Evaluation of chemical phototoxicity, focusing on phosphorylated histone H2AX.

Histone H2AX is a minor component of nuclear histone H2A. The phosphorylation of histone H2AX at Ser 139, termed γ-H2AX, was originally identified as an early event after the direct formation of DNA double-strand breaks (DSBs) by ionizing radiation. Now, the generation of γ-H2AX is also considered to occur in association with secondarily formed DSBs by cellular processing such as DNA replication and repair at the site of the initial damage, including DNA adducts, crosslinks, and UV-induced photolesions. Therefore, γ-H2AX is currently attracting attention as a new biomarker for detecting variou

Familial clustering of breast cancer: possible interaction between DNA repair proficiency and radiation exposure in the development of breast cancer.

The capacity of peripheral blood lymphocytes to repair X-ray-induced DNA damage, manifest as chromatid damage 30-90 min after G2-phase X-irradiation, was measured among available members of a family exhibiting a cluster of breast-cancer cases occurring in one generation. The cancer patients had been exposed to repeated chest fluoroscopic examinations during early childhood and adolescence. The development of breast cancer was correlated with DNA repair proficiency and history of radiation exposure. The results of the family study provide preliminary support for the hypothesis that a deficiency

Nanoscale surface defects of goethite governing DNA adsorption process and formation of the Goethite-DNA conjugates.

In urbanized areas, extracellular DNA (exDNA) is suspected of carrying genes with undesirable traits like virulence genes (VGs) or antibiotic resistance genes (ARGs), which can spread through horizontal gene transfer (HGT). Hence, it is crucial to develop novel approaches for the mitigation of exDNA in the environment. Our research explores the role of goethite, a common iron mineral with high adsorption capabilities, in exDNA adsorption processes. We compare well-crystalline, semi-crystalline, and nano goethites with varying particle sizes to achieve various specific surface areas (SSAs) (18.

A whole-cell bioreporter approach for the genotoxicity assessment of bioavailability of toxic compounds in contaminated soil in China.

A whole-cell bacterial bioreporter Acinetobacter baylyi strain ADP1_recA_lux that responds to genotoxins was employed to directly assess the adverse effects of the bioavailable fraction of mitomycin C (MMC), benzo[a]pyrene (BaP), chromium (VI) and lead (II) in amended soils and soil samples from two fragile areas in China without soil pre-treatment. The amended soils containing pollutants with the concentrations as low as 0.4 mg/kg MMC, 0.5 mg/kg BaP, 520 mg/kg Cr (VI) and 2072 mg/kg Pb (II) were found to be toxic. Soil particle-associated pollutants accounted for 86%, 100%, 29%, and 92% of th

Human health perspectives on environmental exposure to benzidine: a review.

Benzidine, an odorless, white to slightly reddish-white crystalline organic compound, is an environmental contaminant that has been identified at about 30 National Priorities List (NPL) hazardous waste sites in the United States. In the environment, it is usually found attached to suspended particles either in its "free" state or as chloride or sulfate salts. In the past, U.S. industries used large quantities of benzidine to produce dyes for paper, clothes, and leather. Since the ban on its production and use in the United States in the 1970s, this compound is imported for specialty uses. Peop

MGMT, a risk factor for both genetic and environmental forms of dementia.

MGMT, the gene coding for the DNA-repair protein O6 -methylguanine methyltransferase, which has been recently shown to be a risk factor for inherited forms of Alzheimer's disease (AD), notably among women, might also be linked to Western Pacific amyotrophic lateral sclerosis and Parkinsonism-dementia complex (ALS/PDC), one phenotype of which is an AD-like dementia. Guam ALS/PDC is strongly considered to be an environmental disorder caused by oral exposure to natural toxins (i.e., genotoxic/epigenotoxic chemicals), notably methylazoxymethanol (MAM) that alkylates guanine to form O6 -methylguani

Photoprotection in human skin--a multifaceted SOS response.

Human skin has developed elaborate defense mechanisms for combating a wide variety of potentially damaging environmental factors; principal among these is UV light. Despite these defenses, short-term damage may include painful sunburn and long-term UV damage results in both accelerated skin aging and skin cancers such as basal cell carcinoma, squamous cell carcinoma and even malignant melanoma. While UV radiation damages many cellular constituents, its most lasting effects involve DNA alteration. The following sections briefly review UV-inducible protective responses in bacteria and in skin, t

Associations between in utero exposure to polybrominated diphenyl ethers, pathophysiological state of fetal growth and placental DNA methylation changes.

BACKGROUND: Polybrominated diphenyl ethers (PBDEs) are environmental chemicals with harmful effects on pregnancy, but their effects on adverse developmental outcomes are not fully understood. The placental DNA methylation is strongly influenced by prenatal environmental factors and has been linked to fetal growth. OBJECTIVE: To evaluate the association between in utero PBDEs exposure, placental DNA methylation changes (growth regulatory genes), and pathophysiology of fetal growth (birth outcomes, fetal growth retardation) in a population-based pregnancy cohort study. METHODS: This was a nest

Interspecific differences in oxidative DNA damage after hydrogen peroxide exposure of sea urchin coelomocytes.

Interspecific comparison of DNA damage can provide information on the relative vulnerability of marine organisms to toxicants that induce oxidative genotoxicity. Hydrogen peroxide (H2O2) is an oxidative toxicant that causes DNA strand breaks and nucleotide oxidation and is used in multiple industries including Atlantic salmon aquaculture to treat infestations of ectoparasitic sea lice. H2O2 (up to 100 mM) can be released into the water after sea lice treatment, with potential consequences of exposure in nontarget marine organisms. The objective of the current study was to measure and compare d

Human DNA Mutations and their Impact on Genetic Disorders.

DNA is a remarkably precise medium for copying and storing biological information. It serves as a design for cellular machinery that permits cells, organs, and even whole organisms to work. The fidelity of DNA replication results from the action of hundreds of genes involved in proofreading and damage repair. All human cells can acquire genetic changes in their DNA all over life. Genetic mutations are changes to the DNA sequence that happen during cell division when the cells make copies of themselves. Mutations in the DNA can cause genetic illnesses such as cancer, or they could help humans b

Long-Term Environmental Persistence and Phylogenetic Evidence of Burkholderia pseudomallei in a Melioidosis Case Cluster in Taiwan.

OBJECTIVES: Melioidosis, caused by Burkholderia pseudomallei , is an environmentally acquired disease endemic to Southeast Asia and northern Australia, and increasingly recognised as an important but underdiagnosed tropical infection. In Taiwan, the spatial distribution and environmental reservoirs of this pathogen remain poorly understood. METHODS: We analysed 796 melioidosis cases reported between 2003 and 2024 and identified a recurrent, statistically significant cluster zone using SaTScan, especially during 2009-2013 and again in 2024. To investigate environmental persistence, we conducte

Adaptation and changing phenotypes through transgenerational epigenetics.

In this article collection, we describe how noncoding epigenetic changes in DNA are transmitted across multiple generations in eukaryotic organisms including plants and animals. And such environmentally induced biochemical alterations of DNA and histones result in profound changes in gene expression. In plants and invertebrate animals, transgenerational epigenetic inheritance has been well documented, and it continues to be substantiated in humans and other vertebrates. These exciting new discoveries have profound consequences for changing as well as maintaining phenotypes expressed by various

Sensitive and label-free electrochemical lead ion biosensor based on a DNAzyme triggered G-quadruplex/hemin conformation.

Lead ion (Pb2+) is a common environmental contaminant, which causes serious bioaccumulation and toxicity in human body. In this work, we developed a novel Pb2+ electrochemical biosensor using the specific DNAzyme on a DNA tetrahedron probe, in the presence of Pb2+, the substrate strand was cleaved into two parts and released a "G-rich" oligo which subsequently formed a G-quadruplex/hemin complex, generating a detectable catalysis current signal with the assistant of H2O2. The 3-D DNA tetrahedron regulated the density and orientation of the probe and thus improved the DNAzyme reaction, and faci

Green synthesis of gold nanoparticles for staining human cervical cancer cells and DNA binding assay.

Gold nanoparticles have been functionalized by non-ionic surfactants (polysorbates) used in pharmaceutical formulations. This results in the formation of more well-dispersed gold nanoparticles (GNPs) than the GNPs formed in neat water. The synthesized GNPs show good temporal stability. The synthesis conditions are mild and environmentally benign. The GNPs can bind to ct-DNA and displace bound dye molecules. The DNA-binding assay is significant as it preliminarily indicated that DNA-GNP conjugates can be formed. Such conjugates are extremely promising for applications in nanobiotechnology. The

Acute exposure to UVB has a more profound effect on plant genome stability than chronic exposure.

Environmental factors that damage DNA have various lengths of exposure and intensity levels. Although the results of increasing the intensity of a DNA damaging agent is often predictable, it is not clear whether the stage during development when the exposure is received has any influence on the amount of DNA damage. In this paper we analyzed the influence of UVB on the stability of Arabidopsis thaliana and the Nicotiana tabacum genomes. Our experiments showed that the acute exposure to UVB produces a significantly greater increase in homologous recombination frequency (HRF) and recombination r

Nitrosamines, alcohol, and gastrointestinal tract cancer: recent epidemiology and experimentation.

Recent epidemiological and experimental data continues to implicate nitrosamines in causation of gastrointestinal cancers. The evidence is strong for pharynx, esophagus, and stomach, and more problematic for liver, pancreas, and colorectum. Substantial levels of the promutagenic DNA adduct, Ob-methylguanine, in DNA from these organs in patas monkeys after a low dose of N-nitrosodimethylamine confirms the capacity for activation of environmental nitrosamines in these primate tissues. Alcohol is both an independent and a tobacco-interactive risk factor, influencing cancer incidence for oropharyn

Parental Characteristics Associated With Children Born With Neural Tube Defects in Puebla, Mexico.

BACKGROUND: Neural tube defects (NTDs) are one of the most common congenital anomalies. NTDs are caused by multiple factors that develop in the intrauterine stage and represent a critical birth defect morbidity and mortality. NTDs may depend on a risk factor modifiable by parents; hence, paternal health is important for transgenerational health, as it contributes half of the genetic material for the development of a new individual. OBJECTIVE: This study aims to describe demographic, biochemical parameters, and seminal characteristics of parents of offspring with NTDs. MATERIAL AND METHODS: A

Impact of Epigenetic Modifications in Airway Diseases. Role of Inflammation, Environmental Factors and Aging.

BACKGROUND: Airway diseases, including asthma and chronic obstructive pulmonary disease (COPD) are global public health burden due to their high prevalence, morbidity, and mortality. Although these conditions are clinically distinct, they share key pathogenic mechanisms, including chronic inflammation, oxidative stress, and airway remodeling. Continuous exposure of the lungs to environmental agents and pathogens increases their susceptibility to injury and dysregulated immune responses, thereby promoting chronic disease progression. Environmental factors, including cigarette smoke and air poll

Molecular exploration of mosquitoes and their vector-borne pathogens in urban water environments in Benin.

Malaria and dengue fever, both transmitted by mosquitoes, are major health concerns in (sub)tropical regions, with expanding endemic ranges. Monitoring mosquito populations and the pathogens they carry is crucial for effective vector control. Here, we explored molecular-based surveillance using environmental water samples as an alternative to conventional mosquito collection methods. A total of 154 samples were collected from eight environment types, including septic tanks, urban drainage, lake, groundwater, puddles, and tire tracks, in urban Benin. Polymerase chain reaction and high-throughpu
