Cyclooxygenase 2 expression in pterygium.

PURPOSE: Following the recent discovery of an abnormal expression of the p53 gene in the epithelium in pterygium, some researchers felt that pterygium is a tumor rather than a degenerative disease. Ultraviolet (UV) radiation has been reported to be associated with pterygium formation, however the mechanism whereby UV induces uncontrolled proliferation in pterygial cells is unclear. Because cyclooxygenase 2 (COX 2) was reported to exist and play an important role in UV-related cutaneous carcinogenesis, it is logical to suspect that COX 2 existed in pterygium. This study was designed to investig

High resolution Ca/P maps of bone architecture in 3D synchrotron radiation microtomographic images.

The Ca/P ratio was measured in cortical bone samples from the femoral neck and tibia of different animal species, using synchrotron radiation microtomography. Use of a monoenergetic X-ray beam, as provided by the synchrotron facility, generates accurate 3D maps of the linear attenuation coefficient within the sample and hence gives the ability to map different chemical components. Also, by comparing normal and abnormal bones, i.e. osteoporotic (induced by inflammation), changes in the Ca/P ratio brought about by bone diseases can be detected. MicroCT data sets were collected at 20 and 28 keV f

DDB2 gene disruption leads to skin tumors and resistance to apoptosis after exposure to ultraviolet light but not a chemical carcinogen.

Mutations in the human DDB2 gene give rise to xeroderma pigmentosum group E, a disease characterized by increased skin tumorigenesis in response to UV-irradiation. Cell strains derived from xeroderma pigmentosum group E individuals also have enhanced resistance to UV-irradiation due to decreased p53-mediated apoptosis. To further address the precise function(s) of DDB2 and the consequence of non-naturally occurring DDB2 mutations, we generated mice with a disruption of the gene. The mice exhibited significantly enhanced skin carcinogenesis in response to UV-irradiation, and cells from the DDB2

Detection of stable chromosome aberrations by FISH in A-bomb survivors: comparison with previous solid Giemsa staining data on the same 230 individuals.

PURPOSE: To evaluate the relative abilities of the solid Giemsa staining (conventional) and fluorescence in situ hybridization (FISH) methods in the detection of stable chromosome aberrations in the peripheral blood lymphocytes of A-bomb survivors. MATERIALS AND METHODS: Lymphocytes from a total of 230 A-bomb survivors for whom prior chromosome aberration data had been obtained by the conventional method were recently examined afresh using FISH in which chromosomes 1, 2 and 4 were painted with composite probes. RESULTS: It was found that the early use of the solid Giemsa staining method had

The original shaker-with-syndactylism mutation (sy) is a contiguous gene deletion syndrome.

Tests for allelism among mice with four different mutant alleles at the shaker-with-syndactylism locus on mouse Chromosome (Chr) 18 provide evidence that the original radiation-induced mutation, sy, is a deletion including at least two genes associated with distinct phenotypes. Mice homozygous for sy have syndactylous feet and other skeletal malformations, are deaf, and exhibit abnormal behavior characteristic of vestibular dysfunction. Two less severe spontaneous mutations, shown to be allelic with sy, cause syndactylism when homozygous (hence named fused phalanges, sy(fp) and sy(fp-2J)), but

Fasciculation-myokymic activity and prolonged nerve conduction block. A physiopathological relationship in radiation-induced brachial plexopathy.

Fourteen radiation-induced brachial plexus neuropathies in 12 patients suffering from cancer were studied. Neurophysiological evaluation showed a diffuse neurogenic lesion with muscular denervation signs associated with motor and sensory nerve conduction impairment of axonal type in the distal segments of the arm. Somatosensory evoked potentials were frequently abnormal, with absence of N9 in 9 out of the 10 extremities explored. The most characteristic findings were, however, the presence of fasciculation potentials--single and grouped--and myokymic discharges in 78.5% of cases (11 out of 14

Radiation-induced femoral head necrosis.

There are very few cases of radiation-induced femoral head necrosis described in the literature, therefore, this case will add new knowledge and highlights important aspects in the diagnosis and management of this uncommon condition. Our patient was 74 years old and presented with left hip and groin pain for 8 months, with no previous history of trauma or osteoarthritis. However, he had been treated for metastatic prostate cancer, to the pelvis and roof of the left acetabulum, with androgen ablation, and radiotherapy 5 years before presentation. Examination of the left hip revealed painful mov

Luteolin from Gastrodia elata Ameliorates Solar Dermatitis via Inhibition of the MAPK/JUN Signalling Pathway.

Background: Solar dermatitis (SD) is an inflammatory skin disease caused by excessive exposure to ultraviolet (UV) radiation. Gastrodia elata (GE) is a medicinal and edible plant with broad pharmacological activities; however, the polarity distribution of its active components and their specific mechanisms of action in SD remain incompletely understood. Methods: Different polarity fractions of GE-petroleum ether extract (PEE), ethyl acetate extract (EAE), and n-butanol extract (NBAE) were prepared and evaluated in a UVB-induced SD mouse model. We integrated metabolomics, network pharmacology,

Non-erythropoietic tissue-protective peptides derived from erythropoietin: WO2009094172.

Erythropoietin (EPO) is a cytokine with erythropoietic and tissue protective activities. Its action as a tissue protective agent requires, however, high dosage that results in limiting side effects associated with abnormally augmented erythropoiesis. Elimination of the erythropoietic activity of EPO while preserving its tissue protective properties was nevertheless achieved in carbamoylated EPO (CEPO), whose therapeutic activity and apparent safety were documented in experimental models of nervous, heart, kidney and other tissue damage, justifying ongoing clinical trials. Here, we review paten

Molecular mechanisms involved in chemoresistance in paediatric acute lymphoblastic leukaemia.

Acute lymphoblastic leukaemia (ALL) is the most common paediatric cancer. Despite cure rates approaching 80%, resistance to treatment and disease relapse remain a significant clinical problem. Identification of the genes and biological pathways responsible for chemoresistance is therefore crucial for the design of novel therapeutic approaches aiming to improve patient survival. Mutations in the membrane transporter P-glycoprotein genes, genetic variations in drug-metabolising enzymes and defects in apoptotic pathways are mechanisms of chemoresistance common to a wide spectrum of cancers and al

Expression of bcl-2, p53, and p21 in benign and malignant prostatic tissue before and after radiation therapy.

Abnormalities in genes of the apoptotic pathway might contribute to survival in prostatic cancer (PCa) cells after radiation therapy (RT). We investigated the immunohistochemical expression of the products of the p53, p21WAF1, and bcl-2 genes in pre-RT and post-RT biopsy specimens from 38 patients with locally advanced PCa. All of the 38 patients underwent a uniform protocol of RT with or without neoadjuvant hormonal therapy. Immunohistochemical staining for expression of the products of the p53, p21WAF1, and bcl-2 genes was performed on material from pre-RT and post-RT specimens. Sufficient t

Algal and Fungal Antioxidants Alleviate Oxidative Stress-Induced Reproductive Defects.

The intrinsic and extrinsic factors (natural aging, diseases, infections, chemicals, radiation, heavy metals etc.) create oxidative stress, thereby causing reproductive defects in males and females. Similarly, oxidative stress causes the abnormalities in sperm and oocytes, and thus reduces the success of in vitro fertilization. Fungi and/or algae-derived metabolites (polysaccharides, carotenoids, ergothioneine, phycobiliproteins, phlorotannins, and cordycepin) alleviate the damages in ovaries and testis and correct the impaired reproductive functions (spermatogenesis, ovulation, fertilization

Regulation of Mre11/Rad50 by Nbs1: effects on nucleotide-dependent DNA binding and association with ataxia-telangiectasia-like disorder mutant complexes.

The Mre11/Rad50 complex is a critical component of the cellular response to DNA double-strand breaks, in organisms ranging from archaebacteria to humans. In mammalian cells, Mre11/Rad50 (M/R) associates with a third component, Nbs1, that regulates its activities and is targeted by signaling pathways that initiate DNA damage-induced checkpoint responses. Mutations in the genes that encode Nbs1 and Mre11 are responsible for the human radiation sensitivity disorders Nijmegen breakage syndrome (NBS) and ataxia-telangiectasia-like disorder (ATLD), respectively, which are characterized by defective

The Potential Role of Lung Ultrasound B-Lines for Detection of Lung Radio-Induced Toxicity in Breast Cancer Patients after Radiation Therapy.

BACKGROUND AND PURPOSE: Breast cancer patients exposed to doses of radiation after radiotherapy could develop toxicity to lung. Lung ultrasound (LUS) is able to detect interstitial lung disease by the evaluation of B-lines. The aim of our study was to assess the number of B-lines to diagnose lung involvement after chest radiotherapy. MATERIALS AND METHODS: We measured LUS B-lines in the treated and contralateral lung of 20 breast cancer patients, 1-3 months after the end of radiotherapy and 1 year after previous LUS. The sum of the B-lines number in the 72 sites on anterior and posterior ches

Recurrent severe hypercalcemia in a nasopharyngeal carcinoma survivor.

Hypercalcemia is a potentially life-threatening metabolic disorder with diverse etiologies. In oncology patients, it is often attributed to malignancy-related mechanisms, such as parathyroid hormone-related peptide (PTHrP) secretion. However, non-malignant causes, like calcium-alkali syndrome (CAS) and adrenal insufficiency, should not be overlooked. We report a 60-year-old man with nasopharyngeal carcinoma (NPC) treated with cranial radiotherapy, stage 3 chronic kidney disease (CKD), and multiple comorbidities, who presented with recurrent severe hypercalcemia (11.0-14.9 mg/dL) and hyperphosp

Physiological motion of the optic chiasm and its impact on stereotactic radiosurgery dose.

OBJECTIVE: Avoidance of radiation-induced optic neuropathy (RION) from stereotactic radiosurgery (SRS) requires precise anatomical localization; however, no prior studies have characterized the physiologic motion of the optic chiasm. We measured the extent of chiasm motion and its impact on SRS dose. METHODS: In this cross-sectional study, serial MRI was performed in multiple planes in 11 human subjects without optic pathway abnormalities to determine chiasm motion across time. Subsequently, the measured displacement was applied to the hypothetical chiasm dose received in 11 patients treated
