Closing the research-practice gap: factors affecting adoption and implementation of a children's mental health program.

Despite the availability of effective interventions, they are not widely used in community mental health centers. This study examined the adoption and implementation of The Arson Prevention Program for Children (TAPP-C), a program for juvenile firesetters developed at a teaching hospital and disseminated to community settings. Questionnaire data from mental health professionals were used to evaluate the roles of adopter, innovation, and dissemination characteristics in TAPP-C adoption and implementation. Results indicate that different factors are important at different diffusion stages. Moreo…

Discontinuity patterns in stages of the precaution adoption process model: meat consumption during a livestock epidemic.

OBJECTIVES: Patterns of changes in social-cognitive variables were investigated in order to test selected stages of the precaution adoption process model (PAPM). It was hypothesized that non-linear trends (discontinuity patterns) in perceived vulnerability, positive and negative outcome expectancies, procrastination, and self-efficacy might be observed across the stages. DESIGN: Cross-sectional data from 808 respondents were employed. METHOD: Questionnaire data were collected in an on-line study on meat consumption during a livestock epidemic in Germany. Polynomial trends and analyses of var…

A description of the adoption of the 'Fresh start' smoking cessation program by antenatal clinic managers.

This paper reports the dissemination by the Cancer Education Research Program (CERP) of a previously tested smoking cessation program called 'Fresh start' to 23 antenatal clinics. The program was specifically designed for use by staff in antenatal clinics. The aim of the study was to investigate the factors that influenced midwifery managers' adoption of the program. Clinics were randomly assigned to groups that received the program by simple dissemination (mail-out), or intensive dissemination (a mail-out, plus personal contact with midwifery facilitators). A case history approach was used to…

Why CD4+ T cells matter in adoptive cell therapy for solid cancers.

Cytotoxic CD8+ T cells have been the focus of research efforts to improve the efficacy of anticancer adoptive cell therapies. However, emerging evidence is highlighting the critical role of CD4+ T cells in these treatments, including chimeric antigen receptor-T cells. Although the presence of CD4+ T cells is known to be important for efficacy of cell therapies, the mechanisms by which CD4+ chimeric antigen receptor-T cells exert their beneficial effects remain incompletely understood. In this article, we review the roles of CD4+ T cells in adoptive cell therapy, highlighting understudied areas

Microtubules in the axon are GDP bound but adopt a stable GTP-like expanded state.

Microtubules scaffold cells, supporting signaling and cargo transport. They assemble from GTP-tubulin, which hydrolyzes to GDP-tubulin during polymerization. GTP-microtubule lattices are stable; GDP lattices depolymerize rapidly. In vitro, hydrolysis triggers lattice compaction. Lattice spacing regulates motors and microtubule-associated proteins; however, the conformation of tubulin in microtubules in cells is unknown. Here, we present the atomic-resolution cryo-electron microscopy structure of human microtubules in situ, in the axons of human cortical neurons derived from induced pluripotent

Targeting metabolic reprogramming to enhance adoptive immunotherapy: emerging mechanisms and translational perspectives.

BACKGROUND: Adoptive cell therapy (ACT) has achieved breakthrough advances in the treatment of malignancies, demonstrating particularly remarkable efficacy in hematologic cancers. However, the therapeutic efficacy of ACT remains limited by multiple factors, including low tumor antigen expression, the limited in vivo persistence of infused immune cells, and the immunosuppressive characteristics of the tumor microenvironment (TME). Meanwhile, metabolic reprogramming, a hallmark of cancer, plays a pivotal role in immune cells by directly regulating their differentiation, functional states, and lo

Nano-bionic bacteria armed adoptive macrophage therapy against bladder cancer.

Bladder cancer is a highly malignant tumor with limited treatment options. Although adoptive cell therapy has shown promise in oncology, its efficacy is often constrained by poor intrinsic antitumor activity and a highly immunosuppressive tumor microenvironment. To address these challenges, this study develops a novel living adoptive macrophage therapy armed with nano-bionic bacteria for bladder cancer treatment. Specifically, hollow manganese dioxide (MnO2) nanoparticles were synthesized, loaded with an adenosine inhibitor, and coated with E. coli membranes. Under mild conditions, these nano-

Adoptive transfer of ILC2s reveals tumor homing in glioblastoma: a proof-of-concept study.

INTRODUCTION: Glioblastoma (GBM) is an aggressive brain tumor with limited treatment options and poor immune cell infiltration. Although cellular immunotherapies have transformed cancer treatment, they remain largely ineffective against GBM due to the restrictive blood-brain barrier (BBB) and a profoundly immunosuppressive tumor microenvironment. Innate lymphoid cells type 2 (ILC2s) have recently emerged as potential candidates for immune-based approaches because of their regenerative and immunomodulatory functions. METHODS: Bone marrow-derived ILC2s from C57BL/6 mice were fluorescently label

Salvage Metastasectomy After Adoptive Cell Transfer for Melanoma: Long-Term Updates.

BACKGROUND: Adoptive cell transfer (ACT) can mediate durable regression in patients with metastatic melanoma, and some patients with initial response may progress, requiring salvage therapy. METHODS: We report updated follow-up on a retrospective review of patients with metastatic melanoma receiving ACT at a single institution from 2000 to 2022. We identified patients with objective response (RECIST 1.0) or stable disease for at least 6 months (SD6) before experiencing oligoprogression in up to three extracranial sites managed with surgery. The primary outcomes were progression-free survival

Making it rain: Adopting soil science techniques for studies of land-water interactions.

Use of buffer zones or intact riparian areas is common practice to mitigate nutrient losses from agricultural areas that can impair water quality. However, much of the work assessing the impact of these conservation practices comes from watershed or regional-scale analyses linking water quality to land-use/land-cover metrics. We offer a finer-scale approach that utilizes rainfall simulators, a common soil science technique to assess sediment and nutrient loss in agricultural and developed land, to test the efficacy of riparian buffer strips to minimize nutrient or material losses. We replicate

Adoptive Cell Transfer of Tumor-Infiltrating Lymphocytes for Metastatic Acral Lentiginous Melanoma.

PURPOSE: Acral lentiginous melanoma is a subtype of cutaneous melanoma arising from palmar, plantar, or subungual skin. These tumors are characterized by aggressive biology, a low tumor mutational burden (TMB), and diminished sensitivity to immune checkpoint blockade. It is unknown whether adoptive cell transfer of tumor-infiltrating lymphocytes (ACT-TIL) has efficacy in patients with acral melanoma. METHODS: We analyzed prospectively collected data from 442 patients with metastatic cutaneous melanoma who were treated on clinical trials of ACT-TIL at a single institution between 1999 and 2018

Coping strategies adopted when faced with stress and eating disorders: A systematic review.

Eating disorders (EDs) are a group of disorders characterized by altered eating behaviors and/or the emergence of behaviors pertaining to weight control. Some studies report a relationship between coping strategies adopted in the face of stress and EDs. However, the latest review on this topic was carried out in 2008 by Quiles & Terol. A systematic review of studies that have investigated the relationship between coping strategies and EDs in clinical (anorexia nervosa, bulimia nervosa, or binge eating disorder) and non-clinical populations, aged 12-25 years has been conducted. Optimization of In Vitro Th17 Polarization for Adoptive Cell Therapy in Chronic Lymphocytic Leukemia.

Although preclinical investigations have shown notable efficacy in solid tumor models utilizing in vitro-differentiated Th17 cells for adoptive cell therapy (ACT), the potential benefits of this strategy in enhancing ACT efficacy in hematological malignancies, such as chronic lymphocytic leukemia (CLL), remain unexplored. CLL is a B-cell malignancy with a clinical challenge of increased resistance to targeted therapies. T-cell therapies, including chimeric antigen receptor (CAR) T cells, have demonstrated limited success in CLL, which is attributed to CLL-mediated T-cell dysfunction and skewin
