotics to avoid the complication of cellulitis. Most commonly, lymphoedema occurs as a result of damage to the lymphatic system following surgery, trauma, radiation or infection. However, it can be primary, often associated with a genetic defect that causes disruption to the development of the

Skin cancer is the most common cancer in the United States, and most cases are preventable. Persons with certain genetic risk factors, including having a lighter natural skin color; blue or green eyes; red or blonde hair; dysplastic nevi or a large number of common moles; and skin that burns, freckles, or reddens easily or becomes painful after time in the sun, have increased risk for skin cancer. Persons with a family or personal history of skin cancer, especially melanoma, are also at increased risk. Although these genetic factors contribute to individual risk, most skin cancers are also str

Melanin provides a crucial filter for solar UV radiation and its genetically determined variation influences both skin pigmentation and risk of cancer. Genetic evidence suggests that the acquisition of a highly stable melanocortin 1 receptor allele promoting black pigmentation arose around the time of savannah colonization by hominins at some 1-2 Ma. The adaptive significance of dark skin is generally believed to be protection from UV damage but the pathologies that might have had a deleterious impact on survival and/or reproductive fitness, though much debated, are uncertain. Here, I suggest

BACKGROUND: Basal cell carcinoma (BCC) of the skin is the most common cancer in humans. Epidemiologic studies implicate sunlight exposure as one risk factor, but the limited association between BCCs and UVB radiation (i.e., UV radiation of a wavelength of 280-320 nm) suggests that additional factors must be involved. At the molecular level, not much is known about the role of specific environmental agents in the pathogenesis of BCCs. Point mutations of the types produced by UVB radiation are seen in the p53 gene (also known as TP53; chromosome 17p) of 40%-56% of BCCs. Loss of heterozygosity (L

PURPOSE: Recurrent and metastatic human papillomavirus (HPV)-associated head and neck squamous cell carcinoma (R/M HPV+ HNSCC) remains largely incurable, with genetic drivers incompletely defined. We profiled the genetic landscape of R/M HPV+ HNSCC and functionally characterized genetic alterations strongly enriched in this cancer type.
EXPERIMENTAL DESIGN: We identified genetic alterations uniquely enriched in 159 R/M HPV+ tumors. High-priority alterations were functionally modeled, examining proliferation, clonogenicity, migration/invasion, apoptosis, therapy response, in vivo growth, metas

Vitamin D is crucial for full functioning in many genes, and vitamin-D deficiency interferes with many processes, including brain development and DNA repair. Several lines of evidence suggest that prenatal and early postnatal vitamin-D deficiency increases risk for autism, probably through multiple effects that include impaired brain development and increased de novo mutations. High rates of autism in several genetically based hypomelanotic skin disorders present a puzzle, because ultraviolet-B (UVB) radiation acting on skin is the major natural source of vitamin D, and lighter skin, which inc

The rationale for and importance of research on effects after radiation at "low doses" are outlined. Such basic radiobiological studies on induction of repair enzymes, protective mechanisms, priming, and hypersensitivity are certainly all relevant to treatment of cancer (see Section 1, Studies at low doses - relevance to cancer treatment). Included are examples from many groups, using various endpoints to address the possibility of an induced resistance, which has been compared to the adaptive response [M.C. Joiner, P. Lambin, E.P. Humanity will eventually inhabit extraterrestrial environments, yet crucial questions remain regarding the feasibility of reproduction and long-term survival away from the Earth. Exposure to space radiation might induce DNA damage, potentially resulting in harmful mutations in future generations. Altered gravity on other planets or within space colonies could also interfere with embryonic and fetal development. For sustainable colonization beyond Earth, humans and essential animal species, e.g., livestock and companion animals, must be transported. To prevent inbreeding and genetic deteriorati
