Detection of Chlamydia psittaci using DNA probes and the polymerase chain reaction.

Fewer than 10(5) elementary bodies of Chlamydia psittaci could be detected by using DNA hybridisation with a plasmid probe specific for avian chlamydial strains. PCR amplification of chlamydial DNA using primers specific for conserved regions of the major outer membrane protein gene enabled the detection of fewer than 10 elementary bodies. DNA could be amplified from 22 of the 24 chlamydial strains tested including avian, feline, ovine, caprine, koala and lymphogranuloma venereum strains.

Molecular characterization of sylvatic isolates of Trichinella spiralis.

Genetic relationships of 20 Trichinella isolates from Indiana wildlife were assessed and compared to Trichinella isolated from an infected swine herd. Trichinella larvae were isolated from coyotes, mink, raccoons, and red foxes. The larvae were maintained and amplified in white mice (ICR) and wild mice (Peromyscus leucopus). Differences in phenotypic characters of sylvatic isolates in the 2 laboratory hosts included an approximately 10-30-fold increase in parasite fecundity in wild mice. DNA for each isolate was extracted from Trichinella larvae and analyzed by dot-blot hybridization using a r

Localization of measles virus antigens in subacute sclerosing panencephalitis in ferrets.

Young adult male ferrets were inoculated intracerebrally (i.c.) with a cell-associated encephalitogenic subacute sclerosing panencephalitis (SSPE) virus strain to study the pathogenesis of the disease at the ultrastructural level. Most became acutely ill in 8-13 days. Areas of the brain were examined with indirect immunoperoxidase labeling techniques to detect measles antigen. None of these animals showed the characteristic viral nucleocapsids or marked inflammatory response associated with SSPE. However, all had positive immunolabeling of unstructured virus antigen, especially in post-synapti

The dissemination of rabies virus into cranial nerves and other tissues of experimentally infected goats and dogs and naturally infected skunks.

Immunofluorescence examination for rabies virus antigen performed on skin specimens acquired from experimentally inoculated rabid goats and some dogs, often show little or not fluorescence. This observation is contrary to results obtained by comparable examination of the skin of naturally infected animals and most experimentally infected dogs. In an effort to elucidate some factors associated with this experience, an experiment was performed to similarly examine the cranial nerves to see if the limited amounts of fluorescing antigen observed in the skin were also found there. Direct immunofluo

New genicular joint angle criteria for flexor muscle (Musculus Semimembranosus) during the terrestrial mammals walking.

BACKGROUND: The genicular or knee joint angles of terrestrial mammals remain constant during the stance phase of walking; however, the angles differ among taxa. The knee joint angle is known to correlate with taxa and body mass among extant mammals, yet several extinct mammals, such as desmostylians, do not have closely related descendants. Furthermore, fossils lose their soft tissues by the time they are unearthed, making body mass estimates difficult. These factors cause significant problems when reconstructing the proper postures of extinct mammals. Terrestrial mammals use potential and kin

Muscular anatomy of the forelimb of Leopardus geoffroyi: Functional and phylogenetic aspects in Feliformia, Part II: Forearm, autopod, and quantitative analysis.

For cats (Felidae), the forelimb plays a crucial role in their lifestyle, both for locomotion and for hunting. While there are studies on the anatomy of large and medium-sized felids, there are few on small Neotropical felids such as Leopardus geoffroyi. The present work complements a previous study by describing in detail the muscles of the forearm and autopodium, comparing them in a functional and phylogenetic context with other Feliformia species, and providing the first muscle maps of these regions for L. geoffroyi. For this purpose, both forelimbs of three adult specimens of L. geoffroyi

A molecular survey of spotted fever group rickettsiae in introduced raccoons (Procyon lotor).

BACKGROUND: The raccoon Procyon lotor (Linnaeus, 1758) (Carnivora; Procyonidae) is one of the most important and most intensively studied invasive mammal species in Europe. Within the last 30 years the raccoon has spread at an increasing rate, resulting in the establishment of local populations in various regions of Europe. In these newly colonised areas, gaps in knowledge of the raccoon's biology concern not only most aspects of its ecology in a broad sense, but also its pathogens and parasites. Most micropathogens recorded hitherto in the raccoons that have colonised Europe have documented e

Living with wildlife and mitigating conflicts around three Indian protected areas.

Crop and livestock losses to wildlife are a concern for people neighboring many protected areas (PAs) and can generate opposition to conservation. Examining patterns of conflict and associated tolerance is important to devise policies to reduce conflict impacts on people and wildlife. We surveyed 398 households from 178 villages within 10 km of Ranthambore, Kanha, and Nagarahole parks in India. We compared different attitudes toward wildlife, and presented hypothetical response scenarios, including killing the problem animal(s). Eighty percent of households reported crop losses to wildlife and

Molecular inferences suggest multiple host shifts of rabies viruses from bats to mesocarnivores in Arizona during 2001-2009.

In nature, rabies virus (RABV; genus Lyssavirus, family Rhabdoviridae) represents an assemblage of phylogenetic lineages, associated with specific mammalian host species. Although it is generally accepted that RABV evolved originally in bats and further shifted to carnivores, mechanisms of such host shifts are poorly understood, and examples are rarely present in surveillance data. Outbreaks in carnivores caused by a RABV variant, associated with big brown bats, occurred repeatedly during 2001-2009 in the Flagstaff area of Arizona. After each outbreak, extensive control campaigns were undertak

Multiplex PCR identification of Taenia spp. in rodents and carnivores.

The genus Taenia includes several species of veterinary and public health importance, but diagnosis of the etiological agent in definitive and intermediate hosts often relies on labor intensive and few specific morphometric criteria, especially in immature worms and underdeveloped metacestodes. In the present study, a multiplex PCR, based on five primers targeting the 18S rDNA and ITS2 sequences, produced a species-specific banding patterns for a range of Taenia spp. Species typing by the multiplex PCR was compared to morphological identification and sequencing of cox1 and/or 12S rDNA genes. Seroprevalence rates and transmission of plague (Yersinia pestis) in mammalian carnivores.

Exposure to plague (Yersinia pestis) by flea-bites or consumption of infected rodents is common in mammalian carnivores in North America. Most carnivore species exhibit seroprevalence rates ranging from 3% to 100% in areas where plague occurs. Seroprevalence is highest in mustelids, intermediate in ursids, felids, and canids, and lowest in procyonids, probably reflecting variation in exposure rates as a function of dietary habits. Although conventional wisdom suggests that carnivores may only be important in plague ecology as vectors of infective fleas, animal-to-human (zoonotic) transmission

Immunohistochemical characterization of tuberculous and non-tuberculous lesionsin naturally infected European badgers (Meles meles).

A panel of species cross-reactive antibodies was established for the immunohistochemical labelling of phagocytic and lymphoid cells in formalin-fixed normal badger tissues. These reagents were used to investigate the immunopathogenesis of both tuberculous and non-tuberculous granulomas in badgers. In normal badger tissues, antisera specific for the CD79a and CD79b epitopes strongly labelled follicular B lymphocytes and plasma cells in lymph nodes, bronchus-associated lymphoid tissue and Peyer's patches. Semen characteristics of the captive Indian leopard, Panthera pardus.

Semen samples from 11 Indian leopards (Pantherapardus) from 3 different zoos in India were collected by electroejaculation. A computer-aided semen analyzer (CASA) was used for assessing the quality of the semen vis-à-vis sperm motility. The volume of the ejaculate, sperm density, and the number of motile and morphologically normal spermatozoa were found to be 1.57 +/- 1.26 mL, 55.78 million +/- 38.67 million per mL, 57.05% +/- 16.96% and 71.92% +/- 15.32%, respectively. Although the spermatology varied between individuals in the study, Box-Whisker-plot analysis suggested that the distribution

Hormonal correlates of 'masculinization' in female spotted hyaenas (Crocuta crocuta). 1. Infancy to sexual maturity.

This report is concerned with hormone concentrations accompanying sexual maturation in a highly 'masculinized' female mammal, the spotted hyaena, Crocuta crocuta. Plasma concentrations of testosterone, androstenedione and oestrogen were determined by radioimmunoassay in a longitudinal study of 12 female and eight male hyaenas 2.5-62.5 months old. Concentrations of testosterone were significantly higher in males than in females after 26.5 months of age, but earlier measurements did not differ between sexes. Mean testosterone concentrations in adult female hyaenas (0.4-0.5 ng ml-1) were similar

Vagally mediated gastric motor and emetic reflexes evoked by stimulation of the antral mucosa in anaesthetized ferrets.

1. In the urethane-anaesthetized ferret chemical (NaCl, NaOH or HCl) or mechanical (stroking) stimulation of the gastric antral mucosa evoked a decrease in corpus pressure and inhibition of contractions in the presence of guanethidine, adrenalectomy and sectioned greater splanchnic nerves. 2. The fall in corpus pressure was present following administration of atropine but was abolished by vagotomy. 3. Preliminary evidence, using 100 mM-NaOH as the stimulus, is presented that the effects on corpus motility are due to simultaneous activation of the vagal efferents supplying the intramural non-ad

Aleutian disease in ferrets.

When 32 antibody-free ferrets were inoculated with the highly mink-virulent Utah-1 strain of Aleutian disease virus (ADV), most developed ADV antibody starting 15 days after infection, but the antibody titers were much lower than those seen in mink. Relatively small amounts of ADV were demonstrated in CRFK cell culture, using ferret spleen and lymph node homogenates only 4 to 10 days after experimental infection, but low-level viral persistence for 180 days was shown by mink inoculation. The ferrets inoculated with the Utah-1 strain of ADV did not develop elevated gamma globulin levels, but di
