A Deletion in the Canine POMC Gene Is Associated with Weight and Appetite in Obesity-Prone Labrador Retriever Dogs.

Sequencing of candidate genes for obesity in Labrador retriever dogs identified a 14 bp deletion in pro-opiomelanocortin (POMC) with an allele frequency of 12%. The deletion disrupts the β-MSH and β-endorphin coding sequences and is associated with body weight (per allele effect of 0.33 SD), adiposity, and greater food motivation. Among other dog breeds, the deletion was only found in the closely related flat-coat retriever (FCR), where it is similarly associated with body weight and food motivation. The mutation is significantly more common in Labrador retrievers selected to become assistance

Inhibition of eating behavior: negative cognitive effects of dieting.

This study tested the hypothesis that dieters would score higher than nondieters in terms of food rumination. Two hundred and thirty one col … ility. These results are consistent with previous theory and research suggesting that inhibition of appetitive behaviors can have negative cognitive effects. Moreover, they indicate a potential target for therapeutic intervention.

Ghrelin receptor antagonism of morphine-induced accumbens dopamine release and behavioral stimulation in rats.

RATIONALE AND OBJECTIVES: Ghrelin, an orexigenic (appetite stimulating) peptide activates binding sites in the ventral tegmental area (a structure linked with the neural reward system) allowing it to participate in reward-seeking behavior. An increasing number of studies over the past few years have demonstrated ghrelin's role in alcohol, cocaine, and nicotine abuse. However, the role of ghrelin, in opioid effects, has rarely been examined. The aim of the present study was to ascertain whether a ghrelin antagonist (JMV2959) was able to inhibit markers of morphine-induced activation of the neur

A rodent model of enhanced anticipation of positive events: sex-specific modifications in cognitive bias and emotional resilience.

While it is known that chronic unpredictable stress and negative events adversely affect neurobiological outcomes, much less is known regarding the neurobiological impact of positive emotions such as chronic anticipation of appetitive events. From a translational perspective, an enhanced understanding of the impact of extended exposure to positive emotions may provide novel insights into effective non-pharmacological, behavior-based approaches to enhance mental resilience. Here, we investigate a novel rodent model of chronic Unpredictable Positive Event Response (UPER) training in male and fem

Corticostriatal and Dopaminergic Response to Beer Flavor with Both fMRI and [(11) C]raclopride Positron Emission Tomography.

BACKGROUND: Cue-evoked drug-seeking behavior likely depends on interactions between frontal activity and ventral striatal (VST) dopamine (DA) transmission. Using [(11) C]raclopride (RAC) positron emission tomography (PET), we previously demonstrated that beer flavor (absent intoxication) elicited VST DA release in beer drinkers, inferred by RAC displacement. Here, a subset of subjects from this previous RAC-PET study underwent a similar paradigm during functional magnetic resonance imaging (fMRI) to test how orbitofrontal cortex (OFC) and VST blood oxygenation level-dependent (BOLD) responses

Dopamine and the biology of creativity: lessons from Parkinson's disease.

BACKGROUND: Parkinson's disease (PD) is characterized by reduced flexibility, conceptualization, and visuo-spatial abilities. Although these are essential to creativity, case studies show emergence of creativity during PD. Knowledge about the role of dopamine in creativity so far only stems from a few case reports. We aim at demonstrating that creativity can be induced by dopaminergic treatments in PD, and tends to disappear after withdrawal of dopamine agonists. METHODS: Eleven consecutive creative PD patients were selected from candidates for subthalamic nucleus deep brain stimulation (STN

Neuropeptide Y (NPY) in the central nucleus of the amygdala (CeA) does not affect ethanol-reinforced responding in binge-drinking, nondependent rats.

The central nucleus of the amygdala (CeA) has been implicated as having a significant role in mediating alcohol-drinking behavior. Neuropeptide Y (NPY) has been investigated as a potential pharmacotherapeutic due to its ability to attenuate ethanol intake, particularly when administered into the CeA. Previous research suggests, though the evidence is somewhat conflicting, that the efficacy of NPY is contingent upon genetic background and/or prior history of ethanol dependence in rats. However, studies looking at the effects of NPY in nonselected animals lacking a history of ethanol dependence

Assessment of sucrose and ethanol reinforcement: the across-session breakpoint procedure.

We have demonstrated previously that the use of an across-session progressive ratio procedure yields breakpoint values for 10% ethanol (10E) that are stable and comparable to those measured for other drugs of abuse [Alcohol. Clin. Exp. Res. 23 (1999) 1580]. The aims of the present experiment were twofold: (1). to determine whether this procedure is sensitive to changes in reinforcer magnitude using a reinforcer previously demonstrated to affect operant responding in a predictable fashion and (2). to determine whether ethanol reinforcement produced similar changes in behavior. Circadian feeding promotion by Ninjin'yoeito counteracts frailty in aged mice.

Frailty in aging is a major health challenge, requiring solution. Older people with frailty often exhibit malnutrition and dysregulated feeding. Feeding behavior displays circadian rhythm, while aging and frailty involve rhythm disorders, suggesting possible role of circadian feeding in frailty and treatment. Herbal medicine, Ninjin'yoeito (NYT), reportedly ameliorates frail symptoms. The present study explored impacts of NYT on circadian feeding and psychological/physical functions in aged mice. Here, we report that oral NYT independent of administration timing increases food intake specifica

Seasonal variations in mood and behavior associate with common chronic diseases and symptoms in a population-based study.

The purpose of this study was to assess how seasonality is associated with some of the most common non-communicable diseases (NCDs) in the general Finnish population. The global seasonality score (GSS) was used to measure the magnitude of seasonality in 4689 participants, in addition to which they reported the extent to which the seasonal variations in mood and behavior were experienced as a problem. Regression models and the odds ratios were adopted to analyze the associations adjusted for a range of covariates. Seventy percent of the participants had seasonal variations in sleep duration, so

The pharmacological treatment and management of obesity.

Obesity is a pandemic with many complications that increase the societal disease burden and cost of health care, and decrease longevity and quality of life. Currently, 1 in 3 adults in the United States is obese. Physicians must therefore regularly confront obesity and its consequent diseases, and develop strategies for effective treatment and management. This article summarizes current lifestyle modifications, pharmacological treatment, and surgical options for the management of obesity and discusses the benefits, limitations, and risks of each. As insights are gained into the pathophysiology

Smoking-related correlates of depressive symptom dimensions in treatment-seeking smokers.

INTRODUCTION: The symptomatic heterogeneity of depression poses a barrier to understanding the smoking-depression relationship. Different types of depressive symptoms have evidenced disparate relations to smoking. Yet, depression measures employed in past depression-smoking research yield symptom subdimensions, which may not be sufficiently comprehensive or distinct. Exploring automatic approach-avoidance tendencies: the impact of self-relevant social feedback on behavior.

Previous studies have reported automatic approach-avoidance tendencies toward various stimuli, such as words, facial expressions, and images in the appetitive or aversive valence domain. This work investigates whether self-relevant evaluative feedback affects these behavioral tendencies using a touchscreen-based approach and avoidance task, in which participants responded to two-colored fish icons either by pulling toward or by pushing away from themselves. Evaluative feedback on participants' personality traits, provided by the fish, served as a task-irrelevant feature. A pronounced valence-c

Composite impulsivity-related domains in college students.

Impulsivity is a complex, multidimensional construct with prior theoretically and empirically derived characterizations of impulsivity-related behaviors varying considerably among studies. We assessed college students (N = 440) longitudinally with five impulsivity-related self-reported assessments and two computerized behavioral measures. Using a combination of exploratory and confirmatory factor analysis (CFA), we derived then validated several composite impulsivity-related domains (CIRDs). These factors replicated, in large part, findings from a previous study conducted by our group in an in

Levetiracetam as adjunctive antiepileptic therapy for patients with tuberous sclerosis complex: a retrospective open-label trial.

Seizures are a common neurologic symptom of tuberous sclerosis complex. The use of levetiracetam as adjunctive antiepileptic therapy was assessed in 20 patients with tuberous sclerosis complex aged 2 to 19 years. In this retrospective evaluation, 40% of patients treated with levetiracetam achieved a seizure reduction of more than 50%. Levetiracetam was generally well tolerated, and adverse events were relatively uncommon in patients who responded to treatment. The most commonly reported adverse events were behavioral problems. Unstable gait, insomnia, poor appetite, and increased seizure frequ
