Identification of sibling species status of Anopheles culicifacies breeding in polluted water bodies in Trincomalee district of Sri Lanka.

BACKGROUND: Anopheles culicifacies s.l., the major vector of malaria in Sri Lanka, is known to breed in clean and clear water. However, recent findings have confirmed breeding from waste water bodies in urban and semi-urban areas. No study has been conducted to identify whether it is vector or non-vector siblings. The objective of the study was to identify the sibling species status of An. culicifacies s.l. METHODS: Anopheles culicifacies s.l. adult samples (reared from larvae) were obtained from the Padavisiripura Entomological team attached to Tropical and Environmental Diseases and Health …

Establishment of a free-mating, long-standing and highly productive laboratory colony of Anopheles darlingi from the Peruvian Amazon.

BACKGROUND: Anopheles darlingi is the main malaria vector in the Amazon region and is among the most efficient malaria vectors worldwide. However, due to the lack of a well-established laboratory colony, key control-relevant aspects of the bionomics, behaviour, genetics, and vector-parasite relationships of An. darlingi remain unknown. Here, biological parameters that had been successful in initiating other Anopheles colonies were optimized and improved for An. darlingi, with the aim of establish a free-mating, stable, and highly productive laboratory colony. METHODS: Wild An. darlingi adult …

Environmental management through sluice gated bed-dam: a revived strategy for the control of Anopheles fluviatilis breeding in streams.

BACKGROUND & OBJECTIVES: Integrated vector management (IVM) emphasizes sustainable eco-friendly methods and minimal use of chemicals. In this context, the present study highlights the environmental control of breeding of Anopheles fluviatilis, the primary malaria vector, through water management in a natural stream in Koraput district, Odisha, India. METHODS: The District Rural Development Agency (DRDA), Koraput, constructed two bed-dams across streams, one in Barigaon and the other in Pipalapodar village. The bed-dam in the former village was fitted with two sluice gates whereas the bed dam …

Molecular evidence for historical presence of knock-down resistance in Anopheles albimanus, a key malaria vector in Latin America.

BACKGROUND: Anopheles albimanus is a key malaria vector in the northern neotropics. Current vector control measures in the region are based on mass distributions of long-lasting insecticidal nets (LLINs) and focal indoor residual spraying (IRS) with pyrethroids. Resistance to pyrethroid insecticides can be mediated by increased esterase and/or multi-function oxidase activity and/or mutations in the voltage-gated sodium channel gene. The aim of this work was to characterize the homologous kdr region of the voltage-gated sodium channel gene in An. albimanus and to conduct a preliminary retrospec…

Multiplex assay for species identification and monitoring of insecticide resistance in Anopheles punctulatus group populations of Papua New Guinea.

Anopheles punctulatus sibling species (An. punctulatus s.s., Anopheles koliensis, and Anopheles farauti species complex [eight cryptic species]) are principal vectors of malaria and filariasis in the Southwest Pacific. Given significant effort to reduce malaria and filariasis transmission through insecticide-treated net distribution in the region, effective strategies to monitor evolution of insecticide resistance among An. punctulatus sibling species is essential. Mutations in the voltage-gated sodium channel (VGSC) gene have been associated with knock-down resistance (kdr) to pyrethroids and…

The dominant Anopheles vectors of human malaria in the Asia-Pacific region: occurrence data, distribution maps and bionomic précis.

BACKGROUND: The final article in a series of three publications examining the global distribution of 41 dominant vector species (DVS) of malaria is presented here. The first publication examined the DVS from the Americas, with the second covering those species present in Africa, Europe and the Middle East. Here we discuss the 19 DVS of the Asian-Pacific region. This region experiences a high diversity of vector species, many occurring sympatrically, which, combined with the occurrence of a high number of species complexes and suspected species complexes, and behavioural plasticity of many of t

Molecular evidence for the occurrence of a new sibling species within the Anopheles (Kerteszia) cruzii complex in south-east Brazil.

BACKGROUND: Anopheles cruzii (Diptera: Culicidae) has long been known as a vector of human and simian malaria parasites in southern and south-eastern Brazil. Previous studies have provided evidence that An. cruzii is a species complex, but the status of the different populations and the number of sibling species remains unclear. A recent analysis of the genetic differentiation of the timeless gene among An. cruzii populations from south and south-east Brazil has suggested that the population from Itatiaia, Rio de Janeiro State (south-east Brazil), is in a process of incipient speciation. METH…

Recombinant expression and biochemical characterization of the catalytic domain of acetylcholinesterase-1 from the African malaria mosquito, Anopheles gambiae.

Acetylcholinesterases (AChEs) and their genes from susceptible and resistant insects have been extensively studied to understand the molecular basis of target site insensitivity. Due to the existence of other resistance mechanisms, however, it can be problematic to correlate directly a mutation with the resistant phenotype. An alternative approach involves recombinant expression and characterization of highly purified wild-type and mutant AChEs, which serves as a reliable platform for studying structure-function relationships. We expressed the catalytic domain of Anopheles gambiae AChE1 (r-AgA…

Larvivorous fish in wells target the malaria vector sibling species of the Anopheles culicifacies complex in villages in Karnataka, India.

Malaria was a major problem in a sericulture area of Karnataka, south India, where Anopheles culicifacies s.l. and A. fluviatilis s.l. were considered to be the main vectors. Sibling species complexes of these two species were analysed in three ecologically different villages. Among A. culicifacies, only sibling species A and B were found. In Puram, a village with 22 wells, species A predominated; species B predominated in a village with four wells and a stream, and in a village with a stream and no wells. Poecilia reticulata fish were introduced into all wells and streams in the villages, and…

A single multiplex assay to identify major malaria vectors within the African Anopheles funestus and the Oriental An. minimus groups.

The African Anopheles funestus and the Oriental An. minimus groups are closely related and composed of major malaria vectors in Africa and Southeast Asia, respectively. None of the species of either the An. funestus or the An. minimus group can be identified with absolute certainty using the adult morphology. Polymorphisms present on the internal transcribed spacer 2 (ITS2) of ribosomal DNA allowed the development of 10 primers that combined with an universal forward primer lead to a simple and sensitive multiplex allele-specific polymerase chain reaction (AS-PCR). Moreover, the possible addit…

Reflections on the Anopheles gambiae genome sequence, transgenic mosquitoes and the prospect for controlling malaria and other vector borne diseases.

The completion of the Anopheles gambiae Giles genome sequencing project is a milestone toward developing more effective strategies in reducing the impact of malaria and other vector borne diseases. The successes in developing transgenic approaches using mosquitoes have provided another essential new tool for further progress in basic vector genetics and the goal of disease control. The use of transgenic approaches to develop refractory mosquitoes is also possible. The ability to use genome sequence to identify genes, and transgenic approaches to construct refractory mosquitoes, has provided th…

A comparative genomic analysis of two distant diptera, the fruit fly, Drosophila melanogaster, and the malaria mosquito, Anopheles gambiae.

Genome evolution entails changes in the DNA sequence of genes and intergenic regions, changes in gene numbers, and also changes in gene order along the chromosomes. Genes are reshuffled by chromosomal rearrangements such as deletions/insertions, inversions, translocations, and transpositions. Here we report a comparative study of genome organization in the main African malaria vector, Anopheles gambiae, relative to the recently determined sequence of the Drosophila melanogaster genome. The ancestral lines of these two dipteran insects are thought to have separated approximately 250 Myr, a long…

Effect of anti-mosquito hemolymph antibodies on fecundity and on the infectivity of malarial parasite Plasmodium vivax to Anopheles stephensi (Diptera:Insecta).

Rabbit antibodies to hemolymph antigens (102.5, 101, 100, 96, 88, 80, 64, 55, 43, 29, and 23 kDa) of Anopheles stephensi reduced fecundity as well as viability in An. stephensi. However, ingestion of these antibodies was not associated with a marked effect on the engorgement of mosquitoes but egg laying was significantly delayed. Antisera raised against hemolymph proteins were also used to identify cross reactive antigens/epitopes present in other tissues by Western blotting, as well as by in vivo ELISA. In addition, a significant reduction in oocyst development was also observed in An. stephe…

Vectorial capacity and entomological inoculation rates of Anopheles gambiae in a high rainfall forested area of southern Sierra Leone.

We report the first study of gonotrophic cycle duration, survival rates, pre-gravid rates, vectorial capacity and chromosomal polymorphism of Anopheles gambiae s.s. in Sierra Leone. In the village of Bayama in the Southern Province, An. gambiae was the only species found to be naturally infected with Plasmodium falciparum and it constituted 99.7% of 22,541 anopheline mosquitoes caught. Chromosomal studies revealed only An. gambiae s.s. out of 66 females examined for chromosomal polymorphism, 61 (92.4%) had the 2LA inversion in the standard arrangement. Other inversions observed in low frequenc…

Efficacy of Bacillus sphaericus against the malaria vector Anopheles gambiae and other mosquitoes in swamps and rice fields in Zaire.

The microbial control of Anopheles gambiae and other mosquitoes with a granular formulation of Bacillus sphaericus (Vectolex) was evaluated in rice fields and swamps, located around the suburban region of Kingabwa-village in Kinshasa, Zaíre. Ten treatment cycles with 15-day intervals were carried out with the same application rate, 10 kg/ha, during the dry season (May to September 1991). The treatments reduced larval populations of An. gambiae by 98% after 48 h, but repetitive applications were required every 15 days to maintain control. The persistence of B. sphaericus spores was more apparen…

Prioritizing urban water-storage containers for molecular surveillance of invasive Anopheles stephensi in Isiolo, Kenya: An evidence-informed opinion.

Anopheles stephensi is an invasive urban malaria vector whose expansion in the Horn of Africa has raised concern because of its ability to exploit human-made water-holding habitats. In Kenya, molecular surveillance first confirmed the presence of An. stephensi in Marsabit and Turkana counties in 2022, and subsequent nationwide work documented spread into additional counties including Isiolo. Published Kenyan field data show larvae in dumped tires, plastic containers, water-storage tanks, concrete cisterns, spillover around community water points, and water pits at mining sites. Although no hab…

Anopheles mosquitoes exposed to long-acting antimalarials via drug-supplemented bloodmeal absorb drug but do not suffer fitness costs.

BACKGROUND: The World Health Organization's recommendations for antimalarial chemoprevention and mass treatment use in endemic areas have greatly expanded, allowing more flexibility in demographic groups and regions targeted. An overlooked aspect of expanding human population-level drug exposure is the downstream impact of drug on the vector. Anopheles mosquitoes re-feed often with ≥4 bloodmeals during their lifespan. This provides repeated opportunities for mosquitoes to ingest drug via bloodmeals taken from people with antimalarials in the bloodstream and raises questions as to drug impact o…
