Abstract Chronic myeloid leukemia (CML) is a malignant myeloproliferative tumor which is originating from hematopoietic stem cells. Chemotherapy is the preferred made of treatment for the disease. However, in recent years, more and more patients have multidrug resistance (MDR) during treatment, which is the main cause of failure treatment, therefore, the search for effective reversal agents has important clinical significance. Traditional Chinese medicine derived reversal agents have the characteristics of multiple targets, low toxicity and high efficiency, which can reverse the drug resistan

Antibiotics have always been considered as one of the most relevant discoveries of the twentieth century. Unfortunately, the dawn of the antibiotic era has sadly corresponded to the rise of the phenomenon of antimicrobial resistance (AMR), which is a natural process whereby microbes evolve in such a way to withstand the action of drugs. In this context, the identification of new potential antimicrobial targets and/or the identification of new chemical entities as antimicrobial drugs are in great demand. To date, among the many possible approaches used to deal with antibiotic resistance is the

Eukaryotic microbial pathogens are major contributors to illness and death globally. Although much of their impact can be controlled by drug therapy as with prokaryotic microorganisms, the emergence of drug resistance has threatened these treatment efforts. Here, we discuss the challenges posed by eukaryotic microbial pathogens and how these are similar to, or differ from, the challenges of prokaryotic antibiotic resistance. The therapies used for several major eukaryotic microorganisms are then detailed, and the mechanisms that they have evolved to overcome these therapies are described. The

This paper is the fourth in a series on the search for new antibacterial therapies, and covers new compounds belonging to the aminoglycoside, macrolide and tetracycline groups of antibiotics. The article describes eight new substances at the clinical trial stage of development. One of them is an aminoglycoside (plazomicin), four are macrolides, collectively known as ketolides (cethromycin, solithromycin, EDP-420 and EDP-788), and the remaining three are members of the tetracycline group (omadacycline, eravacycline, sarecycline). Despite the long-term and very expensive process of collecting do

Treatment of extensively drug-resistant tuberculosis (XDR-TB) is a challenge. We describe a young child with pulmonary XDR-TB who did not respond to an aggressive multidrug-resistant TB treatment regimen, but was cured with linezolid in combination with other reserve antituberculous drugs. No serious adverse events were observed during 19 months of treatment for XDR-TB.

Abstract Outbreaks of extensively drug-resistant tuberculosis (XDR-TB) in developing countries and recent headlines of an American traveling with a resistant variant of tuberculosis have brought XDR-TB into the spotlight. The World Health Organization and the United States Centers for Disease Control and Prevention have identified XDR-TB as a serious public health threat and are mandating increased efforts at control of tuberculosis. Although XDR-TB is believed to be no more infectious than other variants of tuberculosis, infection with and spread of XDR-TB are concerning because of the ineffe

Chronic myeloid leukaemia (CML) is a disease induced by the BCR-ABL oncogene. Tyrosine kinase inhibitors (TKIs) were introduced in the late 1990s and have revolutionized the management of CML. The majority of such patients can now expect to live a normal life providing they continue to comply with TKI treatment. However, in a significant proportion of cases, TKI resistance develops over time, requiring a change of therapy. Over the past few years, multiple molecular mechanisms of resistance have been identified and some common themes have emerged. One is the development of resistance mutations

Cambodia's first-line artemisinin combination therapy, dihydroartemisinin-piperaquine (DHA-PPQ), is no longer sufficiently curative against multidrug-resistant Plasmodium falciparum malaria at some Thai-Cambodian border regions. We report recent (2008 to 2013) drug resistance trends in 753 isolates from northern, western, and southern Cambodia by surveying for ex vivo drug susceptibility and molecular drug resistance markers to guide the selection of an effective alternative to DHA-PPQ. Over the last 3 study years, PPQ susceptibility declined dramatically (geomean 50% inhibitory concentration

The need to discover and develop alternative therapies to treat multidrug-resistant bacterial infections is timely. The aim of this study was to examine the antimicrobial potential of propolis, as a purified and concentrated special extract GH 2002, against clinical isolates of Streptococcus pyogenes, methicillin-resistent Stapylococcus aureus (MRSA), vancomycin-resistant Enterococcus faecium (VRE) and Candida. Minimal inhibitory concentrations (MICs) and minimal microbicidial concentrations (MMCs) of propolis against microbial pathogens were evaluated using the broth microdilution method. Pro

Antimicrobial resistance (AMR) has consistently been linked to antibiotic use. However, the roles of commonly prescribed non-antimicrobial drugs as drivers of AMR may be under-appreciated. Here, we studied a cohort of patients with community-acquired pyelonephritis and assessed the association of exposure to non-antimicrobial drugs at the time of hospital admission with infection with drug-resistant organisms (DRO). Associations identified on bivariate analyses were tested using a treatment effects estimator that models both outcome and treatment probability. Exposure to proton-pump inhibitors

Hypericum monogynum L. is a plant used in traditional Chinese medicine for treating inflammatory disorders and infectious diseases. Seven new polyprenylated acylphloroglucinols (PPAPs) and seven known PPAPs were isolated from the flowers of H. monogynum. Structurally, compound 1 features the first rearranged PPAP with a fascinating spirocyclic 5/6/6 ring system, and compound 2 is the first example of a highly modified PPAP featuring a rare 6/5/5/5-tetracyclic system. Compound 7 is the first highly degraded PPAP. Their structures were elucidated through comprehensive analysis of HRESIMS, NMR sp

Humans have encountered many epidemics caused by pathogenic microorganisms since the turn of the century. The prevalence of fungal infections has significantly increased on a global scale, adversely impacting human health. Candida species remain among the most prevalent and widely spread opportunistic fungi, with Candida auris emerging as a notorious hospital-acquired pathogen. This fungal pathogen causes nosocomial bloodstream infections (BSI), coupled with a high in-hospital death rate and significant multidrug-resistance (MDR). Standard quaternary ammonium compounds (Quats), are often ineff

Antimicrobial resistance (AMR) in wildlife-associated bacteria represents a growing One Health concern. This study described the occurrence and antimicrobial susceptibility of Mammaliicoccus sciuri and Staphylococcus spp. isolated from reptiles undergoing rehabilitation in northeastern Brazil. Oropharyngeal and cloacal samples from 87 reptiles yielded 87 isolates identified by MALDI-TOF MS, and antimicrobial susceptibility was assessed by disk diffusion. Five bacterial species were detected, with a predominance of M. sciuri (79/87; 90.8%). Resistance to oxacillin was the most frequent finding

The emergence of antimicrobial resistance in environmental bacteria threatens therapeutic efficacy in clinical settings. Shewanella algae, historically regarded as a marine saprophyte, is increasingly recognized as an emerging opportunistic pathogen. In this study, we analyzed 86 S. algae isolates from Spain (19 clinical and 67 environmental) and integrated them with 178 publicly available genomes to explore antimicrobial susceptibility patterns and genomic diversity. Penicillins and fosfomycin consistently showed poor activity, whereas piperacillin/tazobactam, third- and fourth-generation cep

Acinetobacter baumannii (A. baumannii) is a major cause of multidrug-resistant infections, yet resistance is often interpreted from a gene-centered perspective that does not explain inconsistent treatment outcomes. This narrative review redefines resistance as a treatment-associated phenotype arising from interactions among molecular resistance mechanisms, bacterial physiology, and the infection environment. Unlike previous reviews that examine determinants in isolation, this work integrates efflux, permeability, enzymatic activity, and target modification with phenotypic states such as struct

Enterotoxigenic Escherichia coli (ETEC) is a major cause of diarrhea in developing countries and is regularly imported into developed countries as a major cause of traveler's diarrhea. ETEC is usually self-limiting and not necessarily treated with antimicrobials, although antimicrobial treatment is recommended in malnourished children, severe cases, and traveler's diarrhea. However, resistant strains to representative therapeutic agents such as ciprofloxacin and azithromycin have been reported in recent years, and multidrug-resistant ETEC has also emerged. This review discusses the recent anti

The rise of antibiotic-resistant bacteria has become an alarming global health challenge. Disinfectants, such as alcohols, aldehydes, chlorine compounds, phenols, quaternary ammonium compounds (QACs), peroxides, and chlorhexidine, are widely used in healthcare settings as a critical line of defense against infection. Nevertheless, their overuse or misuse, especially at subinhibitory concentrations, can promote the emergence of bacterial resistance, potentially leading to cross-resistance to antibiotics. Several mechanisms, including efflux pump activation, alterations in membrane permeability,
