A highly basic sequence at the N-terminal region is essential for targeting the DNA replication protein ORC1 to the nucleus in Leishmania donovani.

The conserved eukaryotic DNA replication protein ORC1 is one of the constituents of pre-replication complexes that assemble at or very near origins prior to replication initiation. ORC1 has been shown to be constitutively nuclear in Leishmania major. This study investigates the sequences involved in nuclear localization of ORC1 in Leishmania donovani, the causative agent of visceral leishmaniasis. Nuclear localization signals (NLSs) have been reported in only a few Leishmania proteins. Functional analyses have delineated NLSs to regions of ~60 amino acids in length in the tyrosyl DNA phosphodi

Optical fluid and biomolecule transport with thermal fields.

A long standing goal is the direct optical control of biomolecules and water for applications ranging from microfluidics over biomolecule detection to non-equilibrium biophysics. Thermal forces originating from optically applied, dynamic microscale temperature gradients have shown to possess great potential to reach this goal. It was demonstrated that laser heating by a few Kelvin can generate and guide water flow on the micrometre scale in bulk fluid, gel matrices or ice without requiring any lithographic structuring. Biomolecules on the other hand can be transported by thermal gradients, a m

HIV rebounds from latently infected cells, rather than from continuing low-level replication.

Rapid rebound of plasma viremia in patients after interruption of long-term combination antiretroviral therapy (cART) suggests persistence of low-level replicating cells or rapid reactivation of latently infected cells. To further characterize rebounding virus, we performed extensive longitudinal clonal evolutionary studies of HIV env C2-V3-C3 regions and exploited the temporal relationships of rebounding plasma viruses with regard to pretreatment sequences in 20 chronically HIV-1-infected patients having undergone multiple 2-week structured treatment interruptions (STI). Rebounding virus duri

Biomodel-guided stereotaxy.

OBJECTIVES: To simplify the practice of stereotactic surgery by using an original method, apparatus, and solid anatomic replica for trajectory planning and to validate the method and apparatus in a laboratory and clinical trial. METHODS: The patient is marked with fiducials and scanned by using computed tomography or magnetic resonance imaging. The three-dimensional data are converted to a format acceptable to stereolithography. Stereolithography uses a laser to polymerize photosensitive resin into a solid plastic model (biomodel). Stereolithography can replicate blood vessels, soft tissue, t

Viroporins: discovery, methods of study, and mechanisms of host-membrane permeabilization.

The 'Viroporin' family comprises a number of mostly small-sized, integral membrane proteins encoded by animal and plant viruses. Despite their sequence and structural diversity, viroporins share a common functional trend: their capacity to assemble transmembrane channels during the replication cycle of the virus. Their selectivity spectrum ranges from low-pH-activated, unidirectional proton transporters, to size-limited permeating pores allowing passive diffusion of metabolites. Through mechanisms not fully understood, expression of viroporins facilitates virion assembly/release from infected

High-Throughput Sequencing Reveals Three Rhabdoviruses Persisting in the IRE/CTVM19 Cell Line.

Cell cultures derived from ticks have become a commonly used tool for the isolation and study of tick-borne pathogens and tick biology. The IRE/CTVM19 cell line, originating from embryos of Ixodes ricinus, is one such line. Previously, reovirus-like particles, as well as sequences with similarity to rhabdoviruses and iflaviruses, were detected in the IRE/CTVM19 cell line, suggesting the presence of multiple persisting viruses. Subsequently, the full genome of an IRE/CTVM19-associated rhabdovirus was recovered from a cell culture during the isolation of the Alongshan virus. In the current work,

Mutations in the phosphorylation sites of SARS-CoV-2 encoded nucleocapsid protein and structure model of sequestration by protein 14-3-3.

SARS-CoV-2 is the etiologic agent of COVID-19. There is currently no effective means of preventing infections by SARS-CoV-2, except through restriction of population movement and contact. An understanding of the origin, evolution and biochemistry (molecular biology) of SARS-CoV-2 is a prerequisite to its control. Mutations in the phosphorylation sites of SARS-CoV-2 encoded nucleocapsid protein isolated from various populations and locations, are described. Mutations occurred in the phosphorylation sites, all located within a stretch which forms a phosphorylation dependent interaction site, inc

Selective expression of the transcription elongation factor ELL3 in B cells prior to ELL2 drives proliferation and survival.

B cell activation is dependent on a large increase in transcriptional output followed by focused expression on secreted immunoglobulin as the cell transitions to an antibody producing plasma cell. The rapid transcriptional induction is facilitated by the release of poised RNA pol II into productive elongation through assembly of the super elongation complex (SEC). We report that a SEC component, the Eleven -nineteen Lysine-rich leukemia (ELL) family member 3 (ELL3) is dynamically up-regulated in mature and activated human B cells followed by suppression as B cells transition to plasma cells in

Investigation of common, low-frequency and rare genome-wide variation in anorexia nervosa.

Anorexia nervosa (AN) is a complex neuropsychiatric disorder presenting with dangerously low body weight, and a deep and persistent fear of gaining weight. To date, only one genome-wide significant locus associated with AN has been identified. We performed an exome-chip based genome-wide association studies (GWAS) in 2158 cases from nine populations of European origin and 15 485 ancestrally matched controls. Unlike previous studies, this GWAS also probed association in low-frequency and rare variants. Sixteen independent variants were taken forward for in silico and de novo replication (11 com

Isolation of a monoclonal antibody that recognizes the origin binding domain of JCV, but not SV40, large T-antigen.

Within immunocompromised populations, the JC polyomavirus is the cause of the often-fatal disease Progressive Multifocal Leukoencephalopathy (PML). JC virus encodes a protein, termed T-antigen (T-ag), which is essential for its replication and pathogenicity. Previous studies of JCV T-ag have, in general, used antibodies raised against SV40 T-ag. Unfortunately, SV40 T-ag is also detected in humans and therefore there have been concerns about cross-reactivity. To address this issue, we have isolated a monoclonal antibody that binds to the JCV, but not the SV40, T-ag origin-binding domain (OBD).

Prolonged gene expression in primary porcine pancreatic cells using an Epstein-Barr virus-based episomal vector.

Epstein-Barr virus (EBV)-based plasmids containing the origin of replication (oriP) and EBV nuclear antigen 1 (EBNA-1) are well known for the stable episomal maintenance in human cells. In order to clarify whether an EBV-based plasmid can be maintained stably in the porcine pancreatic cells which are the primary candidate sources of islet xenotransplantation, we constructed pEBVGFP encoding the green fluorescent protein (GFP). Monolayer culture of the porcine neonatal pancreatic cells was lipofected with pEBVGFP or pGFP which was derived from pEBVGFP by deleting out oriP and EBNA-1. pEBVGFP si

HIV-1 infection of primary effusion lymphoma cell line triggers Kaposi's sarcoma-associated herpesvirus (KSHV) reactivation.

Kaposi's sarcoma-associated herpesvirus (KSHV) is a gammaherpesvirus etiologically linked to primary effusion lymphoma (PEL), to a subset of multicentric Castleman's disease and to Kaposi's sarcoma (KS), the most common neoplasm associated with AIDS. Among KSHV-infected individuals, the risk of KS is much higher in those with human immunodeficiency-1 (HIV-1) infection than among those with other types of immunosuppression, suggesting a direct action of HIV-1 on KSHV replication. We show in our report that BC-3 cells, a chronically KSHV-infected B-cell line of a PEL origin, are permissive to HI

[Electrophysiology of transmissible encephalopathies].

The purpose of electrophysiological studies during the evolution of spongiform encephalopathies or prion diseases is to describe the changes of spontaneous or evoked electroencephalographic activity in natural and experimental diseases (natural and experimental scrapie, Kuru and Creutzfeldt-Jakob (CJ) disease in man and in different animal models) and to establish or to propose an action mechanism of the infectious agents. In the aim, the changes of electroencephalogram (EEG) and evoked potentials are described. Generally these modifications constitute an excellent and early marker of the init

High catabolism of BrdU may explain unusual sister chromatid differentiation and replication banding patterns in cancer cells.

Two T-cell acute lymphoblastic leukemia (ALL) cell lines, PEER and CCRF-CEM, were studied by various chromosome banding techniques, including 5-bromodeoxyuridine (BrdU) incorporation methods. Although of very similar origin, these two cell lines behave quite differently. In particular, CEM cell line exhibited an abnormal replication banding pattern (RBP) and poor sister chromatid differentiation (SCD). Study of their thymidylate synthase and thymidine kinase activities indicated that CEM had a more active salvage pathway for thymidylate synthesis than did PEER cell line, which may suggest an e

Slow growth but intense hypertrophy of thyrocytes in long-standing Grave's goitres.

In order to evaluate the relative contribution of true cell replication, in comparison to mere cell hypertrophy, to the notoriously slow growth of toxic Graves' goitres persistently exposed to TSH receptor antibodies and requiring continuous thyrostatic treatment for more than one year, we have determined the fraction of dividing cells in 8 such goitres, using the monoclonal antibody Ki-67. This antibody identifies cells in late G1-, S-, G2- and M-phase. The fraction of Ki-67 positive cells ranged from 0.1 to 4.1% with a mean of 2.3 +/- 1.6. The apparently low absolute number of dividing cells

Isolation and characterization of polyoma virus mutants which grow in murine embryonal carcinoma and trophoblast cells.

Mouse trophoblast cell lines established from cultured midterm placenta and a cell line obtained from cultured blastocyst resemble trophectoderm cells. These cells are resistant to infection by wild-type polyoma virus. We have isolated six polyoma virus mutants capable of growing in trophoblast cell lines. Restriction enzyme analyses and marker rescue experiments revealed that the genetic changes necessary for the growth of these mutants ( PyTr mutants) in trophoblast cells were located in a regulatory region of the genome between the origin of viral DNA replication and the region encoding the

CD8 T lymphocytes deploy embryonic cell cycle control mechanisms for rapid cell proliferation.

Rapid proliferation of CD8 T cells is crucial for adaptive immunity against viral infection. CD8 T cells can complete division cycles in les … erestingly, CD8 T cells and mESCs differ in the frequency of G2&M phase cells, the abundance of DNA replication origin licensing and initiation factors, and the abundance of APC/C substrates. Thus, somatic T cells have both unique and shared cell cycle control mechanisms to promote rapid cell cycles.

Non-Coding Transcripts From Diversified Members of IgLec Family Protect Antiviral Effectors From Viral miRNA.

Immune gene families frequently produce numerous non-coding transcripts, yet their biological functions remain largely unexplored. Here, using the arthropod immunoglobulin domain-containing lectin (IgLec) family as a paradigm, we provide evidence that this complexity may reflect a critical defensive function for non-coding transcripts in antiviral immunity. IgLec is restricted to decapod infraorders (Brachyura and Astacidea), and a structurally related bacterial homolog suggests its origin may involve horizontal gene transfer. The IgLec family generates extraordinary transcript diversity throu

Swine acute diarrhea syndrome coronavirus nsp10 antagonizes IFN-λ1 production via synergizing with hnRNPU and blocking IRF1 NUclear Translocation.

Swine acute diarrhea syndrome coronavirus (SADS-CoV) is a bat-origin porcine enteric alphacoronavirus first identified in Guangdong, China in 2017, which causes severe watery diarrhea and high mortality in neonatal piglets, leading to in substantial economic losses to the global swine industry. Here, we report that SADS-CoV nsp10 functions as an IFN-λ1 antagonist. Specifically, nsp10 suppressed IFN-λ1 production independently of its zinc finger domains. Furthermore, nsp10 inhibits MAVS- and IRF1-driven IFN-λ1 expression within the RLR signaling pathway. Nsp10 blocks poly (I:C)-induced nuclear

Mapping transcriptional patterns of MPXV in human epithelial cells.

The 2022 global outbreak of the monkeypox virus (MPXV) clade IIb highlighted changes in transmission dynamics, clinical features, and tissue tropism, yet the impact of host cell origin on viral transcription remains unclear. We performed comparative transcriptomic profiling of MPXV clade IIb in human epithelial cell lines from vaginal, intestinal, ectocervical, and renal tissues. Temporal analysis in vaginal cells revealed two major transcriptional clusters: early genes enriched for host-virus interaction factors in terminal genome regions, and intermediate-late genes encoding transcription, r
