Water-soluble vitamins, amino acids, and nontoxic pharmaceutical excipients were studied as solubilizing agents for poorly water-soluble adenine (nucleic acid base), guanosine (nucleoside), and structurally related drugs (acyclovir and triamterene). A method for the quantitation of three metabolites of theophylline, 1-methylxanthine (1 MX), 3-methylxanthine (3MX), and 1,3-dimethyluric acid (13DMU) in human liver microsomes has been developed. The method is based on a simple one-step extraction followed by isocratic, reversed-phase high-performance liquid chromatography with uv detection (detection wavelength: 273 nm). A high performance liquid chromatographic (HPLC) method has been developed for the determination of levomepromazine in human breast milk and serum. The levomepromazine was extracted by a rapid and simple extraction method using a Sep-Pak C18 cartridge. Cyanide and formate induce spectral changes in E. coli cytochrome bo which are similar to those induced in bovine heart cytochrome-c oxidase (cytochrome aa3). The influence of lipophilic counter ions on the transport of the hydrophilic drugs pholedrine and bretylium across artificial lipid membranes has been studied using a permeation model system. The transport of both drugs was markedly enhanced by hexylsalicylate and by salts of fatty acids with a maximum at decanoate. The transport of pholedrine was additionally increased by bile salts such as desoxycholate.

The effects on theophylline oxidative metabolism of 3 inhibitors of liver microsome activity--cimetidine, troleandomycin and ketoconazole--were investigated in 6 healthy volunteers. A quantitative clean-up procedure for drugs in plasma and urine in preparation for high-performance liquid chromatographic analysis is discussed. Samples are mixed with zinc sulfate, barium hydroxide, and acetonitrile (or methanol). Resultant solutions are clear and free of visible residue. A quantitative high-pressure liquid chromatographic method that uses a reversed-phase column coupled to UV and fluorescence detectors was developed to determine aspirin and salicylic acid in bulk aspirin and in plain, buffered, and enteric-coated tablets. The aspirin was dissolved, filtered, and injected into the chromatograph. The UV absorbance of aspirin was determined at 254 nm, and the fluorescence of salicylic acid was measured at 425 nm. A simple and rapid method is described for the quantitative determinations of free and conjugated oestrogens in pregnancy urine. The oestrogens are precipitated with ammonium sulphate and freed from non-oestrogenic compounds by solvent extraction. The conjugated oestrogens are hydrolysed by a beta-glucuronidase from Escherichia coli, and the total free oestrogens are extracted into ether and their fluorescence intensity at 310 nm in this solvent is determined.
