Radical-Stabilizing Supramolecular Metallacage for Near-Infrared Photothermal Therapy.

The advancement of photothermal therapy (PTT) hinges on developing photothermal agents that combine high efficiency with exceptional chemical and thermal stability. Naphthalene diimide (NDI) derivatives are promising candidates due to their ability to generate radical anions that absorb in the near-infrared (NIR) region. However, these materials generate intrinsically unstable radical species under ambient conditions, hindering their widespread application. In this work, we report the design and synthesis of a core-substituted NDI-based tetrahedral metallacage that exhibits satisfactory radica

Photoinduced electrochemiluminescence: mechanistic insights and emerging applications.

Photoinduced electrochemiluminescence (PECL) has emerged as a light-electrochemical emission platform that integrates photogenerated charge carriers with electrochemiluminescent reaction pathways, enabling optical modulation and readout of interfacial electrochemical processes. By coupling optical excitation with redox chemistry at photoactive interfaces, PECL provides access to operational regimes beyond conventional electrochemiluminescence (ECL), including reduced-bias operation, anti-Stokes emission, and spatially confined activation. Recent advances have demonstrated that PECL can transdu

Beyond Nanoporosity: A CO2-Conditioned Glassy Matrix Contributes to the Brittle-to-Ductile Transition of Nanocellular Polyetherimide.

Nanocellular polyetherimide (PEI) exhibits enhanced toughness and impact resistance compared to the initial solid precursor, an effect usually attributed to the presence of nanometric cells within a confined glassy polymer matrix. In this work, we examine whether the CO2 saturation step used in gas dissolution foaming can affect the mechanical response by modifying the thermodynamic state of the glassy polymer. Thermal annealing without CO2 increases the yield stress and reduces ductility, whereas solid PEI saturated with CO2 and subsequently fully desorbed shows lower yield stress, an increas

Hollow carbon shell-confined CoSe2/Ni3Se4 nanoparticles anchored on rGO for enhanced charge storage and stability in supercapacitors.

Simultaneously achieving high charge-storage capability and long-term cycling stability remains a major challenge for supercapacitor electrodes. Herein, a hierarchical CoSe2/Ni3Se4@HCS-rGO electrode was rationally designed by confining CoSe2/Ni3Se4 nanoparticles within hollow carbon shells anchored on reduced graphene oxide (rGO). The hollow carbon shell (HCS) provides accessible inner and outer surfaces and shortened ion-transport pathways, while the CoSe2/Ni3Se4 heterostructure offers abundant redox-active sites for enhanced Faradaic charge storage. Meanwhile, the rGO framework establishes c

Structural insights into a substrate translocation pathway revealed by the RND efflux pump complex MexJK from Pseudomonas aeruginosa.

Resistance-nodulation-cell division (RND) efflux pumps are the major cause of multidrug resistance in Pseudomonas aeruginosa. The inner membrane protein MexK from P. aeruginosa is a narrow-spectrum RND transporter. It assembles with membrane fusion protein MexJ and outer membrane proteins to form a tripartite efflux complex that efficiently exports drugs like triclosan. Here, we resolve the cryo-EM structures of apo-MexK at 3.4-angstrom resolution and the triclosan-bound MexJK complex at 2.6-angstrom resolution. Besides the unique architectural features of MexK, our structural data reveal a tr

Expanding the Living Polymerization Toolkit: Mechanistic Insights and Optimization of Vinyl-Addition Polymerization of Norbornenes.

The living vinyl-addition polymerization of norbornenes is a robust method for accessing high-molecular-weight, solvent-processable materials with a chemically inert backbone and varied block copolymer architectures. However, the extent of its livingness and the underlying mechanisms of the loss of molecular weight control remain underexplored and unoptimized. In this work, we provide a functional guide to achieving improved molecular weight control through the systematic exploration of vinyl-addition polymerization by a tri-tert-butylphosphine-ligated cationic palladium initiator. We develope

CHD8 dysregulation in neurodevelopment: Emerging insights into autism pathophysiology.

Autism is considered a multifaceted neurodevelopmental condition, marked by impairments in social interaction, communication, and repetitive behaviours. Recent genetic research has highlighted mutations in the CHD8 (Chromodomain Helicase DNA-binding Protein 8) gene as one of the most significant risk factors associated with ASD. CHD8 encodes a chromatin remodeler protein that regulates gene expression by altering chromatin accessibility. This review aims to comprehensively review current findings on CHD8 mutations, with a focus on the gene's role in brain development, synaptic function, and AS

Sequential Topotactic Transformations Traverse Metastable Phases in Complex Aluminophosphate Frameworks with Tunable Pore Architectures.

Crystalline porous frameworks are irreplaceable platforms for catalysis, separation, new energy, sensing, and biomedicine, yet the majority exist as inaccessible metastable structures across an extensive structural landscape. Here we introduce a sequential topotactic transformation (STT) strategy, in which a simple steaming treatment induces a cascade of structurally correlated topotactic transformations, systematically uncovering a family of hidden metastable frameworks. Using this STT approach, the small-pore aluminophosphate molecular sieve (AlPO MS) DNL-17 undergoes sequential crystalline-

A cross-species single-cell kidney epigenome atlas reveals epithelial-dominant aging-like states in disease.

Epigenetic aging is a hallmark of chronic diseases. While such epigenetic changes can arise following tissue injury, the cell types most affected remain largely unknown. Here we built a cross-species single-cell multiomics atlas of DNA methylation, chromatin accessibility and transcription profiles from healthy, injured (human) and aged (mouse) kidneys. We found that tubular epithelial cells in diseased kidneys exhibit pronounced accelerated epigenetic aging and showed that this pathological state mirrors transcriptional trajectories observed during aging, driven by preferential dysregulation

Applied spintronics: From spin transport to spin-charge interconversion and emerging devices.

Spintronics exploits the electron's spin degree of freedom to go beyond the capabilities of conventional charge-based electronics. Since the discovery of giant magnetoresistance, the field has evolved from the study of spin-polarized transport in ferromagnets to a broader framework encompassing spin-orbit coupling, collective spin dynamics, and electric-field control of magnetic states. In this Review, we discuss the physical mechanisms that underpin modern applied spintronics, with particular emphasis on spin-charge interconversion, magnetoresistive effects, and the electrical detection and m

Human induced pluripotent stem cell-derived inner ear organoids reveal hair cell damage and plasticity after cisplatin and gentamicin exposure.

Ototoxicity is a leading cause of sensory deficits, including hearing loss and balance disorders. Predicting ototoxicity is challenging owing to translatability issues of animal models and limited access to human inner ear tissue. Known ototoxic drugs, such as cisplatin (a chemotherapeutic) and gentamicin (an aminoglycoside antibiotic), cause irreversible damage to sensory hair cells and neurons. Here, we establish human induced pluripotent stem cell (hiPSC)-derived inner ear organoids as an in vitro model for studying ototoxicity. Exposure to cisplatin and gentamicin led to hair cell and neur

Mapping brain-wide monosynaptic inputs to single neurons with ROInet-seq.

Viral projection tracing strategies help build regional connectomes of mammalian brains. G-deleted rabies virus (ΔG-RV) establishes monosynaptic input connectivity but cannot distinguish networks at cell resolution. Here, we implement a barcoded ΔG-RV for network tracing by quantitative RNA-sequencing. At optimized library complexity and uniformity, barcode detection reliably distinguished individual monosynaptic input networks of multiple infected neurons in parallel. To scale this approach to hundreds of cells and full-brain inputs, we develop regions-of-interest network sequencing (ROInet-s

Hydrogel-Based Vat Photopolymerization of Ceramics and Metals with Low Shrinkages via Repeated Infusion Precipitation.

Vat photopolymerization (VP) is a powerful tool for the fabrication of architected ceramic and metal structures. However, conventional methods of ceramic/metal VP, such as with the use of slurries or organic-inorganic hybrid resins, have challenges with viscosities, light-scattering, and limited material compositions. Recently, the use of metal-salt solutions has emerged as a promising approach for the VP of ceramics and metals. While versatile and accessible, the process is accompanied by a significant amount of shrinkage, which causes warping, porosity, and structural damage. Here, a versati

The regulatory code of injury-responsive enhancers enables precision cell-state targeting in the CNS.

Enhancer elements direct cell-type-specific gene expression programs. After injury, cells change their transcriptional state to adapt to stress and initiate repair. Here we investigate how injury-induced transcriptional programs are encoded within enhancers in the mammalian CNS. Leveraging single-nucleus transcriptomics and chromatin accessibility profiling, we identify thousands of injury-induced, cell-type-specific enhancers in the mouse spinal cord after a contusion injury. These are abundant in glial cells and retain cell-type specificity, even when regulating shared wound response genes.

Structural insights into nonpeptide antagonist inhibition of somatostatin receptor subtype 5.

The somatostatin receptor subtype 5 (SSTR5) is a critical pharmacological target involved in neuroendocrine signaling, metabolic regulation, and tumorigenesis. Despite its therapeutic potential, the structural mechanisms underlying SSTR5 inhibition by nonpeptide antagonists remain largely unresolved. In this study, we present high-resolution cryoelectron microscopy structures of human SSTR5 in complex with two selective small-molecule antagonists, antagonist 1 and S5A1, revealing the receptor's inactive conformation. Both antagonists induce a unique remodeling of extracellular loop 2, which ad

Photocatalytic construction of N-acyl-N,O-acetal-linked pyridines via aminocyclopropane ring opening.

Pyridine and quinoline linkers are privileged motifs in medicinal chemistry, yet their site-selective installation into complex scaffolds remains challenging. Here, we report that aminocyclopropanes serve as precursors to N-acyl-N,O-acetal linkers installed onto pyridines via visible-light-driven ring opening under oxidant-free conditions. The ring-opened radical is captured by N-aminopyridinium salts to forge C(sp3)-C(aryl) bonds at the C4-selective site of the pyridine core, while the concomitantly released N-centered radical oxidizes the reduced photocatalyst, enabling efficient turnover. Synthetic Antiferromagnetic Designer Nanodisks for High-Performance Magnetic Separation.

Magnetic separation of magnetic particles offers an appealing route to rapid and selective target capturing and isolation. However, to leverage such an approach to its full potential, high colloidal stability of the nanoparticles in the absence of a magnetic field for optimal binding, and rapid and quantitative recovery upon magnetic gradient field application are imperative. While these properties are mutually exclusive for conventional nanoparticles synthesized by wet-chemistry approaches, sputter deposition gives access to layered architectures featuring the properties of a synthetic antife
