Rodent models of depression: forced swim and tail suspension behavioral despair tests in rats and mice.

The development of antidepressants requires simple rodent behavioral tests for initial screening before undertaking more complex preclinical tests and clinical evaluation. Presented in the unit are two widely used screening tests used for antidepressants, the forced swim (also termed behavioral despair) test in the rat and mouse, and the tail suspension test in the mouse. These tests have good predictive validity and allow rapid and economical detection of substances with potential antidepressant-like activity. The behavioral despair and the tail suspension tests are based on the same principl

Role of potassium channels in the antidepressant-like effect of folic acid in the forced swimming test in mice.

Potassium (K(+)) channels have been implicated in depressive disorders and in the mechanism of action of antidepressants. Considering that several studies have indicated that folic acid plays an important role in the pathophysiology of depression, the present study investigated the involvement of potassium channels in the antidepressant-like effect of this vitamin. For this aim, the effect of the combined administration of different types of K(+) channel blockers and folic acid in the forced swimming test (FST) was investigated. Treatment of mice by intracerebroventricular (i.c.v.) route with …

Automated experimental system capturing three behavioral components during murine forced swim test.

AIMS: An automated experimental system applying a commercially available video image analyzer was developed for the simultaneous detection and measurement of three behavioral components; immobility, swimming (horizontal movements) and climbing (vertical movements) that occur in the murine forced swim test (FST). The system was validated using four typical antidepressants. MAIN METHODS: System validity was confirmed by demonstrating no significant difference in 6 min time course of control group and imipramine-dosed group (30 mg/kg) between manual examinations and automated digital analysis fo…

Delta(9)-tetrahydrocannabinol enhances an increase of plasma corticosterone levels induced by forced swim-stress.

The present study was designed to determine the effect of delta(9)-tetrahydrocannabinol (THC) on susceptibility to stress. We reported that THC significantly prolonged the immobility time during the forced swim-stress. The selective cannabinoid CB(1) receptor antagonist O-2050 significantly reduced the enhancement of immobility by THC. We investigated the effect of THC on levels of stress hormone corticosterone under non-stress and forced swim-stress conditions. THC did not affect plasma corticosterone levels under non-stress conditions. However, THC, together with forced swim-stress, signific

Calcium metabolism in bone and teeth of rats during exposure to restriction of motor activity and to swimming exercise.

The effects of motor activity restriction for 90 days (hypokinesia, HK) and swimming training (T) on calcium metabolism in rat bones and teeth were evaluated. Male Wistar rats were distributed in four groups: untrained vivarium control rats (UVCR), untrained hypokinetic rats (UHKR), trained hypokinetic rats (THKR) and trained vivarium control rats (TVCR). Hypokinesia was obtained keeping the animals for 90 days in small individual cages which restricted their movements in all directions without hindering food and water intakes. Rats of THKR and TVCR were forced to swim for 15 to 90 minutes eve…

Benzodiazepine prevention of swim stress-induced sensitization of cortical biogenic amines: an in vivo microdialysis study.

In vivo microdialysis was used to determine the effect of diazepam, flumazenil and FG-7142 upon the biogenic amine response to acute and repeated swim stress in the medial prefrontal cortex of the rat. Acute swim stress increased norepinephrine levels, although dopamine and serotonin levels remained stable. Upon re-exposure to swim stress twenty-four hours later, sustained increases (200-300% of baseline) in all three biogenic amines were detected. This enhanced response to re-stress was not seen in rats pretreated with either a benzodiazepine: agonist (diazepam, 2 mg/kg), an antagonist (fluma

The effect of agonists at the GABA-benzodiazepine receptor complex on the duration of immobility of mice in the forced swimming test.

In the present study, we examined the effect of various agents which affect in a different manner the GABA-benzodiazepine receptor-chloride ionophore complex system in relation to the immobile behavior of mice in the forced swimming test. The benzodiazepines diazepam and flurazepam, the barbiturates pentobarbital and phenobarbital, zopiclone and beta-CCP (propyl-beta-carboline-3-carboxylate) enhanced the immobile behavior in a dose-dependent manner. In the doses used here, these agents produced almost no muscle relaxant action. Ro15-1788 and beta-CCM (methyl-beta-carboline-3-carboxylate) thems…

Cholinergic receptor blockade in the rat impairs locale but not taxon strategies for place navigation in a swimming pool.

Normal rats and rats drugged with atropine sulfate, a cholinergic muscarinic blocker, were evaluated in the Morris water task for their use of spatial navigation strategies. Atropine-treated rats were impaired on a place response of swimming to a platform hidden in a pool filled with opaque water. With extended training, they did learn the place response, though not with control precision, as shown by better than chance heading angles upon swim initiation, searches in the old location for a platform that had been moved, and by correct swims from novel starting locations. Acquisition could not …

Animove: a low-cost, easy-to-use, fast and high-resolution system to evaluate swimming performance in zebrafish.

BACKGROUND AND OBJECTIVES: Imaging devices are increasingly present in daily life and are becoming widely used in the record of animal experiments. It is easy to register movement and change in position with video, which is the basis of several methodologies. Yet most of this data is either only qualitatively analysed or their quantitative analysis is based on expensive, proprietary systems, depending on specialized devices and closed software. Here, we present a novel, low-cost system for quantifying swimming performance in both adult and larval zebrafish (Danio rerio), using smartphones for …

Effects of Swimming at Different Water Temperatures on Muscle and Adipose Tissue Adaptation in Diet-Induced Obese Mice.

BACKGROUND: Obesity is a major global health concern, leading to increased risk of metabolic diseases and mortality. Swimming, as a low-impact exercise, may provide metabolic benefits. However, the influence of water temperature on the metabolic and physiological responses remains underexplored. This study investigated the effects of water temperatures on muscle and adipose tissue adaptations in high-fat diet-induced obese mice. METHODS: Obese mice were subjected to swimming at different water temperatures: 15°C, 25°C, or 32°C. Changes in body and tissue weight, grip strength, exhaustive swim…

Mechanical resonance conditions in insect flapping wing apparatus: insights from flight and swimming of a miniature waspTiphodytes gerriphagus.

Evidence suggests that insects may utilize resonant mechanics during flight to optimize energetic efficiency, though whether this mechanism is universal across all insect species remains debated. Microinsects appear particularly intriguing in this context: they exhibit agility comparable to larger species despite experiencing higher aerodynamic damping forces on their wings. We investigated mechanical resonance dynamics focusing on the miniature waspTiphodytes gerriphagus-a remarkable species capable of both aerial flight and underwater locomotion, using wings in both cases. This dual-mode mob

Dietary DHA limitation did not affect swimming and metabolic performance, but reduced growth in wild European sea bass.

Long-chain polyunsaturated fatty acids (LC PUFA), particularly docosahexaenoic acid (DHA), are essential for cell membrane structure and function, impacting overall fish performance. These molecules, produced primarily by phytoplankton, are transferred up the trophic chain; however, climate change is predicted to modify phytoplankton communities with a cascading effect on the global DHA production and thus availability for consumers such as fish. This study aimed to evaluate the effects of dietary DHA limitation on i) the fatty acid composition in fish tissues ii) somatic growth, swimming perf…

Blood Lactate and Maximal Lactate Accumulation Rate at Three Sprint Swimming Distances in Highly Trained and Elite Swimmers.

We examined the blood lactate response, in terms of the maximal post-exercise concentration (Lamax), time to reach Lamax, and maximal lactate accumulation rate (VLamax), to swimming sprints of 25, 35, and 50 m. A total of 14 highly trained and elite swimmers (8 male and 6 female), aged 14-32, completed the 3 sprints in their specialization stroke with 30 min of passive rest in between. The blood lactate was measured right before and continually (every minute) after each sprint to detect the Lamax. The VLamax, a potential index of anaerobic lactic power, was calculated. The blood lactate concen…

Within-day variation and health risk assessment of trihalomethanes (THMs) in a chlorinated indoor swimming pool in China.

Trihalomethanes (THMs) are the most common species of disinfection by-products (DBPs) in swimming pools and have received widespread attention due to their risk to public health. However, studies examining within-day variation and the carcinogenic health risks from exposure to THMs in indoor swimming pools are limited. Our study aimed to detect the within-day variation of four THMs categories and carcinogenic health risk in indoor swimming pool water in Taiyuan, China, and to examine the correlations between THMs and environmental parameters. Our results showed chloroform (TCM) was the most ab…

The effect of fasting period on swimming performance, blood parameters and stress recovery in Atlantic salmon post smolts.

In this study, Atlantic salmon post smolts (~250 g, ~29 cm) were fasted for four weeks at 12 °C in full strength seawater. During this period, the critical swimming speed (Ucrit) was measured after 1, 2 and 4 weeks of fasting, as well as in a fed control group. Furthermore, blood samples were taken in each treatment group prior to the swim test, at fatigue, and following 3 h and 24 h of subsequent recovery. Four weeks of fasting gradually reduced the condition factor from 1.03 to 0.89. However, the Ucrit remained statistically unaffected at 3.5 body lengths s-1. Exhaustive exercise stress caus…

Regulatory protein HilD stimulates Salmonella Typhimurium invasiveness by promoting smooth swimming via the methyl-accepting chemotaxis protein McpC.

In the enteric pathogen Salmonella enterica serovar Typhimurium, invasion and motility are coordinated by the master regulator HilD, which induces expression of the type III secretion system 1 (T3SS1) and motility genes. Methyl-accepting chemotaxis proteins (MCPs) detect specific ligands and control the direction of the flagellar motor, promoting tumbling and changes in direction (if a repellent is detected) or smooth swimming (in the presence of an attractant). Here, we show that HilD induces smooth swimming by upregulating an uncharacterized MCP (McpC), and this is important for invasion of …
