Structural Evolution of Paramagnetic Lanthanide Compounds in Solution Compared to Time- and Ensemble-Average Structures.

Anisotropy in the magnetic susceptibility strongly influences the paramagnetic shifts seen in nuclear magnetic resonance (NMR) and magnetic resonance imaging (MRI) experiments. A previous study on a series of C3-symmetric prototype MRI contrast agents showed that their magnetic anisotropy was highly sensitive to changes in molecular geometry and concluded that changes in the average angle between the lanthanide-oxygen (Ln-O) bonds and the molecular C3 axis due to solvent interactions had a significant impact on the magnetic anisotropy and, consequently, the paramagnetic shift. However, this st

Characterisation of circulating endothelial microparticles in Behçet's disease: new markers of chronic endothelial damage?

Multiple sclerosis (MS) has been recognised as a disease since the mid-19th century. The delineation of its CNS pathology, revealing the presence of inflammatory demyelination and relative sparing of axons, was originally interpreted as evidence of infection. Despite many studies, a primary infectious aetiology of MS has not been found. However, the occurrence of acute demyelinating disease following a variety of infections and vaccinations, leading to MS in about a third of cases, provides evidence for the existence of an auto-allergic pathogenesis for the disease. Improved understanding of t

A new EGFR and c-Met bispecific NIR-II fluorescent probe for visualising colorectal cancer and metastatic lymph nodes.

BACKGROUND: The aim of the study was to increase the specificity and targeting of tumour imaging, targeting molecules that enable the simultaneous recognition and binding of multiple tumour-associated receptors. We constructed a NIR-II fluorescence probe based on a bispecific antibody to epidermal growth factor receptor (EGFR) and cellular mesenchymal-epithelial transition factor (c-Met) for visualising colorectal cancers (CRCs) and metastatic lymph nodes. METHODS: The expression of EGFR and c-Met in tumour and metastatic lymph node specimens from patients with CRC was examined using immunohi

Cooperative Membrane Recognition by Lipid-Sensing and Kinase Domains Shapes Membrane-Bound States of AKT.

The PI3K/AKT signaling pathway is triggered by the recruitment of AKT to cellular membranes. Although AKT is a multidomain serine/threonine kinase composed of an N-terminal pleckstrin homology (PH) domain and a C-terminal kinase domain (KD), how these domains cooperate to regulate AKT activation on membranes remains unclear at the molecular level. Here, using molecular dynamics simulations of full-length AKT on phosphatidylinositol (3,4,5)-trisphosphate (PIP3)-containing lipid bilayers, we identify four distinct membrane-binding modes that differ in the orientations and membrane contacts of th

Gene mutation screening using single-molecule force spectroscopy: a novel way of PCR-independent genetic testing.

Detection of systemic pathologies during their subclinical phases remains a global challenge, as current diagnostics often fail to correctly identify diseases in asymptomatic individuals. While conventional radiological imaging and histopathological tissue biopsies are considered clinical benchmarks, they are frequently hindered by ionizing radiation risks, invasiveness, and the inability to account for intratumoral heterogeneity. Consequently, the diagnostic landscape is shifting toward liquid biopsy, with salivary diagnostics emerging as a promising, non-invasive alternative that facilitates

Seeing the unseen: Eiichi Nakamura on chemistry, life, and a new chapter in China.

Walking into the meeting room for the interview with National Science Review (NSR), Prof. Eiichi Nakamura () first played us a documentary about himself that ran over 20 minutes. The footage was, of course, related to chemistry, but it was far more about art and life: his passion for the Baroque flute, his small concerts with his wife and friends, his travel footprints spanning the globe since the age of 20, and his friendships with numerous fellow scientists. Over the 2-hour interview that followed, the 75-year-old professor left a profound impression on us-romantic yet rigorous, restrained y

Multimodal sensing platform based on MnO2 nanozyme and carbon dots for colorimetric and fluorescent detection of peroxynitrite.

Peroxynitrite (ONOO-), as a crucial bioactive molecule, is closely associated with various diseases when present at abnormal levels, making reliable and accurate detection urgently needed. This study developed a colorimetric and fluorometric multi-mode sensing platform based on manganese dioxide nanosheets (MnO2 NSs) and carbon dots (CDs) for the quantitative detection of ONOO-. Under acidic conditions, MnO2 NSs oxidized 3,3',5,5'-tetramethylbenzidine (TMB) to a blue oxidized product (TMB+), which was subsequently further oxidized by ONOO- to the diimine form (oxTMB). The resulting oxTMB effec

Randomized Cross-Over Study to Examine Associations Between Dialyzer Clotting, Uremic Toxin Removal, and Hemocompatibility During Post-Dilution Hemodiafiltration With Reduced Anticoagulation.

To date, it remains unclear to what extent dialyzer performance is affected by clotting, and which hemocompatibility parameters can reliably predict this phenomenon. This study investigated the relationship between dialyzer clotting, performance, and hemocompatibility in four commercially available dialyzers under conditions of intentionally reduced anticoagulation. This crossover study included 10 chronic hemodialysis patients, who were randomized over four dialyzers: FX CorAL800, FX CorDiax800, xevonta Hi20 (all polysulfone-based), and Solacea-19H (cellulose-based). Patients received a singl

Role of Hydrogen Sulfide Regulation of Programmed Cell Death: Implications for Cardiovascular Diseases.

Significance: Hydrogen sulfide (H2S) is an important signaling molecule involved in cardiovascular diseases (CVDs). Although it is important, the precise mechanisms underlying the diverse functions of H2S in CVDs are not known and need to be elucidated. Recent Advances: Studies have shown the importance of different programmed cell death (PCD) modalities, such as NETosis, apoptosis, necroptosis, pyroptosis, ferroptosis, and cuproptosis, in the pathogenesis of CVDs. An overview of the role of H2S in regulating PCD in diabetic cardiomyopathy (DCM), cardiac hypertrophy and fibrosis, hypertension,

Single-cell RNA sequencing reveals the mechanism of sonodynamic therapy combined with a RAS inhibitor in the setting of hepatocellular carcinoma.

BACKGROUND: Ras activation is a frequent event in hepatocellular carcinoma (HCC). Combining a RAS inhibitor with traditional clinical therapeutics might be hampered by a variety of side effects, thus hindering further clinical translation. Herein, we report on integrating an IR820 nanocapsule-augmented sonodynamic therapy (SDT) with the RAS inhibitor farnesyl-thiosalicylic acid (FTS). Using cellular and tumor models, we demonstrate that combined nanocapsule-augmented SDT with FTS induces an anti-tumor effect, which not only inhibits tumor progression, and enables fluorescence imaging. Pathological Mechanism of a Constitutively Active Form of Stromal Interaction Molecule 1 in Skeletal Muscle.

Stromal interaction molecule 1 (STIM1) is the main protein that, along with Orai1, mediates store-operated Ca2+ entry (SOCE) in skeletal muscle. Abnormal SOCE due to mutations in STIM1 is one of the causes of human skeletal muscle diseases. STIM1-R304Q (a constitutively active form of STIM1) has been found in human patients with skeletal muscle phenotypes such as muscle weakness, myalgia, muscle stiffness, and contracture. However, the pathological mechanism(s) of STIM1-R304Q in skeletal muscle have not been well studied. To examine the pathological mechanism(s) of STIM1-R304Q in skeletal musc

A phase I study of the VEGFR kinase inhibitor vatalanib in combination with the mTOR inhibitor, everolimus, in patients with advanced solid tumors.

Purpose Combining small-molecule inhibitors of different targets was shown to be synergistic in preclinical studies. Testing this concept in clinical trials is, however, daunting due to challenges in toxicity management and efficacy assessment. This study attempted to evaluate the safety and efficacy of vatalanib plus everolimus in patients with advanced solid tumors and explore the utility of dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) studies as a predictive biomarker. Patients and Methods This single-center, phase I trial containing 70 evaluable patients consisted of a do

Nanoscale reorganization of sarcoplasmic reticulum in pressure-overload cardiac hypertrophy visualized by dSTORM.

Pathological cardiac hypertrophy is a debilitating condition characterized by deleterious thickening of the myocardium, dysregulated Ca2+ signaling within cardiomyocytes, and contractile dysfunction. Importantly, the nanoscale organization, localization, and patterns of expression of critical Ca2+ handling regulators including dihydropyridine receptor (DHPR), ryanodine receptor 2 (RyR2), phospholamban (PLN), and sarco/endoplasmic reticulum Ca2+-ATPase 2A (SERCA2A) remain poorly understood, especially during pathological hypertrophy disease progression. In the current study, we induced cardiac

Highly Specific and Sensitive Fluorescent Nanoprobes for Image-Guided Resection of Sub-Millimeter Peritoneal Tumors.

A current challenge in the treatment of peritoneal carcinomatosis is the inability to detect, visualize, and resect small or microscopic tumors of pancreatic, ovarian, or mesothelial origin. In these diseases, the completeness of primary tumor resection is directly correlated with patient survival, and hence, identifying small sub-millimeter tumors (i.e., disseminated disease) is critical. Thus, new imaging techniques and probes are needed to improve cytoreductive surgery and patient outcomes. Highly fluorescent rhodamine-labeled expansile nanoparticles (HFR-eNPs) are described for use as a vi

Plasma Biomarkers of Inflammation Reflect Seizures and Hemorrhagic Activity of Cerebral Cavernous Malformations.

The clinical course of cerebral cavernous malformations (CCMs) is highly variable. Based on recent discoveries implicating angiogenic and inflammatory mechanisms, we hypothesized that serum biomarkers might reflect chronic or acute disease activity. This single-site prospective observational cohort study included 85 CCM patients, in whom 24 a priori chosen plasma biomarkers were quantified and analyzed in relation to established clinical and imaging parameters of disease categorization and severity. We subsequently validated the positive correlations in longitudinal follow-up of 49 subjects. P

Delayed emergence of subdiffraction-sized mutant huntingtin fibrils following inclusion body formation.

Aberrant aggregation of improperly folded proteins is the hallmark of several human neurodegenerative disorders, including Huntington's Disease (HD) with autosomal-dominant inheritance. In HD, expansion of the CAG-repeat-encoded polyglutamine (polyQ) stretch beyond ~40 glutamines in huntingtin (Htt) and its N-terminal fragments leads to the formation of large (up to several μm) globular neuronal inclusion bodies (IBs) over time. We report direct observations of aggregating Htt exon 1 in living and fixed cells at enhanced spatial resolution by stimulated emission depletion (STED) microscopy and

Cediranib for metastatic alveolar soft part sarcoma.

PURPOSE: Alveolar soft part sarcoma (ASPS) is a rare, highly vascular tumor, for which no effective standard systemic treatment exists for patients with unresectable disease. Cediranib is a potent, oral small-molecule inhibitor of all three vascular endothelial growth factor receptors (VEGFRs). PATIENTS AND METHODS: We conducted a phase II trial of once-daily cediranib (30 mg) given in 28-day cycles for patients with metastatic, unresectable ASPS to determine the objective response rate (ORR). We also compared gene expression profiles in pre- and post-treatment tumor biopsies and evaluated th

Imaging protein aggregates in the serum and cerebrospinal fluid in Parkinson's disease.

Aggregation of α-synuclein plays a key role in the development of Parkinson's disease. Soluble aggregates are present not only within human brain but also the CSF and blood. Characterizing the aggregates present in these biofluids may provide insights into disease mechanisms and also have potential for aiding diagnosis. We used two optical single-molecule imaging methods called aptamer DNA-PAINT and single-aggregate confocal fluorescence, together with high-resolution atomic force microscopy for specific detection and characterization of individual aggregates with intermolecular β-sheet struct
