The role of the Armanni-Ebstein lesion, hepatic steatosis, biochemical analysis and second generation anti-psychotic drugs in fatal diabetic ketoacidosis.

Diabetic ketoacidosis (DKA) is an acute severe complication of diabetes and characterized by a complex disordered metabolic state due to an absolute or relative insulin deficiency, leads to hyperglycaemia, ketoacidosis and ketonuria. DKA can cause sudden unexpected death and often yields minimal and/or subtle autopsy findings or a negative autopsy and the diagnosis mainly depends upon biochemical analysis of body fluids. This communication highlights the role of Armanni-Ebstein lesion, hepatic steatosis, biochemical analysis and second generation anti-psychotic drugs in 25 adult cases of fatal

The long and winding road for the development of tasquinimod as an oral second-generation quinoline-3-carboxamide antiangiogenic drug for the treatment of prostate cancer.

IMPORTANCE OF THE FIELD: Prostate cancer is the mostly commonly diagnosed non-skin cancer in males. The culmination of the last 70 years of clinical drug development has documented that androgen ablation plus taxane-based systemic chemotherapy enhances survival, but is not curative, in metastatic prostate cancer. To effect curative therapy, additional drugs must be developed that enhance the response when combined with androgen ablation/taxane therapy. AREAS COVERED IN THIS REVIEW: The history of the discovery and development of tasquinimod as a second-generation oral quinoline-3-carboxamide

HIV drug resistance after the use of generic fixed-dose combination stavudine/lamivudine/nevirapine as standard first-line regimen.

Early failures to stavudine/lamivudine/nevirapine used as a generic fixed-dose combination in Mali showed resistance mutations in 50% of cases (mostly M184V and Y181C). No thymidine analogue mutations were seen, suggesting that most nucleoside reverse transcriptase inhibitors could be used in a second-line regimen. This highlights the importance of the accessibility of HIV-RNA assays for monitoring treated patients in resource-poor countries to detect early virological failure in order to preserve future therapeutic options.

Artificial intelligence-guided green and circular nanotechnology: designing sustainable, biodegradable, and bioresponsive nanocarriers for next-generation precision drug delivery.

The convergence of artificial intelligence (AI), green chemistry, and nanocarrier-based drug delivery is reshaping pharmaceutical research toward a more sustainable and precision-oriented paradigm. Conventional nanoparticle design strategies have traditionally prioritized biological efficacy while largely overlooking environmental sustainability, circular manufacturing, and life-cycle accountability, thereby challenging translational pipelines increasingly faced with regulatory, economic, and ecological concerns. This review evaluates how AI methodologies, including machine learning (ML), rein

Parental income and drug use disorder among second-generation individuals in Sweden: effect modification by immigrant background and region of origin.

BACKGROUND: Evidence is limited for the association between parental income during childhood and adolescence and the subsequent risk of drug use disorder (DUD) in the second generation and whether the association is modified by region of origin. Identifying the association and effect modification by immigrant background and region of origin would provide an important knowledge base to consider tailored policies. METHODS: We used nationwide longitudinal Swedish data on 1,137,721 non-immigrants and 293,636 s-generation individuals who turned 15 years of age between 2005 and 2020. Cox proportion

From General-Purpose to Disease-Specific Features: Aligning LLM Embeddings on a Disease-Specific Biomedical Knowledge Graph for Drug Repurposing.

Identifying new therapeutic uses for existing drugs is a major challenge in biomedicine, especially for complex neurodegenerative conditions such as Alzheimer disease and related dementias (ADRD), where treatment options remain limited and relevant data are often sparse, heterogeneous, and difficult to integrate. Although general-purpose Large Language Model (LLM) embeddings encode rich semantic information, they often lack the task-specific biomedical context needed for inference tasks such as computational drug repurposing. We introduce Contextualizing LLM Embeddings via Attention-based gRap

Targeted next-generation sequencing for comprehensive diagnosis and drug resistance detection in pulmonary and extrapulmonary tuberculosis: a single-center retrospective study.

Tuberculosis (TB), including drug-resistant forms, is a significant global health issue requiring accurate and rapid diagnostic tools. Traditional diagnostic methods suffer from low sensitivity and slow results, while nucleic acid amplification tests (NAATs) like Xpert MTB/RIF provide faster but incomplete solutions. Targeted next-generation sequencing (tNGS) has the potential to simultaneously detect Mycobacterium tuberculosis, drug resistance mutations, and co-infecting pathogens. This study evaluates the effectiveness of tNGS for TB detection across various pulmonary and extrapulmonary TB s

CRB-701: A Second-generation Nectin-4-Targeted Antibody-Drug Conjugate with Optimized Stability and Pharmacokinetics for the Treatment of Solid Tumors.

UNLABELLED: The adhesion protein nectin-4 is a clinically validated tumor-selective antigen that is overexpressed across multiple cancer types and has been successfully exploited as a target for antibody-drug conjugates (ADCs), most notably enfortumab vedotin (EV), which is approved for the treatment of urothelial carcinoma. In this study, we report the design and preclinical characterization of a second-generation nectin-4-targeted ADC, CRB-701 (also identified as SYS6002). CRB-701 was synthesized using microbial transglutaminase technology to conjugate two molecules of monomethyl auristatin

Next-generation antibody-based therapeutics in cancer: antibody-drug conjugates bispecific antibodies across hematologic malignancies and solid tumors.

The therapeutic paradigm in oncology is undergoing a profound transformation driven by antibody-drug conjugates (ADCs) and bispecific antibodies (bsAbs). This review comprehensively summarizes recent clinical advances of these platforms across both hematologic malignancies and solid tumors. ADCs such as trastuzumab deruxtecan have expanded the concept of targetable HER2 expression, demonstrating meaningful intracranial activity and redefining standards of care in HER2-low breast cancer and beyond. The landscape continues to broaden with novel ADC targets (TROP2, CLDN18.2, B7-H3, HER3) and bisp
