Functional characterization of the adenosine receptor mediating inhibition of intestinal secretion.

1. Previous studies have shown that the mixed A1/A2 adenosine agonist 5'-N-ethylcarboxamido-adenosine (NECA) inhibits intestinal fluid secretion which is thought to contribute to its antidiarrhoeal effect in the rat. The aim of this study was to characterize the adenosine receptor mediating this antisecretory effect via functional studies using a range of selective agonists and antagonists and by applying the pharmacological criteria of relative agonist and antagonist potencies. 2. Adenosine agonists and antagonists were administered i.v. to anaesthetized rats. Intestinal secretion was then st…

Therapeutic and diagnostic utility of adenosine during tachycardia evaluation in children.

Adenosine has become the drug of choice for termination of regular, normal QRS tachycardia. Initial studies in adult and pediatric patients have shown that the drug is effective for tachycardias using the atrioventricular (AV) node as an integral part of the tachycardia circuit and has few serious side effects. Experience with adenosine administration in children was reviewed to examine the diagnostic and therapeutic usefulness, effective dose, and adverse effects of adenosine. Adenosine was administered to 38 children during 50 separate electrophysiologic evaluations. Eleven patients had stru…

Effects of chronic caffeine on adenosine, dopamine and acetylcholine systems in mice.

Chronic ingestion of caffeine by male NIH Swiss strain mice leads in about 3 days to a significant increase in A1-adenosine, nicotinic and muscarinic receptors, and a significant decrease of beta 1-adrenoceptors in cerebral cortical membranes. Plasma levels of caffeine in the chronically treated mice range from 0.70 to 5.7 micrograms/ml. The changes in receptors reverse after withdrawal of caffeine within 7 days. An increase in nitrendipine binding sites, associated with L-type calcium channels, also occurs within 4 days and has reversed in 7 days after withdrawal. There is no change in the le…

[Efficacy and safety of adenosine triphosphate in the control of supraventricular paroxysmal tachycardia].

Our purpose was to assess the efficacy and safety of intravenous ATP for the acute termination of paroxysmal supraventricular tachycardia. There were 14 women and 10 men, aged 38 +/- 15 years. Three patients had evidence of structural heart disease (Ebstein's anomaly associated to atrial septal defect, operated mitral stenosis with insertion of a mechanical heart valve and CAD respectively). Twelve patients had Wolff-Parkinson-White syndrome and another had undergone surgical ablation of an accessory pathway. At the time of electrophysiologic testing, ATP was administered during episodes of pa

[Effectiveness of combined use of verapamil and adenosine triphosphate in experimental animals].

A wide application of ATP to abolish reciprocal paroxysmal tachycardia is limited by frequent and adverse effects of the drug. Experiments on 49 rats have revealed that successive intravenous injections of verapamil and ATP have a more pronounced and prolonged action than the drugs alone. It is assumed that decreased pacemaker activity of sinus nodal cells and reduced atrioventricular conduction ("binodal dysfunction") are as a result of drug calcium channel block in the cellular membranes of myocytes. Potentiation of the pharmacological action of the drugs in question may be used in the clini

Effects of adenosine on electrical activity of isolated guinea pig hearts.

Negative chronotropic and dromotropic effects of adenosine seem to be responsible for its antiarrhythmic action on supraventricular tachyarrhythmias. To further characterize the effects of adenosine on supraventricular arrhythmias heart rate, conduction, refractoriness, the time to steady-state of AV-nodal conduction slowing and of sinus rate reduction were evaluated. Changes of heart rate, conduction intervals and effective refractory periods were determined by the use of a high-resolution ECG recording technique in isolated guinea pig hearts perfused by the method of Langendorff. Adenosine i…

Reduced transport of adenosine in erythrocytes from patients with beta-thalassaemia.

The transport of adenosine into blood from beta-thalassaemia subjects was measured to provide a background to the relationship between resistance of malaria infection and beta-thalassaemia. Adenosine transport was significantly reduced in the abnormal cells in the blood samples. As adenosine is one of the major purines salvaged by P. falciparum malaria, we suggest that the resistance to malaria in beta-thalassaemia subjects may be due to a nutrient deficiency in the abnormal red cells.

Hepatic and renal phosphomonoesterases and adenosine triphosphatases in chronic hyperprolactinaemic rats.

Influence of hyperprolactinaemia, induced endogenously by anterior pituitary transplantation on rat hepatic and renal cortical and medullary phosphomonoesterases and adenosine triphosphatases (ATPases) has been investigated. Although prolactin has a stimulatory effect on phosphomonoesterases and ATPases, it exhibits a specific and temporal influence on each subtype of hepatic and renal ATPases and phosphomonoesterases at different durations of pituitary transplantation. The specific activities of alkaline phosphatase and Na(+)-K+ dependent ATPases are activated in all the regions of different …

Adenosine receptor antagonism accounts for the seizure-prolonging effects of aminophylline.

The mechanism of action of aminophylline in prolonging seizures was tested in amygdala-kindled rats. Aminophylline prolonged the afterdischarge duration of kindled seizures. This seizure-prolonging action of aminophylline was strongly antagonized by the adenosine A1 agonist cyclohexyladenosine and partially antagonized by the benzodiazepine partial agonist RO 15-1788. However, the specific benzodiazepine antagonist CGS 8216 did not affect the seizure-prolonging action of aminophylline. Also, the potent anticonvulsant effect of diazepam on kindled seizures, which was completely antagonized by C…

[Adenosine and methylxanthines in the central nervous system--on their significance for psychiatry and neurology].

In the last 15 years the central effects of adenosine and its importance for neurology and psychiatry have increasingly become an object of investigation. Physiology and metabolism of adenosine have been examined by different research groups. The effects of adenosine in the central nervous system and the various mechanisms of these effects have been described by electrophysiologic methods. The inhibition of the release of several neurotransmitters through adenosine has already been described 10 years ago. In this review studies on binding sites for adenosine receptor ligands are discussed thor…

Effects of some adenosine analogs on morphine-induced analgesia and tolerance.

1. The analogs of adenosine D- and L-phenylisopropyladenosine (D- and L-PIA) and chloroadenosine (CADO) induced analgesia in mice (hot-plate test). 2. The antinociceptive effects of the three adenosine agonists were antagonized by caffeine but were unaffected by naloxone. 3. Morphine-induced antinociception was increased by pretreatment with adenosine agonists. 4. Whereas CADO significantly attenuated the induction of morphine tolerance, D- and L-PIA did not affect the process.

Effect of adenosine on melatonin and norepinephrine release in rat pineal explants.

To study adenosine effect on melatonin production, rat pineal explants were incubated for 6 h with 10(-4) M adenosine or 2-chloroadenosine in the presence or absence of 5 x 10(-5) M norepinephrine (NE). Melatonin content in pineal gland and medium was measured by RIA. Both adenosine and 2-chloroadenosine increased melatonin production rate by 3-4-fold, and augmented NE stimulation by 30-40%. Addition of adenosine deaminase prior to NE reduced stimulated melatonin release by 40-46%. 2-Chloroadenosine counteracted the inhibition of NE response given by adenosine deaminase. Either adenosine or it…

Superoxide generation and its modulation by adenosine in the neutrophils of subjects with asthma.

Airway inflammation with neutrophil infiltration may play a role in airway hyperreactivity. Neutrophils may exert their effects through the generation of superoxide O2- anion and other oxygen-derived free radicals. O2- generation by neutrophils has been demonstrated to be modulated by adenosine at physiologic concentrations. Therefore, we have investigated the function of peripheral blood neutrophils with respect to O2- anion generation and its regulation by adenosine in both subjects with asthma and normal subjects and also the relationship between O2- anion generation and airway hyperrespons…

Prognostic significance of adenosine deaminase determinations in subjects with the lymphoadenopathy syndrome.

The association between human immunodeficiency virus type I (HIV-I) infection and high levels of erythrocyte adenosine deaminase (ADA) has been suggested by Cowan et al [1986]. We have analyzed the specific activities of the same enzyme during different stages of acquired immunodeficiency syndrome (AIDS), including asymptomatic subjects at high risk and patients with lymphoadenopathy syndrome (LAS), AIDS-related complex (ARC), full-blown AIDS, and AIDS encephalopathy (AIDS enc). The ADA activities were significantly higher (P less than .05) in asymptomatic HIV-I serum-positive individuals (13.…

Adenosine: a clinical experience and comparison with verapamil for the termination of supraventricular tachycardias.

The efficacy and side-effects of adenosine for treatment of supraventricular arrhythmias were compared to verapamil therapy in patients presenting to the emergency room. Clinical variables and the time interval from the initiation of treatment to the termination of the supraventricular tachycardia, as well as the time from the initial effective dose of medication to the termination of supraventricular tachycardia were compared for adenosine and verapamil. Adenosine was given to a total of 44 patients, 16 patients in the electrophysiology laboratory, and 28 patients in the emergency room for ev…

Effects of pulmonary oedema on pharmacokinetics of adenosine in rat isolated lungs.

1 Pulmonary oedema, assessed by decreases in the lung dry weight:wet weight ratio, was induced in rats by a single i.p. injection of alpha-naphthylthiourea (ANTU). The oedema reached a peak at 4 h after ANTU and had completely resolved after 28 h. 2 Pulmonary pharmacokinetics of adenosine were measured in isolated, perfused lungs using radiolabelled adenosine and sucrose, injected into the perfusate as a single bolus. 3 By 1 h after ANTU the 1 min efflux of tracer for adenosine increased to over 60% and remained high until 16 h after ANTU. The time for 50% of injected radioactivity to appear i…
