The effect of oxygen on vasoformative cell division. Evidence that 'physiological hypoxia' is the stimulus for normal retinal vasculogenesis.

PURPOSE: To assess the role of oxygen in normal retinal vasculogenesis. METHODS: A new preparation for studying cytogenesis in retinal wholemounts was developed. Nuclei of dividing cells were labeled with a monoclonal antibody to bromodeoxyuridine (BrdU), and vascular cells were covisualized with Griffonia simplicifolia lectin. The topography and time course of vasculogenic cell division and vessel formation were determined in the kitten retina during normal development and under experimental hyperoxia and hypoxia. RESULTS: During normal development, vasculogenic cell division was maximal at

Structural and causal links between retinal vascular geometry and neural layer thickness.

PURPOSE: To investigate structural relationships between retinal vasculometry from color fundus photography (CFP) and neural layers obtained from Optical Coherence Tomography (OCT) scans. METHODS: This cross-sectional study used the Retina-based Microvascular Health Assessment System (RMHAS) to extract retinal vascular measurements in the 6*6 mm area centered on the macular region and analyzed their associations with OCT parameters. We investigated both pairwise correlations between individual retinal layers and vascular parameters and associations between sets of variables. Mendelian randomi

The retinal vasculature pathophysiological changes in vision recovery after treatment for indirect traumatic optic neuropathy patients.

PURPOSE: To evaluate the retinal vasculature pathophysiological changes of indirect traumatic optic neuropathy (ITON) patients after effecti … y after effective treatment of ITON patients was associated with the increased oxygen saturation of retinal vessels, better availability of oxygen in the retina, greater vessel density, and thicker retinas, which might further underlie the vasculature mechanism of vision recovery in ITON patients.

Tubercular Retinitis and Retinal Vasculitis: Manifestations of a Shared Disease Spectrum.

Isolated retinal involvement in ocular tuberculosis (TB) is not well understood and remains largely underreported. TB retinal lesions can be classified into two broad categories: retinal vasculitis and isolated retinal granuloma. TB retinal vasculitis is the more recognized phenotype and may present with perivascular infiltrates alone or alongside other ocular signs such as subvascular lesions, focal vascular tortuosity, and occlusive vasculitis. These additional signs are crucial for diagnosing TB retinal vasculitis in TB-endemic regions. However, isolated retinal granulomas due to TB are rar

Spontaneous retinal changes in Sprague Dawley rats.

Routine ophthalmological examination of over 6,000 untreated Crl:CD(SD)BR rats up to 2 years old, used in toxicologic studies from 1989 to 1992, has provided information on spontaneous retinal changes and their incidence with age. Focal linear retinopathy and coloboma were the most common findings; retinal hemorrhage, saccular aneurysm of the retinal vessels, retinal fold, absence of optic disk and retinal vascularization, and myelination of optic nerve fibers were also observed. Such accumulated data in untreated animals are of prime necessity in assessing possible drug- and chemical-induced …

Retinal imaging for cerebrovascular risk stratification: evidence, limitations, and practical thresholds for the ophthalmologist.

BACKGROUND: The retina shares developmental origin, microvascular anatomy, and barrier physiology with the brain, making non-invasive retinal imaging a candidate window onto cerebrovascular burden. Despite a growing body of association data, translational claims in this field have frequently outpaced the evidence, and clinically actionable guidance for the practising ophthalmologist remains sparse. This review asks a deliberately narrow question: which retinal imaging domains provide evidence mature enough to inform clinical decisions about cerebrovascular risk, and which remain investigationa

Inner ocular blood flow responses to an acute decrease in blood pressure in resting humans.

Whether inner ocular vessels have an autoregulatory response to acute fluctuations in blood pressure is unclear. We tried to examine the validity of acute hypotension elicited by thigh-cuff release as to assess the dynamic autoregulation in the ocular circulation. Blood flow velocity in the superior nasal and inferior temporal retinal arterioles, and in the retinal and choroidal vasculature were measured with the aid of laser speckle flowgraphy before and immediately after an acute decrease in blood pressure in 20 healthy subjects. Acute hypotension was induced by a rapid release of bilateral

Colocalization of neuropilin-1 and Flk-1 in retinal neovascularization in a mouse model of retinopathy.

PURPOSE: To investigate the mechanisms of the development of retinal neovascularization, the localizations of vascular endothelial (VEGF) receptors Flk-1 and neuropilin (NP)-1 mRNAs were examined. METHODS: The model of retinopathy of prematurity (ROP) was produced by ischemia-induced ocular neovascularization, by exposing postnatal day-7 mice to 75% oxygen for 5 days and then returning them to room air for 5 days. Retinal neovascularization was visualized by injection of fluorescein-dextran. Expression of Flk-1 and NP-1 mRNAs were examined by in situ hybridization with flatmount and serial se

Congenital optociliary shunt vessels.

Congenital optociliary shunt vessels have rarely been reported in the literature. Three cases of congenital optociliary shunt vessels are presented: two with unilateral shunt vessels and one with bilateral shunt vessels. All three patients were asymptomatic with good visual acuity and essentially normal visual fields with no apparent change in visual function over time. Associated with congenital optociliary shunt vessels is an anomalous bifurcation of the vasculature. Diagnostic implications of optociliary shunt vessels include 1) congenital variations, or 2) acquired secondary to a long-stan

Structural and vascular retinal changes and visual outcomes after treatment of compressive anterior visual pathway lesions: A prospective longitudinal study.

There is limited evidence regarding longitudinal postoperative changes in retinal vascular density in patients with compressive anterior visual pathway lesions. In this study, we aimed to investigate the changes in retinal vasculature after decompression surgery based on optical coherence tomography (OCT) and OCT angiography (OCTA) and the predictive utility of OCT and OCTA parameters for postoperative visual outcomes. This prospective study included 22 eyes (15 patients) with a compressive anterior visual pathway lesion, which were assessed preoperatively as well as at 1, 3, and 6 postoperati

Pathogenesis and therapy of traction detachment in various retinal vascular diseases.

We examined six patients with retinal vasculopathies (two cases each of angiomatosis retinae [von Hippel-Lindau disease], exudative vitreoretinopathy, and Coats' disease), who developed preretinal and vitreal membranes, retinal traction detachment, and exudation under the retina. The membranes consisted of a core of collagen of various diameter fibers usually covered on both sides by cells that displayed glial cell characteristics. The retina reattached spontaneously and exudates disappeared when traction was eliminated by vitreous surgery. Membrane formation did reoccur after vitrectomy if th

Hypoxic-ischemic injury causes functional and structural neurovascular degeneration in the juvenile mouse retina.

Ischemic stroke is a major cause of long-term disabilities, including vision loss. Neuronal and blood vessel maturation can affect the susceptibility of and outcome after ischemic stroke. Although we recently reported that exposure of neonatal mice to hypoxia-ischemia (HI) severely compromises the integrity of the retinal neurovasculature, it is not known whether juvenile mice are similarly impacted. Here we examined the effect of HI injury in juvenile mice on retinal structure and function, in particular the susceptibility of retinal neurons and blood vessels to HI damage. Our studies demonst

[Guidance of Medical Care for Familial Exudative Vitreoretinopathy:Research on Rare and Intractable Diseases, Health and Labour Sciences Research Grants].

Familial exudative vitreoretinopathy is a hereditary insufficiency of retinal vascularture, which manifests a variety of vitreoretinal abnormalities, including nonvascularlized retina, abnormality of retinal vessel growing, dragged retina, retinal folds and total retinal detachment. While causative genes have been identified, cases are often sporadic. Periodic examination is necessary to find recurrence of the disease and late complications, including rhegmatogenous retinal detachment, cataract and glaucoma.

Short-term treatment with VEGF receptor inhibitors induces retinopathy of prematurity-like abnormal vascular growth in neonatal rats.

Retinal arterial tortuosity and venous dilation are hallmarks of plus disease, which is a severe form of retinopathy of prematurity (ROP). In this study, we examined whether short-term interruption of vascular endothelial growth factor (VEGF) signals leads to the formation of severe ROP-like abnormal retinal blood vessels. Neonatal rats were treated subcutaneously with the VEGF receptor (VEGFR) tyrosine kinase inhibitors, KRN633 (1, 5, or 10 mg/kg) or axitinib (10 mg/kg), on postnatal day (P) 7 and P8. The retinal vasculatures were examined on P9, P14, or P21 in retinal whole-mounts stained wi

Retinal Vascular Occlusion and Underlying Generalized Vascular Diseases.

Retinal vascular occlusion not only threatens vision loss but is also associated with other systemic risk factors and vascular diseases. Interdisciplinary cooperation is of great importance in these patients. The risk factors hardly differ between arterial and venous retinal occlusions, which is due to the special anatomy of retinal vessels. Major underlying conditions associated with retinal vascular occlusion include arterial hypertension, diabetes mellitus, dyslipidemia, cardiac disease, particularly atrial fibrillation, or vasculitis of large- and middle-sized arteries. Every new diagnosed…
