[A new method for the disruption of cell walls of gram-positive bacteria and mycobacteria on the point of nucleic acid extraction: sand method].

Nowadays molecular methods are widely used in the rapid diagnosis of infectious agents. Polymerase chain reaction (PCR) is the most preferre … like cetyltrimethyl ammonium bromide for the Mycobacterium genus. In this study, we planned to use sand particles for the mechanical elimination of the cell wall without any need for chemicals and we called this procedure as "sand method". For the purpose of DNA extraction, the fine-grained sand was washed with ddH(2)O without losing small particles a …

Polymerase chain reaction-based assay for the detection and identification of sand fly gregarines in Lutzomyia longipalpis, a vector of visceral leishmaniasis.

Gregarines that parasitise phlebotomine sand flies belong to the genus Psychodiella and, even though they are highly host-specific, only five species have been described to date. Their most outstanding features include the unique localisation of the oocysts in the accessory glands of the female host, which ensures contamination of the egg surface during oviposition, and the fact that they naturally parasitise the vectors of Leishmania, causal agent of leishmaniasis. The type species, Ps. chagasi, was first described in Lutzomyia longipalpis, vector of visceral leishmaniasis (VL), from Brazil. …

Influence of ionic strength and pH on the limitation of latex microsphere deposition sites on iron-oxide coated sand by humic acid.

This study, for the first time, investigates and quantifies the influence of slight changes in solution pH and ionic strength (IS) on colloidal microsphere deposition site coverage by Suwannee River Humic Acid (SRHA) in a column matrix packed with saturated iron-oxide coated sand. Triple pulse experimental (TPE) results show adsorbed SRHA enhances microsphere mobility more at higher pH and lower IS and covers more sites than at higher IS and lower pH. Random sequential adsorption (RSA) modelling of experimental data suggests 1 μg of adsorbed SRHA occupied 9.28 ± 0.03 × 10(9) sites at pH7.6 and…

Study of phlebotomine sand flies (Diptera: Psychodidae) collected in a Leishmania-endemic area of the metropolitan region of Belo Horizonte, Brazil.

Phlebotomine sand flies are distributed across nearly all faunal regions of the world, represented by over 800 species, of which many are important vectors of human pathogens. Brazil is currently faced with the expansion and urbanization of leishmaniases, with an increase in the numbers of human cases and seropositive dogs in various medium-sized to large cities. The objective of the current study was to survey the phlebotomine sand fly species in an area endemic for American cutaneous leishmaniasis (ACL) and American visceral leishmaniasis (AVL), i.e., the municipal district of Santa Luzia, l…

Geochemical interactions between process-affected water from oil sands tailings ponds and North Alberta surficial sediments.

In Northern Alberta, the placement of out-of-pit oil sands tailings ponds atop natural buried sand channels is becoming increasingly common. Preliminary modeling of such a site suggests that process-affected (PA) pond water will infiltrate through the underlying clay till aquitard, reaching the sand channel. However, the impact of seepage upon native sediments and groundwater resources is not known. The goal of this study is to investigate the role of adsorption and ion exchange reactions in the clay till and their effect on the attenuation or release of inorganic species. This was evaluated u…

Phytotoxicity and naphthenic acid dissipation from oil sands fine tailings treatments planted with the emergent macrophyte Phragmites australis.

During reclamation the water associated with the runoff or groundwater flushing from dry stackable tailings technologies may become available to the reclaimed environment within an oil sands lease. Here we evaluate the performance of the emergent macrophyte, common reed (Phragmites australis), grown in chemically amended mature fine tailings (MFT) and simulated runoff/seepage water from different MFT drying treatments. The present study also investigated the phytotoxicity of the concentration of oil sands naphthenic acids (NAs) in different MFT drying chemical treatments, in both planted and u…

Field trial of the efficacy of a combination of imidacloprid and permethrin against Tunga penetrans (sand flea, jigger flea) in dogs in Brazil.

In a field trial in Brazil 17 dogs penetrated by females of the jigger flea, Tunga penetrans, were topically treated with a combination of 10% imidacloprid and 50% permethrin (Advantix), while 17 dogs remained untreated. The follow-up controls on days 7, 14, 21 and 28 post-treatment clearly showed that, beginning from day 7, the flea load in treated dogs decreased, so that most of the dogs became free of tungiasis lesions, while in the untreated group the flea load remained high. Since the dogs distribute the flea eggs throughout the village, leading to a high incidence of tungiasis in humans,

Preliminary description of a new entomoparasitic nematode infecting Lutzomyia longipalpis sand fly, the vector of visceral leishmaniasis in the New World.

Phlebotomine sandflies are vectors of important pathogens world-wide, including Leishmania spp. in the Neotropics. Entomoparasites have been described from phlebotomines, including virus, bacteria, protozoa, fungi, nematodes, and mites, some of which are capable of killing the host. In the present study, interference, fluorescence, and scanning electron microscopies were used for the first time to detect and morphologically characterize a new entomoparasite infecting Lutzomyia longipalpis. Several filiform larvae and eggs in different stages were encountered in the abdomen of female and male i…

The identity of Leishmania isolated from sand flies and vertebrate hosts in a major focus of cutaneous leishmaniasis in Baturite, northeastern Brazil.

During a field investigation carried out in Baturite, Brazil from 1989 to 1991, sand flies, sympatric rodents, domestic dogs and humans were surveyed for leishmaniasis. Twenty strains of Leishmania were isolated by in vitro culture from Lutzomyia whitmani, three strains were obtained from Rattus rattus, two strains from dogs, and five strains from humans. The isolates were characterized by isoenzyme electrophoresis by hybridization with kinetoplast DNA-specific probes. All the samples were identified as L. (Viannia) braziliensis. The importance of these results in the dynamics of the Leishmani…

[Quantitative histologic studies of mast cells in the parathyroid gland of sand rats (Psammomys obesus) during the development of a mellitus syndrome].

Mast cells in the parathyroid glands of sand rats with different disturbances of the carbon hydrate tolerance (reference, impaired-glucose-tolerance, and diabetic group) are quantitative-histologically investigated. In the IGT-group (impaired glucose tolerance), mast cells are significantly increased. However, the number of this cells in the diabetic group is significantly decreased. This investigations demonstrate that the mellitus-syndrome is not only a complex neuroendocrine disturbance but also a disturbance of biogenic amines and the organ of the connective tissue.

Enhanced weathering of lizardite-rich sand in seawater: effects of pH and CO₂ pressure on carbonate formation for climate change mitigation.

Enhanced weathering (EW) of serpentinized peridotites is being investigated as a potential approach for ex situ CO₂ sequestration in both coastal and open-ocean environments. Previous studies on serpentine minerals have provided important insights into their geochemical reactivity, highlighting their preferential dissolution behavior and enhanced weathering potential under natural conditions. This study examines the CO₂ sequestration potential of lizardite-rich sand and evaluates the formation of hydrated and carbonated mineral phases under varying CO₂ conditions. Mineralogical analyses indica…

Rapid Oil Sands Extraction Processing Stream Composition Determination by a 19Fluorine Nuclear Magnetic Resonance Spectrometer with Fluorinated Tracers.

A benchtop 19F-NMR spectrometer, coupled with the addition of fluorine-containing tracers (hereafter referred to as the 19F-NMR method), was explored as an alternative to the conventional Dean-Stark method for determining the composition of the oil sands slurry. Trifluorotoluene and sodium trifluoroacetate were identified as oleophilic and hydrophilic tracers, respectively, enabling accurate quantification of bitumen and water in both synthetic and real oil sands samples. Bitumen (0.5-90 wt %) and water (5-95 wt %) were accurately measured, demonstrating the method's suitability for a broad ra…

Probing the mechanisms of Fe(III)/As(V) reduction and As mobilisation using mineral-coated sands; impact of electron donor treatments.

The release of geogenic arsenic into groundwater, driven by reductive dissolution of Fe(III)/As(V) oxide phases, poses a severe health risk to millions in South and Southeast Asia. However, the microbes and electron donors responsible for the reductive dissolution remain unclear, due to complex a(biotic) interactions in sediments (traditionally used in microcosm incubation studies). In this study, indigenous microbial communities were sampled from arsenic-prone aquifers in Kandal Province, Cambodia, by filtering groundwater through sands coated with Fe(III)/As(V) minerals. This provided a stre…
