[The brain, movement and space].

The combination, in humans, of methods of experimental Psychology, of brain imagery by position tomography, and functional MRI, and in animals of fine methods of neuronal recordings, has allowed us to reveal the neural mechanisms of the control of gaze. Thus is now available as important knowledge which can be used in Neuro-Ophthalmology for the understanding of the deficits of gaze control and in Neuro-Otology for the understanding of the vestibular disorders. We have also explored the neural basis of spatial memory and spatial disorientation such as observed in psychiatric symptoms as agorap…

Hypoglycaemia and the neonatal brain.

UNLABELLED: There has been much controversy and confusion regarding potential damage caused to the neonatal brain by low blood glucose levels. Previous studies of outcome after neonatal hypoglycaemia are flawed by many factors including retrospective data collection and inability to control for co-existing clinical complications. There is no doubt that hypoglycaemic brain damage does occur but the severity and duration of low blood glucose levels required to cause lasting harm varies between subjects and is related to the ability of each baby to mount a protective response such as the producti…

Language and the infant brain.

Three logically and empirically independent issues are often conflated in theory and research on brain and language: localization, innateness, and domain specificity. Research on adults and infants with focal brain injury support the following conclusions: (a) linguistic knowledge is not innate, and it is not localized in a clear and compact from in either the infant or adult brain; (b) the infant brain is not, however, a tabala rasa-it is already highly differentiated at birth, and certain regions are biased from the beginning toward modes of information processing that are particularly usefu…

[Aging brains and the dementias].

Dementia has been defined generally as an acquired persistent impairment of intellectual function caused by organic brain diseases. The rapidly increasing incidence of dementias has been claimed on the basis of epidemic studies in Japan as well as in Western developed countries. These observations are justified by the facts that most dementias are found in people over the age of 65 years, and that the elderly population is increasing rapidly resulting in both absolute numbers and percentage of the population. In Japan, it is estimated that 1.5 million of demented persons are present. The purpo…

The history of iron in the brain.

Brain iron research began in the late nineteenth century when Zaleski (1886) made a quantitative analysis of one human brain and correlated iron levels with observations on stained slices and some microscopic sections. Gradually, the realization grew that the central nervous system (CNS) contained iron which was different from hemoglobin-iron. This non-heme iron was found in highest concentrations in globus pallidus, substantia nigra, red nucleus, and dentate nucleus. The enhancement of the traditional histochemical stain, potassium ferrocyanide in hydrochloric acid, by incubating the reacted …

Gene therapy and the brain.

In this article we describe the application of the emerging concepts of gene therapy to 4 different neurologic disorders. The first of these is Lesch-Nyhan disease, a genetically-determined neurodevelopmental disorder caused by a defect in the gene which encodes the purine salvage enzyme hypoxanthine-guanine phosphoribosyltransferase (HPRT). Two additional disorders, Parkinsonism and Alzheimer's disease, are both neurodegenerative diseases of unknown etiology which affect the elderly. The final disorder involves malignant brain tumors. In each of these disorders, basic research with in vitro s…

VGF expression in the brain.

VGF is a neuronal polypeptide first identified as a cDNA clone in a gene library from nerve growth factor-stimulated PC12 cells. In the present paper, the expression of VGF is examined for the first time throughout the adult rat central nervous system with immunocytochemistry and Northern blot analysis. VGF RNA was found in all brain regions studied, including hypothalamus, hippocampus, cerebellum, olfactory bulb, and cortex. In contrast to the relatively strong immunostaining of hypothalamic neurons, the level of VGF RNA expression in the hypothalamus was relatively low in comparison with oth…

Ischaemia of the preterm brain.

Much has been learned about the diagnosis and prognosis of periventricalar leucomalacia during the past 10 years. The understanding of its causation, however, has not advanced much. Still, the candidate for the principal role is focal ischaemia in the particularly vulnerable regions of the brain. It has been difficult to provide direct supportive evidence for this hypothesis. It has not been possible to define a threshold for (global) cerebral blood flow (CBF) under which brain damage or electrical dysfunction occurs. Three hypothetical explanations for the lack of clear relation between globa…

Brain death in the newborn.

The clinical courses of 18 preterm and term infants less than 1 month of age in whom brain death was diagnosed were retrospectively reviewed. Clinical diagnosis was determined neurologically and included (1) coma, (2) apnea, manifested by inability to sustain respiration, and (3) absent brainstem reflexes. Electroencephalograms were performed in all patients; 17 patients had adequate cerebral blood flow as estimated by radionuclide imaging. The results indicate that (1) neurodiagnostic tests such as electroencephalograms and radionuclide scanning reconfirmed clinically determined brain death i…

Adrenocortical steroids and the brain.

In summary, a wide variety of effects of adrenal steroids on the brain have been reported and have been recently and exhaustively reviewed. From the viewpoint of endocrine physiology, however, what is often forgotten is the extraordinary difference in signal level between the two unique products of the adrenal cortex, the mineralocorticoid and glucocorticoid hormones. Levels of cortisol or corticosterone are 2-3 orders of magnitude higher than those of aldosterone, a difference that is tempered by perhaps one order of magnitude by the much higher binding of glucocorticoids to plasma protein. T…

NMR imaging of the brain.

The basic features of an NMR imaging system are outlined and three pulse sequences which produce images with varying dependence on proton density T1 and T2 are described. The first of these sequences, Repeated Free Induction Decay produces images which demonstrate changes in proton density as well as flow effects. The second sequence, Inversion-recovery, produces images which are dependent on T1 and show a high level of grey, white matter contrast giving considerable anatomical detail. In addition pathological processes such as infarction, haemorrhages, demyelination and malignancy, produce ch

Zinc, the brain and behavior.

The total content of zinc in the adult human body averages almost 2 g. This is approximately half the total iron content and 10 to 15 times the total body copper. In the brain, zinc is with iron, the most concentrated metal. The highest levels of zinc are found in the hippocampus in synaptic vesicles, boutons, and mossy fibers. Zinc is also found in large concentrations in the choroid layer of the retina which is an extension of the brain. Zinc plays an important role in axonal and synaptic transmission and is necessary for nucleic acid metabolism and brain tubulin growth and phosphorylation. …
