Immunohistochemical study of the orthokeratinized odontogenic cyst: a comparison with the odontogenic keratocyst.

OBJECTIVE: Orthokeratinized odontogenic cyst (OOC) is a developmental cyst that occurs in the maxilla and the mandible and is defined by the World Health Organization as the uncommon orthokeratinized type of odontogenic keratocyst (OKC). However, studies have shown that OOC has peculiar clinicopathologic aspects and biologic behavior when compared with other developmental odontogenic cysts, especially OKCs. Therefore, in this study, the immunohistochemical profile of the OOC was delineated and compared with that of the OKC. STUDY DESIGN: Twelve cases of OOC were submitted to a panel of antibo

Collagen of breast with benign dysplasia.

Benign dysplasia is a pathological process in the female breast associated with hypertrophy of interlobular and periductal connective tissue. It leads to disappearance of the lobular structure of this organ. It was found that the mammary tumours contain more collagen in comparison to the normal breast. In the normal breast collagen type I and type III constitute 70% and 25% of the total collagen content, respectively. The investigated tumors contain mainly a complex of collagen type I and type III which cannot be dissociated by the methods used for collagen preparation.

All-trans retinoic acid diminishes collagen production in a hepatic stellate cell line via suppression of active protein-1 and c-Jun N-terminal kinase signal.

Following acute and chronic liver injury, hepatic stellate cells (HSCs) become activated to undergo a phenotypic transformation into myofibroblast-like cells and lose their retinol content, but the mechanisms of retinoid loss and its potential roles in HSCs activation and liver fibrosis are not understood. The influence of retinoids on HSCs and hepatic fibrosis remains controversial. The purpose of this study was to evaluate the effects of all-trans retinoid acid (ATRA) on cell proliferation, mRNA expression of collagen genes [procollagen α1 (I), procollagen α1 (III)], profibrogenic genes (TGF

Increased deposition of types III and V collagen in neurofibroma tissue from patients with von Recklinghausen disease.

Collagen components in neurofibroma tissue from patients with von Recklinghausen disease were investigated, in comparison with those in normal skin and peripheral nerve tissue. Biochemical analysis of collagen isolated from the tissues by limited pepsin digestion indicated that the neurofibroma tissue contained type I collagen as the major constituent and increased amounts of types III and V collagen. The relative ratios of alpha 1(III)/alpha 1(I) and alpha 1(V) + alpha 2(V)/alpha 1(I) in the tissue were 0.87-0.92 and 0.16-0.17, respectively, while in normal skin, these ratios were 0.36-0.45 a

Modified dendrimer cross-linked collagen-based matrices.

Dendrimer cross-linking has been achieved with pepsin digested over 80% type-I bovine collagen to create strong hydrogels with good cell compatibility. Herein we investigate the use of commercially available collagen-based products with the dendrimer cross-linking technology. Specifically PureCol(®) (PC), a 97% bovine type-I collagen, human collagen (HC) and human extracellular matrix (hECM) were concentrated, and then cross-linked with polypropyleneimine octaamine generation two dendrimers using 1-ethyl-3-(3-dimethyl aminopropyl) carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS)

Tuneable microfibrillar collagen structures within dense chitosan hydrogels.

Chitosan-type I collagen hydrogels are paradigms of polysaccharide-protein assemblies with applications as biomaterials. However, preparing physical hydrogels combining them at comparable, high concentrations (>20 mg mL-1) within interpenetrated networks remains challenging. Here, we could combine chitosan and collagen solutions at 25 mg mL-1 to prepare two different types of concentrated hydrogels. When neutralized under ammonia vapours, mixed solutions form composite hydrogels, where collagen fibers exhibiting an unusual, branched morphology occupy a chitosan network porosity. In contrast, n

Effect of Sparc knockout on the extracellular matrix of mouse periodontal ligament cells.

The periodontal ligament (PDL) is a critical component in maintaining tooth stability. It is composed of cells and an extracellular matrix (ECM), each with unique roles in tissue function and homeostasis. Secreted protein acidic and rich in cysteine (SPARC), a calcium-binding matricellular glycoprotein, plays a crucial role in regulating ECM assembly and turnover, alongside facilitating cellular-ECM interactions. In the present study, mass spectrometry-based proteomics was used to assess the impacts of Sparc-knockout (KO) on PDL-derived cells. Results demonstrated that Sparc-KO significantly r

Collagen V deficiency during murine tendon healing results in distinct healing outcomes based on knockdown severity.

Tendon function is dependent on proper organization and maintenance of the collagen I tissue matrix. Collagen V is a critical regulator of collagen I fibrils, and while prior studies have shown a negative impact of collagen V deficiency on tendon healing outcomes, these studies are confounded by collagen V deficiency through tendon development. The specific role of collagen V in regulating healing tendon properties is therefore unknown. By using inducible Col5a1 knockdown models and analyzing gene expression, fibril and histological tendon morphology, and tendon mechanical properties, this stu

Collagens I and V differently regulate the proliferation and adhesion of rat islet INS-1 cells through the integrin β1/E-cadherin/β-catenin pathway.

Extracellular matrix (ECM) plays an important role in tissue repair, cell proliferation, and differentiation. Our previous study showed that collagen I and collagen V differently regulate the proliferation of rat pancreatic β cells (INS-1 cells) through opposite influences on the nuclear translocation of β-catenin. In this study, we investigated the β-catenin pathway in INS-1 cells on dishes coated with collagen I or V. We found that nuclear translocation of the transcription factor Yes-associated protein (YAP) was enhanced by collagen I and suppressed by collagen V, but had no effect on INS-1

Presence of type III collagen in bone from a patient with osteogenesis imperfecta.

Samples of bone from a patient with osteogenesis imperfecta were found to synthesize and contain type III collagen as well as type I collagen. Normal bone contains only type I collagen except in the lining cells of the bone marrow cavities. In the patient's tissue, type III collagen was localized in nonfibrillar structures in discrete areas of the bone. These and previous studies indicate that certain types of osteogenesis imperfecta may be caused by a failure of normal bone maturation and the sites in which the type III collagen is found appear to be defects in the bone.

Mechanisms of Local Aggressiveness Without Metastasis in Dermatofibrosarcoma Protuberans.

BACKGROUND/AIM: Dermatofibrosarcoma protuberans (DFSP) is a rare cutaneous sarcoma characterized by marked local invasiveness and a low but definite risk of metastasis. The biological basis for this discrepancy remains incompletely understood. This review synthesizes current evidence regarding local invasion, metastatic behavior, tumor microenvironment, vascular and lymphatic involvement, and the role of the collagen type I alpha 1 (COL1A1)-platelet-derived growth factor beta (PDGFB) fusion. MATERIALS AND METHODS: A narrative review of peer-reviewed literature was conducted, prioritizing PubM…

The Rarity in the Rarity: A Rare Case of Bednar Tumor (Pigmented Dermatofibrosarcoma Protuberans) Localized on Acral Skin With Negative COL1A1-PDGFB Fusion on Fluorescence In Situ Hybridization.

Dermatofibrosarcoma protuberans (DFSP) is a rare, low-grade fibroblastic tumor that typically exhibits CD34 positivity and a storiform growth pattern. Although commonly found on the trunk and proximal extremities, DFSP can also present in less frequent locations, such as the head, neck, and acral sites. This report discusses a rare case of pigmented DFSP, also known as Bednar tumor, located on the back of the left hand in a 48-year-old woman. Histological examination revealed a mesenchymal proliferation with a storiform pattern and occasional dendritic melanocytes. Immunohistochemical analysis

A case of osteogenesis imperfecta diagnosed after subchondral insufficiency fracture of bilateral femoral heads.

Osteogenesis imperfecta (OI) is a heterogeneous disorder characterised by bone fragility. Herein, we report a case of OI diagnosed after subchondral insufficiency fracture (SIF) of bilateral femoral heads. A 37-year-old woman was referred to Saitama Medical University Hospital due to left hip pain without any trauma that lasted for 2 months. She was subsequently diagnosed with SIF of the left femoral head. After 3 months, she further developed SIF of the right hip without any trauma. Magnetic resonance imaging of the bilateral hips showed linear low-signal changes of the subchondral bone and b

RNA Sequencing of Urine-Derived Cells for the Characterization and Diagnosis of Osteogenesis Imperfecta.

DNA sequencing is a reliable tool for identifying genetic variants in osteogenesis imperfecta (OI) but cannot always establish pathogenicity, particularly in variants altering splicing. RNA sequencing can provide functional evidence of the effect of a variant on the transcript but requires cells expressing the relevant genes. Here, we used urine-derived cells (UDC) to characterize genetic variants in patients with suspected or confirmed OI and provide evidence on the pathogenicity of variants of uncertain significance (VUS). Urine samples were obtained from 45 children and adolescents; UDC cul

STRO-1 confers myofibroblast transdifferentiation in fibroblasts derived from oral submucous fibrosis.

BACKGROUND: STRO-1 is a mesenchymal cell marker present on all clonogenic stromal precursors. Current evidence has indicated that the pathogenesis of fibrotic changes may be mediated by stemness properties. The aim of this study was to investigate the role of STRO-1 in areca quid chewing-associated oral submucous fibrosis (OSF). METHODS: Thirty OSF specimens and ten normal buccal mucosae were examined by immunohistochemistry. The activity of STRO-1 from fibroblasts cultured from normal buccal mucosa (BMFs) and OSF (OSFFs) were measureed and the effect of arecoline, a major areca nut alkaloid,

Mutations in COL1A1 Gene Change Dentin Nanostructure.

Although many studies have attempted to associate specific gene mutations with dentin phenotypic severity, it remains unknown how the mutations in COL1A1 gene influence the mechanical behavior of dentin collagen and matrix. Here, we reported one osteogenesis imperfecta (OI) pedigree caused by two new inserting mutations in exon 5 of COL1A1 (NM_000088.3:c.440_441insT;c.441_442insA), which resulted in the unstable expression of COL1A1 mRNA and half quantity of procollagen production. We investigated the morphological and mechanical features of proband's dentin using atomic force microscope (AFM)

A novel splicing mutation in COL1A1 gene caused type I osteogenesis imperfecta in a Chinese family.

Osteogenesis imperfect (OI) is a heritable connective tissue disorder with bone fragility as a cardinal manifestation, accompanied by short stature, dentinogenesis imperfecta, hyperlaxity of ligaments and skin, blue sclerae and hearing loss. Dominant form of OI is caused by mutations in the type I procollagen genes, COL1A1/A2. Here we identified a novel splicing mutation c.3207+1G>A (GenBank ID: JQ236861) in the COL1A1 gene that caused type I OI in a Chinese family. RNA splicing analysis proved that this mutation created a new splicing site at c.3200, and then led to frameshift. This result fu
