Effects of dietary methionine on growth performance, muscle nutritive deposition, muscle fibre growth and type I collagen synthesis of on-growing grass carp (Ctenopharyngodon idella).

In the current research, a 60-d experiment was conducted with the purpose of exploring the impacts of methionine (Met) on growth performance, muscle nutritive deposition, muscle fibre growth and type I collagen synthesis as well as the related signalling pathway. Six diets (iso-nitrogenous) differing in Met concentrations (2·54, 4·85, 7·43, 10·12, 12·40 and 15·11 g/kg diets) were fed to 540 grass carp (178·47 (SD 0·36) g). Results showed (P < 0·05) that compared with Met deficiency, optimal level of dietary Met (1) increased feed intake, feed efficiency, specific growth rate and percentage wei

Molecular cloning, expression analyses of polymeric immunoglobulin receptor gene and its variants in grass carp (Ctenopharyngodon idellus) and binding assay of the recombinant immunoglobulin-like domains.

The Polymeric Immunoglobulin Receptor (pIgR) gene has been proved to play an important role in transporting polymeric immunoglobulin (Ig) in the mucosal tissues of mammals. pIgR gene also exists in teleost, but the genetic diversity and functions of this gene still need to be further explored. We obtained seven grass carp pIgR splicing transcripts, a full-length pIgR (CipIgR-1) and six truncated variants (CipIgR-2 to CipIgR-7). The full-length pIgR contained two immunoglobulin-like domains (ILD), a transmembrane domain (TMD) and a cytoplasmic domain (CyD). The CipIgR-2 lacked a small part in C

Expression pattern of potential biomarker genes related to growth, ion regulation and stress in response to ammonia exposure, food deprivation and exercise in common carp (Cyprinus carpio).

Waterborne ammonia has become a persistent pollutant of aquatic habitats. During certain periods (e.g. winter), food deprivation may occur simultaneously in natural water. Additionally, under such stressful circumstances, fish may be enforced to swim at a high speed in order to catch prey, avoid predators and so on. Consequently, fish need to cope with all these stressors by altering physiological processes which in turn are controlled by their genes. In this present study, toxicogenomic analyses using real time PCR was used to characterize expression patterns of potential biomarker genes cont

Histamine directly acts on beta-adrenoceptors as well as H1-histaminergic receptors, and causes positive inotropic effects in isolated ventricular muscles of carp heart (Cyprinus carpio).

1. The mechanism for positive and negative inotropic effects of histamine was studied in electrically stimulated ventricular strips of carp heart. 2. A high concentration of histamine (1 mM) caused a transient negative, and subsequent positive inotropic effects. The positive effect was significantly reduced by pyrilamine, diphenhydramine or dl-propranolol, but was not affected by cimetidine or d-propranolol. 3. Prior treatment with reserpine significantly decreased epinephrine and norepinephrine contents in ventricular muscles, and also almost completely abolished the positive inotropic effect

Upregulated expression of cardiac ankyrin repeat protein in human failing hearts due to arrhythmogenic right ventricular cardiomyopathy.

AIMS: Expression of cardiac ankyrin repeat protein (CARP) is augmented in heart failure due to dilated or ischaemic cardiomyopathy. It is unclear whether CARP is upregulated in heart failure due to arrhythmogenic right ventricular cardiomyopathy (ARVC). In the present study, we investigated the expression pattern of CARP and the correlation between CARP and the well-known heart failure marker pro-atrial natriuretic peptide (proANP) in ARVC failing hearts. METHODS AND RESULTS: Gene microarray analysis demonstrated increased CARP expression in ARVC failing hearts compared with non-failing contr

Genome-Wide SNP Discovery and Population Genomic Characterisation in Indian Major Carp Labeo catla (Hamilton, 1822) Through Low-Depth Whole-Genome Resequencing.

Single nucleotide polymorphisms (SNPs) are key markers for population and conservation genetics in aquaculture species. Using low-depth whole-genome resequencing of 35 Labeo catla individuals from seven populations (three wild riverine, four hatchery), we generated the first genome-wide SNP catalogue for this Indian major carp. Filtering against the L. catla reference genome (GCA_012976165.1) yielded 2.05 million high-quality bi-allelic SNPs (467 291-1 221 268 per population), with 27 000 shared across all populations. Heterozygosity (He = 0.4477-0.4532) and nucleotide diversity (π = 0.4703-0.

Screening and characterization of a probiotic fish-derived Enterococcus faecium and its co-fermentation with Acanthopanax senticosus extract modulate growth and immunity in crucian carp(Carassius auratus).

Under intensive aquaculture practices, the frequent occurrence of diseases exacerbates the risk of antibiotic overuse. Therefore, screening safe and efficacious probiotics originating from the host fish themselves represents one of the key strategies to address this issue. In this study, lactic acid bacteria were isolated from the intestinal contents of healthy crucian carp and identified by morphological, physiological, and biochemical characterization, as well as 16SrDNA sequencing. Their probiotic potential was comprehensively evaluated by antimicrobial assays, in vitro tolerance assays, an

Integrated analysis of the miRNA-mRNA regulatory network and role of the miRNA 1971-x in poly-β-hydroxybutyrate (PHB) immunoenhancement in gibel carp (Carassius auratus gibelio).

Poly-β-hydroxybutyrate (PHB), a bacterial-derived homopolymer, has recently emerged as a promising prebiotic with immunostimulatory properties in aquatic animals. However, the immunomodulatory molecular remains unclear. MicroRNAs (miRNAs) play a crucial role in post-transcriptional gene regulation, particularly in immune function. In this study, we investigated the immunoenhancement effects of dietary PHB supplementation in gibel carp (Carassius auratus gibelio) by analyzing miRNA and mRNA expression profiles in the spleen. Results showed that a total of 948 known miRNAs and 471 novel miRNAs i

Mass mortality events associated with cyprinid herpesvirus 2 (CyHV-2) infection in wild Prussian carp Carassius gibelio in the Netherlands, and molecular biology of virus strains.

In 2011 and 2015, four mass mortalities of Prussian carp (Carassius gibelio) were observed in a recreational freshwater lake and open freshwater in the western part of the Netherlands. Cyprinid herpesvirus 2 (CyHV-2) infection was suspected in these cases, based on presumptive gross diagnosis. To elucidate the cause of the mass mortalities diagnostic PCR assays were performed for CyHV-2, based on the helicase gene. Furthermore, the viral isolates were genotyped by sequencing the enlarged marker A and marker B sequences. Diagnostic PCR revealed that three of four samples were positive for CyHV-

Micro-algal astaxanthin ameliorates polystyrene microplastics-triggered necroptosis and inflammation by mediating mitochondrial Ca2+ homeostasis in carp's head kidney lymphocytes (Cyprinus carpio L.).

Polystyrene microplastics (PM) is a pressing global environmental concern, posing substantial risks to aquatic ecosystems. Microalgal astaxanthin (MA), a heme pigment, safeguards cells against oxidative damage induced by free radicals, which contributes to various health conditions, including aging, inflammation and chronic diseases. Herein, we investigated the potential of MA in ameliorating the immunotoxicity of PM on carp (Cyprinus carpio L.) based on head kidney lymphocytes treated with PM (250 μM) and/or MA (100 μM). Firstly, CCK8 results showed that PM resulted in excessive death of head
